A fluorescent “turn off–on” pattern for the detection of herring sperm DNA (hsDNA) had been designed through utilizing the interaction between polymyxin B sulfate (PMBS) and hsDNA as an inherent performance and the fluorescent transformation of glutathione (GSH)-capped CdTe quantum dots (QDs) as an external manifestation. Due to the occurrence of the photoinduced electron transfer from the QDs to PMBS, the fluorescence of GSH-capped CdTe QDs could be effectively quenched by PMBS, causing the system into “off” state. With the addition of hsDNA, the quenched fluorescence of GSH-capped CdTe QDs could be restored for the reason that PMBS embedded into hsDNA double helix structure to form new complex and peeled off from the surface of GSH-capped CdTe QDs, leading the system into “on” condition. Corresponding experimental results illustrated that the relative recovered fluorescence intensity of GSH-capped CdTe QDs–PMBS system was near proportional to the concentration of hsDNA within the range of 0.059–15.0 μg mL −1 . This proposed method demonstrated a good linear correlation coefficient of 0.9937 and a detection limit (3 σ / K ) of 0.018 μg mL −1 for hsDNA. This dual-directional fluorescent biosensor overcame the selectivity problem commonly existed in the traditional mono-directional fluorescence detection mode and owned perfect analysis applications in biochemical DNA monitoring.

Bacterial infections cause high morbidity and mortality worldwide, and the emergence of drug-resistant bacteria further complicates the treatment of infections. Therefore, it is necessary to continuously develop new treatment methods. Polymyxin B (PMB), as the last line of defense, can combat most aerobic Gram-negative bacilli including common drug-resistant bacteria in clinical practice. However, the suboptimal lung tissue concentration of PMB and dose-dependent nephrotoxicity and neurotoxicity limit its clinical application. The nanodrug delivery system offers several key advantages, including high drug loading capacity, excellent biocompatibility, controlled release mechanisms, and targeted delivery. These features enhance the bioavailability of drugs while simultaneously reducing their toxicity and minimizing side effects. In this study, we designed a targeted nanodrug delivery system (PMB@HMnO2@NM) consisting of hollow mesoporous manganese dioxide (HMnO2) coated with neutrophil membrane (NM). In a mouse model of acute pneumonia induced by multidrug-resistant Pseudomonas aeruginosa, treatment with PMB@HMnO2@NM demonstrated the ability to target bacterial aggregation and specifically deliver the drug to the infected lung tissue. This targeted approach resulted in improved survival rates and reduced inflammatory damage without causing adverse effects. The findings of this study suggest the potential for developing a new class of multifunctional nanodrugs, providing new therapeutic strategies for multidrug-resistant (MDR) bacterial infections. Furthermore, these results provide a solid foundation for the design of biomimetic nanosized antibacterial drugs.

Objectives This study investigated the mechanisms underlying polymyxin B (PMB)-induced resistance and examined the role of PMB in combination with amikacin (AMK) in delaying the development of resistance. Methods In vitro continuous induction of three Klebsiella pneumoniae strains was performed using PMB alone or in combination with AMK, and variations in the minimum inhibitory concentration (MIC) were determined via the microdilution method. The Kp81 strain, which presented the most significant delay in resistance development, was selected for analysis of the expression of relevant genes via qRT–PCR. Homologous recombination techniques were employed to construct nlpE and cpxR gene knockout and complemented Kp81 strains, and the expression levels of these genes, as well as changes in the MIC following continuous induction, were assessed. Additionally, the biofilm-forming abilities of the strains were analyzed via crystal violet staining and confocal laser scanning microscopy. Results The results indicate that the combination of PMB and AMK can delay the development of resistance to PMB in K. pneumoniae . qRT–PCR analysis demonstrated a significant increase in the expression of phoP , nlpE , cpxR , and acrA in the resistant strain Kp81·R, whereas these genes were significantly downregulated in the resistant delay strain Kp81·DR. Notably, after 20 h of PMB treatment, the expression of nlpE was markedly elevated in the nlpE knockout strain. cpxR exhibited synchronized dynamic changes with nlpE in both the nlpE knockout and complemented strains, whereas acrA showed a similar expression pattern in the cpxR knockout and complemented strains. Additionally, the absence of nlpE or cpxR was found to delay PMB resistance. Furthermore, biofilm formation was significantly increased in both the resistant strains and those treated with PMB, while the combination of PMB with AMK inhibited biofilm formation. Conclusions The combination of PMB and AMK can delay the development of resistance in K. pneumoniae to PMB. The nlpE gene, as a key regulatory factor, can influence PMB resistance by modulating the CpxA/R two-component system and biofilm formation.

Nanofiber sponge, which showed high porosity and excellent blood compatibility, was successfully prepared by electrospinning and freeze-drying technology. The APTT (activated partial thromboplastin time) and PT (prothrombin time) of human plasma with nanofiber sponge were a little longer than control groups, which revealed the nanofiber sponge has excellent blood compatibility. By activating the carboxyl groups on the fibers, PMB (polymyxin B) was successfully immobilized on the nanofiber sponge. The adsorption performance was researched in different solutions. The adsorption capacity in physiological saline solution was around 17,889 EU/mL and the adsorption equilibrium time was about 40 min. The adsorption behavior not only obeyed to second-order kinetic equation, but also was conformed to Langmuir model, which turned out to be chemical adsorption and monolayer adsorption. The endotoxin removal rate in BSA (bovine serum albumin) physiological saline solution was about 96.6%, and the BSA recovery rate was around 75.6%, which indicated the PMB immobilized nanofiber sponge adsorbed endotoxin selectively. The endotoxin removal rate of normal nanofiber sponge was about 17.3%, while it was around 99.6% when it comes to PMB immobilized-nanofiber sponge, which manifested PMB played an important role in endotoxin adsorption. The endotoxin removal rate in human plasma reached 90%, and the adsorption reached equilibrium within 60 min, which proved the PMB immobilized-nanofiber sponge would have great potential for applying in clinical blood purification.

This paper proposes synergistic photodynamic antimicrobial therapy and chemotherapy for enhanced wound healing using polymyxin B-functionalized phthalocyanine (Pc-PB) as a new photosensitive antibacterial agent. The rapid healing of a bacteria-infected wound was achieved via combining the photodynamic antibacterial activity of phthalocyanine with the innate antibacterial ability of polymyxin B. After the functionalization of polymyxin B, the new phthalocyanine derivative can effectively target the bacterial surface and generate singlet oxygen to kill Gram-positive and Gram-negative bacteria under 680 nm light irradiation. Additionally, the prepared Pc-PB can form colloidal nanoparticles to significantly increase the solubility and dispersion of phthalocyanine in water. For the antimicrobial action mechanism, the antibacterial agent can interact with bacteria via binding to the Ca2+ and Mg2+ ions of the bacterial surface and then disrupting the bacterial membrane. Pc-PB shows good biocompatibility and can promote the healing of bacteria-infected wounds. These results suggest that phthalocyanine combined with bioactive molecules has potential for application in photodynamic therapy.

Hemoperfusion is an important strategy for liver disease treatment. Polystyrene-divinylbenzene (PS-DVB) microspheres are widely applied as absorbents in hemoperfusion to efficiently remove the important toxin bilirubin. However, as another common toxin, endotoxin will remain during this process and cause endotoxemia. Therefore, simultaneous removal of both bilirubin and endotoxin is highly desirable. In the present study, we engineered PS-DVB microspheres with polymyxin B sulfate (PMB) to meet this goal. After modification, the novel PMB-engineered (P-PMB) microspheres displayed excellent biocompatibility and hemocompatibility. Notably, compared to PS-DVB microspheres, P-PMB microspheres exhibited markedly stronger detoxification of both bilirubin and endotoxin, increasing by 17.03% and 42.57%, respectively. Overall, we believe that the novel P-PMB microspheres have considerable potential for liver disease treatment in clinical practice.

Polymyxin B is increasingly employed all over the world to treat patients who affected by multidrug-resistant Gram-negative bacteria. Although the mechanism of resistance to polymyxin B is well known, the metabolic role of bacteria in stress response to polymyxin B remains an important task and may help to better understand polymyxin B-related stress response. In this study, the proteome changes of Escherichia coli ( E. coli ) continuously induced in concentrations of 1.0 mg/L and 10.0 mg/L polymyxin B were revealed. Compared to E. coli (PMB 0 ), E. coli exposed to polymyxin B at 1.0 mg/L (PMB 1 ) and 10.0 mg/L (PMB 10 ) resulted in 89 and 314 differentially expressed proteins (DEPs), respectively. Such differences related to fatty acid degradation, quorum sensing and two-component regulatory system pathways. Based on absolute quantitative (iTRAQ) proteomics analysis, this study comprehensively studied the changes of E. coli proteome in culture with concentrations of 1.0 mg/L and 10.0 mg/L polymyxin B through confocal laser scanning microscopy observation, cell viability detection and reactive oxygen species analysis. The results showed that E. coli cultured at concentration of 10.0 mg/L polymyxin B increased the expression levels of multidrug-resistant efflux transporters and efflux pump membrane transporters, which might further improve the pathogens of polymyxin B-resistant bacteria lastingness and evolution. It has emerged globally to resist polymyxin B. The reuse of polymyxin B should be aroused public attention to avoid causing more serious environmental pollution . These findings could provide new insights into polymyxin B-related stress.

Chronic inflammation is critical in the onset and progression of atherosclerosis (AS). The lipopolysaccharide (LPS) level in the circulation system is elevated in AS patients and animal models, which is correlated with the severity of AS. Inspired by the underlying mechanism that LPS could drive the polarization of macrophages toward the M1 phenotype, aggravate inflammation, and ultimately contribute to the exacerbation of AS, LPS in the circulation system was supposed to be the therapeutic target for AS treatment. In the present study, polymyxin (PMB) covalently conjugated to PEGylated liposomes (PLPs) were formulated to adsorb LPS through specific interactions between PMB and LPS. In vitro , the experiments demonstrated that PLPs could adsorb LPS, reduce the polarization of macrophages to M1 phenotype and inhibit the formation of foam cells. In vivo , the study revealed that PLPs treatment reduced the serum levels of LPS and pro-inflammatory cytokines, decreased the proportion of M1-type macrophages in AS plaque, stabilized AS plaque, and downsized the plaque burdens in arteries, which eventually attenuated the progression of AS. Our study highlighted LPS in the circulation system as the therapeutic target for AS and provided an alternative strategy for AS treatment.

Benzalkonium chloride (BAC) is widely employed as a broad-spectrum biocide and has emerged as a significant environmental pollutant. Polymyxin B (PB) serves as the last-line defense for the treatment of Gram-negative pathogens. Previous studies reported that BAC-adapted Pseudomonas aeruginosa increased the tolerance to PB. Herein, we present the novel finding that the combination of BAC and PB exhibited synergistic antibacterial effects against P. aeruginosa . Time-killing assay demonstrated a significant reduction in bacterial cell viability. Scanning electron microscopy, zeta potential analysis, hydrophobicity measurements, and fluorescence probe analyses collectively revealed severe disruption of the cell envelope and membrane potential induced by the combination of BAC and PB. Transcriptomic analysis revealed that the BAC-PB combination notably downregulated the expression of genes involved in lipid A modification and cell envelope production, including phoPQ, pmrAB, bamABCDE, lptABCDEG, lolB, yidC , and murJ . Additionally, the combination group exhibited augmented production of reactive oxygen species and diminished ATP synthesis. The expression of the genes associated with substance metabolism and energy generation was significantly impeded. This study provides significant implications for the interactions of biocides and antibiotics on Gram-negative pathogens, while also addressing antibiotic resistance and developing the external treatment strategy for Pseudomonas -infected wounds and burns.

The epidemic of multidrug-resistant Gram-negative bacteria is an ever-growing global concern. Polymyxin B (PMB), a kind of “old fashioned” antibiotic, has been revived in clinical practice and mainly used as last-line antibiotics for otherwise untreatable serious infections because the incidence of the resistance to PMB is currently relatively low in comparison with other antibiotics in vivo owing to the unique bactericidal mechanism of PMB. However, serious adverse side effects, including nephrotoxicity and neurotoxicity, hamper its clinical application. Herein, we describe the development of a nanoparticle that can target sites of inflammation and forcedly release PMB specifically in the area of Gram-negative bacteria. This particle was constructed through the electrostatic self-assembly of hyaluronic acid (HA) and PMB molecules in order to realize the safe and effective treatment of pneumonia. After systemic administration, PMB-HA nanoparticles were found to actively accumulate in the lungs, precisely target the CD44 receptors over-expressed on the membrane of activated endothelial cells in inflammatory sites, and then come into contact with the bacteria resident in the damaged alveolar-capillary membrane. Due to the electrostatic and hydrophobic interactions between PMB and the lipopolysaccharide (LPS) in the outer membranes of bacteria, the PMB molecules in the PMB-HA nanoparticles are expected to escape from the nanoparticles to insert into the bacteria via competitive binding with LPS. Through shielding the cationic nature of PMB, PMB-HA nanoparticles also possess outstanding biosafety performance in comparison to free PMB. It is thus believed that this smart delivery system may pave a new way for the resurrection of PMB in the future clinical treatment of bacterial inflammatory diseases.

Background Acinetobacter baumannii is a notorious clinical pathogen that predominantly causes nosocomial infections. The multidrug resistant Acinetobacter baumannii clinical isolates are becoming more prevalent on a global scale. In this study, we aimed to determine the potential synergistic antibacterial activity of contezolid in combination with polymyxin B nonapeptide (PBNP) against various A. baumannii strains. Results Contezolid, a commonly-used agents targeting Gram-positive bacteria, in combination with PBNP (CP) was used to explore the potential synergistic antibacterial activity against A. baumannii in vitro and Caenorhabditis elegans models. We found that CP treatment exhibited a strong synergistic effect on A. baumannii or multidrug resistant A. baumannii . CP also inhibited the biofilm formation and alter morphology of A. baumannii compared to the control group. Moreover, the residual bacteria were significantly decreased in the CP-treated murine alveolar macrophages (MH-S). Importantly, CP treatment improved the survival of C. elegans compared to the control or monotherapy group. Conclusions This strategy improved antibacterial activity spectrum of agents targeting Gram-positive bacteria like contezolid. Nevertheless, CP combination therapy requires further validation in mammalian infection models.

Endotoxemia, a life-threatening disease affecting people worldwide, can be treated by hemoperfusion alone. New hemoperfusion materials with high biocompatibility and endotoxin-combination ability are always in demand. Herein, polymyxin B (PMB), a specific endotoxin binding molecule, was loaded onto gellan–polylysine polyion complex, and the obtained material was used in preparing wet-spun fibers. The tensile strength of the as-spun yarns (100 fibers) ranged from 1.49 N to −1.58 N and that of the dried and rewetted yarns ranged from 1.45 N to 1.56 N. The adsorption ability of the fibers with lipopolysaccharides from E. coli was 2.784 ± 0.036 EU/mg in simulated human body fluid and 2.452 ± 0.107 EU/mg in mouse plasma. The fibers showed no cytotoxicity toward U2OS cells and no hemolysis toward mouse blood. The influence of the fibers on the clotting time of mouse blood was negligible, and the blood cells were not adhesive to the fibers. Thus, the PMB-loaded gellan–polylysine complex fiber and its derivate fabrics can be used in hemoperfusion.
