Polyporus umbellatus (Pers.) Fries (PU), a medicinal fungus, contains polysaccharides (PUPs) as its primary bioactive components. In this paper, physicochemical properties, biological activities and in vitro fermentation prebiotic properties of PUPs, extracted by 90 °C hot water (PUP-W), 0.9 % NaCl (PUP-N), citric acid (PUP-S) and 0.1 M NaOH (PUP-A) were compared. The PUPs were composed of multiple monosaccharides, with significant differences in chemical composition and structure. The extracted PUPs demonstrated notable antioxidant, antitumor and prebiotic activities. With the highest yield (2.96 ± 0.12), PUP-A exhibited better biological activities due to its small molecular weight. The antioxidant ability (clearing DPPH (IC 50 = 0.64) and ABTS free radicals (IC 50 = 1.23)) of PUP-A was strong, so was the ability of ROS clearing. PUP-A significantly decreased the cell survival rate of HepG2 (IC 50 = 1090). In terms of prebiotic properties, both PUP-N and PUP-A showed substantial promoting effects on some beneficial bacteria, like Bacteroidetes at the phylum level and Lactobacillus at the genus level. In summary, our study suggests that alkali is the better solvent and provides new insights into the relationship between the structure, biological activities, and gut microbiota of PUPs, laying the foundation for better development of the role of PU.

M1 polarization of tumor-associated macrophages (TAMs) is a promising approach to breaking through therapeutic barriers imposed by the immunosuppressive tumor microenvironment (TME). As a clinically-used immunopotentiator for cancer patients after chemotherapies; however, the immunomodulatory mechanism and potential of polyporus polysaccharide (PPS) remains unclear. Here, we present mannose-decorated PPS-loaded superparamagnetic iron-based nanocomposites (Man/PPS-SPIONs) for synergistic M1 polarization of TAMs and consequent combinational anti-breast cancer therapy. Once internalized by M2-like TAMs, PPS released from Man/PPS-SPIONs induces the M1 polarization via IFN-γ secretion and downstream NF-κB pathway activating. The SPIONs within the nanocomposites mediate a Fenton reaction, producing OH· and activating the subsequent NF-κB/MAPK pathway, further facilitating the M1 polarization. The Man/PPS-SPIONs thereby establish a positive feedback loop of M1 polarization driven by the “IFN-γ-Fenton-NF-κB/MAPK” multi-pathway, leading to a series of anti-tumoral immunologic responses in the TME and holding promising potential in combinational anticancer therapies. Our study offers a new strategy to amplify TME engineering by combinational natural carbohydrate polymers and iron-based materials.

Objective Poria cocos and Polyporus umbellatus are similar medicinal fungi in traditional Chinese medicines . A method for fingerprint analysis of monosaccharide composition of polysaccharides by HPLC combined with chemometrics methods has been developed for characterization and discrimination of them in this research. Methods The polysaccharides were extracted by decocting in water, and then completely hydrolyzed with hydrochloride. Monosaccharides in the hydrolyzates were derivatized with 1-phenyl-3-methyl-5-pyrazolone (PMP) for HPLC analysis. More than 20 batches of P. cocos and P. umbellatus from different regions were analyzed. Results The fingerprints of P. cocos showed five common characteristic peaks, which were identified by comparing with the reference substances. The five peaks corresponded to the derivatives of mannose , ribose , glucose, galactose , and fucose . At the same time, the fingerprints of P. umbellatus showed eight common characteristic peaks, of which seven were identified as the derivatives of mannose, ribose, rhamnose , glucose, galactose, xylose , and fucose. Moreover, the similarity of their fingerprints was respectively calculated by the Similarity Evaluation System for Chromatographic Fingerprint of TCM published by China Pharmacopoeia Committee (Version 2004A). And the data were further processed by hierarchical cluster analysis (HCA) and principal component analysis (PCA). The similarity evaluation and HCA indicated that there were no significant difference in P. cocos or P. umbellatus samples from different geographical regions, but PCA was performed to characterize the difference in monosaccharide constituents between P. cocos and P. umbellatus , and linear discriminant analysis (LDA) showed the overall correct classification rate was 100%. Conclusion The fingerprint analysis method of monosaccharide composition of water-soluble polysaccharides can distinguish P. cocos and P. umbellatus , and can be applied for the authentication or quality control for P. cocos and P. umbellatus.
