In vitro assessment of the effect of Porphyra haitanensis polysaccharides on the intestinal flora of allergic mice

Peilin Chen,YuShan Huang,Hongliang Zeng,Mingjing Zheng,Juanjuan Guo

This study systematically investigated the modulatory effects of Porphyra haitanensis polysaccharides (PHP) and its two P. haitanensis polysaccharide components (PHP40 and PHP80) on gut microbiota in ovalbumin (OVA)-sensitized mice via in vitro fermentation. PHP40 and PHP80 exhibited sulfate contents of 5.94 ± 0.05 % and 11.72 ± 0.03 %, respectively, with galactose and glucose as dominant monosaccharides. The different polysaccharide components had different effects on the aforementioned changes in the species composition and structure of the gut microbiota in allergic mice, with distinct dominant microbial profiles across groups. PHP could promote Bacillus and Enterococcus proliferation while inhibiting Staphylococcus . PHP40 and PHP80 could promote Enterococcus and Enterobacter growth but suppressed Staphylococcus and Bacillus . Functional prediction indicated PHP significantly improved galactose metabolism and primary/secondary bile acid biosynthesis, potentially alleviating allergic responses.

Novel angiotensin-I-converting enzyme (ACE) inhibitory peptides from Porphyra haitanensis: Screening, digestion stability, and mechanistic insights

Xianjiang Ye,Ran Yang,Zhizhi Yang,Baote Huang,Talha Riaz,Chao Zhao,Jicheng Chen

Food-derived peptides with angiotensin-I-converting enzyme inhibitory (ACEI) activity have garnered significant research attention. This study focused on examining ACEI peptides from Porphyra haitanensis and investigating their potential inhibitory mechanisms through a combination of in silico and in vitro methods. Five novel potential ACEI peptides (ASF: Ala-Ser-Phe, EIIL: Glu-Ile-Ile-Leu, LCVPR: Leu-Cys-Val-Pro-Arg, LDVVN: Leu-Asp-Val-Val-Asn, and LSPSW: Leu-Ser-Pro-Ser-Trp) were identified through a combination of peptidomics and in silico bioinformatics analysis, of which LSPSW showed the highest activity (IC 50 : 5.27 ± 0.53 μM). After simulated gastrointestinal digestion (SGD), the activities of LCVPR and LSPSW remained almost unchanged. The results from inhibition kinetics and molecular docking demonstrated that ACEI peptides with differing inhibition mechanisms could successfully bind to ACE through hydrogen bonds. Based on network pharmacology analysis and cellular experiments, LSPSW exerted antihypertensive effects by regulating the expression levels of peroxisome proliferator activated receptor gamma (PPARG), prostaglandin-endoperoxide synthase 2 (PTGS2), and renin (REN). This research provided a novel theoretical foundation for the high-value application of P. haitanensis .

Structure and Antidiabetic Activity of a Glycoprotein from Porphyra haitanensis

Yujia Ou,Lijingting Xu,Mingrong Chen,Xiaodan Lu,Zebin Guo,Baodong Zheng

A novel antidiabetic glycoprotein (PG) was isolated and purified from Porphyra haitanensis, and its structure and inhibiting activity on α-amylase and α-glucosidase were analyzed. The purity of the PG was 95.29 ± 0.21%, and its molecular weight was 163.024 ± 5.55 kDa. The PG had a tetramer structure with α- and β-subunits, and it contained 54.12 ± 0.86% protein (with highly hydrophobic amino acids) and 41.19% ± 0.64% carbohydrate (composed of galactose). The PG was linked via an O-glycosidic bond, exhibiting an α-helical structure and high stability. In addition, the PG inhibited the activities of α-amylase and α-glucosidase, by changing the enzyme’s structure toward the PG’s structure in a noncompetitive inhibition mode. Molecular docking results showed that the PG inhibited α-amylase activity by hydrophobic interaction, whereas it inhibited α-glucosidase activity by hydrogen bonds and hydrophobic interaction. Overall, the PG was linked to polysaccharides via O-glycosidic bonds, showing an α-helical configuration and a hydrophobic effect, which altered the configuration of α-amylase and α-glucosidase and exerted hypoglycemic activity. This study provides insights into analyzing the structure and antidiabetic activity of glycoproteins.

Exploring the therapeutic potential of porphyran extracted from Porphyra haitanensis in the attenuation of DSS-induced intestinal inflammation

Kit-Leong Cheong,Xu-Ting Xie,Tao Zhou,Sathuvan Malairaj,Suresh Veeraperumal,Saiyi Zhong,Karsoon Tan

Ulcerative colitis is a chronic, spontaneous inflammatory bowel disease that primarily affects the colon. This study aimed to explore how Porphyra haitanensis porphyran (PHP) modulates the immune response and the associated mechanisms that alleviate dextran sulphate sodium-induced colitis in mice. Histological assessments via H&E staining and AB-PAS staining revealed that PHP intervention partially restored the number of goblet cells and improved intestinal mucosal function. Immunohistochemical and Western blot analyses of claudin-1, occludin, and MUC-2 demonstrated that PHP could repair the intestinal barrier and reduce colon damage by upregulating the expression of these proteins. PHP intervention was associated with a decrease in pro-inflammatory cytokine expression and an increase in anti-inflammatory cytokine expression. Moreover, the expression of proteins involved in intestinal immune homing, such as CCR-9, CCL-25, MAdCAM-1, and α4β7, was significantly suppressed in response to PHP treatment. Conversely, PHP upregulates the expression of CD40 and TGF-β1, both of these can promote healing and reduce inflammation in the gut lining. This study demonstrates that PHP can ameliorate ulcerative colitis by enhancing the intestinal barrier and modulating immune responses. These findings offer valuable insights into the potential utility of P. haitanensis as a promising natural product for managing ulcerative colitis.

Physicochemical characterization and antioxidant activity of sulphated polysaccharides derived from Porphyra haitanensis

Bilal Muhammad Khan,Hua-Mai Qiu,Shu-Ying Xu,Yang Liu,Kit-Leong Cheong

This study was designed to fully characterize Porphyra haitanensis polysaccharides, and to evaluate their antioxidant activity. The polysaccharides primarily contained galactose and 3,6-anhydrogalactose in a molar ratio of 1.2:1.0, respectively and sulfate content about 3.8%. The molecular weight of polysaccharides is 2.5 × 10 5 Da. Scanning electron microscopy and atomic force microscopy of the polysaccharides pointed towards an irregular network with more or less hexagonal and a few rectangular pores. The chemical structure was confirmed through Fourier transform infrared spectroscopy, and 1D and 2D nuclear magnetic resonance structural characterization wherein → 4–3,6-anhydro-α-L-galactopyranose-(1 → 3)-β-D-galactopyranose segments. The extracted polysaccharides revealed relatively high 2, 2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity (53.16% at 2 mg/mL), moderate 2,2-diphenyl-1-picrylhydrazyl radical scavenging efficacy (34.63% at 2 mg/mL), and low hydroxyl radical scavenging potential (23.80% at 2 mg/mL). Further purification of these polysaccharides, hence, is advised for their potential role as antioxidants in the food, pharmaceutical and cosmeceutical industry.

Microbial catabolism of Porphyra haitanensis polysaccharides by human gut microbiota

Shu-Ying Xu,Jude Juventus Aweya,Na Li,Rui-Yan Deng,Wei-Yi Chen,Jie Tang,Kit-Leong Cheong

This study applied an in vitro fermentation model, whereby the catabolism of Porphyra haitanensis polysaccharides (PHP) was monitored coupled with the variations of microbiota composition and the concentration of short-chain fatty acids (SCFAs), so as to assess the effects of PHP on human intestinal microbiota. After 24 h anaerobic incubation, the level of microflora diversity increased significantly, as the microbiome structure was reshaped through promotion of intestinal probiotics proliferation and inhibition of pathogens growth. Besides, the final concentration of total SCFAs increased to 32.32 ± 1.81 mmol/L, and contained high amounts of acetic, propionic and butyric acid. Furthermore, the molecular weight of PHP decreased from 2.623 × 10 5  g/mol to 2.308 × 10 4  g/mol. The degree of polymerization of oligosaccharide products ranged from 2 to 9, with the main linkage patterns being 1 → 3 and 1 → 4 linked Gal p . The current study provides new insight on the probiotic activity of PHP within the human gastrointestinal tract.

Purification, characterization and antioxidant activity of polysaccharides from Porphyra haitanensis

Yun-Tao Wu,Yun-Feng Huo,Lin Xu,Ying-Ying Xu,Xiao-Ling Wang,Tao Zhou

In order to study the antioxidant activity of Porphyra haitanensis polysaccharides and the effect of their structure on the antioxidant activity, the Porphyra haitanensis polysaccharide (PHP), prepared by hot water extraction and alcohol precipitation, was separated and purified by Cellulose DEAE-52 ion exchange and Sephadex G-100 column chromatography. Three purified components including PHP1, PHP2 and PHP3 were obtained, and chemical composition analysis and structural characterization were performed. The in vitro free radical scavenging activity of the purified polysaccharide fractions and their ability to relieve oxidative stress in macrophage RAW264.7 were investigated. The results indicated that all the three fractions possessed appreciable DPPH radical, superoxide anion radical, and hydroxyl radical scavenging ability and reducing power. They were also found to effectively reduce the MDA content and ROS level, and to improve the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) in H 2 O 2 -treated RAW264.7 cells. Among the three polysaccharides, PHP3 possessed the most potent activity. Porphyra haitanensis polysaccharides have potential to develop as natural non-toxic antioxidants and may find application as the ingredients of functional foods.

Resveratrol-loaded ovalbumin/Porphyra haitanensis polysaccharide composite nanoparticles: Fabrication, characterization and antitumor activity

Ying-Ying Xu,Yun-Feng Huo,Lin Xu,Yu-Zhu Zhu,Yun-Tao Wu,Xiao-Yi Wei,Tao Zhou

Resveratrol (RES) possesses a variety of health-promoting effects. However, the application of RES in food and pharmaceutical industries is limited due to the poor water solubility and bioavailability, and low photostability. In order to overcome the drawback of RES, in the present work, RES-loaded nanoparticles were prepared using ovalbumin (OVA) and Porphyra haitanensis polysaccharide (PHP) as encapsulating materials. Response surface methodology was employed to optimize the fabrication of OVA-PHP-RES nanoparticles (OPR). It was found that OVA-PHP possessed superior effects on both stability of the dispersion and RES protection to those of OVA alone. In vitro simulated digestion tests demonstrated that RES in OPR was released slower in stomach and more rapidly in intestine when compared to OVA-RES nanoparticles (OR). In addition, OPR was found to effectively inhibit the growth of two tumor cell lines (HeLa and HepG2), indicating that OPR nanoparticles could deliver effectively RES in biological system, thereby improving its bioavailability. The finding in this study demonstrated OVA-PHP system could improve the stability and bioavailability of RES.

Porphyran from Porphyra haitanensis Enhances Intestinal Barrier Function and Regulates Gut Microbiota Composition

Sathuvan Malairaj,Suresh Veeraperumal,Wanzi Yao,Mugesh Subramanian,Karsoon Tan,Saiyi Zhong,Kit-Leong Cheong

In this study, the effects of a homogenous porphyran fromPorphyra haitanensis(PHP) on the intestinal barrier and gut microbiota were investigated. The results showed that oral administration of PHP resulted in a higher luminal moisture content and a lower pH environment for the growth of beneficial bacteria in the colon of mice. PHP significantly increased the production of total short-chain fatty acids during the fermentation process. PHP made the intestinal epithelial cells of mice arrange more tidily and tightly with a significant increase in mucosal thickness. PHP also increased the amount of mucin-producing goblet cells and the expression of mucin in the colon, which maintained the structure and function of the intestinal mucosal barrier. Moreover, PHP up-regulated the expression of tight junctions including ZO-1 and occludin, improving the intestinal physical barrier function. The results of 16S rRNA sequencing showed that PHP regulated the composition of gut microbiota in mice, increasing the richness and diversity of gut microbiota and the ratio of Firmicutes to Bacteroidetes. This study revealed that the intake of PHP is beneficial for the gastrointestinal tract and PHP could be a potential source of prebiotics in the functional food and pharmaceutical industries. Keywords:Porphyra haitanensis;porphyran;intestinal barrier;gut microbiota

Temperature-controlled steam explosion modulates structural and functional characteristics of Porphyra haitanensis dietary fiber

Yujia Ou,Binhui Zhou,Yesong Wang,Tao Hong,Zedong Jiang,Hui Ni,Mingjing Zheng

Developing efficient modification methods for seaweed dietary fiber (DF) is hindered by its sulfated galactan backbone, which differs fundamentally from terrestrial lignocellulosic matrices. In this study, temperature-controlled steam explosion (SE) was employed to modulate the structural and functional characteristics of Porphyra haitanensis TDF. Moderate SE (140 °C) promoted an effective IDF-to-SDF conversion, evidenced by pronounced increases in SDF (127.67 %), galactose (37.71 %), and uronic acids (22.73 %–25.65 %), accompanied by reductions in IDF (17.95 %), acid-insoluble residue (28.54 %), and sulfate groups (47.40 %) ( p  < 0.05). Multiscale characterization revealed the formation of a honeycomb-like porous network, strengthened hydroxyl/carboxyl signals, elevated crystallinity, and enhanced thermal stability. Correspondingly, hydration capacity, lipid adsorption, cholesterol binding, and inhibition of α-amylase, α-glucosidase, and pancreatic lipase increased significantly. Excessive SE (180 °C) led to structural densification and diminished functionality. These findings suggest that controlled SE may restructure sulfated seaweed DF, offering a green strategy for its functional utilization.

Structural characterization and antioxidant activity of polysaccharides extracted from Porphyra haitanensis by different methods

Mingjing Zheng,Menghan Ma,Yuanfan Yang,Zhiyu Liu,Shuji Liu,Tao Hong,Hui Ni,Zedong Jiang

This study was to investigate the structure and antioxidant activity of Porphyra haitanensis polysaccharides (PHPs) extracted by different methods, including water extraction (PHP), ultra-high pressure (UHP-PHP), ultrasonic (US-PHP) and microwave assisted water extraction (M-PHP). Compared with water extraction, the total sugar, sulfate and uronic acid contents of PHPs was enhanced by ultra-high pressure, ultrasonic and microwave assisted treatments, especially those of UHP-PHP were increased by 24.35 %, 12.84 % and 27.51 %, respectively ( p  < 0.05). Meanwhile, these assisted treatments affected the monosaccharide ratio of polysaccharides and significantly reduced the protein content, molecular weight as well as particle size of PHPs ( p  < 0.05), and resulted in a loose microstructure with more porosity and fragments. PHP, UHP-PHP, US-PHP, and M-PHP all possessed in vitro antioxidant capacity. Among them, UHP-PHP had the strongest oxygen radical absorbance capacity , DPPH and ·OH radicals scavenging capacities, which increased by 48.46 %, 116.24 %, and 14.98 % respectively. Moreover, PHPs particularly UHP-PHP effectively increased the cell viability and reduced ROS levels of H 2 O 2 induced RAW264.7 cells ( p  < 0.05), indicating their good effects against cell oxidative damage. The findings suggested that PHPs with ultra-high pressure assisted treatments has the better potential to develop natural antioxidant.

Optimization of Ultrasound-Assisted Deep Eutectic Solvent Extraction and Bioactivities of Polysaccharide from Porphyra haitanensis

Wendie Hu,Bing Liu,Yao Wen

In this paper, ultrasound-assisted method and deep eutectic solvent (DES) are used to extract polysaccharides from Porphyra haitanensis . The single-factor experiment is carried out first, and then the response surface experiment is designed to obtain the optimal process conditions. The optimal extraction conditions are the molar ratio of choline chloride to oxalate of 1:1, the water content of DES of 60%, the solid–liquid ratio of 1:60 g mL −1 , the time of 120 min, the power of 200 W, and the temperature of 80 °C, with the extraction yield of 4.42%. And the yield of P. haitanensis polysaccharides (PHPs) is 3.14% and 3.31% by water immersion and ultrasonic water extraction, respectively, indicating that the DES extraction method is superior to these two methods, and the optimization effect is good. The extracted polysaccharides are tested for biological activity after separation and purification on DEAE and Sephadex G-75 columns. The results of α-glucosidase inhibitory activity and α-amylase inhibitory activity show that PHPs have certain antioxidant activity. 1,1-Diphenyl-3-nitrophenylhydrazine (DPPH) and 2,2-diazo-di-3-ethylbenzothiazolin-6-sulfonic acid (ABTS) free radical scavenging assays show that PHPs have certain antiglycemic activity.
