Periodontitis is caused by oral flora imbalance, which leads to immune imbalance. Porphyromonas gingivalis is a keystone pathogen in periodontitis, causing the blooming of inflammophilic microbes, and becoming dormant to resist antibiotics. Targeted interventions are needed to destroy this pathogen and collapse its inflammophilic flora. Therefore, a targeting nanoagent antibody-conjugated liposomal drug carrier with ginsenoside Rh2 (A-L-R) was developed for pleiotropic benefits. A-L-R showed high quality in high-performance liquid chromatography (HPLC), Fourier transform infrared (FTIR), and transmission electron microscope (TEM) detection. Only P. gingivalis was influenced by A-L-R, as shown by live/dead cell staining and a series of antimicrobial effects assays. With fluorescence in situ hybridization (FISH) staining and in propidium monoazide-quantitative polymerase chain reaction (PMA-qPCR), the clearance of P. gingivalis by A-L-R was more than for other groups, and only the proportion of P. gingivalis was reduced by A-L-R in monospecies culture. Moreover, in a periodontitis model, A-L-R targeted P. gingivalis with high efficiency and low toxicity, maintaining homeostasis with a relatively stable oral microflora. This targeting nanomedicine offers new strategies for periodontitis therapy, providing a foundation for the prevention and treatment of periodontitis.

ABSTRACT Porphyromonas gingivalis ( P. gingivalis, P. g ) is the main pathogen of periodontal disease, which is treated with egg yolk immunoglobulin Y ( IgY ) against P. gingivalis . In order to quickly obtain IgY, 30 hens were immunized with inactivated P. gingivalis . The purification of IgY was carried out by the oleic acid ( OA ) method and the classical method ( AS ), respectively. The IgY antibody characteristics and antibacterial effects in HPDLF cells were detected by SDS-PAGE, indirect ELISA, Western blot and viability/toxicity assays. SDS-PAGE and Western blot analysis showed that IgY molecules which were rapidly purified by OA method were complete and specific to P. gingivalis . In addition, the results of crystal violet staining and bacterial staining indicated that IgY could agglutinate with P. gingivalis , inhibiting bacterial invasion of host cells. This study is the first to rapidly and efficiently purify IgY by OA method, and the purified IgY is expected to be used in the detection and treatment of P. gingivalis .

Periodontitis is a microbe-driven infectious disease, in which Porphyromonas gingivalis (Pg) plays a keystone role. As the front line to eliminate dysbiotic microbiota, macrophages are critical for recognition, phagocytosis and digestion of bacteria. However, deficiencies in the antimicrobial function of periodontal macrophages lead to diminished Pg clearance and destructive periodontal inflammation. Cold atmospheric plasma (CAP) enables non-invasive treatment by producing reactive species including reactive oxygen species (ROS), reactive nitrogen species (RNS) and electro-magnetic field, and is of great interest for infectious diseases. These radicals have a significant influence on cellular biochemistry and are crucial components of the immune system. The CAP jet using helium gas was developed and driven by the bipolar pulse high voltage. The negative voltage was 5 kV and the positive voltage was 10 kV. The irradiation time was set to 120 s for in vivo experiments and 80 s for in vitro experiments. In vivo experiments demonstrated that CAP significantly alleviated periodontitis. In addition to the directly antimicrobial effects, in vitro experiments demonstrated that CAP enhanced intracellular killing of Pg by bone marrow-derived macrophages (BMMs) and murine macrophage cell line RAW 264.7 in a ROS-dependent manner. BMMs were collected from the tibias and femurs of healthy C57BL/6 mice aged 6–8 weeks old. Mechanistically, it is found that CAP promotes microtubule-associated protein 1A/1B-light chain 3 (MAP1LC3, LC3)-associated phagocytosis (LAP) in macrophages to defend against Pg. Therefore, CAP is proposed a potential therapy for effectively alleviating periodontitis through regulating the bactericidal activity of macrophages.

Periodontitis, a chronic inflammatory oral disease resulting from plaque biofilms, affects about 743 million individuals worldwide. However, the efficacy of current treatments is hampered by challenges in delivering antibiotics to recalcitrant oral biofilms and bacterial resistance, thereby impeding successful treatment of infectious diseases. To address the issues, an antibacterial photothermal material was designed, comprising a spherical structure of zinc oxide (ZnO) wrapped with triiron tetraoxide (Fe3O4). The outer layer of the material adsorbed epsilon-polylysine (EPL) by electrostatic action, ultimately leading to the fabrication of Fe3O4/ZnO/EPL nanoparticles (FZE NPs). The Fe3O4 core endowed the nanoparticles with efficient photothermal properties, facilitating the dispersion of dense biofilms, which dramatically promoted the adsorption and penetration of ZnO and EPL into the biofilms to effectively kill bacteria in biofilms in vitro with enhanced sterilization ability. Additionally, upon dissolution in aqueous media, EPL acts as a positively charged antimicrobial peptide that adsorbs onto the surface of negatively charged bacterial membranes, thereby effectively modulating inflammatory responses. In order to ascertain the efficacy of FZE NPs, an investigation was conducted into their antimicrobial effects against the periodontitis-associated pathogen Porphyromonas gingivalis (P. gingivalis) in vitro. Furthermore, the antiperiodontitis potential of FZE NPs was evaluated in Sprague–Dawley (SD) rats of ligamentous periodontitis. In addition, toxicity evaluations indicated that the material had an acceptable biosafety profile in vitro and in vivo. In summary, the nanospheres (FZE NPs) represent a promising therapeutic strategy for the treatment of periodontitis.

Porphyromonas gingivalis has been demonstrated to have the strongest association with periodontitis. Within the host, P. gingivalis relies on acquiring iron and heme through the aggregation and lysis of erythrocytes, which are important factors in the growth and virulence of P. gingivalis. Additionally, the excess obtained heme is deposited on the surface of P. gingivalis, protecting the cells from oxidative damage. Based on these biological properties of the interaction between P. gingivalis and erythrocytes, this study developed an erythrocyte membrane nanovesicle loaded with gallium porphyrins to mimic erythrocytes. The nanovesicle can target and adhere with P. gingivalis precisely, being lysed and utilized by P. gingivalis as erythrocytes. Ingested gallium porphyrin replaces iron porphyrin in P. gingivalis, causing intracellular metabolic disruption. Deposited porphyrin generates a large amount of reactive oxygen species (ROS) under blue light, causing oxidative damage, and its lethality is enhanced by bacterial metabolic disruption, synergistically killing P. gingivalis. Our results demonstrate that this strategy can target and inhibit P. gingivalis, reduce its invasion of epithelial cells, and alleviate the progression of periodontitis.

Periodontitis is an oral immunoinflammatory disease, and macrophages play a crucial role in its pathophysiology. However, macrophage death during antibacterial activities will exacerbate inflammation and tissue damage. Porphyromonas gingivalis is a major constituent of subgingival biofilm plaques in periodontitis, but the effects and precise molecular mechanisms by which it triggers macrophage death remain unknown. Here we found that P. gingivalis infection notably activated multiple death pathways in bone-marrow-derived macrophages, including pyroptosis, apoptosis and necrosis. Furthermore, using RNA sequencing, we identified that P. gingivalis infection markedly increased the expression of Z-DNA binding protein 1 (Zbp1) in bone-marrow-derived macrophages. Initially identified as an interferon-induced tumor-associated protein, Zbp1 serves as an upstream sensor that regulates cell death by activating PANoptosis. Mechanistically, P. gingivalis induced a mitochondrial stress response, prompting the release of mitochondrial DNA. This mitochondrial DNA then interacted with Zbp1, consequently augmenting its downstream PANoptosis signals. In addition, P. gingivalis stimulated macrophage Zbp1 expression through the Tlr2/4–JNK–Stat3/5 pathway, exacerbating macrophage death. Importantly, blocking the biosynthesis of endogenous Zbp1 by pharmacological delivery with microneedles improved the survival of P. gingivalis -infected macrophages and inhibited periodontal tissue destruction. These findings highlight Zbp1 as a potential therapeutic target for P. gingivalis -induced periodontitis.

A great challenge in esophageal squamous cell carcinoma (ESCC) research lies in the scarcity of a model that precisely replicates the tumor microenvironment, pathological conditions, and therapeutic responses. To address these limitations, we propose an innovative biomimetic esophageal cancer model utilizing tumor-cell-laden hollow microfibers fabricated through microfluidic spinning. This approach enables precise fluid manipulation within microchannels, facilitating versatile integration, diverse flow configurations, and rapid response times. On this basis, hydrogel microfibers were developed to possess adjustable shell thicknesses, offering a three-dimensional (3D), biocompatible, and porous matrix that supports sustained cell adhesion, proliferation, and spatial cellular interactions. This model elucidates the interaction between Porphyromonas gingivalis (Pg) and the malignant progression of ESCC. Overall, by enabling the co-culture of tumor stromal cells and ESCC cells, this study provides an innovative and promising platform for investigating the pathology of esophageal cancer, promoting drug discovery, and exploring other pertinent clinical applications for ESCC.

Antibiotics can kill bacteria, but their continued use can easily lead to drug resistance, particularly the main pathogenic bacteria of periodontitis, Porphyromonas gingivalis. However, to avoid drug resistance, carbon quantum dots (CDs) have great potential as a bioactive material in antimicrobial therapy. Herein, we use ornidazole as raw material to prepare CDs of different sizes by microwave irradiation and screen CDs with fluorescence and bacteriostatic properties. The inhibition experiments and live/dead assays of P. gingivalis exhibited outstanding antibacterial effects. This research aimed to develop nano-level antibacterial active materials that also have fluorescence traceability. This study offers a different method for the development of multifunctional CDs, provides valuable strategies for the treatment of diseases associated with P. gingivalis, and predicts great application prospects in the field of biomedicine.

Porphyromonas gingivalis(P. gingivalis) is a primary pathogen in periodontitis, yet its elimination is limited by complex anatomical structures. Photodynamic therapy (PDT) is a promising adjunct, but its antimicrobial efficacy is compromised in the acidic microenvironment induced byP. gingivalis. Given that 2′,4′,5′,7′-tetraiodofluorescein (TIF) exhibits robust and stable photodynamic activity under acidic conditions, this study investigated the antibacterial effect of TIF-mediated PDT (TIF-PDT) againstP. gingivalis.P. gingivalisATCC 33277 was treated with TIF (5, 10, 20, and 40 μM), light (525 nm), or both. Reactive oxygen species (ROS) generation was assessed at pH 4.5 or 7.4. Bacterial viability and membrane integrity were evaluated by colony-forming unit (CFU) assay and LIVE/DEAD staining. CFU assays demonstrated that TIF-PDT groups achieved an approximately 4-log reduction in bacterial viability compared to the DEMI, Light, and TIF groups, with no dark cytotoxicity and light-alone effects. LIVE/DEAD staining revealed bright yellow fluorescence in the TIF-PDT (40 μM), indicating membrane damage and significantly lower survival rates than controls. TIF-PDT at 10, 20, and 40 μM produced ROS under both neutral and acidic conditions, exhibited low dark cytotoxicity, and demonstrated potent antibacterial activity againstP. gingivalisin vitro, suggesting its potential as an acid-tolerant photosensitizer for periodontitis adjunctive therapy.

Objectives Ferroptosis is associated with multiple inflammatory diseases. Periodontitis is an inflammatory disease mainly caused by oral opportunistic pathogens. However, the ferroptosis–periodontitis relationship has not been thoroughly described. We here analyzed whether ferroptosis is involved in periodontitis. Materials and methods Human gingival fibroblasts (HGFs) were stimulated with P. gingivalis -LPS and ferrostatin-1 (Fer-1, a ferroptosis inhibitor), and changes in mitochondrial morphology, ferroptosis-related factors, and inflammation levels were detected. After the rat experimental periodontitis model was established, changes in ferroptosis-related factors and inflammation levels were re-evaluated in the same manner. Results Porphyromonas gingivalis -LPS-induced mitochondrial shrinkage, an increase in mitochondrial membrane density, and upregulation of reactive oxygen species in HGFs. The expression of prostaglandin-endoperoxide synthase 2, transferrin receptor 1, and malondialdehyde and inflammation levels were upregulated, whereas the expression of solute carrier family seven member 11, glutathione peroxidase 4, superoxide dismutase, and glutathione were downregulated. Fer-1 attenuated these aforementioned changes and inflammation levels induced by P. gingivalis -LPS. The in vivo experiment results were consistent with the in vitro experiment results. Conclusions Ferroptosis is involved in inflammatory processes in HGFs upon P. gingivalis -LPS stimulation. Ferroptosis is observed in the gingival tissue of periodontitis rats.

Oral diseases stemming from oral pathogenic bacteria pose a significant global health concern, and current methods for managing these pathogens have limitations. Plasma-activated water (PAW), containing various reactive species, emerges as a promising disinfectant with impressive inactivation capabilities. In this study, PAW prepared by mixed-mode plasma-activated gases was applied to inactivate oral pathogenic bacteria, including Streptococcus mutans and Porphyromonas gingivalis . The PAW could reduce more than 6.1-log 10 planktonic bacteria and 4.1-log 10 bacteria within biofilm, respectively, and PAW treatment of planktonic bacteria effectively inhibited biofilm formation. Compared to chlorhexidine, PAW exhibited superior inactivation effects in both planktonic bacteria and biofilm. This study presented a potent strategy for bacteria eradication to reduce the incidence of oral diseases.

Introduction Modulating the inflammatory response of human gingival fibroblasts (hGFs) is important for the control of periodontal inflammation because it is a key event in the pathogenesis of periodontitis. Here, we aimed to determine whether polyglucose sorbitol carboxymethyl ether (PSC)-coated superparamagnetic iron oxide nanoparticles (SPIONs) protect hGFs against invasion and inflammatory stimulation by Porphyromonas gingivalis (P. gingivalis). Methods First, we determined the cytotoxicity and antimicrobial activity of PSC-SPIONs. Then, their effects on invasion of hGFs by P. gingivalis were evaluated by counting invading P. gingivalis, fluorescence staining, and transmission electron microscopy. The effect of PSC-SPIONs on inflammation in hGFs induced by P. gingivalis lipopolysaccharide was evaluated by measurement of reactive oxygen species (ROS), and enzyme-linked immunosorbent assays, quantitative reverse transcription-polymerase chain reaction, and Western blotting of key indicator molecules. The effects of dimercaptosuccinic acid (DMSA)-coated SPIONs and the free form of PSC alone were also tested and compared with those of PSC-SPIONs.Results PSC-SPIONs (25 μg/mL) are cytocompatible with hGFs and exhibit no antimicrobial effects on P. gingivalis. However, they inhibit invasion of hGFs by P. gingivalis at 15 μg/mL. They also decrease ROS production and inflammatory cytokine secretion by hGFs at 5, 15, and 25 μg/mL, by downregulating activation of the nuclear factor-kappa B signaling pathway. Furthermore, PSC alone does not inhibit inflammation, while DMSA-SPIONs do. This indicates that the nanosize effects of PSC-SPIONs, rather than their coating material, play the dominant role in their anti-inflammatory activity. Conclusion PSC-SPIONs protect hGFs against P. gingivalis invasion and inflammatory stimulation. Thus, they have potential for clinical application in control of periodontal inflammation.
