Preparation of metal–organic framework combined with Portulaca oleracea L. extract electrostatically spun nanofiber membranes delayed release wound dressing

Yize Wang,Hua Kang,Jao Hu,Heming Chen,Huimin Zhou,Ying Wang,Huizhen Ke

In this study, we prepared a polyacrylonitrile (PAN) composite nanofiber membrane comprising Portulaca oleracea L. extract (POE) and a zinc-based metal–organic framework (MOF) by an in situ growth method as a potentially new type of wound dressing with a slow drug-release effect, to solve the problem of the burst release of drugs in wound dressings. The effects of the MOF and POE doping on the nanofiber membranes were examined using scanning electron microscopy (SEM) and FTIR spectroscopy. SEM analysis revealed the dense and uniform attachment of MOF particles to the surface of the nanofiber membrane, while FTIR spectroscopy confirmed the successful fusion of MOF and POE. Furthermore, investigations into the water contact angle and swelling property demonstrated that the incorporation of the MOF and POE enhanced the hydrophilicity of the material. The results of the in vitro release test showed that the cumulative release rate for PAN/MOF/POE60 decreased from 66.5 ± 2.34% to 32.18 ± 1.31% in the initial 4 h and from 90.54 ± 0.79% to 65.92 ± 1.95% in 72 h compared to PAN/POE, indicating a slowing down of the drug release. In addition, the antimicrobial properties of the fiber membranes were evaluated by the disc diffusion method, and it was evident that the PAN/MOF/POE nanofibers exhibited strong inhibition against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). The antioxidant properties of the nanofiber membranes loaded with POE were further validated through the DPPH radical scavenging test. These findings highlight the potential application of the developed nanofiber membranes in wound dressings, offering controlled and sustained drug-release capabilities.

Portulaca oleracea L. Extract and Its Alkaloid Oleracein E Alleviates Cardiac Remodeling and Heart Failure in Mice by Targeting STAT2 Through the MAPK Signaling Inflammatory Pathway

Risheng Zhao,Min Zhang,Gege Yang,Wei Liu,Hui Yu,Shuang Yan,Ting Ren,Linxin Zhang,Mengyang Wang,Haiming Sun

Hypertensive heart failure pathogenesis involves angiotensin II (Ang II)-mediated mechanisms through both hemodynamic overload and direct cellular signaling pathways. This study investigates the therapeutic potential of Portulaca oleracea L. (POL) extract and its active constituent Oleracein E (OE) in attenuating Ang II-induced pathological cardiac remodeling and subsequent heart failure progression. An experimental model of hypertensive heart failure was established in C57BL/6 mice through continuous subcutaneous infusion of Ang II for 4 weeks, with Oleracein E (OE) intervention administered during the final 2 weeks of the protocol. To elucidate the molecular mechanisms underlying OE's cardioprotective effects, we employed an integrated approach combining RNA sequencing analysis, molecular docking simulations, and target validation through drug affinity response target stability (DARTS) and cellular thermal shift assay (CETSA) techniques. The experimental results demonstrated that both POL extract and its active component OE exerted significant cardioprotective effects against Ang II-induced cardiac dysfunction in murine models, primarily through attenuation of pathological cardiac hypertrophy and suppression of inflammatory responses. Transcriptomic profiling via RNA sequencing identified the MAPK signaling pathway as a critical mediator of these protective effects. Subsequent transcription factor analysis revealed STAT2 as a key regulatory component in this pathway. Importantly, OE treatment effectively mitigated inflammatory responses in both in vivo cardiac tissues and in vitro cultured cardiomyocytes by specifically inhibiting the Ang II-activated MAPKs/STAT2 signaling cascade. Genetic ablation experiments further confirmed the essential role of this pathway, as the anti-inflammatory efficacy of OE was completely abolished in cardiomyocytes with MAPKs or STAT2 deficiency. Molecular interaction studies employing in silico docking simulations combined with experimental validation through DARTS and CETSA techniques provided direct evidence of physical binding between OE and the STAT2 protein. Our findings demonstrate that Oleracein E (OE) confers cardioprotection against Ang II-induced myocardial injury through a novel mechanism involving STAT2 targeting and subsequent suppression of MAPK signaling-mediated inflammatory cascades. These results not only elucidate a previously unrecognized pharmacological pathway but also suggest that OE and its derivatives hold significant therapeutic promise for the clinical management of hypertensive heart failure, potentially offering a targeted approach to modulate pathological cardiac remodeling.

Protective Effects of Different Molecular Weights of Purslane (Portulaca oleracea L.) Aqueous Extract on DSS-Induced Ulcerative Colitis in Mice

Ke Ning,Yameng Duan,Weiwei Tong,Yue Chen,Qinghui Zhang,Qiuhong Xie,Hongyu Xiang

Purslane, a common wild vegetable, contains active substances with various biological functions. However, its effects have been under-investigated in ulcerative colitis (UC). Therefore, this study investigated the therapeutic effects of purslane macromolecular (POEM) and small molecular extracts (POES) on dextran sulfate sodium (DSS)-induced UC in mice. Membrane separation was used to obtain extracts of different molecular weights, and their compositional differences were compared using liquid chromatography–mass spectrometry (LC/MS). POEM contained more proteins and polysaccharides, whereas POES contained more organic acids and alkaloids. These differences in composition were directly responsible for the different degrees of remission of the alleviated UC in model mice. POEM may alleviate UC by regulating the antioxidant capacity and the gut microbiota, whereas the major alleviatory effect of POES was primarily related to the regulation of antioxidant capacity. The POEM and POES effects identified in this study provide a theoretical basis for the development of purslane as a functional food. Keywords:ulcerative colitis;macromolecular extract;purslane;oxidative stress;small molecular extract;gut microbiota;Portulaca oleraceaL.

Purification, structural characterization, and immunomodulatory activity of two polysaccharides from Portulaca oleracea L.

Yanxi Li,Mengjie Ren,Huan Yan,Lan Luo,Xin Fang,Li He,Wenyi Kang,Mingyi Wu,Haiyang Liu

In present study, two water-soluble polysaccharides designated as POL-1 and POL-2 were purified from purslane and their structural characteristics as well as immunomodulatory activity were investigated. The weight-average molecular weight ( M w ) of POL-1 and POL-2 were determined to be 64,100 Da and 21,000 Da, respectively. Comprehensive techniques including UV, IR, GC–MS, and NMR were applied to deduced that POL-1 was a pectin polysaccharide homogalacturonan (HG) consisting of →4)- α -Gal p A-(1→ with methyl ester degree of 9.71 % and acetylation degree of 0.34 %, while POL-2 was composed of a 1, 4-linked β -Gal p backbone substituted by short side chain →4)- α -Glc p -(1→ and →6)- α -Glc p -(1→. The →4)- α -Glc p -(1→ was attached at the O -6 position of →4)- β -Gal p -(1→. TEM further revealed that POL-1 was non-branched single chains, while POL-2 was entangled microstructure with side chains. Moreover, POL-2 significantly promoted macrophage phagocytosis as well as the secretion of NO and cytokines (TNF- α , IL-6) through activating NF-κB signaling pathway, thus demonstrating potential immunomodulatory activity. These findings suggested that purslane may be exploited as a potential adjuvant and dietary supplement with immunostimulatory purpose.

Structural characteristics of a highly branched and acetylated pectin from Portulaca oleracea L.

Wei Tang,Dan Liu,Yu Li,Ming-Yue Zou,Yu-Chen Shao,Jun-Yi Yin,Shao-Ping Nie

The water-extracted polysaccharide from Portulaca oleracea L. mainly consisted of two pectic polysaccharides designated as POWP-H and POWP-L. The POWP-L was rich in linear homogalacturonan with GalA content of 77.6%, while POWP-H might be a rhamnogalacturonan I (RG-I) pectin composed of Rha (15.7%), Ara (31.7%), Gal (42.5%) and GalA (10.1%). The structure of POWP-H was elucidated by partial degradation, methylation analysis combined with GC-MS method and NMR spectroscopy. It had a weight-average molecular weight of 1.2 × 10 6  g/mol and high degree of branch of 0.57. The presence of abundant arabinogalactan-II and certain galactan side chains in POWP-H was verified by the T-Ara f (17.4%), 1,3,6-Gal p (11.8%), 1,3-Gal p (8.1%), T-Gal p (13.4%) and 1,4-Gal p (14.3%). The evidence for the RG-I backbone of POWP-H was demonstrated by the structure analysis of its degraded product POWP-H-H, which had the backbone of →4)-α-Gal p A-(1 → 2)-α-Rha p -(1 → 4)-α-Gal p A-(1 → 2,4)-α-Rha p -(1→ substituted by short side chain of β-Gal p -(1 → 4)-β-Gal p -(1→ at O -4 position of →2,4)-α-Rha p -(1 → . Moreover, the POWP-H-H was mainly O -acetylated at O -3 position of α-Rha p with 24.8% acetylation degree. In conclusion, POWP-H was mainly the highly branched and acetylated RG-I pectin with relative short RG-I backbone and abundant arabinogalactan II and certain galactan side chains. Besides, it adopted a flexible chain conformation in 0.1 M NaNO 3 solution.

Portulaca oleracea L. polysaccharide alleviates dextran sulfate sodium-induced ulcerative colitis by regulating intestinal homeostasis

Ke Ning,Chao Shi,Yan-Yu Chi,Yong-Fei Zhou,Weiwei Zheng,Yameng Duan,Weiwei Tong,Qiuhong Xie,Hongyu Xiang

Portulaca oleracea L. (purslane) is a vegetable that contains a variety of active compounds with nutritional properties and has the potential to treat ulcerative colitis (UC). However, the mechanisms underlying the effects of Portulaca oleracea L. polysaccharide (POP) in alleviating UC remain unclear. In this study, we prepared an aqueous extract of purslane and separated a fraction with molecular weight >10 kDa using membrane separation. This fraction was used to isolate POP. The effect of POP on gut microbiota and colon transcriptome in dextran sulfate sodium-induced UC model mice was evaluated. POP treatment reduced inflammation and oxidative stress imbalance in UC mice. In addition, POP improved the intestinal barrier and regulated intestinal homeostasis . Importantly, POP was found to regulate gut microbiota, maintain the levels of retinol and short-chain fatty acids in the gut, promote the proliferation and differentiation of B cells in the colon, and increase the expression of immunoglobulin A. These results provide novel insights into the role of POP in regulating intestinal homeostasis, which should guide further development of POP as a functional food.

Portulaca oleracea L.-Derived Extracellular Vesicles Alleviate Renal Lipotoxicity in Diabetic Kidney Disease by Regulating the NLRP3 Pathway

Yao Zhou,Huijia Zhang,Yijing Li,Yingying Li,Yupeng Tao,Kejia Zhang,Buhui Liu,Qi Wu,Kun Gao

Background Diabetic kidney disease (DKD) remains a leading indication for renal replacement therapy, with existing drugs having many limitations. Portulaca oleracea L.-derived extracellular vesicles (PO-EVs) exhibit anti-inflammatory, antioxidant, and immune-regulatory properties. However, their function in DKD has yet to be fully elucidated. Objective This investigation sought to examine the interplay between lipid metabolism and inflammation in renal tubular epithelial cells in DKD, and to clarify the role and underlying mechanism of PO-EVs in modulating lipid metabolism–inflammation crosstalk to mitigate DKD progression. Methods PO-EVs underwent isolation via differential ultracentrifugation and characterization through transmission electron microscopy (TEM), particle size analysis, zeta potential measurement, and untargeted metabolomics. In vivo, db/db mice (DKD model) were given PO-EVs intragastrically, with renal function and tissue indicators detected, and mechanisms explored by transcriptomics and proteomics. In vitro , palmitic acid (PA)-induced lipotoxicity in normal rat kidney tubular epithelial cells (NRK-52E) was used to explore the effects of PO-EVs. Results PO-EVs significantly enhanced renal function and mitigated renal tissue lipid deposition in db/db mice. They also alleviated PA-induced cellular lipid accumulation and mitochondrial damage, inhibited NLRP3 inflammasome-related proteins, and enhanced fatty acid oxidation (FAO) and ATP synthesis. Suppressing NLRP3 expression markedly improved mitochondrial morphology, reduced lipid accumulation and increased peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α) expression in NRK-52E. Conclusion PO-EVs disrupt the metabolism-inflammation vicious cycle by inhibiting NLRP3 and upregulating PGC-1α, thereby mitigating renal lipotoxicity and delaying DKD progression, providing a basis for DKD therapy. Application of choline chloride-based deep eutectic solvents for the extraction of dopamine from purslane (Portulaca oleracea L.)

Kunlin Liu,Jia-Neng Tan,Yulei Wei,Changkun Li,Yuqing Dou,Zhongfeng Zhang

Deep eutectic solvents (DESs) are a recently developed class of green, efficient, and versatile alternatives to traditional organic solvents in natural product extractions. Inspired by the emergence of natural DESs and their green nature, we herein optimized the experimental variables for the extraction of dopamine from purslane ( Portulaca oleracea L., POL) using DESs. Nine choline chloride (ChCl)-based DESs were tested to select the optimum solvent for ultrasound-assisted extraction (UAE), and the combination of ChCl:glycerol (1:2) with 30% water extracted more dopamine from POL than ethanol. The amount of dopamine in the extracts was quantified by HPLC equipped with a fluorescence detector. The influence of UAE treatment on the morphology of plant material was studied by SEM. Based on single-factor experiments, the UAE conditions were optimized by response surface methodology using the Box-Behnken design. The optimum conditions were: extraction time of 30 min, water content in DESs of 32%, temperature of 58 °C, and plant material-liquid ratio of 40 mg/mL, which resulted in 2.96 mg of dopamine per 1 g of POL. The percentage of water in DESs and the extraction temperature were the most significant factors influencing the extraction efficiency of dopamine from POL. The results of this study promote the application of DESs for green and efficient extractions of bioactive alkaloids and other target molecules from plant materials.

Mechanism of interaction between Portulaca oleracea L. extract and pork myofibrillar protein: Insights from spectroscopic and molecular docking analyses

Haijun Chang,Xin Wen,Yu Huang,Yu Hu,Yuanwei Shi

The research aimed to elucidate the interaction and mechanism between the Portulaca oleracea L. extract (POE) and the pork myofibrillar proteins (MPs) from the perspectives of fluorescence spectroscopy and molecular docking. The results showed that POE significantly increased intermolecular hydrogen bonds and hydrophobic interactions in MPs ( P  < 0.05). Among the non-covalent forces between POE and MPs, hydrophobic interactions were dominant followed by hydrogen bonds. POE would cause a decrease in the endogenous fluorescence intensity of MPs, and the quenching type was static quenching. The binding molar ratio of POE to MPs was 1:1, and the formed complex exhibited high stability. The thermodynamic constants ∆G < 0, ∆H > 0, and ∆S > 0 indicated that the binding process between POE and MPs was spontaneous, with hydrophobic interactions being the primary driving force. Additionally, the nine most abundant compounds in POE were selected for molecular docking with myosin, and the binding energies were all negative, confirming that POE could spontaneously interact with MPs. The molecular docking results further demonstrated that hydrophobic interactions and hydrogen bonding were the main binding forces, with Leu270 potentially serving as a key amino acid residue in the interaction between POE and MPs. This study provides valuable insights into the molecular interactions between Portulaca oleracea L. extract and pork myofibrillar proteins, offering a theoretical basis for phenolic small molecule substances in regulating protein oxidation and potential applications in improving meat product quality.

Purslane (Portulaca oleracea L.) polysaccharide attenuates carbon tetrachloride-induced acute liver injury by modulating the gut microbiota in mice

Jiahui Li,Yuyang Chen,Shuang Zhang,Yuehan Zhao,Demeng Gao,Jiaying Xing,YuYan Cao,Guangyu Xu

This study investigated the preventive and protective effects of Portulaca oleracea polysaccharides (PP) on Acute liver injury (ALI) in mice and its regulatory effects on intestinal microorganisms, and explored the underlying protective mechanisms. Initially, PP was administered, and then CCl4 was used to induce the mouse ALI model. Serum and liver markers were measured by ELISA. The fecal microbiota was analyzed by 16S rRNA sequencing. The results showed that PP significantly decreased the expression levels of ALT and AST in the serum of mice. The expression levels of MDA, TNF-α, and IL-6 in liver tissue were found to be reduced, while the levels of GSH and SOD increased. At the same time, PP increased the number of Bacteroides , reduced the number of Proteobacteria , activated the GAG degradation pathway, protected the integrity of the intestinal barrier, inhibited oxidative stress and reduced inflammation, thereby assisting the prevention and protection of ALI.

Laccase-Catalyzed the Polymerization of Portulaca Oleracea L. Extract as a Precursor for Ecological Dyeing of Pure Cotton Fabric

Zhang Qi,Yang Ce,Gu Mingqi,Qin Shaoxuan,Yin Jianhua,Wang Xiangrong,Zhang Wei

The integration of natural dyes and biomass-based dyeing auxiliaries holds huge potential to address the environmental sustainability challenges, which the textile industry encounters. This research employed Portulaca oleracea L. extract as a natural dye, utilizing laccase as a catalyst for polymerizing the dye, thereby creating a precursor that enhances its eco-friendly dyeing attributes on cotton fabrics. The initially exhibited suboptimal direct dyeing performance on cotton fabrics, resulting in a faint yellow hue and a K/S value of 0.66. However, following laccase catalysis, the depth of color on the dyed fabric significantly increased, with a K/S value reaching 4.41, and the dye uptake rate increased from 14.54% to 33.69%. The resulting dyed fabric exhibited excellent fastness to soap washing, light, and rubbing, all scoring above levels 4 to 5. Analysis of dyeing kinetics and adsorption models indicated that the dyeing process was consistent with both the pseudo-first-order and pseudo-second-order kinetic models, suggesting physical and chemical bonding between the precursor and the fiber. Further investigations employing ultraviolet spectrum analysis, matrix-assisted laser desorption/ionization time of flight mass spectrometry, and gas chromatography-mass spectrometer revealed the fundamental composition of the natural dye, the molecular weight, and structural characteristics of the precursor, substantiating that the catalytic mechanism involves laccase-catalyzed oxidation. Additionally, the dyed cotton yielded an ultraviolet protection factor value of 230 and a long-lasting herbal fragrance, accurately identified by an electronic nose. These results confirm the viability of intensifying fabric color using the laccase-catalyzed system. This technique significantly expands the environmental applications of its extract.
