Full-length normalization subtractive hybridization analysis provides new insights into sexual precocity and ovarian development of red swamp crayfish Procambarus clarkii

Hong-Yue Tan,Guang-Ming Shao,Peng-Fei Kang,Yu-Feng Wang

The red swamp crayfish Procambarus clarkii is an important freshwater economic crayfish in Chinese inland due to its high market value and consumer demand. The sexual precocity in this species has received considerable attention in recent years because more and more individuals matured at small sizes, which devalues the commercial production. In this study, by the full-length normalization subtractive hybridization (FNSH) method we identified 16 differently expressed sequences between precocious and normal grown P . clarkii ovaries. Of the down-regulated sequences in precocious ovaries, one encodes a peptide which shared the highest identity to C1q-like family and contained a typical C1q domain at the C-terminal, thus it was named PcC1q-like . QRT-PCR analysis showed that PcC1q-like was transcribed specifically in ovary, with higher expression level in vitellogenic stages. In situ hybridization exhibited specific expression of PcC1q-like in follicular cells around the vitellogenic oocytes . Knockdown of PcC1q-like by injection of double strand RNA (dsRNA) resulted in disruption of follicular cells, decreased transcripts of vitellogenin ( Vg ) and vitellogenin receptor ( VgR ), and reduced progesterone content. These results suggest that PcC1q-like may promote ovarian development by stimulating the synthesis and transportation of vitellogenin into oocytes. These data provide insights into the molecular regulatory mechanisms of sexual precocity and ovarian development in crustaceans. Statement of relevance The red swamp crayfish Procambarus clarkii is one of the most important freshwater aquaculture species in Chinese inland. We screened 16 sequences that are potentially involved in sexual precocity of P. clarkii . One candidate was demonstrated to be a novel ovary-specific C1q-like gene of P. clarkii , thus named PcC1q-like . Knockdown of PcC1q-like resulted in disruption of follicular cells, decreased transcripts of Vg and VgR , and reduced progesterone, suggesting its essential role in vitellogenic ovarian development of P. clarkii .

Identification and expression of an ecdysteroid-responsive amylase from red crayfish Procambarus clarkii

Peng Tao,Wang Daojun,Yu Yingying,Liu Chaoliang,Zhu Baojian

Amylases catalyze the hydrolysis of starch material and play important roles in the regulation of many physiological processes. Here, we report an amylase gene denoted Pc - Amy in the crayfish Procambarus clarkii (Decapoda: Cambaridae). The Pc - Amy gene encodes a polypeptide of 514 amino acids, and the deduced amylase protein is highly homologous to those from crustaceans. Real-time quantitative reverse transcription PCR (qRT-PCR) analysis showed that Pc - Amy is highly expressed in hepatopancreas during the intermolt period. SDS-PAGE and western blot analysis demonstrated that a 58-kDa recombinant protein is successfully expressed in Escherichia coli cells. The expression of Pc - Amy was significantly downregulated in hepatopancreas at 48 h after ecdysteroid induction, as confirmed by qRT-PCR and western blot analyses. In addition, RNA interference with the ecdysteroid receptor had an impact on the expression of Pc - Amy . These results suggest that Pc - Amy is an ecdysteroid-responsive gene which plays a role in the developmental process of P. clarkii .

Effects of Aminomethylphosphonic Acid on the Transcriptome and Metabolome of Red Swamp Crayfish, Procambarus clarkii

Tao Mao,Jinhua Gan,Keping Yuan,Li He,Yali Yu,Ziduo Liu,Yuntao Zhou,Gaobing Wu

Red swamp crayfish,Procambarus clarkii(P. clarkii), is an important model crustacean organism used in many types of research. However, the effects of different doses of aminomethylphosphonic acid (AMAP) on the transcriptome and metabolites ofP. clarkiihave not been explored. Thus, this study investigated the molecular and metabolic mechanisms activated at the different exposure dosages of AMAP inP. clarkiito provide new insights into the strategies ofP. clarkiiin response to the high concentrations of AMAP in the environment. In the present study, theP. clarkiiwere divided into three groups (control group; low-dosage AMAP exposure; high-dosage AMAP exposure), and hepatopancreatic tissue samples were dependently taken from the three groups. The response mechanisms at the different dosages of AMAP were investigated based on the transcriptome and metabolome data ofP. clarkii. Differentially expressed genes and differentially abundant metabolites were identified in the distinct AMAP dosage exposure groups. The genes related to ribosome cell components were significantly up-regulated, suggesting that ribosomes play an essential role in responding to AMAP stress. The metabolite taurine, involved in the taurine and hypotaurine metabolism pathway, was significantly down-regulated.P. clarkiimay provide feedback to counteract different dosages of AMAP via the upregulation of ribosome-related genes and multiple metabolic pathways. These key genes and metabolites play an important role in the response to AMAP stress to better prepare for survival in high AMAP concentrations.Keywords:AMAP;P. clarkii;transcriptomics;metabolome

Transcriptomic analysis of the effects of dietary chitosan on the molting process in Procambarus clarkii

Jie Wang,Xiang-Ming Chang,Chen-Yang Zhang,Rui-Geng Niu,Xiao-Tong Cao,Jiang-Feng Lan

Molting is a critical physiological process for growth and development in arthropods. After molting, Procambarus clarkii (red swamp crayfish) exhibits a distinctive behavior of ingesting its exuviae. However, the functional significance of this behavior during the molting process remains unclear. Histological analysis revealed distinct intestinal remodeling during molting, characterized by the sequential detachment of the peritrophic membrane (PM) and the sloughing of the degenerating intestine (DI) into the lumen. To simulate the physiological effects of exuviae ingestion, chitosan, a key bioactive derivative of the exoskeleton, was administered via oral gavage. Transcriptomic analysis was performed to identify key events in the intestine and hemocytes following chitosan supplementation. The results showed that chitosan supplementation upregulates the expression of genes associated with immune responses and enzyme inhibitors in the intestine. Meanwhile, transcriptomic analysis of hemocytes reveals the enrichment of pathways involved in cell motility and exoskeleton hardening-related processes. This process is potentially linked to the enhanced expression of immune-related genes and shapes the microbial community during molting, thereby providing a novel foundation for investigating disease management strategies in crayfish during the molting cycle.

Gallic acid and luteolin-based self-assembled nanoparticles alleviate white spot syndrome virus infection in Procambarus clarkii

Songlin Luo,Fansheng Zeng,Guohao Chen,Shunqiang Wei,Chushun Yi,Xuemin Wei,Yinghui Wang,Aiguo Huang

White Spot Syndrome Virus (WSSV) is a major viral pathogen severely threatening crustacean aquaculture. Although natural products possess antiviral potential, their application remains limited due to stability and utilization constraints. This study employed a self-assembly strategy to construct a natural product-based metal-organic nanostructure of luteolin-loaded magnesium-gallic acid (Mg-GA) metal-organic framework (LM), to enhance the anti-WSSV efficacy of luteolin. Results indicate that the prepared LM exhibits a regular prismatic nanostructure with uniform particle size distribution, stable coordination structure, and good biocompatibility. In vivo experiments demonstrated that compared to single luteolin or Mg-GA treatment groups, LM significantly reduced WSSV viral copy numbers in infected crayfish Procambarus clarkii and markedly improved host survival rates. Gene expression analysis revealed that this self-assembled nanomaterial significantly downregulated expression of key viral genes IE1 , DNApol , and VP28 , while upregulating host antioxidant-related genes CAT and SOD , apoptosis-regulating genes BI-1 , and downregulating apoptosis-regulating genes BAX , inflammation-related genes COX1 and COX2 . This indicates its ability to synergistically inhibit viral replication and enhance host defense capabilities. Histopathological results further confirmed that LM effectively alleviated pathological damage caused by WSSV to gill and intestinal tissues, with no apparent toxicity observed in healthy individuals. In summary, the self-assembled natural product metal-organic nanostructure significantly enhances the anti-WSSV efficacy of single compounds, offering a novel nanostrategy for controlling viral diseases in aquaculture.

Umifenovir protects Procambarus clarkii against white spot syndrome virus by suppressing viral replication and modulating innate immunity

Cheng Chen,Bo-Han Li,Chao-Yu Wang,Er-Long Wang,Tian-Qiang Liu,Zhe Zhao

White spot syndrome virus (WSSV) remains one of the most destructive viral pathogens in crustacean aquaculture, yet effective antiviral agents are still lacking. Umifenovir is a clinically approved broad-spectrum antiviral drug with a favorable safety profile and immunomodulatory properties; however, its antiviral potential in aquatic invertebrates has not been explored. In this study, we systematically evaluated the antiviral efficacy and immunological mechanisms of umifenovir against WSSV infection in the crayfish Procambarus clarkii . Toxicity assessment and ADMET predictive analysis confirmed a wide safety window of umifenovir in crayfish. In vivo infection experiments demonstrated that umifenovir significantly suppressed WSSV replication in a dose-, tissue-, and time-dependent manner, achieving up to 97.26% inhibition at 30 mg/kg after 24 h. The treatment also reduced the transcription of viral genes from immediate-early to late stages and improved the survival rate of infected hosts. Both preventive and therapeutic administration conferred robust antiviral protection. Mechanistically, umifenovir inhibited the transcription of key viral genes required for WSSV replication and disrupted the virus-mediated manipulation of innate immune signaling pathways. Umifenovir further enhanced antioxidant capacity, alleviated oxidative stress and inflammatory responses, and attenuated virus-induced apoptosis. In addition, umifenovir reduced abnormal protein accumulation associated with viral replication while increasing lysosomal enzyme activity, indicating restoration of protein homeostasis and strengthened innate immune function. Collectively, these findings provide the first evidence that umifenovir exerts potent anti-WSSV activity in crustaceans by coordinately suppressing viral replication and modulating host immune responses, offering new mechanistic insights into its immunomodulatory effects. Given its favorable safety profile and feasibility for industrial synthesis, umifenovir represents a promising antiviral candidate for immunological intervention and provides a potential translational basis for future disease control strategies in aquaculture.

Luteolin in Lonicera japonica inhibits the proliferation of white spot syndrome virus in the crayfish Procambarus clarkii

Hai-Feng Jiang,Cheng Chen,Xin-Yuan Jiang,Jing-Lei Shen,Fei Ling,Peng-Fei Li,Gao-Xue Wang

White spot disease caused by the white spot syndrome virus (WSSV) is a highly lethal infectious disease of crustacean aquaculture, with no effective means of control. Active compounds from herbal medicines are promising candidates for the development of antiviral agents for WSSV. We evaluated the efficacy of 12 herbal medicines in inhibiting WSSV proliferation in a WSSV infection model ( Procambarus clarkii ), Lonicera japonica extract had the strongest inhibitory effect on WSSV replication. Luteolin (LUT), the major bioactive compound of L. japonica extract, was further investigated and revealed to have a strong inhibitory effect on WSSV (96.04%, 50 mg/kg). LUT treatment decreased the WSSV load and the expression of important viral genes (immediate-early gene ie1 , DNA polymerase DNApol and envelope protein Vp28 ) in a dose-dependent manner and increased the survival of WSSV-infected crayfish by 60.00% (50 mg/kg). Co-incubation and time-of-addition assays revealed that LUT did not affect the infectivity of viral particles or viral absorption and entry. Expression of immune-related genes such as barrier-to-autointegration ( BAF ), heat shock protein 70 ( Hsp70 ) and signal transducer and activator of transcription ( STAT ), which may be beneficial for viral replication, were significantly decreased ( P  < 0.01) in LUT-treated crayfish. LUT also modified the expression of antioxidants ( SOD , CAT and GST ), pro-inflammatory factor ( COX-1 and COX-2 ), and apoptosis-related ( BAX and BI-1 ) genes ( P  < 0.01) to enhance antioxidant defenses, mitigate inflammation and induce apoptosis, respectively. Overall, LUT could be used as a prophylactic or therapeutic agent against WSSV infection in crustacean aquaculture.

Herbicide atrazine exposure induce oxidative stress, immune dysfunction and WSSV proliferation in red swamp crayfish Procambarus clarkii

Hui Yang,Yinan Jiang,Kaiyuan Lu,Haoran Xiong,Yingying Zhang,Wenzhi Wei

Atrazine is considered as a potential environmental endocrine disruptors and exhibits various toxic effects on animals. It has a great impact in the aquatic ecosystems , but there are few studies on its immunotoxicity in crustaceans. In the present study, the Procambarus clarkii were utilized to assess the immune toxicity after 0.5 mg/L and 5 mg/L atrazine exposure. A significant decrease in total hemocytes count (THC) was observed at 5 mg/L atrazine exposure throughout the experiment. The activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) were significantly inhibited, but the content of reactive oxygen species (ROS) and malondialdehyde (MDA) were up-regulated, indicating the potential oxidative stress . The analysis of the integrated biomarker response (IBR) showed the induction of oxidative stress biomarkers and the inhibition of antioxidants. After 5 mg/L atrazine exposure for 144 h, the integrity of crayfish hepatopancreas was destroyed with disappeared connections between tubules and increased liver tubules vacuoles. The relative expression levels of different immune genes in hepatopancreas after atrazine exposure were measured. Most of these genes were suppressed and exhibited a certain dose-dependent effect. The results of crayfish white spot syndrome virus (WSSV) replication shown the amount of virus in muscle was significantly higher and exhibited a higher mortality rate at 5 mg/L group than other groups. The present study determined the impact of atrazine exposure on WSSV outbreaks, and also provide an important basis for further assessing the occurrence of pesticides on diseases of P. clarkii.

Complex organic acids treatment on crayfish (Procambarus clarkii): the relationship between myofibrillar protein structure and meat quality

Ruyi Zhu,Zhouyi Xiong,Dan Li

Summary The effect of complex organic acids (COA) treatment at different concentrations (0%, 1%, 5%, 10% and 15%) on the myofibrillar protein (MP) structure and meat quality of crayfish ( Procambarus clarkii ) was investigated. The results showed that with increasing concentrations of COA treatment, the sulfhydryl content of MP decreased and dityrosine content increased. The MP secondary structure has a reduced α-helix content and an increased β-fold content. The MP tertiary structure unfolded, exposing hydrophobic and other amino acids to a polar environment. MP aggregation was induced in the 1% and 5% COA treatment groups and MP degradation was induced in the 10% and 15% COA treatment groups. Crayfish meat in the 5% COA treatment group showed improved textural properties due to protein aggregation. It also enhanced the colour, aroma and micromorphology of the crayfish meat.

The antiviral effect and potential mechanism of matrine against white spot syndrome virus infection in crayfish (Procambarus clarkii)

Jia Liu,Cheng Chen,Hui Du,Dexin Wang,Haixiang Ma,Gaoxue Wang,Tao Liu,Erlong Wang

White spot syndrome virus (WSSV), the causative agent of White spot disease (WSD), has caused high mortality and great economic losses in worldwide crustacean industries including shrimp and crayfish. Many bioactive components of Chinese herbs have been reported to be effective to control aquatic viral diseases including WSD. In this study, the antiviral effects of twelve herbal extracts were evaluated in WSSV-infected crayfish model, and Sophora flavescens extract exhibited the highest inhibition rate (87.63%) of WSSV replication. The anti-WSSV effect of matrine , the main bioactive component of S. flavescens, was further investigated. The results showed matrine could significantly inhibit WSSV replication in a dose-dependent manner and exhibited a robust inhibition rate (96.16%) with 50 mg/kg. Matrine both possessed potential preventive and therapeutic effects against WSSV infection and the preventive effect was better than the therapeutic effect. Besides, compared with the control, matrine significantly ( p  < 0.01) improved the survival rate of WSSV-infected crayfish (from 0% to 48.57% on 10th day), notably ( p  < 0.01) suppressed the expression of virus replication-related genes ie1 , DNApol and Vp28 , remarkably ( p  < 0.01) activated C-type lectin ( CTL ) and prophenoloxidase ( ProPo ) and inhibited barrier-to-autointegration ( BAF ) expression to improve crayfish innate immunity against WSSV infection, up-regualated the CAT and GST expression and down-regulated the expression of NF-κB , cyclooxygenases COX-1 and COX-2 genes to balance the oxidative stress and inflammatory injury caused by WSSV. This study will provide a theoretical basis for the development of matrine as an efficient and practical anti-WSSV drugs and meaningful reference for the study of antiviral effect of matrine on other aquatic animal viruses.

Effects of Different Sediment Improvers on the Growth Environment, Innate Immune Responses, and Intestinal Health of Procambarus clarkii

Xinyu Wu,Hao Wu,Yifan Wu,Zhiqiang Xu,Hong Shan,Tianheng Gao

Simple SummarySediment improvers are important mediators of aquatic animals’ growth performance and the surrounding environmental quality. However, the physiological responses of crayfish (Procambarus clarkii) to different sediment improvers remain unclear. Here, we cultivated crayfish using two chemical (potassium monopersulfate and potassium ferrate) and two biological (purple nonsulfur photosynthetic bacteria andBacillus subtilis) sediment improvers at low and high concentrations. After 42 days, we found that the addition of chemical sediment improvers were more effective in improving water quality than biological sediment improvers. By contrast, the application of biological sediment improvers resulted in considerably enhanced final weight, weight gains, and survival rates. In all low-concentration groups, the activity of immune-related enzymes in the hemolymph and hepatopancreas considerably increased, whereas the malondialdehyde activity and mRNA expression of AMP genes (PcALF and PcCru) considerably decreased. Crayfish exposed to low concentrations of sediment improvers exhibited enhanced intestinal and hepatopancreatic integrity. Additionally, the composition of the gut microbiota varied after the addition of different sediment improvers. In summary, our research indicated that different types of sediment improvers not only improved the farming environment but also had varying effects on crayfish. Therefore, an appropriate sediment improver based on specific aquaculture conditions is needed.Sediment improvers are important mediators of aquatic animals’ growth performance and the surrounding environmental quality. Multifunctional electrospun nanofiber films toward efficient deodorization and preservation of crayfish meat (Procambarus clarkii) via a triple-synergistic strategy

Cheng Wang,ZiHan Wang,MengHuan Liu,Ning Jiang,Jia Kan,LiFang Liu,MingZhu Li,ZeFeng Yue,Hao Yu

Although aquatic products are nutritious, they are highly perishable and tend to develop undesirable odors. Traditional methods of preservation and deodorization often suffer from limitations including inadequate efficacy and safety issues. Here, we propose a triple-synergistic strategy that integrates host-guest complexation by β-cyclodextrin, physical adsorption enabled by a highly porous nanofiber film, and the bioactivity of grape seed extract. The multifunctional nanofiber film, composed of polyvinyl alcohol, β-cyclodextrin, and grape seed extract, was prepared via electrospinning. At a 10 % grape seed extract loading level, the triple-synergistic strategy endowed the film with the remarkable adsorption rate (74.08 %), along with excellent antibacterial and antioxidant performance. This multi-mechanism action effectively extended the shelf life of crayfish meat ( Procambarus clarkii ) and minimized the production of undesirable odors during storage. Ultimately, the underlying mechanism of these excellent performances was expected to inspire the design of other aquatic products packaging films.
