Determination of procymidone residues in rapeseed oil based on olfactory visualization technology

Hui Jiang,Mingxing Zhao,Quansheng Chen

A new means about olfactory visualization technique for the quantitative analysis of procymidone residues in rapeseed oil has been proposed. First, an olfactory visualization system was set up to collect volatile odor information from rapeseed oil samples containing different concentrations of procymidone residues. Then, we utilized four intelligent optimization algorithms, namely particle swarm optimization (PSO), genetic algorithm (GA), ant colony optimization (ACO) and simulated annealing (SA), to optimize the characteristics of the sensors. Finally, support vector machine regression (SVR) models employing optimized features were constructed for the quantitative detection of procymidone residues in rapeseed oil. The study demonstrated that the SA-SVR model demonstrated superior prediction results, achieving a high determination coefficient of prediction ( R P 2 RP2 ) at 0.9894. As indicated by the results, it is possible to successfully conduct non-destructive detection of procymidone residues in edible oil by the olfactory visualization technology.

Identification of procymidone in rapeseed oils based on olfactory visualization technology

Mingxing Zhao,Hui Jiang,Quansheng Chen

A new method for qualitative identification of pesticide residues in edible oil based on olfactory visualization technology is proposed. Firstly, according to the pre-experimental experience, 12 suitable chemical dyes were selected to prepare sensor arrays to form an olfactory visualization system. The system was used to gather volatile odor information in rapeseed oil samples that had different levels of procymidone residues. Subsequently, the olfactory information was translated into a visual representation. Then, the visual data, i.e. color features, were preprocessed and underwent principal component analysis to extract and reduce the dimensionality of sensor image information, resulting in the optimal number of principal components (PCs). Finally, three different models based on the sensor image features were constructed for analysis. These models included the support vector machine (SVM), the random forest (RF) and the kernel based extreme learning machine (KELM). In process of model correction, the parameters of the three models were optimized by dung beetle optimizer (DBO) algorithm, resulting in the identification of the optimal parameters. The results show that the cumulative variance contribution increases slowly when the number of PCs starts from 6, and the three models (i.e. DBO-SVM, DBO-RF, and DBO-KELM) have the best prediction results when PCs = 6. Compared to the DBO-RF and DBO-KELM models, the training and prediction performance of the SVM model based on the DBO optimization algorithm is more stable and has the higher recognition accuracy of the samples, reaching 100%. The results show that it is feasible to use olfactory visualization technology to achieve high-precision qualitative identification of edible oil pesticide residues.

A molecule-imprinted electrochemiluminescence sensor based on self-accelerated Ru(bpy)32+@ZIF-7 for ultra-sensitive detection of procymidone

Xin Zhang,Li Tian,Zhuo Sun,Qian Wu,Xiangyu Shan,Yingjie Zhao,Ruizhan Chen,Juan Lu

A molecular-imprinted electrochemiluminescence sensor for procymidone (PCM) detection was reported based on Ru(bpy) 3 2+ @ZIF-7. A novel self-accelerated Ru(bpy) 3 2+ @ZIF-7 was prepared by one-step synthesis method, which exhibited high electrochemiluminescence (ECL) emission as a coreactant in the presence of tri- n -propylamine (TPrA). Concretely, ZIF-7 was not only the carrier of Ru(bpy) 3 2+ , but also the co-reaction promoter of Ru(bpy) 3 2+ /TPrA ECL system. The introduction of molecular imprint polymer (MIP) with PCM gave new characteristics of specific recognition of analyte PCM. The change value of ECL intensity (ΔI) was proportional to the logarithm of PCM concentration, with a wide linear range and a low detection limit of 1.0 × 10 −10 to 1.0 × 10 −6  mol L −1 and 2.0 × 10 −11  mol L −1 , respectively. The presented MIP-ECL sensor had high sensitivity, selectivity and stability, and had great potential in the field of food safety detection.

A dual-mode sensor for rapid detection of procymidone: “Dark box” qualitative analysis and electrochemical quantification mediated by PdHPCN-222/PEI-rGO and CRISPR/Cas12a

Ruonan Wang,Baoshan He,Zhengyong Liang,Yao Liu,Jun Yang,Huali Jin,Min Wei,Wenjie Ren,Zhiguang Suo,Yiwei Xu

This study presents an integrated dual-mode sensing strategy, in which qualitative fluorescence screening is followed by quantitative electrochemical detection, improving detection efficiency and enabling rapid PCM analysis. It develops a novel fluorescence electrochemical aptasensor that combines in vitro “Dark-box” applications with CRISPR/Cas12a system electrode surface sensing technology. PCM activates the DNA walker, and the DNAzyme induces cyclic cleavage of DNA strands bearing the Carboxyfluorescein (FMA) group. After magnetic separation, the fluorescence reaction combined with the “Dark box” enables the preliminary qualitative analysis of procymidone (PCM). Following the preliminary qualitative detection, the solution is introduced to the electrochemical aptasensor platform integrated with the CRISPR/Cas12 system. The Cas12a system triggers cyclic cleavage, producing a signal change that enables the electrochemical quantitative detection of PCM. An fluorescence (FL) response occurs when the PCM concentration in spiked samples is at or above 1 pg·mL −1 , allowing for qualitative fluorescence detection. The EC platform's detection limit is 8.51 × 10 −6  ng·mL −1 , with a range of 1 × 10 −2  ng·mL −1 to 1 × 10 4  ng·mL −1 . The designed dual-mode sensor provides reliable monitoring of PCM in real samples.

Two Methods for Detecting PCM Residues in Vegetables Based on Paper-Based Sensors

Jiazheng Sun,Shiling Li,Xijun Shao,Mingxuan Fang,Heng Zhang,Zhiheng Zhu,Xia Sun

Procymidone (PCM) is an effective, low-toxicity fungicide commonly used to control plant diseases in grains, vegetables, and fruits. Its usage has significantly increased in recent years, resulting in higher residues in vegetables. This study developed a sensitive and rapid immunoassay method utilizing a gold- and fluorescence-labeled monoclonal antibody (mAb) for detecting PCM residues in vegetable samples. Under optimal conditions, the fluorescent microsphere-labeled monoclonal antibody immunochromatographic strips achieved a limit of detection (LOD) of 1.67 ng/mL, with a visual LOD of 50 ng/mL. Intra-batch accuracy ranged from 94.98% to 103.82%, with a coefficient of variation (CV) of 1.97% to 8.26%. Inter-batch accuracy ranged from 96.16% to 102.51%, with a CV of 4.62% to 8.91%. The visual detection range of the gold nanoparticle-labeled monoclonal antibody immunochromatographic strips was 50 to 200 ng/g. The method demonstrated excellent performance in actual vegetable samples, confirming its applicability across various matrices. This dual-method approach enables rapid screening of negative samples with gold test strips, followed by accurate quantitative analysis of positive samples using fluorescent test strips, thereby enhancing efficiency and addressing diverse detection needs. Consequently, this method holds significant market potential for practical applications.

Terahertz metamaterial sensor-based detection of procymidone and chlorpyrifos residues in Ligusticum chuanxiong

Yuting Shao,Xinyu Zhu,Bingwei Liu,Yan Peng,Yiming Zhu,Wenchao Tang,Yongni Shao

Pesticide residues in Traditional Chinese Medicine (TCM) have raised significant concerns due to their potential health risks. Conventional pesticide residues detection methods are often limited by prolonged detection times and complex sample pretreatment procedures. Therefore, developing a rapid and highly sensitive method for pesticide residue detection is essential. In this paper, we present a terahertz (THz) metamaterial sensor for rapid detection of trace procymidone and chlorpyrifos residues in Ligusticum chuanxiong . The sensor exhibits a theoretical sensitivity of 360 GHz/RIU and offers a minimum detectable level of 1 μg/L, while the entire workflow can be completed within 30 min. Based on the THz metamaterial sensor, quantitative concentration-frequency shift models for chlorpyrifos and procymidone in organic Ligusticum chuanxiong were established (fitting coefficient R 2  > 0.98). These models, combined with the proposed Leave-One-Out Cross-Validation (LOO-CV) based calibration method, were applied to the detection of pesticide residues in commercial Ligusticum chuanxiong . The final average prediction error was approximately 3.8%, providing a novel and rapid testing solution for the quality control of TCM.

Effects of maternal exposure to procymidone on hepatic metabolism in the offspring of mice

Xiaofang Wang,Lingyu Hu,Cuiyuan Jin,Mingrong Qian,Yuanxiang Jin

As an effective fungicide widely used in agricultural production, the excessive procymidone (PRO) residue has been detected in the environment and food. Our previous study demonstrated that PRO could destroy the intestinal barrier in mice and has a joint toxic effect. To explore the cross-generational impact of maternal exposure, 10-week-old C57BL/6 female mice were orally administrated to 10 and 100 mg/kg body weight/day of PRO during pregnancy and lactation. The offspring obtained nutrients from the maternal through the placenta and breast milk, and PRO residues were detected in the liver, intestine, and feces of F1 generation. Fecal examination found that the residual PRO had been completely metabolized when the offspring mice grew to 35 days. The drug residue of F1 generation male mice was higher than that of female mice. We attributed this result to the difference in cytochrome P450 (CYP450) enzyme expression between male and female mice. The transcriptional levels of CYP1A1, CYP1A2, CYP2D9, and CYP3A4, and CYP450 protein expression levels, were higher in female mice. Furthermore, targeted MS of plasma revealed abnormal amino acid levels. In addition, PRO-induced hepatic metabolite changes in F0 and F1-7w mice. KEGG pathway analysis further showed that PRO jointly changed the amino acid biosynthesis pathway of the maternal and offspring. In summary, these results indicated that maternal exposure to PRO during a special period would interfere with self metabolism, and offspring will also have metabolic disorders.

Prussian blue nanoparticles-based signal amplification in aptamer-based lateral flow assay for procymidone detection

Haowei Dong,Jingcheng Huang,Zhen Guo,Haifang Wang,Lingjun Geng,Ziping Cai,Ibrahim A. Darwish,Yemin Guo,Xia Sun,Yanfang Wu

Procymidone is frequently detected in vegetables, prompting stringent regulatory limits in many countries and regions. Despite this, the development of rapid and sensitive detection methods remains limited. In this study, an aptamer-based lateral flow assay (LFA) was developed for procymidone detection, utilizing Prussian blue nanoparticles (PBNPs) as amplified signal markers, chosen for their intrinsic color and enzyme-like properties. An aptamer was coupled with the PBNPs to form a signal probe, enabling the construction of a competitive LFA. Under optimized conditions, the LFA achieved a limit of detection (LOD) of 0.35 ng/mL and a visual limit of detection (vLOD) of 0.64 ng/mL. The enzyme-mimetic activity of PBNPs was harnessed and such a catalytic reaction strategy further improved the assay sensitivity, leading to a further improvement of the LOD by 14.3-fold. The assay demonstrated high sensitivity, selectivity, and reproducibility, performing effectively in typical vegetable samples. This study provides a rapid, sensitive and reliable tool for procymidone detection, offering valuable insights and a new strategy for monitoring pesticides.

Combination of supercritical fluid elution and resin adsorption for removal of procymidone from ginseng extracts

Li Guangtao,Sun Liwei,Tang Shaokun

We propose a new method of resin adsorption (RA) coupled with supercritical fluid elution (SFE) for removal of pesticide residue and recovery of ginsenosides from ginseng extracts. D-101-1 resin was selected as the proper adsorption resin, acetone- n -hexane (4 : 6, v : v) served as the modifier with the flow rate of 1.5 mL/min during supercritical CO 2 elution of procymidone at 25 MPa, 55°C for 2 h, and absolute ethanol as the modifier at a flow rate of 1 mL/min for supercritical CO 2 elution of ginsenosides at 20 MPa, 60°C and 1 h. The results showed that the content of procymidone in the final products was only 0.0089 mg/kg. Meanwhile, the recovery rate of ginsenosides reached up to 92.5%. RA-SFE procedure provides an efficient approach to remove pesticide residue traces with little loss of active ingredients. The used resin can be recycled without any additional regeneration.

Paper-based biosensors relying on core biological immune scaffolds for the detection of procymidone in vegetables

Gaozhen Li,Jiashuai Sun,Jiahuan Li,Yaoli Zhang,Jingcheng Huang,Fengling Yue,Haowei Dong,Falan Li,Huihui Xu,Yanyin Guo,Yemin Guo,Xia Sun

In order to achieve a highly sensitive detection of procymidone in vegetables, three paper-based biosensors based on a core biological immune scaffold (CBIS) were developed, which were time-resolved fluorescence immunochromatography strips with Europium (III) oxide (Eu-TRFICS). Goat anti-mouse IgG and europium oxide time-resolved fluorescent microspheres formed secondary fluorescent probes. CBIS was formed by secondary fluorescent probes and procymidone monoclonal antibody (PCM-Ab). The first type of Eu-TRFICS (Eu-TRFICS-(1)) fixed secondary fluorescent probes on a conjugate pad, and PCM-Ab was mixed with a sample solution. The second type of Eu-TRFICS (Eu-TRFICS-(2)) fixed CBIS on the conjugate pad. The third type of Eu-TRFICS (Eu-TRFICS-(3)) was directly mixed CBIS with the sample solution. They solved the problems of steric hindrance of antibody labeling, insufficient exposure of antigen recognition region and easy loss of activity in traditional methods. They realized multi-dimensional labeling and directional coupling. They replaced the loss of antibody activity. And the three types of Eu-TRFICS were compared, among which Eu-TRFICS-(1) was the best detection choice. Antibody usage was reduced by 25% and sensitivity was increased by 3 times. Its detection range was 1–800 ng/mL, the limit of detection (LOD) was 0.12 ng/mL with the visible LOD (vLOD) of 5 ng/mL.

Maternal procymidone exposure has lasting effects on murine gut-liver axis and glucolipid metabolism in offspring

Xiaofang Wang,You Weng,Shinan Geng,Caiyun Wang,Cuiyuan Jin,Liyun Shi,Yuanxiang Jin

There is increasing evidence that maternal exposure to environmental pollutants can cause intestinal and metabolic diseases , and these disease risks still exist in offspring . Here, female C57BL/6 mice were orally treated with procymidone (PRO) (10 and 100 mg/kg body weight/day) by dietary supplementation during the gestation and lactation periods. Then, we discovered PRO changed the physiology, intestinal barrier and metabolism both in the generations of F 0 and different developmental stages of F 1 (7 weeks and 30 weeks old, respectively). Maternal PRO exposure affected the growth phenotypes and the glucolipid metabolism related indicators and genes of mice, especially the male mice of F 1 generations. The changes in bile acids (BAs) metabolism demonstrated that PRO disordered glucolipid metabolism through enterohepatic circulation. Furthermore, PRO reduced mucus secretion in the gut and altered the composition of gut microbiota , leading more bacteria to disseminate in the gut and inflammatory responses both in F 0 and F 1 regenerations. And PRO-induced gut microbiota dysbiosis was tightly related to BAs metabolites. Together, the results indicated that PRO destructed the functional integrity of intestinal barrier and the inflammatory reaction was triggered. And then, the disorder of glucolipid metabolism was induced through the BAs enterohepatic circulation. This study indicated that the cross-generation effects of PRO could not be ignored.

Determination of residual levels of procymidone in rapeseed oil using near-infrared spectroscopy combined with multivariate analysis

Mingxing Zhao,Hui Jiang,Quansheng Chen

The issue of pesticide residues has always been a hot topic at home and abroad. A method for the quantitative detection of procymidone residues in grain and oil products using near-infrared (NIR) spectroscopy has been proposed. First, a NIR spectrometer was used to collect spectral data from rapeseed oil samples with different concentrations of procymidone residues. Based on full-spectrum data, the wavelength points selected by bootstrapping soft shrinkage (BOSS) algorithm, competitive adaptive reweighted sampling (CARS) algorithm, and variable combination population analysis (VCPA) algorithm then were compared and were quantified using support vector regression (SVR) model. Simultaneously, the prediction results of the SVR model optimized by dung beetle optimizer (DBO) algorithm and pigeon-inspired optimization (PIO) algorithm were compared using the full-spectrum data. Finally, the wavelength selection algorithms and parameter optimization algorithms with the best prediction results were selected for comparison and combination. In light of the outcomes, the three spectral characteristic wavelength selection algorithms and the two optimization algorithms can improve the coefficient of determination ( R P 2 ) and reduce the root mean square error of prediction (RMSEP). The SVR model that utilizing CARS and PIO algorithm demonstrates the best generalization performance among all models evaluated, and the R P 2 is 0.9939 with a RMSEP of 2.3435 mg·kg −1 . The results indicate that the high-precision and rapid detection of procymidone in edible oil can be achieved using the SVR model optimized by input feature and parameter based on NIR spectral data. This has great significance in ensuring the safety of grain and oil food.
