The extensive application of cyhalofop-butyl, an aryloxyphenoxypropionate (AOPP) herbicide, for weed control in rice fields inevitably results in environmental pollution and ecological toxicity. Microbial degradation of cyhalofop-butyl is garnering immense research attention. This study revealed that Sphingobium yanoikuyae B1 can efficiently degrade the cyhalofop-butyl, completely breaking down 30 mg/L within 12 h via ester bond hydrolysis, forming cyhalofop acid. A novel hydrolase gene cbeH (37.39% identity) was identified in B1. CbeH exhibited catalytic activity by hydrolyzing the ester bond (C-O) in cyhalofop-butyl and other AOPP herbicides, yielding AOPP acids. Additionally, it catalyzed the cleavage of the C-N bond in amide herbicides propanil and diuron, as well as the C-C bond in the aromatic compound 2-hydroxy-6-oxo-6-phenylhexa-2,4-dienoic acid. Sequence alignments and site-directed mutagenesis indicated the catalytic triad Ser-His-Asp in CbeH serves as active center cleaving C-O, C-C, and C-N bonds. Comparative analysis revealed the gene cbeH , highly conserved within Sphingomonads , was located within multiple insertion sequences on the plasmid of Rhodococcus strains. CbeH was widespread in Proteobacteria and Actinobacteria, with its host strains prevalent in aromatic-polluted environments. This study provides novel enzyme resources for remediating combined pollution and enhances our understanding of the genetic organization and environmental distribution of trifunctional pollutant-degrading genes.

Municipal wastewater treatment plants (MWTP) are the main gathering places for antibiotic resistance genes (ARGs) and pathogenic bacteria. In this study, plasma oxidation was applied to remove the ARGs and inhibit pathogenicity in the effluent. The experimental results demonstrated that approximately 1.32 log, 1.34 log, 2.39 log, and 2.80 log of aac (3)-II , bla TEM-1 , Tet W, and Tet C were removed within 25 min of the plasma treatment, respectively. OH, 1 O 2 , O 3 , H 2 O 2 , NO 2 − , and NO 3 − all contributed to ARG removal. Coexisting dissolved organic matter was also oxidized, which was positively related to ARG removal. Plasma oxidation led to a decrease in the diversity and richness of microbial communities; Parcubacteria and Proteobacteria were the main potential hosts of ARGs and mobile genetic elements. Integron gene IntI 1 decreased by 2.46 log after plasma treatment, which was positively related to ARG removal. Gene expression related to ATP binding, DNA repair, gene transfer, and the cell membrane was reduced after plasma treatment, resulting in effective ARG removal. Furthermore, pathogenic genes related to lumpy skin disease, Niemann-Pick disease, polycystic kidney and hepatic disease were all down-regulated by plasma treatment. Overall, the plasma treatment inhibited the transmission risks of ARGs and pathogenicity in the effluent of MWTP.

Lentil hulls, a by-product of lentil processing, are rich in polyphenols and dietary fiber. In this study, a mouse model of nonalcoholic fatty liver disease (NAFLD) induced by dextran sulfate sodium (DSS) and high-fat diet (HFD) was used to investigate the intervention effect of green lentil hulls (GLH) from the perspective of intestinal and hepatic metabolism. Results suggest that DSS-induced gut inflammation and barrier disruption aggravate NAFLD; GLH intervention lowered body weight, liver weight gain and lipid deposition. It also inhibited liver fat accumulation and inflammation as well as intestinal inflammation. In addition, it enhanced liver antioxidant capacity, glucose tolerance and intestinal barrier integrity. Analysis of intestinal flora indicated that GLH advanced the abundance of Akkermansia-muciniphila and Verrucomicrobiaceae , which led to increased content of short-chain fatty acids (SCFAs) and inhibited the Firmicutes and Proteobacteria . Moreover, GLH intervention regulated taurine and hypotaurine metabolism, as well as the biosynthesis of unsaturated and saturated fatty acids in the liver. The relief by GLH on NAFLD may be a comprehensive effect of its rich dietary fiber and polyphenols on intestinal and hepatic metabolism via the gut-liver axis. This research also suggests that the low-value lentil hulls can serve as a valuable food supplement for enhanced gut health and reduced risk of NAFLD.
