Study on chemical characterization and sleep-improvement function of Prunella vulgaris L. based on the functional components

Chang Zhou,Yi-fan Hu,Yan Zhang,Cheng-hao Wang,Xue-jing Liao,Fa-feng Cheng,Yan-yan Jiang

Prunella vulgaris L. (P. vulgaris) has great application value and development prospects in improving sleep. In this study, we continued to evaluate the sleep-improvement function and mechanism of P. vulgaris from both chemical characterization and function based on sleep-improvement functional ingredients, rosmarinic acid and salviaflaside, screened out in the previous stage as the index components. The chemical constituents of P. vulgaris and its phenolic acid fraction were characterized by the UPLC-MS n technology. The quality of the sleep-improvement phenolic acid fraction of P. vulgaris was scientifically evaluated by fingerprints combined with quantitative analysis of rosmarinic acid and salviaflaside. The function of phenolic acid parts of P. vulgaris in improving sleep was verified by different insomnia models including the PCPA-induced insomnia model and surface platform sleep deprivation model. HE staining was used to observe the effect of P. vulgaris on the morphology of nerve cells in different brain regions. In vivo experiments and molecular docking explored the sedative-hypnotic effects of functional ingredients of P. vulgaris . All these results investigated the material basis and mechanism of P. vulgaris to improve sleep from multiple perspectives, which contribute to providing a basis for the development of functional food to improve sleep.

Vulgarisin-type diterpenoids from self-heal (Prunella vulgaris) and their neuroprotective effects against ischemia/reperfusion (I/R) via a mitochondria-related pathway

Fengwei Ma,Qingfang Deng,Huayong Lou,Jinyu Li,Su Xu,Wenneng Wu,Qihua Wen,Lei Tang,Xin Wang,Weidong Pan

Self-heal (Prunella vulgaris L.) is a perennial edible plant that is widely distributed across the world and is traditionally consumed as a food additive in soft drink beverages. In this study, to explore the functional components of P. vulgaris, an investigation of its ethanol extracts has been conducted by our group. As a result, twelve (1–12) vulgarisin-type diterpenoids with a special 5/6/4/5-fused ring skeleton, including six new ones (1–6), were obtained. Their structures including the absolute configuration were elucidated based on comprehensive spectroscopic evidence, ECD calculations, as well as single-crystal X-ray diffraction analyses. All the isolates were tested for neuroprotective effects against ischemia/reperfusion (I/R) on primary neuron cells through the oxygen and glucose deprivation and reperfusion (OGD/R) induced injury model. The results showed that all twelve vulgarisin-type diterpenoids possess promising neuroprotective activity at a concentration of 10 μM. Among them, compound 3 can significantly suppress cell apoptosis by regulating Bax/Bcl-2 protein expression and inhibiting cleaved caspase-3 and caspase-9 expression with a western blotting assay. Further research revealed that compound 3 could improve mitochondrial function by inhibiting mitochondrial cytochrome c release, reducing ROS levels, and maintaining the membrane potential. This work firstly reports vulgarisin-type diterpenoids possessing neuroprotective activity. These findings also suggest that daily consumption of P. vulgaris might prevent cerebral disorders via a mitochondria-related pathway.

Characterization, antioxidant and immunomodulatory activities of polysaccharides from Prunella vulgaris Linn

Chao Li,Qiang Huang,Xiong Fu,Xiu-Jie Yue,Rui Hai Liu,Li-Jun You

Water-soluble polysaccharides from Prunella vulgaris Linn ( P. vulgaris ) were fractionated using DEAE-Sepharose fast-flow column to obtain several eluents of water (PV-P1), 0.1   M NaCl (PV-P2) and 0.2   M NaCl (PV-P3). Structural analyses showed that PV-P1 had a higher molecular weight and degree of branching as compared to PV-P2 and PV-P3. Tertiary structure analyses indicated that PV-P1, PV-P2 and PV-P3 did not have triple-helical conformation. PV-P2 and PV-P3 showed stronger antioxidant activities than PV-P1, as measured radical scavenging capacities. PV-P1 showed stronger immunomodulatory activities than PV-P2 and PV-P3 in term of stimulation of the production of pro-inflammatory cytokines, including nitric oxide (NO), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in murine macrophage RAW 264.7 cells. PV-P1, PV-P2 and PV-P3 did not exhibit cytotoxicities against RAW 264.7 at the concentrations tested. These results suggest that P. vulgaris polysaccharides could be explored as potential antioxidant and immunomodulatory agents for the complementary medicine or functional foods.

Prunella vulgaris Compound Improves Sleep Architecture and Attenuates Hippocampal Injury in Insomnia Mice

Kai Wang,Haojia Zhang,Chang Zhou,Zijin Sun,Wei Shao,Kunjing Liu,Wenyuan Ma,Chunyu Wang,Xueqian Wang,Qingguo Wang,Yanyan Jiang,Fafeng Cheng

Insomnia is associated with disrupted sleep architecture, hippocampal pathological alterations, neurotransmitter imbalance, and neuroinflammatory responses. This study evaluated the effects of the Prunella vulgaris compound (PVC) on sleep architecture and hippocampal alterations in p-chlorophenylalanine (PCPA)-induced insomnia model mice and examined their association with NLRP3-related pyroptotic signaling. PVC was characterized using UPLC-Q-Exactive Orbitrap MS and targeted HPLC quantification. PCPA-induced insomnia models were established in ICR and C57BL/6N mice for pharmacodynamic and molecular assessments and EEG/EMG recording, respectively. Sleep outcomes were assessed using the pentobarbital-induced sleep test and EEG/EMG; hippocampal histomorphology, neurotransmitters, and inflammasome- and pyroptosis-related markers were evaluated using histological, biochemical, RT-qPCR, and Western blot analyses. UPLC-MS tentatively characterized 198 constituents, and salviaflaside, rosmarinic acid, and linarin were quantified by HPLC. PVC shortened sleep latency, prolonged sleep duration, reduced wakefulness, increased NREM and REM sleep, and improved sleep continuity. It also attenuated hippocampal histomorphological alterations; modulated 5-HT, GABA, glutamate, and dopamine levels; and reduced NLRP3 inflammasome- and pyroptosis-related markers together with IL-1β and IL-18. PVC improved insomnia-like phenotypes and attenuated hippocampal pathological alterations in PCPA-induced insomnia model mice. These effects were associated with improved sleep architecture and neurotransmitter homeostasis and with reduced NLRP3/caspase-1/GSDMD-related signaling.

Preparation, characterization and in vitro antioxidant activities of a homogeneous polysaccharide from Prunella vulgaris

Jiaqi Huang,Mengyao Wang,Yu Wang,He Sun,Yadan Zou,Xiaojin Tian,Dianxin Cui,Peng Zhang,Xue Li,Wenzhi Yang

Prunella vulgaris is a medicinal and edible homologous plant, commonly used as a folk medicine to treat diseases. The Prunella vulgaris polysaccharides (PVPs) are reported with the antioxidant activity. This work was designed to isolate, characterize, and test the antioxidant activity of purified PVPs from P. vulgaris . A new homogeneous polysaccharide (PVP-1) was prepared by the DEAE column from PVPs, and diverse chromatography/spectroscopy and chemical methods were simultaneously employed to characterize the fine structure of PVP-1. The results showed PVP-1 had a triple helix structure, and the repeating structural unit of PVP-1 was composed of →6)- β -D-Gal p -(1→6)- β -D-Gal p -(3,1→6)- β -D-Gal p -(1→6)- β -D - Gal p -(1→ as the main chain, together with →6)- β -D-Gal p -(1,3→1)- α -D-Ara f -(5→1)- β -D-Gal p -(4→1)- α -D-Gal p -(2→ and →6)- β -D-Gal p -(1,3→1)- α -D-GlcA p -(4→1)- α -D-Glc p -(4→1)- α -D-Gal p as the branch chains. The main monosaccharides of PVP-1 were galactose (Gal, 41.25 %), galactose-OMe (Gal-OMe, 27.73 %), arabinose (Ara, 10.63 %), mannose (Man, 9.86 %), glucose (Glc, 3.88 %), glucuronic acid (GlcA, 2.86 %), ribose (Rib, 1.79 %), and xylose (Xyl, 1.76 %). In addition, the scanning electron microscopy (SEM) displayed that the surface of PVP-1 was rough and porous. PVP-1 gave the scavenging rates of the DPPH, ABTS, and hydroxyl radical lower than vitamin C at the same concentration, with the highest scavenging rate of DPPH radical at 82.71 % ± 4.19 % (5 mg/mL).

Multi-level chemical characterization and anti-inflammatory activity evaluation of the polysaccharides from Prunella vulgaris

He Sun,Jia Lou,Bo-Xue Chen,Jia-Qi Huang,Qi-Long Wang,Shao-Fei Song,Zi-Yue Jia,Rong Miao,Shi-Yu Wang,Xue Li,Wen-Zhi Yang

Prunella vulgaris (PV) is a medicine and food homologous plant, but its quality evaluation seldom relies on the polysaccharides (PVPs). In this work, we established the multi-level fingerprinting and in vitro anti-inflammatory evaluation approaches to characterize and compare the polysaccharides of P. vulgaris collected from the major production regions in China. PVPs prepared from 22 batches of samples gave the content variation of 5.76–24.524 mg/g, but displayed high similarity in the molecular weight distribution. Hydrolyzed oligosaccharides with degrees of polymerization 2–14 were characterized with different numbers of pentose and hexose by HILIC-MS. The tested 22 batches of oligosaccharides exhibited visible differences in peak abundance, which failed to corelate to their production regions. All the PVPs contained Gal, Xyl, and Ara, as the main monosaccharides . Eleven batches among the tested PVPs showed the significant inhibitory effects on NO production on LPS-induced RAW264.7 cells at 10 μg/mL, but the exerted efficacy did not exhibit correlation with the production regions. Conclusively, we, for the first time, investigated the chemical features of PVPs at three levels, and assessed the chemical and anti-inflammatory variations among the different regions of P. vulgaris samples.

Ultrasonic extraction and structural identification of polysaccharides from Prunella vulgaris and its antioxidant and antiproliferative activities

Li Chao,Fu Xiong,Huang Qiang,Luo Faxing,You Lijun

Response surface methodology was used to optimize the ultrasonic extraction of Prunella vulgaris polysaccharides (PVP). When the quality of P. vulgaris was 4.0 g, the optimal extraction conditions were ultrasonic power 210 W, time 50 min, and temperature 70 °C. Using this condition, the experimental yield was agreed closely with the predicted value. Compared with conventional extraction, ultrasonic extraction showed a higher polysaccharide yield. The molecular weights were determined by high-performance gel permeation chromatography. Gas chromatography analysis suggested that PVP comprised of rhamnose, arabinose, xylose, mannose, glucose, and galactose with molar percentages of 2.8, 28.2, 38.5, 11.0, 3.0, and 16.5 %, respectively. The glycosidic linkage analysis showed that PVP contained 1→ and 1→6 glycosidic linkages (48.0 %), 1→2 and 1→4 glycosidic linkages (8.1 %), and 1→3 glycosidic linkages (43.9 %). PVP showed good antioxidant activities by the methods of DPPH radical scavenging assay, oxygen radical absorbance capacity assay, and cellular antioxidant activity assay. The antiproliferative activities of PVP (2.0 mg/mL) on the growth of human hepatoma HepG2, gastric carcinoma SGC 901, and human breast cancer MCF-7 cells were 51.2, 35.2, and 31.8 %, respectively. PVP did not exhibit cytotoxicity against normal liver L02 cells within the tested concentrations. These results demonstrated that polysaccharides isolated from P. vulgaris may be helpful to develop potential antioxidant and antitumor agents for food and pharmaceutical industries.

Arabinoxylan from Prunella vulgaris L. by room temperature extraction and the hepatoprotective effect through gut-liver axis

Chenyu Wang,Guannan Yu,Qian Liu,Lichun Ye,Shilei Wang,Guohua Zheng,Junjie Hu,Lei Xu,Yan Meng,Zhaohua Shi

Recently, low-temperature water isolation has been confirmed to better keep the original structure of polysaccharides in plant. Thus, in this work, we obtained a new polysaccharide (WPVP) by water extraction at room-temperature (RT) from Prunella vulgaris L., a famous traditional edible Chinese medicinal herb. Unlike most reported complex heteropolysaccharides from Prunella vulgaris L. by hot water, the polysaccharide we obtained at RT was identified as an arabinoxylan, which is comprised a β -1,4-D-xylopyranose (Xyl p ) backbone substituted at O-2 with linear α -1,5-L-arabinofuranose (Araƒ) side chains. Notably, this new WPVP significantly ameliorated alcohol-induced hepatic damage in mice through gut regulation, as indicated by the improvement on hepatic lipid accumulation, inflammation, and intestinal microecology. Collectively, this work suggests a new strategy for polysaccharides isolation to maintain the original structure, and also proposes a novel therapeutic strategy for alcoholic liver disease (ALD).

Prunella vulgaris polyphenols mediate the gut-liver axis to improve MASLD: regulating cholesterol metabolism and gut microbiota

Chengfeng Li,Ying Liu,Sitong Ye,Haihua Zhang,Miao Sun,Yingyan Liao,Yan Lin,Sha Tian,Kun Luo,Zhimin Zhang

BackgroundMetabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide. The latest research shows that its pathogenesis is closely related to the imbalance of gut microbiota. Prunella vulgaris L. is an edible-medicinal plant containing bioactive compounds such as polyphenols that can lower cholesterol and protect the liver. However, whether it has anti MASLD effects has not been reported. The present study aimed to investigate the effect of Prunella vulgaris polyphenols (PVP) on alleviating MASLD from the perspective of the gut-liver axis.MethodsPVP composition was characterized via UPLC-MS/MS and HPLC. Enzymatic kinetics, fluorescence quenching, and molecular docking were used to study the inhibition of PVP and rosmarinic acid (RA) on cholesterol esterase (CEase). Effects on liver lipid accumulation and intestinal cholesterol transport were assessed using HepG2 and Caco-2 cell models. A MASLD mouse model was evaluated through ELISA, tissue staining, and 16S rRNA sequencing to determine the efficacy and mechanisms.ResultsPVP and RA exhibited anti-competitive inhibition of CEase, with IC50 values of 1.63 ± 0.06 and 0.39 ± 0.17 mg/mL, respectively. RA showed strong binding to CEase. PVP and RA significantly alleviated hepatic lipid accumulation in HepG2 cells, inhibited intestinal cholesterol absorption, and promoted cholesterol efflux in Caco-2 monolayers. In MASLD mice, PVP significantly reduced serum ALT, TBA, and TG, lowered hepatic TC and fecal TBA (P < 0.05), and ameliorated liver pathological damage. In addition, both PVP and RA modified the composition of gut microbiota in the cecum, which characterized by a reduction in bile acid (BA)-related bacteria such as g_UBA7173, g_Bacteroides_H, f_Burkholderiaceae_A, g_Phocaeicola_A, and g_Turicimonas, while increasing f_Lachnospiraceae and f_Oscillospiraceae.ConclusionPVP ameliorates MASLD by inhibiting CEase and intestinal cholesterol absorption, promoting cholesterol efflux, and regulating TBA levels along with intestinal microbiota homeostasis. Our findings suggest that PVP and RA deserve further investigation as potential modulators of cholesterol metabolism in MASLD.

Prunella vulgaris polyphenols alleviate atherosclerosis by regulating lipid metabolism via suppressing STAT3 and activating the PI3K-Akt pathway

Taoli Sun,Wei Yuan,Xinru Li,BoHou Xia,Jianhua Huang,Meihong Piao,Wen Chen,Qinhui Tuo

Atherosclerosis (AS), a leading cause of cardiovascular diseases, remains a major challenge due to the lack of effective preventive and therapeutic strategies. This study investigated the anti-atherosclerotic effects and underlying mechanisms of Prunella vulgaris polyphenols (PVPs). PVPs were found to significantly reduce lipid deposition in the aorta and enhance plaque stability by increasing fibrous cap thickness and collagen content, while simultaneously reducing MMP-9 and CD68 protein expressions. Moreover, PVPs alleviated hepatic lipid accumulation by upregulating ABCA1 and downregulating SREBP1 protein expressions. The PI3K-Akt pathway was identified as a key mechanism underlying the therapeutic effects of PVPs, with STAT3 predicted as a critical target involved in lipid metabolism. These predictions were further validated experimentally, demonstrating that PVPs effectively inhibited STAT3 activation while enhancing the PI3K-Akt pathway. Taken together, these findings reveal that PVPs exert anti-atherosclerotic effects by modulating lipid metabolism through the inhibition of STAT3 and activation of the PI3K-Akt pathway.

Chemoprevention by Prunella vulgaris L. extract of non-small cell lung cancer via promoting apoptosis and regulating the cell cycle.

Feng L,Jia X,Zhu M,Chen Y,Shi F

Chemoprevention is one feasible approach to decreasing morbidity and mortality of non-small cell lung cancer (NSCLC). The present study aimed to explore the mechanisms of chemoprevention of NSCLC by Prunella vulgaris L. (PV) using a PV extract of 60% ethanol (P-60). In an A/J mouse model benzo[a]pyrene induction of lung tumors was significantly reduced difference by P-60 group. In addition, P-60 was found to have the ability to regulate cell cycle and induce apoptosis in SPC-A-1 cells. Therefore, we propose that P-60 has potential as a lung cancer chemopreventive agent.

Dual-mode fluorometric and colorimetric sensor based on prunella vulgaris derived carbon dots for Co2+ detection in real water and food

Xiukun Qi,Zhuoying Li,Tingting Cui,Fang Zhang,XueYang Wang,Huiyi Yang,Fang Chai

Heavy metals exert adverse impacts on human health, ecosystems, and the environment. Accumulated Co 2+ poses severe threats to human health, consequently, the development of portable and reliable Co 2+ detection methods has become an urgent demand in the fields of environmental monitoring and food safety. Here, Chinese herbal medicine (CHM)-inspired carbon dots (P-CDs) were fabricated using prunella vulgaris as a carbon source and o -phenylenediamine (OPD) as a nitrogen source. The yellow emissive P-CDs enabled dual-mode (colorimetric and fluorometric) detection of Co 2+ , with a limit of detection (LOD) of 1.94 μM and 0.57 μM, respectively. The portable smartphone-based RGB mode strategy was also developed and achieved on-site effective detection with visible and intelligent treatment. The recoveries ranged from 90.3%–105.7% and were attained from the practical test in real water and food samples, demonstrating excellent sensing reliability and environmental tolerance. This study advanced the promising systematic development of functional CHM-derived CDs, and provided an innovative dual-modal strategy for the portable detection of heavy metals. This strategy offers a novel perspective for Co 2+ detection in real environmental and food samples; it is of great significance for investigating CHM-derived carbon active sites from unique insights.
