The antimicrobial efficacy of punicalagin against foodborne pathogens has been extensively studied, yet its impact on spoilage microorganisms remains poorly characterized. This study aimed to evaluate punicalagin's inhibitory effects on Pseudomonas fluorescens and explore its potential application in tilapia preservation. Growth curve analysis showend that punicalagin effetively inhibited the growth of Pseudomonas fluorescens . In addition, punicalagin triggered intracellular potassium ion efflux and leakage of nucleic acids and protein. Scanning electron microscopy revealed compromised cell membrane integrity after punicalagin treatment. Furthermore, punicalagin exposure reduced intracellular ATP levels and elevated reactive oxygen species accumulation. Concurrently, it effectively suppressed biofilm formation and destroyed preformed biofilms. In tilapia spiked with Pseudomonas fluorescens , punicalagin markedly attenuated spoilage progression, as evidenced by diminished protein degradation and lipid oxidation. These collective findings indicate that punicalagin exhibit antimicrobial and anti-biofilm activities against Pseudomonas fluorescens , highlighting its potential as a natural preservative for aquatic products.

Purpose: Pseudomonas syringae pv. actinidiae causes bacterial canker of kiwifruit and is responsible for severe economic losses and emergence of drug-resistant bacteria. Bacteriophages are viruses that infect target bacterial hosts and may be the best strategy to prevent and control kiwifruit canker disease. The objective of this experiment was to monitor the prevalence of Pseudomonas syringae pv. actinidiae and provide insight for the use of phages in biological control.Materials and methods: In this study, 52 strains of Pseudomonas syringae pv. actinidiae were isolated from 68 stem samples of kiwi plant (cv. Hongyang & Jinkui). Following polymerase chain reaction (PCR) analysis, 15 isolates belonging to biovar 3 were identified, one of which was named XWY0007 and used as the target strain to isolate the phages. Thirty-six phages were isolated and purified from a total of 51 surface water samples collected in Shanghai. All phages were identified by transmission electron microscopy (TEM) and their host ranges were evaluated. Three phages, designated φXWY0013, φXWY0014 and φXWY0026 were selected and further characterised using one-step growth curve and stability at different temperatures and pH.Results and conclusions: The isolated phages are promising for use as antimicrobials against bacterial canker in kiwi. This report is regarding Pseudomonas syringae pv. actinidiae and its phages from major areas of kiwifruit cultivation.

Pseudomonas lundensisis the main bacterium responsible for meat spoilage and its control is of great significance. 3-Carene, a natural monoterpene, has been proved to possess antimicrobial activities. This study aimed to investigate the antibacterial activity and mechanism of 3-carene against the meat spoilage bacteriumP. lundensis, and explore its application on pork. After 3-carene treatment, cellular structural changes were observed. Cell walls and membranes were destroyed, resulting in the leakage of alkaline phosphatase and cellular contents. The decreased activity of Ca2+-Mg2+-ATPase and Na+-K+-ATPase showed the imbalance of intracellular ions. Subsequently, adenosine triphosphate (ATP) content and oxidative respiratory metabolism characteristics indicated that 3-carene inhibited the metabolism of the tricarboxylic acid cycle inP. lundensis. The results of binding 3-carene with the vital proteins (MurA, OmpW, and AtpD) related to the formation of the cell wall, the composition of the cell membrane, and the synthesis of ATP further suggested that 3-carene possibly affected the normal function of those proteins. In addition, the growth ofP. lundensisand increase in pH were inhibited in pork during the 5 days of cold storage after the samples were pre-treated with 3-carene. These results show the anti-P. lundensisactivity and mechanism of 3-carene, and its potential use in meat preservation under refrigerated conditions. Keywords:Pseudomonas lundensis;3-carene;preservatives;membrane damage

Diazotrophs can produce bioavailable nitrogen from inert N2 gas by bioelectrochemical nitrogen fixation (e-BNF), which is emerging as an energy-saving and highly selective strategy for agriculture and industry. However, current e-BNF technology is impeded by requirements for NH4+ assimilation inhibitors to facilitate intracellular ammonia secretion and precious metal catalysts to generate H2 as the energy-carrying intermediate. Here, we initially demonstrate inhibitor- and catalystless extracellular NH4+ production by the diazotroph Pseudomonas stutzeri A1501 using an electrode as the sole electron donor. Multiple lines of evidence revealed that P. stutzeri produced 2.32 ± 0.25 mg/liter extracellular NH4+ at a poised potential of −0.3 V (versus standard hydrogen electrode [SHE]) without the addition of inhibitors or expensive catalysts. The electron uptake mechanism was attributed to the endogenous electron shuttle phenazine-1-carboxylic acid, which was excreted by P. stutzeri and mediated electron transfer from electrodes into cells to directly drive N2 fixation. The faradaic efficiency was 20% ± 3%, which was 2 to 4 times that of previous e-BNF attempts using the H2-mediated pathway. This study reports a diazotroph capable of producing secretable NH4+ via extracellular electron uptake, which has important implications for optimizing the performance of e-BNF systems and exploring the novel nitrogen-fixing mode of syntrophic microbial communities in the natural environment.IMPORTANCE Ammonia greatly affects global ecology, agriculture, and the food industry. Diazotrophs with an enhanced capacity of extracellular NH4+ excretion have been proven to be more beneficial to the growth of microalgae and plants, whereas most previously reported diazotrophs produce intracellular organic nitrogen in the absence of chemical suppression and genetic manipulation. Here, we demonstrate that Pseudomonas stutzeri A1501 is capable of extracellular NH4+ production without chemical suppression or genetic manipulation when the extracellular electrode is used as the sole electron donor. We also reveal the electron uptake pathway from the extracellular electron-donating partner to P. stutzeri A1501 via redox electron shuttle phenazines. Since both P. stutzeri A1501 and potential electron-donating partners (such as electroactive microbes and natural semiconductor minerals) are abundant in diverse soils and sediments, P. stutzeri A1501 has broader implications on the improvement of nitrogen fertilization in the natural environment.

Bisphenol A (BPA) poses multiple risks to human health, and it has become a critical environmental pollution issue. The advantage of bacterial degradation of BPA has become more prominent, and enhancement methods have become increasingly diverse. Using quorum sensing to regulate functional bacteria and genes to enhance the bacterial degradation efficiency of BPA serves as a highly promising new approach. This study investigated the effect of adding exogenous N-hexanoyl-L-homoserine lactone (C6-HSL) on the degradation of BPA by Pseudomonas asiatica P1 (strain P1) and analyzed the regulatory mechanism of the signaling molecules. The results showed that exogenous homoserine lactone signaling molecules (AHLs) could enhance the BPA degradation efficiency of strain P1, with 10 μM C6-HSL demonstrating the most significant enhancement, increasing the degradation rate by 12.4 % compared with the control group and producing products such as benzoic acid and succinic acid. In terms of the regulatory mechanism, C6-HSL preferentially induced a 5.18-fold upregulation in the expression of the AHL receptor-encoding gene LasR. Subsequently, LasR regulates the upregulation of hub genes such as bvgS and aruS , ultimately achieving an upregulation of BPA degradation genes through hierarchical regulation. This led to an enhancement in the initial degradation step of BPA and the activation of the benzoic acid degradation pathway. This study analyzed the potential of C6-HSL to enhance microbial BPA degradation and, for the first time, clarified the mechanism by which C6-HSL enhanced bacterial BPA degradation efficiency through hierarchical regulation.

This work investigated the effect of Desulfovibrio caledoniensis (D. caledoniensis) and Pseudomonas aeruginosa (P. aeruginosa) on the microbiologically influenced corrosion (MIC) behaviour of 70Cu-30Ni alloy using surface analysis and electrochemical techniques. The results demonstrated that the mixed medium containing D. caledoniensis and P. aeruginosa further accelerated the MIC of 70Cu-30Ni alloy compared to the single species medium. The addition of exogenous pyocyanin (PYO) to the D. caledoniensis medium increased the maximum pit depth on 70Cu-30Ni alloy from 5.40 μm to 6.59 μm, and the corrosion current density (icorr) increased by one order of magnitude. From the perspective of bioenergetics and extracellular electron transfer (EET), the comprehensive MIC mechanism of 70Cu-30Ni alloy induced by D. caledoniensis and P. aeruginosa was proposed.

BACKGROUND Trans -2, 3-dihydro-3-hydroxyanthranilic acid (DHHA) is a valuable metabolic intermediate for the biosynthesis of wide-ranging benzoic acid derivatives with enormous biological or pharmaceutical activities. Pseudomonas chlororaphis GP72 is a non-pathogenic biocontrol strain that displays unique capability to produce phenazines. Nevertheless, DHHA production is quite low by the wild type strains, which necessitates yield improvement by constructing engineered strains for large-scale biotechnological applications. RESULTS In this study, two negative regulatory genes namely rsmE and lon were successively deactivated in the DA4 strain. The resulting engineered DA6 strain produced a significantly high titer of DHHA (20.6% higher than that of DA4). The influence of varying pH from 6.2 to 8.2 on DHHA production was studied in a 6-L benchtop fermenter. A controlled broth pH of 7.2 stimulated maximum DHHA production by the engineered DA6 strain. A DO-stat based fed-batch system maintaining a constant DO level (about 20%) accompanied by a glycerol feeding strategy was applied. Highest DHHA production using this approach was recorded to be 10.06 g L -1 , which was 31% higher than with traditional batch cultivation. CONCLUSION The production of DHHA in metabolically engineered GP72 was considerably improved by inactivating two negative regulatory genes in a fed-batch mode. It is concluded that fed-batch fermentation with intermittent glycerol feeding and pH-control was an effective approach to improve DHHA production yield. © 2017 Society of Chemical Industry

An important environmental issue is the pollution caused by poly(ethylene terephthalate) (PET) and the emitted monomers terephthalic acid (TPA) and ethylene glycol (EG). The TPA biodegradation pathway has been well documented for the microbial biodegradation and assimilation of PET; however, the PET biodegradation process by marine microorganisms via the EG biodegradation pathway is less understood. Here, we demonstrate how the marine bacteria Pseudomonas chengduensis BC1815’s membrane-anchored PET esterase (OF113_10420) breaks down PET. We also explain the characteristics of this enzyme and demonstrate that the strain assimilates PET via the EG biodegradation pathway. In addition to depolymerizing PET, PET esterase (OF113_10420) hydrolyzes BHET and MHET to generate EG more efficiently in alkaline circumstances. It has been discovered that P. chengduensis BC1815 cells exhibiting surface-displayed PET esterase (OF113_10420) have the potential to function as cell biocatalysts for the breakdown of PET. Additionally, we look into the microbial communities that have PET esterase linked to the EG biodegradation pathway, primarily those belonging to the phyla Proteobacteria and Actinobacteriota. Consequently, research on the microbial absorption and biodegradation of PET via the EG metabolism route increases our knowledge of the bioremediation of PET pollution in the marine environment and contributes to our understanding of the environmental destiny of PET pollutants in the marine environment.

A Pseudomonas sp. strain WL2 that is able to efficiently metabolize ethyl mercaptan (EM) into diethyl disulfide (DEDS) through enzymatic oxidation was isolated from the activated sludge of a pharmaceutical wastewater plant. One hundred percent removal of 113.5 mg L −1 EM and 110.3 mg L −1 DEDS were obtained within 14 and 32 h, respectively. A putative EM degradation pathway that involved the catabolism via DEDS was proposed, which indicated DEDS were further mineralized into carbon dioxide (CO 2 ), bacterial cells, and sulfate (SO 4
2− ) through the transformation of element sulfur and ethyl aldehyde. Degradation kinetics for EM and DEDS with different initial concentrations by strain WL2 were evaluated using Haldane-Andrews model with maximum specific degradation rates of 3.13 and 1.33 g g −1 h −1 , respectively, and maximum degradation rate constants of 0.522 and 0.175 h −1 using pseudo-first-order kinetic model were obtained. Results obtained that aerobic degradation of EM by strain WL2 was more efficient than those from previous studies. Substrate range studies of strain WL2 demonstrated its ability to degrade several mercaptans, disulfides, aldehydes, and methanol. All the results obtained highlight the potential of strain WL2 for the use in the biodegradation of volatile organic sulfur compounds (VOSCs).

Four uncharacterized ω-transaminases (ωTAs) from Pseudomonas putida NBRC 14164 have been identified and cloned from the pool of fully sequenced genomes. The genes were functionally expressed in Escherichia coli BL21, and the enzymes were purified and characterized. Four TAs showed highly ( S )-selective ωTA activity and converted ( S )-α-methylbenzylamine and pyruvate to acetophenone and l -Ala. The maximum activity of cloned enzymes was in the pH range of 8.0– 8.5 (Pp36420), 8.5 –9. 5 (Pp21050), 9 .0– 9.5 (PpspuC), and 9.5 –1 0 . 5 (PpbauA), and the optimal temperatures were at 35 °C (Pp36420, Pp21050, and PpspuC) and 50 °C (PpbauA), respectively, with K
M of 161.3 mM (Pp21050), 136.7 mM (PpbauA), 398.5 mM (Pp36420), and 130.9 mM (PpspuC) and yielding a catalytic efficiency k
cat / K
M of 0.015, 0.003, 0.012, and 0.023 mM −1 s −1 . Several racemic amines and amino alcohols were resolved by the cloned ωTAs; perfect conversions (48–50 %) were obtained by at least one enzyme, and the residual substrates were left with 97–99 % ee. Kinetic resolution of racemic phenylglycinol was done with PpspuC in a 100-mL scale. Enaniomeric excess of ( S )-phenylglycinol reached 99 % with 45 % isolated yield. The high enantioselectivity and large substrate spectra of the cloned PpTAs showed an attractive potency for biotechnology application in production of chiral amines and amino alcohols.

Soil contamination by PAH and heavy metals is a growing problem. Here, we showed that a new isolate, Pseudomonas brassicacearum strain LZ-4, can simultaneously degrade 98% of 6 mM naphthalene and reduce 92.4% of 500 μM hexavalent chromium [Cr (VI)] within 68 h. A draft genome sequence of strain LZ-4 (6,219,082 bp) revealed all the genes in the naphthalene catabolic pathway and some known Cr (VI) reductases. Interestingly, genes encoding naphthalene pathway components were upregulated in the presence of Cr (VI), and Cr (VI) reduction was elevated in the presence of naphthalene. We cloned and expressed these naphthalene catabolic genes and tested for Cr (VI) reduction, and found that NahG reduced 79% of 100 μM Cr (VI) in 5 minutes. Additionally, an nahG deletion mutant lost 52% of its Cr (VI) reduction ability compared to that of the wild-type strain. As nahG encodes a salicylate hydroxylase with flavin adenine dinucleotide (FAD) as a cofactor for electron transfer, Cr (VI) could obtain electrons from NADH through NahG-associated FAD. To the best of our knowledge, this is the first report of a protein involved in a PAH-degradation pathway that can reduce heavy metals, which provides new insights into heavy metal-PAH contamination remediation.

Indole-3-acetic acid (IAA), known as a common plant hormone, is one of the most distributed indole derivatives in the environment, but the degradation mechanism and cellular response network to IAA degradation are still not very clear. The objective of this study was to elucidate the molecular mechanisms of IAA degradation at the protein level by a newly isolated strain Pseudomonas sp. LY1. Label-free quantitative proteomic analysis of strain LY1 cultivated with IAA or citrate/NH4Cl was applied. A total of 2,604 proteins were identified, and 227 proteins have differential abundances in the presence of IAA, including 97 highly abundant proteins and 130 less abundant proteins. Based on the proteomic analysis an IAA degrading (iad) gene cluster in strain LY1 containing IAA transformation genes (organized as iadHABICDEFG), genes of the β-ketoadipate pathway for catechol and protocatechuate degradation (catBCA and pcaABCDEF) were identified. The iadA, iadB, and iadE-disrupted mutants lost the ability to grow on IAA, which confirmed the role of the iad cluster in IAA degradation. Degradation intermediates were analyzed by HPLC, LC-MS, and GC-MS analysis. Proteomic analysis and identified products suggested that multiple degradation pathways existed in strain LY1. IAA was initially transformed to dioxindole-3-acetic acid, which was further transformed to isatin. Isatin was then transformed to isatinic acid or catechol. An in-depth data analysis suggested oxidative stress in strain LY1 during IAA degradation, and the abundance of a series of proteins was upregulated to respond to the stress, including reaction oxygen species (ROS) scavenging, protein repair, fatty acid synthesis, RNA protection, signal transduction, chemotaxis, and several membrane transporters. The findings firstly explained the adaptation mechanism of bacteria to IAA degradation.
