The antimicrobial efficacy of punicalagin against foodborne pathogens has been extensively studied, yet its impact on spoilage microorganisms remains poorly characterized. This study aimed to evaluate punicalagin's inhibitory effects on Pseudomonas fluorescens and explore its potential application in tilapia preservation. Growth curve analysis showend that punicalagin effetively inhibited the growth of Pseudomonas fluorescens . In addition, punicalagin triggered intracellular potassium ion efflux and leakage of nucleic acids and protein. Scanning electron microscopy revealed compromised cell membrane integrity after punicalagin treatment. Furthermore, punicalagin exposure reduced intracellular ATP levels and elevated reactive oxygen species accumulation. Concurrently, it effectively suppressed biofilm formation and destroyed preformed biofilms. In tilapia spiked with Pseudomonas fluorescens , punicalagin markedly attenuated spoilage progression, as evidenced by diminished protein degradation and lipid oxidation. These collective findings indicate that punicalagin exhibit antimicrobial and anti-biofilm activities against Pseudomonas fluorescens , highlighting its potential as a natural preservative for aquatic products.

Food spoilage by certain species of bacteria is reported to be regulated by quorum sensing (QS). Acinetobacter johnsonii and Pseudomonas fluorescens, the major specific spoilage organisms, are found to be limited in their QS and co-culture interactions. The aim of this study was to determine how QS-regulated proteins affect the spoilage potential of co-cultured A. johnsonii and P. fluorescens obtained from spoiled bigeye tuna (Thunnus obesus) using a proteomics approach. The A. johnsonii, P. fluorescens, and their co-culture tested the N-acyl-homoserine lactone (AHL) activities using reporter Chromobacterium violaceum CV026 and LC-MS/MS in qualitative and quantitative approaches, respectively. These latter showed that, of the 470 proteins and 444 proteins in A. johnsonii (A) and P. fluorescens (P), respectively, 80 were significantly up-regulated and 97 were significantly down-regulated in A vs. AP, whereas 90 were up-regulated and 65 were down-regulated in P vs. AP. The differentially expressed proteins included the AI-2E family transporter OS, 50S ribosomal protein L3, thioredoxin reductase OS, cysteine synthase CysM OS, DNA-binding response regulator, and amino acid ABC transporter ATPase OS. The cellular process (GO:0009987), metabolic process (GO:0008152), and single-organism process (GO:0044699) were classified into the gene ontology (GO) term. In addition, energy production and conversion, amino acid transport and metabolism, translation, ribosomal structure and biogenesis, post-translational modification, protein turnover, and chaperones were distributed into the clusters of orthologous groups of proteins (COG) terms. The KEGG pathways revealed that 84 and 77 differentially expressed proteins were divided into 20 KEGG pathways in A vs. AP and P vs. AP, respectively, and amino acid metabolism, carbohydrate metabolism, energy metabolism, and translation were significantly enriched. Proteins that correlated with the spoilage-related metabolic pathways, including thioredoxin reductase OS, cysteine synthase OS, and pyridoxal phosphate-dependent enzyme family protein OS, were identified. AI-2E family transporter OS and LuxR family transcriptional regulator OS were identified that related to the QS system. These findings provide a differential proteomic profile of co-culture in A. johnsonii and P. fluorescens, and have potential applications in QS and the regulation of spoilage potential.

ABSTRACT Biofilm formation is usually affected by many environmental factors including divalent cations. The purpose of the current work was to analyze how calcium (Ca 2+ ) affects the biofilm formation of dairy Pseudomonas fluorescens isolates by investigating their growth, swarming motility, biofilm-forming capacity, EPS production, and biofilm structures. Moreover, the regulation mechanism of Ca 2+ involved in its biofilm formation was explored through RNA-sequencing analysis. This work revealed that supplementation of 5, 10, 15, and 20 mM Ca 2+ significantly reduced the swarming motility of P. fluorescens strains (P.F 2 , P.F 4 , and P.F 17 ), but the biofilm-forming ability and polysaccharide production were increased after the supplementation of 5 and 10 mM Ca 2+ . By the supplementation of Ca 2+ , complex structures with more cell clusters glued together in P. fluorescens P.F 4 biofilms were confirmed by scanning electron microscopy, and increased biomass and coverage of P. fluorescens P.F 4 biofilms were observed by confocal laser scanning microscopy. In addition, RNA-sequencing results showed that P. fluorescens P.F 4 showed a transcriptional response to the supplementation of 10 mM Ca 2+ , and a total of 137 genes were significantly expressed. The differential genes were represented in 4 upregulated KEGG pathways (nonribosomal peptide structures, quorum sensing, biosynthesis of siderophore group nonribosomal peptides, and phenylalanine metabolism), and 4 downregulated KEGG pathways (flagellar assembly, amino sugar and nucleotide sugar metabolism, nitrotoluene degradation, and cationic antimicrobial peptide (CAMP) resistance). The results indicate that Ca 2+ might serve as an enhancer to substantially trigger the biofilm formation of dairy P. fluorescens isolates in the dairy industry.

Pseudomonas fluorescens is a prevalent spoilage bacterium in various chilled aquatic products, producing toxic biogenic amines (BAs) through the decarboxylation of amino acids via amino acid decarboxylases. Our previous studies revealed that P. fluorescens PF12 regulates BA production through its quorum sensing (QS) system, suggesting that disrupting QS could be a potential approach to control BA generation. In this study, we targeted the three-dimensional structure of the LuxI-type protein in P. fluorescens PF12 to screen for QS inhibitors (QSIs) through high-throughput screening from Seaweed Metabolite Database and Comprehensive Marine Natural Products Database. Nine compounds with high binding scores to the LuxI-type protein were identified. Considering cost-effectiveness and safety, phytol was selected for further investigation. Experimental results demonstrated that sub-inhibitory concentrations of phytol significantly suppressed BA production, siderophore production, motility, extracellular protease secretion, and biofilm formation in P. fluorescens PF12. Specifically, when the salmon fillets were stored for 288 h, the histamine content in the fillets treated with 25 mg/mL phytol decreased by 43.81 %. RT-qPCR revealed that the expression levels of key genes hisD and hisC related to histamine synthesis in the phytol treatment group were reduced by 7.68 ( p < 0.05) and 8.23 % ( p < 0.05), respectively, compared to the control group. Molecular docking analysis revealed that phytol interacts with key residues in the LuxI-type protein, thereby blocking the synthesis of acyl-homoserine lactones (AHLs) and downstream QS signaling. These findings indicate that phytol has great potential for controlling BA production in aquatic products and ensuring food safety.

The spoilage of aquatic products is mainly caused by the bacterial growth, and the specific spoilage organism (SSO) plays an important role. Quorum sensing (QS) is a microbial cell–cell communication system which is coordinated with the population density, and is controlled by N -acyl-homoserine lactone (AHLs) as the Gram-negative bacteria communication signals. In this study, the SSO was Pseudomonas fluorescens (PF-04), isolated from the turbot ( Scophthalmus maximus L. ) during aerobically refrigerated storage. The supernatant extract of PF-04 tested the AHLs activities utilizing biosensor Chromobacterium violaceum CV026. AHL production was influenced by the environment temperature, and AHL production reduced obviously at 10 °C compare with 25 °C. In Luria-Bertani (LB) supplemented with 0.5–1.0% NaCl, AHL production reached the maximum. The AHL production was also regulated by pH of culture medium, acidic condition was conducive to persistent existence of the AHL molecules, but the alkaline environment would cause chemically unstable of AHL molecules. QS system in P. flurosecens played an imperative role in biofilm formation, protease and siderophore production. AHLs could regulate above three factors in PF-04. In summary, this study showed that (1) the influence of different environmental conditions (temperature, NaCl and pH) on AHL production revealed the correlation of QS in foods and (2) that proved the effect of external AHLs to regulate the biofilm formation, protease and siderophore production in PF-04.

LuxR solos sense exogenous signaling molecules and regulate microbial behaviors, thus facilitating intra- and inter-species communication among bacteria. However, studies on the role of LuxR solos in regulating the spoilage potential of specific spoilage organisms are limited. In this study, we screened a LuxR solo EvgA, derived from Pseudomonas fluorescens PF08. qRT-PCR and molecular docking results indicated that EvgA responds and binds most strongly to exogenous N -3-tetradecanoyl-L-homoserine lactone (C14-HSL) through hydrogen bonds and hydrophobic interactions, with a –CDOCKER ENERGY of 45.87 kcal/mol. Subsequently, the evgA knockout (Δ evgA ) and complementary evgA (Δ evgA -com) mutants of P. fluorescens PF08 were constructed through homologous recombination. Δ evgA mutants displayed diminished eavesdropping to exogenous C14-HSL (by 18.99 ± 2.09%), reduced adhesion ability (by 30.33 ± 0.36%), attenuated biofilm formation (by 15.97 ± 0.14%), and decreased motility (swarming zones by 4.34 ± 0.002 mm and swimming zones by 4.17 ± 0.11 mm). In addition, EvgA deletion reduced spoilage factor production (hydrolysis zone), which included extracellular proteases, lipases, and biogenic amines (by 4.45 ± 0.51 mm, 10.12 ± 0.10 mm, and 14.41 ± 0.57 mm, respectively), thereby diminishing the spoilage of salmon fillets stored at 4 °C. Notably, the addition of exogenous C14-HSL promoted the spoilage ability of all strains. This study emphasizes the important role of LuxR solo EvgA in AHLs-mediated quorum sensing and the spoilage potential of P. fluorescens PF08, which is expected to be a new target for controlling the spoilage of aquatic products.

The application of palygorskite (PAL) for potentially toxic trace elements (Cd 2+ , Ni 2+ , etc.) remediation in polluted soil can substantially reduce the bioavailability and toxicity of these hazard materials. However, the secretion of organic acids and siderophores by microorganisms might result in the re-mobilization of cadmium (Cd) in PAL-bound forms (PAL-Cd). In this study, the interactive effects between Cd stabilized by PAL and mobilized by siderophores from Pseudomonas fluorescens were performed with four flask-shaking experimental treatments, namely, strain with or without an ability of siderophores production respectively associated with or without PAL-Cd. The GC-MS and UHPLC-MS test methods were used to analyze the concentrations of metabolites. Results showed that the Cd mobilized by strain with siderophores production was 22.1% higher than that of strain without the ability of siderophores production ( p < 0.05). The mobilization of Cd in PAL in turn significantly reduced the siderophores production of Pseudomonas fluorescens by 25.1% ( p < 0.05). The numbers of metabolites significantly up-regulated and down-regulated were 9 and 22 in strain groups with PAL-Cd addition compared with the groups without PAL-Cd, respectively. Metabolomics analysis revealed that the mobilized Cd affects the signal transduction pathway and primary metabolic processes, reduces the metabolic capacity of pentose phosphate pathway, glycolysis and tricarboxylic acid cycle pathway. These changes inhibit the ability of strain to biosynthesize amino acids during the mobilization processes, further reducing the capacity of Pseudomonas fluorescens to produce siderophores. This study provides a useful information on how to select soil Cd-stabilizing materials in a targeted manner and how to avoid Cd re-mobilization by siderophores.

Proteobacteria primarily utilize acyl-homoserine lactones (AHLs) as quorum-sensing signals for intra-/interspecies communication to control pathogen infections. Enzymatic degradation of AHL represents the major quorum-quenching mechanism that has been developed as a promising approach to prevent bacterial infections. Here we identified a novel quorum-quenching mechanism revealed by an effector of the type IVA secretion system (T4ASS) in bacterial interspecies competition. We found that the soil antifungal bacterium Lysobacter enzymogenes OH11 (OH11) could use T4ASS to deliver the effector protein Le1288 into the cytoplasm of another soil microbiome bacterium Pseudomonas fluorescens 2P24 (2P24). Le1288 did not degrade AHL, whereas its delivery to strain 2P24 significantly impaired AHL production through binding to the AHL synthase PcoI. Therefore, we defined Le1288 as LqqE1 ( L ysobacter q uorum- q uenching e ffector 1 ). Formation of the LqqE1-PcoI complex enabled LqqE1 to block the ability of PcoI to recognize/bind S -adenosy-L-methionine, a substrate required for AHL synthesis. This LqqE1-triggered interspecies quorum-quenching in bacteria seemed to be of key ecological significance, as it conferred strain OH11 a better competitive advantage in killing strain 2P24 via cell-to-cell contact. This novel quorum-quenching also appeared to be adopted by other T4ASS-production bacteria. Our findings suggest a novel quorum-quenching that occurred naturally in bacterial interspecies interactions within the soil microbiome by effector translocation. Finally, we presented two case studies showing the application potential of LqqE1 to block AHL signaling in the human pathogen Pseudomonas aeruginosa and the plant pathogen Ralstonia solanacearum .

Pseudomonas fluorescens , a specific spoilage organism, can control cell density and various physiological processes through quorum sensing (QS). Quorum sensing inhibitors (QSIs) are a crucial strategy for enhancing food quality and safety. In this study, benzyl isothiocyanate (BITC) at subminimum inhibitory concentrations (MICs) promoted protein and nucleic acid leakage, β-galactosidase activity and the relative fluorescence intensity of the membrane potential in P. fluorescens . N-butyryl- l -homoserine lactone (C4-HSL) and N-decanoyl- l -homoserine lactone (C10-HSL) were detected in P. fluorescens by ultra-performance liquid chromatography-mass spectrometry (UPLC-MS/MS). C4-HSL and C10-HSL are known to orchestrate the QS of gram-negative bacteria. The inhibition rates of C4-HSL and C10-HSL by BITC at 1/2 MIC were 54.0%–96.2% and 31.6%–70.5%, respectively, after cultured with P. fluorescens for 6–24 h. The inhibitory effects of BITC at 1/4 MIC on P. fluorescens biofilm formation, extracellular polysaccharide production, and motility were enhanced in the presence of exogenous C4-HSL. Moreover, sub-MICs of BITC markedly downregulated the relative expression of QS-related genes , and the molecular docking results also showed excellent binding ability of BITC to QS proteins. This study demonstrated the feasibility of BITC as a QSI, whereby exogenous C4-HSL promoted the ability of BITC to disturb the QS system in P. fluorescens .

Baeyer–Villiger mono-oxygenases (BVMOs) are enzymes that could insert an oxygen into carbon skeletons adjacent to carbonyl groups to form lactone or ester. Recently, a new method of sebacic acid production from oleic acid based on Pseudomonas fluorescens DSM 50106 (PfBVMO)
catalysis was reported. However, the instable property of PfBVMO limited the further investigation and application of this enzyme. In this study, we aimed to improve the stability of PfBVMO by sitedirected mutagenesis. The sequence of PfBVMO indicates probable factors from the susceptible
oxidizing cysteine residue leading to the instability of PfBVMO. Activity and stability of mutants and the wild-type were characterized to investigate the correlation of cysteine residues and activity. The mutant enzyme with entire cysteine residues replaced by serine residues, showed significantly
higher stability than the wild type. The result would facilitate further work on the enzyme modification and crystal structure determination. Meanwhile, the results of this study provided potential for other enzyme stability evolution.

This study investigated the synergistic effects of lactic acid (0.5%, 1% and 2%) and ultrasound (400 W, 20 kHz) on the inactivation of Pseudomonas fluorescens . Combined ultrasound and 2% lactic acid for 5 min reduced the planktonic cell count below 1.40 log cfu/mL. Ultrasound promoted the inactivation of P. fluorescens biofilms by lactic acid and increased the release of ATP and nucleic acids from bacterial cells by 0.09–0.22 μmol/OD and 0.043–0.113 of OD 260 values, respectively. Results indicated that combined ultrasound and lactic acid can serve as a potential technique to control P. fluorescens biofilms.

In this study, the synergistic antimicrobial effect of allyl isothiocyanate (AITC) and cinnamaldehyde (CAL) against Pseudomonas fluorescens ( P. fluorescens ) was investigated. The fractional inhibitory concentration index (FICI) value of 0.5 in the combination of AITC and CAL indicated the synergistic effect on P. fluorescens by reducing their dosages. Compared with the individual AITC and CAL, the combined treatment group increased protein leakage concentration and intracellular reactive oxygen species content, as well as significantly inhibited the motility of P. fluorescens ( P < 0.05), with a reduction of 6.3 and 10.3 mm in swimming diameter, respectively. Meanwhile, this combination resulted in a 93.4 % reduction in biofilm formation compared to the untreated group. Moreover, combined treatment with AITC and CAL significantly inhibited the growth of P. fluorescens and delayed the increase of total volatile base nitrogen in chilled beef. Therefore, this study provides a reference for the application of a combination of AITC and CAL for beef preservation.
