Purpose: Pseudomonas syringae pv. actinidiae causes bacterial canker of kiwifruit and is responsible for severe economic losses and emergence of drug-resistant bacteria. Bacteriophages are viruses that infect target bacterial hosts and may be the best strategy to prevent and control kiwifruit canker disease. The objective of this experiment was to monitor the prevalence of Pseudomonas syringae pv. actinidiae and provide insight for the use of phages in biological control.Materials and methods: In this study, 52 strains of Pseudomonas syringae pv. actinidiae were isolated from 68 stem samples of kiwi plant (cv. Hongyang & Jinkui). Following polymerase chain reaction (PCR) analysis, 15 isolates belonging to biovar 3 were identified, one of which was named XWY0007 and used as the target strain to isolate the phages. Thirty-six phages were isolated and purified from a total of 51 surface water samples collected in Shanghai. All phages were identified by transmission electron microscopy (TEM) and their host ranges were evaluated. Three phages, designated φXWY0013, φXWY0014 and φXWY0026 were selected and further characterised using one-step growth curve and stability at different temperatures and pH.Results and conclusions: The isolated phages are promising for use as antimicrobials against bacterial canker in kiwi. This report is regarding Pseudomonas syringae pv. actinidiae and its phages from major areas of kiwifruit cultivation.

Bacterial canker caused by Pseudomonas syringae pv. actinidiae (Psa) severely impacts global kiwifruit production. This study shows that 5-fluoroindole exerts significant bactericidal activity against Psa, with a half-maximal effective concentration (EC50) of 15.34 μg/mL, demonstrating a higher efficacy than the positive control copper hydroxide (EC50 = 58.65 μg/mL). 5-Fluoroindole disrupts membrane integrity, induces oxygen species (ROS) accumulation, and triggers apoptosis in Psa. Transcriptome analysis reveals that 5-fluoroindole treatment increased the expression of the methionine synthase II (MetE) gene by 6.28 fold compared with the expression of this gene in the control (CK) group. Microscale thermophoresis and isothermal titration calorimetry show that the dissociation constants of the binding of 5-fluoroindole to MetE protein are 0.33 and 8.55 μM, respectively. Molecular docking experiments indicate that Asp693 is a key binding site and that there is no specific binding between 5-fluoroindole and the Psa MetED693A mutant. This study provides valuable insights for developing effective agents to control kiwifruit bacterial canker.

Pseudomonas syringae pv. actinidiae (Psa), the pathogen that causes bacterial canker disease in kiwifruit, has brought about substantial losses to the kiwifruit industry. Investigating the infection process at both the cellular and microscopic levels is of great significance for the formulation of effective control strategies against this disease. Thus, the establishment of appropriate chemical imaging analysis methods becomes essential. Confocal Raman microspectral imaging (CRMI), combined with chemometrics, provides an intuitive means to visualize and characterize the spatiotemporal changes of biopolymers in the cell walls of both healthy and infected kiwifruit stems. Raman spectra of different infected stems exhibit clustering effects in Principal component analysis (PCA), and a classification model constructed using Support vector machines (SVM) achieves an accuracy of 97%. Multivariate Curve Resolution-Alternating Least Squares (MCR-ALS) is utilized to resolve spectral matrices and concentration profiles from raw Raman imaging signals. The reconstructed concentration data yields accurate molecular imaging maps of High-methylated pectin (HMP), Low-methylated pectin (LMP), cellulose, hemicellulose, and lignin. The results indicate that, three days after Psa infection, the content of cellulose and HMP in the cell wall increases, while the changes in hemicellulose, lignin, and LMP are minimal. However, five days after infection, the contents of HMP, LMP, cellulose, hemicellulose, and lignin decrease significantly, resulting in the disruption of the cell-wall structure. The chemical imaging method proposed in this study shows great promise as an effective means for studying the bacterial infection process in kiwifruit stems at the cellular level.

With the demand for natural polymers, cellulose nanocrystal (CNC)-based Pickering emulsion systems (PE) have been introduced in agricultural control due to their high stability. Surfactants further reduce surface tension and enhance the spreading and deposition of pesticide formulations. In controlling bacterial canker in kiwifruit, conventional formulations often exhibit high-resistance, but poor spreading, and deposition, resulting in unsatisfactory efficacy. Therefore, selecting highly effective and low-toxicity formulations is critical for kiwifruit production. In this study, the plant extract—P-Cymene (PC, identified via laboratory screening) was used as a model compound with strong antibacterial activity to prepare CNC/surfactant synergistically stabilized Pickering emulsions (PC@PE). The CNC employed was the type I cellulose nanorods exhibiting structural integrity, and strong electronegativity, enabling the formation of oil-in-water Pickering emulsions. PC@PE stabilized with CNC and the surfactant SK-551 demonstrated high stability across temperature, pH, and ionic strength variations. Rheological analysis confirmed that PC@PE behaves as a non-Newtonian liquid with gel-like properties. PC@PE exhibited a surface tension of 25.90 mN·m −1 , a contact angle of 9.40° on kiwifruit leaves, a deposition of 48.36 mg·cm −2 , and an EC 50 value of 69.089 mg·L −1 against Pseudomonas syringae pv. actinidiae ( Psa ), indicating enhanced leaf affinity and biological activity. These results highlight the effectiveness of PC against Psa , expand the application of Pickering emulsions in agricultural disease management, and provide a new approach for controlling bacterial canker in kiwifruit.
