The current study aimed to evaluate the in vivo wound healing potential of Psidium guajava L. by wound excision model owing to its ethnomedicinal use as a wound healer since olden times. P. guajava leaves were extracted using methanol-chloroform (1:1). Phytochemical analysis of P. guajava leaves extract (PGLE) was performed using total phenolic content (TPC) assay, total flavonoid content (TFC) assays, and High-performance liquid chromatography (HPLC). In vitro , antioxidant potential (DPPH radical scavenging assay, total antioxidant capacity (TAC) assay, and ferric reducing antioxidant power (FRAP) assay), antibacterial activity (broth microdilution method), and hemolytic analysis of PGLE were performed. In vivo , wound healing proficiency of PGLE (10, 5, and 2.5% w/w) was determined by estimating hydroxyproline and endogenous antioxidant markers. TPC and TFC were found to be 57.5 ± 0.37 µg gallic acid equivalent/mg extract and 32.1 ± 0.24 µg quercetin equivalent/mg extract, respectively. A prominent concentration of polyphenols was demonstrated by HPLC-based quantification with the highest value of rutin which is 12.3 ± 0.12 µg/mg extract. A noteworthy DPPH radical scavenging activity with an IC 50 of 40.7 µg/ml, TAC, and FRAP i.e., 159.3 ± 0.11 and 206.1 ± 0.17 µg ascorbic acid equivalent/mg extract, respectively were exhibited by PGLE. Significant antibacterial activity against Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeroginosa, and Methicillin-resistant Staphylococcus aureus with the minimum inhibitory concentration of 250, 400, 400, and 500 µg/ml respectively was depicted by PGLE and found biocompatible with 1.1 ± 0.16% hemolysis. Significant wound contraction, faster epithelialization (9 ± 1 days), extensive fibroblast proliferation, and substantial neovascularization were observed in the 10% PGLE-treated group also evident by histopathological examination. Significant hydroxyproline (15.7 ± 1.02 µg/mg tissue), catalase, peroxidase (5.4 ± 0.26 and 6.1 ± 0.35 U/mg tissue) and glutathione content (9.7 ± 0.06 mmol/mg tissue) with p-value of less than 0.01 was demonstrated by 10% PGLE. A reduction in lipid peroxidation (0.5 ± 0.07 mmol/mg tissue) was also observed at the same concentration (10%). These findings scientifically validate the potent pharmacological effects of P. guajava phytochemicals and suggest their promising role as wound healing agents.

Hydrodistilled essential oils (EO) of Psidium guajava L. leaves from different regions in China were analyzed by GC and GC/MS. The samples from Guangdong Province displayed high EO yields (0.61 – 0.75%, v / w ). A total of 50 components, representing over 98.00% of the EOs, were identified and semi-quantitatived. The major constituents of EOs included β -caryophyllene (17.17 – 31.38%), γ -gurjunene (9.17 – 15.22%), τ -cadinol (1.35 – 10.02%) and calamenene (2.13 – 7.80%). The terpenoids in all sample oils were dominated by sesquiterpenes hydrocarbons (70.18 – 84.35%), followed by oxygenated sesquiterpenes (9.89 – 22.19%). The similarities and differences among EOs from different samples were evaluated by hierarchical cluster analysis and principal component analysis methods. The IC 50 values of EOs from different regions were between 18.52 – 33.72 mg/ml (DPPH) and 13.12 – 25.15 mg/ml (ABTS + ). The FRAP value of EO from Guangdong Province was 7.34 – 9.13 mmol Vc/g DM, while the FRAP value of EO from Taiwan Province was 2.29 – 2.36 mmol Vc/g DM. The antimicrobial tests revealed that EO had a higher antimicrobial activity against all Gram -positive bacteria and two fungi. Moreover, EO from P . guajava leaves of Guangdong Province showed the highest antimicrobial activity. These properties can be considered in the design of industrial products and for further application in the food, pharmaceutical and cosmetic industries. Graphical

Graphical Psidium guajava leaf extract used as an agent for reducing Ag(I) ions to silver nanoparticles on the surface of hydrotalcite particles. The hybrid material has high antibacterial ability to E. coli , S. aureus and marine bacterium, P. stutzeri B27. Psidium guajava leaf extract is a green, low-cost reducing agent and the biosynthesis process of the hybrid material can be up to large scale. Phenolic compounds are widely known for its well-reduced behavior and surprisingly appeared in many leaf extracts, including Psidium guajava ( P. guajava ) leaf. This leaf extract has been confirmed as an efficiency reducing agent to reduce Ag(I) ions to silver nanoparticles (AgNPs). This work focused on the development of a “green” synthesis of nanosilver-hydrotalcite nanoparticles (HT-AgNPs) based on the bio-reduction of the P. guajava leaf extract and the application of HT-AgNPs as an antibacterial material. To assess factors affecting on synthesis process of the HT-AgNPs, experiments have been carried out with a fixation of HT/AgNO 3 precursors while varying concentration of P. guajava leaf extract as well as solvent for the extraction. The methods, including ultraviolet-visible (UV-Vis) spectroscopy, infrared (IR) spectroscopy, field emission scanning electron microscopy (FESEM), dynamic light scattering (DLS), X-ray diffraction (XRD) and energy dispersive X-Ray (EDX), transmission electron microscopy (TEM) have been used to characterize the HT-AgNPs. The yield of reduction process of Ag(I) ions to AgNPs on HT reached 55.12 to 90.55 % depending on the content of AgNO 3 precursor. In particular, the antibacterial activity of HT-AgNPs against to E. coli and S. aureus was also assessed and discussed.

ABSTRACT Introduction Psidium guajava L. ( P guajava ) and its leaf, one of the most popular plants in traditional medicine and cuisine, were demonstrated to have high flavonoid content with many bioactivities, including antigout activity. However, the correlation between total flavonoid content ( TFC ) and antigout activities and the optimal conditions for extracting P guajava have not been reported. Methods This study established the optimisation of ethanol extracts of P guajava leaves for TFC and antigout activities using the single-factor method and response surface methodology. The antigout activities were determined through the xanthine oxidase inhibition ( XOI ) method. Results The four independent variables (ethanol concentration, plant-solvent ratio, extraction time, and temperature) significantly impacted two response variables (TFC and XOI activity). The optimal extracts for XOI activity reach the maximum TFC of 323.86 ± 2.26 mg quercetin/g and XOI activity of 26.37 ± 0.5 μg/ml. The changes in the chemical structure of extracts were analysed by Fourier transform infrared, which indicated that several flavonoids in P guajava do not have XOI activity. The correlation of TFC and XOI is robust with the Pearson correlation of −0.882 and Spearman Rho test of −0.884. Additionally, the biological activities, such as antioxidant and anti-inflammatory, of optimal extracts for XOI and TFC were determined to extend the application capacity to support the treatment of gout diseases. Conclusions The optimised extraction conditions are critical to isolating flavonoids, which can serve as functional food or medicine for managing gout and its complications.

Six metabolites (1–6), including 1 new fusaric acid derivative, designated atransfusarin (1), and 5 known compounds, (3R,6R)-3-benzyl-6-isopropyl-4-methylmorpholine-2,5-dione (2), daucoterol (3), adenosine (4), cerebroside B (5), and 2,3-dihydroxypropyl (Z,Z)-9,12-octadecadienate (6), were isolated from the solid culture of an endophytic Alternaria atrans MP-7, associated with the medicinal plant Psidium guajava. The structure of the new compound was elucidated by spectral data. The antifungal test indicated that (3R,6R)-3-benzyl-6-isopropyl-4-methylmorpholine-2,5-dione (2) showed obvious inhibitory activity against Alternaria solani, Colletotrichum gloeosporioides, and Phyricularia grisea. Compared to fusaric acid (1a), even though the antifungal activity of atransfusarin (1) against A. solani, C. gloeosporioides, and P. grisea was greatly decreased, its inhibitory effect against Botrytis cinerea was still retained.

Fermentation results in the release and biotransformation of phenolic compounds in guava leaves. This study aimed to evaluate the bioaccessibility, safety and antidiabetic effect of phenolic-rich extract from fermented guava leaves (PE-fgl) using Monascus anka and Bacillus sp. BS2. The total phenolics and flavonoids were 51.18% and 16.60% bioaccessible after gastrointestinal digestion, respectively. High bioaccessibility was observed, particularly for ellagic acid, isoquercitrin, quercetin-3-O-β-D-xylopyranoside, quercetin-3-O-α-L-arabinofuranoside and avicularin, resulted in high recovery of α-glucosidase inhibition activity and antioxidant capacity. Moreover, PE-fgl was practically nontoxic with a median lethal dose (LD 50 ) greater than 5000 mg/kg and was without evidence of toxicity following subchronic administration up to the dose limit of 2000 mg/kg. Furthermore, PE-fgl significantly improved serum glucose and lipid levels as well as antioxidant capacity in diabetic mice. Liver, kidney and pancreas damage was also significantly alleviated. These results suggested that PE-fgl could be a nontoxic candidate for diabetes treatment.

The cuticular membrane covering almost all aerial plant organs has a primary function in limiting uncontrolled water loss . The guava fruits were collected and this work was done to study the potential contribution of cuticular chemical composition to fruit transpiration after harvest. The detailed cuticular chemical composition, based on gas chromatography together with mass spectrometry , and the transpiration rate determined gravimetrically in guava fruit were characterized in the present study. The predominant wax mixtures were fatty acids and primary alcohols with homologous series of C 16 –C 33 , as well as various pentacyclic triterpenoids with abundant amounts of ursolic acid, maslinic acid and uvaol. The most prominent cutin compounds were C 16 and C 18 ‒type monomers dominated by 9(10),16-diOH-hexadecanoic acid and 9,10-epoxy-ω–OH–octadecanoic acid, respectively. Relatively high water permeability with a value of 5.1 × 10 −4 m s −1 was detected for guava fruit. The lower efficiency of the cuticle as barrier to transpiration in guava fruit, as compared to that of other reported fruits, leaves, and petals, was seemingly related to the relatively short average chain-length of acyclic compounds in wax mixtures. These findings provide useful insights linking the chemical composition of the cuticular membrane that covers plant organs to putative physiological roles.

The development of natural anti-hyperglycemic agents with minimal side effects is a critical objective in functional food research. This study comprehensively investigated the anti-diabetic potential and bioactive substances in guava juice fermented with Lacticaseibacillus paracasei NCUH012072 (FM). Using an integrated in vitro and metabolomic approach, the anti-hyperglycemic activity was systematically assessed through a series of bioassays (enzyme inhibition, antioxidant capacity, and insulin-resistant cell models), untargeted metabolomics, and bioinformatic and affinity-based validation. The results demonstrated that FM significantly inhibited key carbohydrate-metabolizing enzymes ( α -amylase, α -glucosidase) as well as the insulin signaling regulator protein tyrosine phosphatase 1B (PTP1B) ( P < 0.05). Additionally, FM exhibited enhanced antioxidant capacity and improved glucose uptake in insulin-resistant HepG2 cells ( P < 0.05). Untargeted metabolomics revealed that fermentation modulated 13 pivotal metabolic pathways. Integrated analysis using Spearman correlation and random forest modeling identified key anti-hyperglycemic metabolites, including flavonoids ( e.g ., liquiritigenin), phenolic acids ( e.g ., cis-caffeic acid), and redox-active compounds such as (R)-lipoic acid. Affinity ultrafiltration combined with liquid chromatography-mass spectrometry (AF-LC-MS) further characterized quercetin 3-galactoside and resveratrol as potent inhibitors of the target enzymes. This study not only systematically demonstrates the anti-hyperglycemic properties of probiotic-fermented guava juice but also proposes a comprehensive strategy for identifying bioactive compounds in complex food matrices. The findings offer a theoretical basis for developing fermented fruit-based functional foods as dietary strategies in hyperglycemia management.

Lycopene-rich guava ( Psidium guajava L.) exhibits significant economic potential as a functional food ingredient, making it highly valuable for the pharmaceutical and agro-food industries. However, there is a need to enhance the extraction methods of lycopene to fully exploit its beneficial uses. In this study, we evaluated various ionic liquids to identify the most effective one for extracting lycopene from guava. Among thirteen ionic liquids with varying carbon chains or anions, 1-butyl-3-methylimidazolium chloride demonstrated the highest productivity. Subsequently, a single-factor experiment was employed to test the impact of several parameters on the efficiency of lycopene extraction using this selected ionic liquid. These parameters included extraction time, ultrasonic power, liquid-solid ratio, concentration of the ionic liquid, as well as material particle size. Moreover, models of artificial neural networks using genetic algorithms (ANN-GA) and response surface methodology (RSM) were employed to comprehensively assess the first four key parameters. The optimized conditions for ionic liquid ultrasound-assisted extraction (IL-UAE) were determined as follows: 33 min of extraction time, 225 W of ultrasonic power, 22 mL/g of liquid-solid ratio, 3.0 mol/L of IL concentration, and extraction cycles of three. Under these conditions, lycopene production reached an impressive yield of 9.35 ± 0.36 mg/g while offering advantages such as high efficiency, time savings, preservation benefits, and most importantly environmental friendliness.
