Twenty glycoside derivatives and nine flavonoids from the leaves of Pueraria (P. thomsonii) were isolated by column chromatography and characterized by nuclear magnetic resonance spectroscopy (NMR) and high performance liquid chromatography (HPLC). The contents of twenty glycosides and nine flavonoids from the extract of P. thomsonii leaf (PL) were 173.3 mg g−1 and 134.7 mg g−1, respectively. Two flavonoids with the highest content were robinin (49.28 mg g−1) and puerarin (42.87 mg g−1). Six flavonoids, i.e. puerarin, robinin, rutin, quercetin, quercitrin, and kaempferol showed more inhibitory effects against α-glucosidase than acarbose. PL could effectively increase the level of insulin, decrease the content of fasting blood glucose, reduce lipid accumulation in plasma, ameliorate oxidative injury and inflammation, and relieve liver and kidney damage in diabetic mice. Moreover, PL could increase intestinal probiotics to improve metabolic disorders caused by diabetes and decrease the level of Clostridium celatum to relieve inflammation. This study suggested that PL or its glycoside derivatives and flavonoids regulating glycolipid metabolism and inflammation levels might have the potential to be used to control type 2 diabetes.

Root plasticity is essential for optimizing crop yields and ensuring survival under stress. However, the regulatory roles of root-enriched secondary metabolites in developmental processes have remained largely unexplored. Pueraria thomsonii , a leguminous species distinguished by its rare tuberous roots, exhibits remarkable isoflavonoid accumulation. Chemical quantification and in vitro functional analyses have identified daidzin as a key regulator of root architectural plasticity in this species. Using an integrated approach combining Arabidopsis thaliana (L.) root phenotyping system, cellular thermal shift assays (CETSA) and thermal proteome profiling (TPP), daidzin was demonstrated to modulate root development via stem cell-related processes and auxin signaling pathways. Specifically, daidzin led to the reduced auxin levels, silenced DR5::GFP expression in the stele, and suppressed PIN protein expression accompanied by a lateral shift in PIN2 polarity. Furthermore, daidzin decreased SHR, SCR, PLT1, and PLT2 protein levels while increasing WOX5 and CYCD6;1 accumulation. In the wox5 mutant, daidzin inhibited root elongation, whereas in the p35S:WOX5-GR transgenic line, it enhanced root elongation and attenuated starch granule degradation. These findings reveal that WOX5 functionally counteracts daidzin-mediated root growth inhibition. Furthermore, daidzin stimulated cell proliferation in Pueraria thomsonii (Benth.) and induced root thickening in Panax quinquefolius (L.). Collectively, these results suggest that daidzin's coordinates root architectural plasticity by modulating auxin transport, polarity, and stem cell niche maintenance, highlighting its potential as a valuable tool for enhancing root development in economically important crops.

A simple and efficient method was developed for the rapid screening and identification of ligands for monoamine oxidase B. A new ionic-liquid-based ultrasound-assisted extraction method for medicinal herbs was also developed and validated. In addition, the hyphenated technique of countercurrent chromatography and semipreparative-LC was developed and applied to the isolation of the chemical constituents for Pueraria thomsonii Benth. Three potent monoamine oxidase B inhibitors, namely, daidzein-4′,7-diglucoside (42.2 mg), puerarin 6′′- O -xyloside (88.3 mg), and 3′-hydroxypuerarin (48.5 mg) with purities of 98.2, 96.3, and 97.1%, respectively, were obtained from 500 g of P. thomsonii raw material using semi-preparative high-performance liquid chromatography, whereas 3′-methoxypuerarin (76.2 mg), daidzein-8-C-apiosyl (1→6) glucoside (84.2 mg), and tectorigenin (75.1 mg) with purities of 98.5, 96.4, and 96.8%, respectively, were obtained from 500 g raw material via countercurrent chromatography using a two-phase solvent system comprising n -hexane–ethyl acetate–methanol–water at a volume ratio of 1.85:1.00:0.86:3.69 (v/v/v/v). Then, the anti-Alzheimer activity of the phytochemicals was assessed using a PC12 cell model. Treatment with tectorigenin, daidzein-4′,7-diglucoside, puerarin 6′′- O -xyloside, 3′-hydroxypuerarin, 3′-methoxypuerarin, and daidzein-8-C-apiosyl (1→6) glucoside (100 μg/mL), resulted in cell viabilities of 69.00, 65.81, 59.69, 57.90, 55.61, and 54.59%, respectively ( p < 0.001). The protocol was proved to be very accurate and efficient.
