Bioinspired pullulan-starch nanoplatelets nanocomposite films with enhanced mechanical properties

Qian Xiao,Miaoqi Dai,Min Huang,Loong-Tak Lim

Inspired by the leaf-vein network structure, the pullulan-starch nanoplatelets (SNPs) bioinspired films with enhanced strength and toughness were successfully fabricated through a water evaporation-induced self-assembly technique. SNPs (SNP200 and SNP600) of two sizes were separated by differential centrifugation . Interactions between SNPs and pullulan during drying resulted in the vein-like network structure in both nanocomposite films when the appropriate amounts of SNP200 or SNP600 were added to pullulan , respectively. The TS and toughness values of pullulan with 1 % w/w SNP200 films reached up to 51.05 MPa and 69.65 MJ·m −3 , which were 86 % and 223 % higher than those of the neat pullulan films, respectively. Moreover, the TS and toughness values of pullulan-SNP200 were significantly higher than those of pullulan-SNP600 films, when SNP content exceeded the 1 % w/w level. By applying a graph theory, the network structures were found to correlate with the mechanical properties of the pullulan-SNPs bioinspired films. The new strategy for designing starch nanoplatelets-based edible films that combine mechanical strength and toughness holds promises for the development of novel biobased composite materials for food packaging application.

Efficient decontamination of heavy metals from aqueous solution using pullulan/polydopamine hydrogels

Qiankun Zeng,Xiaoliang Qi,Mengying Zhang,Xianqin Tong,Ning Jiang,Wenhao Pan,Wei Xiong,Yahui Li,Jianxiong Xu,Jianliang Shen,Lijian Xu

Designing a new adsorbent with recyclability, high efficiency and biodegradability is important for treating heavy metals contamination but remains a severe challenge. In this work, a novel type of hydrogel biosorbents based on pullulan and polydopamine were designed for heavy metal ions removal from aqueous solution. The physicochemical properties of the prepared pullulan/polydopamine (Pu/PDA) hydrogels were fully characterized by thermal gravimetric analysis, Fourier transform infrared spectroscopy, rheology, scanning electron microscopy, swelling and compression tests. We observed that their mechanical strength, pore size, water absorption and retention properties could be nicely controlled by adjusting the PDA concentration in the pre-gel solution. Subsequently, the adsorption ability of designed Pu/PDA hydrogels to Cu 2+ , Co 2+ and Ni 2+ was investigated in detail. These hydrogels presented excellent adsorption capability for heavy metal ions and matched well with the pseudo-second-order kinetic model and Freundlich isotherm model. Overall, having tunable physicochemical properties coupled with the high absorption ability for heavy metal ions, these Pu/PDA hydrogels may be a promising strategy for removal of pollutants from aqueous solution.

The Role of Fe(III) in Selective Adsorption of Pullulan on Calcite Surfaces: Experimental Investigation and Molecular Dynamics Simulation

Kaiwei Ding,Tingsheng Qiu,Xianhui Qiu,Guanfei Zhao,Qinghao Jiao,Jiangjie Fang,Ruisen Lai,Wenhui Yang

The floatability of fluorite and calcite exhibit similar properties, rendering their flotation separation challenging. Macromolecular polysaccharide reagents containing the polyhydroxyl group have shown broad promising application. The selectivity of polysaccharide is relatively low. In this study, the introduction of Fe3+was employed to enhance the selective adsorption capacity of Pullulan polysaccharide towards fluorite and calcite minerals, thereby achieving effective flotation separation. Furthermore, the mechanism underlying intramolecular interactions was elucidated. The DFT calculation and XPS analysis revealed that the adsorption of Fe3+on the calcite surface was more favorable, leading to the formation of a Ca-O-Fe structure. The MD simulation, XPS analysis, and Zeta potential analysis revealed that the Fe-OH groups on the surface of calcite reacted with the -OH groups in Pullulan and formed bonds, resulting in the formation of a Calcite-Fe-Pullulan structure. This facilitated the attachment of a significant number of Pullulan molecules to the calcite surface. The formation of a hydrophilic layer on the outer surface of calcite by Pullulan, in contrast to the absence of such layer on fluorite’s surface, results in an increased disparity in surface floatability between these two minerals, thereby enhancing the efficiency of flotation separation.

Synergistic chemotherapeutic effect of sorafenib-loaded pullulan-Dox conjugate nanoparticles against murine breast carcinoma

Junhui Sui,Yani Cui,Hanxu Cai,Shaoquan Bian,Zhiyi Xu,Ling Zhou,Yong Sun,Jie Liang,Yujiang Fan,Xingdong Zhang

pH-Sensitive pullulan-doxorubicin conjugates encapsulating sorafenib (P-Dox/S) nanoparticles were developed as a synergistic combinatorial delivery system against murine breast carcinoma. The nanoparticles can encapsulate Dox and sorafenib with ultra-high loading capacity (65.34 wt%) through chemical conjugation and physical loading, whereas can remain stable under physiological conditions and gradually release Dox and sorafenib with the decreasing pH. These conjugates can be effectively internalized and clearly suppress 4T1 cell growth in vitro. Furthermore, research data of in vivo animal models revealed that the synergistic combinatorial P-Dox/S nanoparticles heavily accumulated in solid tumor tissue sites to maximize therapeutic efficacy; they also significantly inhibited solid tumor growth, even remarkably reduced solid tumor volume in comparison to the initial volume, and obviously diminished adverse effects. The anti-tumor therapeutic effect obviously outperformed the delivery of combinational chemotherapy of free drugs or single drug-loaded P-Dox nanoparticles at the same concentration. These promising results indicate the high-efficiency synergistic chemotherapeutic effects of these nanoparticles. Combinational chemotherapy using P-Dox/S nanoparticles has important potential in the clinical treatment of malignancy for overcoming drug resistance and heterogeneity.

Separation of hematite and quartz in a cationic collector flotation system using Pullulan as a greener depressant

Panxing Zhao,Wengang Liu,Wenbao Liu,Ying Guo,Mengqiang Chen,Yong Mao,Rui Ding

As the uncertainly of climate and the world situation intensifies, there are more and more pressures on resources and environment faced by human beings, the requirements for the development of mineral resources have become higher and higher. Iron mineral, as the most exploited mineral by mankind at present, the green and efficient purification of iron ore is an important means to realize energy saving and carbon reduction. In this paper, a novel hematite depressant—Pullulan (an environment friendly microbial polysaccharide, represented as PU thereafter), was introduced into the purification of hematite from quartz in cationic collector flotation system. Furthermore, a serious of surface tests were applied to explore the adsorption mechanisms between PU and the mineral surfaces. According to these experiments, it can be concluded as follows. The results of micro-flotation exhibited that PU showed better inhibition for hematite in DDA flotation system, the concentrate product with Fe grade of 65.88 %, and 91.06 % recovery was obtained from the artificial mixed minerals at 20 mg/L DDA, 15 mg/L PU at natural slurry pH. Wettability analysis suggested that PU could enable the hematite surface to become more hydrophilic, while little effect on wettability changes were observed on the quartz surface. The zeta-potential, FTIR and XPS analyses revealed that PU could occur adsorption on the hematite surface relying on hydrogen bonding with the hydrated hematite surface, and chemical complexation formed between the hydroxyl groups on PU and the Fe sites of the hematite surface.

Flexibility, dissolvability, heat-sealability, and applicability improvement of pullulan-based composite edible films by blending with soluble soybean polysaccharide

Fan Zhao,Lijun Jiang,Chenyu Wang,Shuangdie Li,Di Sun,Qingbao Ma,Zhongjie Yu,Yu Liu,Wei Jiang

This study aimed to prepare a heat-sealable, water-soluble, edible film based on pullulan (PUL)/soluble soybean polysaccharide (SSPS) for the portable packaging of convenience foods. The effects of the PUL/SSPS ratios on the microstructure, physicochemical properties, barrier properties, and mechanical properties of the PUL/SSPS films were explored. Fourier-transform infrared spectroscopy and X-ray diffraction showed an apparent hydrogen bonding interaction between SSPS and PUL in the PUL/SSPS films. As the SSPS proportion in the PUL/SSPS films increased, the water contact angle, moisture content, water vapor permeability, solubility, oxygen permeability, flexibility, and antioxidant activity increased, whereas light transmittance decreased. The heat-seal strength of the P9S1 film (PUL/SSPS ratio of 9:1) is slightly lower than that of the PUL film ( p > 0.05). Furthermore, the application potential of the PUL/SSPS films for packaging peptide powder was evaluated. The P9S1 film exhibited a satisfactory packaging effect and was completely dissolved in water with low turbidity. Therefore, the P9S1 film is a promising packaging material for convenient powdered foods.

Effects of pullulan on the biomechanical and anti-collapse properties of dicalcium phosphate dihydrate bone cement

Wenjing Xi,Zhengwen Ding,Haohao Ren,Hong Chen,Yonggang Yan,Qiyi Zhang

In this work, a modified dicalcium phosphate dihydrate (DCPD) bone cement with unique biodegradable ability in a calcium phosphate cement system was prepared by the hydration reaction of monocalcium phosphate monohydrate and calcium oxide and integration with pullulan (Pul), a non-toxic, biocompatible, viscous, and water-soluble polysaccharide that has been successfully used to improve defects in DCPD bone cement, especially its rapid solidification, fragile mechanical properties, and easy collapse. The effect of different contents of Pul on the structure and properties of DCPD were also studied in detail. The modified cement was characterised by X-ray diffraction, Fourier-transform infrared spectroscopy, scanning electron microscopy, ultraviolet–visible absorption, X-ray photoelectron spectroscopy analysis, and rheological property measurements. The results indicated that Pul promoted the hydration formation of DCPD, and interface bonding occurred between Pul and DCPD. With increasing content of Pul, the setting time of the DCPD bone cement increased from 2.6 min to 42.3 min, the compressive strength increased from 0 MPa to 20.4 MPa, and the anti-collapse ability also improved owing to the strong interface bonding, implying that the DCPD bone cement improved by Pul has better potential for application in the field of non-loading bone regenerative medicine compared to unmodified DCPD bone cement.

Fabrication of Abelmoschus manihot gum/pullulan/magnesium L-ascorbate green composite packaging film and its excellent performance in preserving fresh-cut carrots

Kok Bing Tan,Meixia Zheng,Junyan Lin,Yujing Zhu,Guowu Zhan,Jianfu Chen

Pullulan-based composite film can be a potential alternative packing material to non-environmentally friendly plastic wrap (PE) to preserve fresh-cut carrots. However, many developed pullulan-based composites either have high water vapor permeability (WVP) and high mechanical strength or vice versa, which limits the practicality of the developed packaging materials for potential commercialization. Herein, Abelmoschus manihot gum (AMG)/pullulan/magnesium L-ascorbate (MLA) was created as a green composite film (APL) to preserve fresh-cut carrots. The optimal amount of MLA was found to be 10 % (APL10), demonstrating a balance of lower WVP and greater mechanical strength and antioxidant performance than many pullulan-based films. This effectively solved many problems faced by other pullulan-based packaging films. After the fresh-cut carrots were packed with the composite film for 4 days, it was found that APL10 was effective in preserving the quality of carrots, in terms of freshness, weight loss rate, Vitamin C (VC), and malondialdehyde (MDA) content after 4 days of storage, much better than non-biodegradable PE. Thus, based on these findings, it is concluded that APL films have huge potential as a green packaging material for food to replace PE in the future.

Development of electrospun nanofiber films based on pullulan/polyvinyl alcohol incorporating bayberry pomace anthocyanin extract for aquatic products freshness monitoring

Lijun Jiang,Chenyu Wang,Fan Zhao,Shuangdie Li,Di Sun,Qingbao Ma,Zhongjie Yu,Bin Zhang,Yu Liu,Wei Jiang

Aquatic products were susceptible to spoilage during storage due to microorganisms and enzymes , which could cause food safety problems. The electrospun nanofiber films, based on pullulan (PUL) and polyvinyl alcohol (PVA) incorporating bayberry pomace anthocyanin extract (BAE), were developed to monitor the freshness states of aquatic products in this study. The SEM results revealed that the electrospun nanofiber films exhibited a smooth surface, and the average diameter decreased from 481.56 nm to 237.68 nm as anthocyanin content increased from 0 to 20%. The FTIR and XRD results showed that there were no new characteristic peaks, confirming the successful loading of BAE. The incorporation of BAE decreased the water content (from 13.58% to 7.84%), swelling ratio (from 146.75% to 114.24%), water vapor permeability (from 3.38 g·mm·m −2 ·Pa −1 ·s −1 ·10 −5 to 2.00 g·mm·m −2 ·Pa −1 ·s −1 ·10 −5 ), and elongation at break (from 24.67% to 14.90%), but increased water solubility (from 74.99% to 85.74%) and water contact angle (from 51.84° to 63.73°). The electrospun nanofiber films presented obvious color changes in buffer solutions (pH 2.0 to 12.0) or volatile ammonia and showed good reversibility. Besides, the electrospun nanofiber films were stable during storage at 4 °C or 25 °C for 20 days. Furthermore, PUL-PVA-A15 effectively distinguished the freshness states (fresh, sub-fresh and spoiled) of Penaeus vannamei during storage at 4 °C by showing different colors (from pink to grayish purple to green). Therefore, the electrospun nanofiber films showed great potential to real-time monitor the quality of aquatic foods as intelligent packaging.

Fabrication of novel pullulan/carboxymethyl chitosan-based edible film incorporated with ultrasonically equipped aqueous zein/turmeric essential oil nanoemulsion for effective preservation of mango fruits

Jawad Ashraf,Nadia Ismail,Tabussam Tufail,Jiyao Zhang,Muhammad Awais,Qianqian Zhang,Zahoor Ahmed,Yajing Qi,Shuyi Liu,Bin Xu

This study aimed to develop ultrasonically-assisted, alcohol-free, and noncorrosive aqueous zein/turmeric essential oil (TEO)-loaded nanoemulsions (NEs) to stimulate pullulan/carboxymethyl chitosan (P/CMCS)-based edible films for mango fruit preservation. The influence of innovative sonicated zein/TEO-based NEs (ZTNEs) as nanofillers on the physico-mechanical characteristics of the resulting P/CMCS edible films was investigated. A stable and well-dispersed ZTNE was achieved using 20 % zein with 10 min of ultrasound treatment, leading to a reduced droplet size (194.23 ± 0.41 nm) and high ζ-potential (−48.72 ± 0.74 mV). Furthermore, the homogeneity of the ZTNE dispersion, evaluated via confocal laser microscopy (CLSM) analysis, indicated that it can be uniformly incorporated into P/CMCS edible films. In addition, the incorporation of ZTNE into P/CMCS edible films improved barrier properties, mechanical strength, oxidative stability, and antimicrobial properties compared with P/CMCS edible films without ZTNE, possibly due to hydrogen bonding in the film matrix, as confirmed by scanning electron microscopy (SEM) micrographs as well as Fourier transform infrared spectroscopy and X-ray diffraction. Interestingly, the results indicated that P/CMCS/ZTNE/US 10 min and P/CMCS/ZTNE 4 % films demonstrated substantial antibacterial activity against Escherichia coli (11.54 ± 0.72 and 16.62 ± 1.13 mm, respectively) and Staphylococcus aureus (12.81 ± 1.03 and 14.13 ± 0.69 mm), respectively. Moreover, P/CMCS/ZTNE/US 10 min and P/CMCS/ZTNE 4 % films were found to be effective in mango preservation over a 12-day storage period at 25 °C ± 1 °C, as assessed by fruit firmness, weight loss, total soluble solid content, total phenolic content, lipoxygenase activity, and electronic nose analysis. Consequently, these findings indicate that P/CMCS/ZTNE edible films may function as an effective biodegradable packaging solution for fruit preservation within the food industry.

Improved flotation of molybdenite from talc using a selective reagent scheme

Wanjia Zhang,Liming Tao,Luobing Xun,Zhongxu Qi,Stephen Pooley,Wei Sun,Jian Cao,Zhiyong Gao

Talc is one of the most problematic gangue minerals for molybdenite flotation owing to its strong natural hydrophobicity. In this work, pullulan polysaccharide (PU) was evaluated as a potential depressant in the flotation separation of molybdenite from talc. Bench flotation tests of single mineral samples and multi-mineral samples were carried out to evaluate the performance of PU in flotation. And the results show that under the preferable reagent scheme of 40 mg/L PU, 160 mg/L butyl xanthate (BX) and 25 μL/L MIBC, molybdenite can be effectively separated from talc over a wide pH range (from 2.0 to 8.0). Furthermore, FTIR and XPS analyses demonstrate that BX can desorb the pre-adsorbed PU on the surface of the molybdenite by competitive adsorption, but has only a slight effect on the PU adsorbed on surface of the talc. The results also confirm that PU has a strong and stable adsorption on the surfaces of talc and molybdenite, whereas BX adsorbs more strongly on the surface of molybdenite. Hence, the reagent scheme of PU + BX, with biodegradability, low toxicity, low cost and synergistic selectivity, has great potential for industrial application in the flotation separation of molybdenite from talc.

Pullulan fermented by Aureobasidium melanogenum TZ-FC3 for the preparation of self-healing, adhesive, injectable and antibacterial pullulan/PVA/borax hydrogel

Guang Yang,Hongjuan Mo,Bingjie Liu,Yiwen Wu,Guiqin Liu,Yadong Hu,Xue Jiao,Kexin Guo,Xin Wei,Yaowei Fang,Mingxuan Pan,Liang Hao

Natural polymer hydrogels, such as pullulan-based hydrogels, offer significant advantages over synthetic materials due to their thermal stability, film-forming capacity, solubility, adhesiveness, and antioxidant properties. In this study, the strain Aureobasidium melanogenum TZ-FC3, which produces a high level of pullulan, was successfully isolated from the mangrove ecosystems of Guangdong Province, China. 66.01 ± 1.10 g/L pullulan without melanin was produced by the TZ-FC3 strain within 120 h at flask level. Pullulan fermented by A. melanogenum TZ-FC3 was added to enhance the hydrogen bond network within the pullulan/PVA/borax hydrogels (P-2, P-3 and P-4 hydrogels) to improve mechanical strength and crosslinking density of PVA/borax hydrogel (P-1 hydrogel). Compared to the P-1 hydrogel, the P-2 hydrogel exhibited a 65.4 % increase in tensile strain, a remarkable 694.03 % increase in tensile strength and improved the degree of internal crosslinking. Additionally, the pullulan/PVA/borax hydrogels demonstrated excellent self-healing properties, adhesion, injectability, and antibacterial activity. The preparation process of pullulan/PVA/borax hydrogels is straightforward and effective, suggesting broad industrial applicability and underscoring their potential as next-generation materials for advanced healthcare solutions.
