The effect of glutathione metabolism on the release of Wound-Induced Spores from Pyropia yezoensis

Haihong Chen,Ziyan Shi,Zhihai Zhong,Nanjing Ji,Xiaoqian Zhou,Zhijie Dan,Xin Shen

Oxidative stress from mechanical injury can trigger the development of Wound-Induced Spores (WIS) in Pyropia yezoensis , yet the regulatory mechanisms underlying this process require further elucidation. In this study, comparative transcriptomic analysis between injured and uninjured P. yezoensis thalli revealed significant differential gene expression within the glutathione (GSH) metabolism pathway during WIS development. There was a down-regulation in the expression of genes encoding for γ-glutamyl-cystein synthetase (GCL) and glutathione synthetase (GSS), which were pivotal for GSH biosynthesis, during the development of WIS. Moreover, a diminished expression level was also observed in genes associated with oxidative metabolism of GSH during this process, including those encoding glutathione S-transferase (GST), glutathione peroxidase (GPX), glutathione reductase (GR), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), and dihydroascorbate reductase (DHAR). However, this transcriptional regulation does not correspond to a significant decrease in intracellular GSH levels, which may be associated with a reduction in the oxidative metabolism of GSH, leading to decreased cellular utilization of GSH. Treatment injured P. yezoensis with Buthionine sulfoximine (BSO) promoted the release probability of WIS, the release efficiency of WIS and germination rate of asexual spores, while supplementation of GSH produced opposite results, suggesting that modulating GSH levels can regulate the release of WIS. Correlation analysis suggested that there was a significant positive correlation between the release of WIS and the germination rate of asexual spores, suggesting that promoting the release of WIS could lead to the acquisition of a greater number of asexual spore seedlings. Our findings underscore the key role of GSH metabolism in WIS regulation and provide a theoretical foundation for the subsequent development of strategies to obtain asexual spore seedlings.

Effects of periodical dehydration on biomass yield and biochemical composition of the edible red alga Pyropia yezoensis grown at different salinities

Xinshu Li,Xin Sun,Lin Gao,Juntian Xu,Guang Gao

Pyropia is an important marine crop and periodical dehydration could help kill fouling organisms during cultivation. However, our understanding regarding the effects of periodical dehydration on biomass yield and biochemical composition of Pyropia remains very lacking, particularly for thalli grown at different salinities. In this study, Pyropia yezoensis experiencing periodical dehydration or not was cultured at four levels of salinities (17, 25, 32, 39 psu) for 19 days. The highest growth rate was found at salinity 25 psu and lowest was at salinity 39 psu. Dehydration reduced relative growth rate at each salinity except for the insignificant decrease at 39 psu. Dehydration did not affect the maximal photochemical efficiency (Fv/Fm) and the highest salinity decreased it slightly. Dehydration induced more Chl a and carotenoids and highest Chl a content was found at salinity 39 psu. Dehydration did not affect phycoerythrin or phycocyanin content at salinities 25–39 psu but respectively increased them by 28% and 50% at salinity 17 psu. Dehydration did not affect content of any amino acid while salinity interacted with dehydration on most amino acids. The optimal salinity for the production of total amino acids (AA),essential AA, umami AA, and sweet AA was 25 psu for the thalli without dehydration; while it shifted to 17 psu for the thalli with dehydration. Dehydration and salinity interacted on content of the fatty acids C12, C18:1(n-9), C18:2(n-6), and C20:2(n-6). For the synthesis of eicosapentaenoic acid (EPA, C20:5(n-3)) and polyunsaturated fatty acids (PUFA) in thalli without dehydration, the optimal salinity was 39 psu, but the effect of salinity disappeared when thalli experienced periodical dehydration. These findings show the interactive effects of periodical dehydration and salinity on biomass yield and biochemical composition in P. yezoensis and provide insights into optimizing conditions for nori cultivation.

Nitrogen availability modulates the effects of ocean acidification on biomass yield and food quality of a marine crop Pyropia yezoensis

Guang Gao,Qi Gao,Menglin Bao,Juntian Xu,Xinshu Li

Pyropia yezoensis is an important marine crop in the world. We cultured it under two levels of partial pressure of carbon dioxide (pCO 2 ) (408 (LC), 998 (HC) μatm) and nitrate (30 (LN) and 500 (HN) μmol L −1 ) to investigate the effect of ocean acidification on its growth and food quality under changing nitrogen supply. HC decreased growth rate of P. yezoensis under LN but did not affect it under HN. Phycoerythrin and phycocyanin were enhanced by HC, particularly at HN, which contributed to the darker color. HC stimulated the synthesis of sweat amino acids regardless of nitrate condition and umami amino acid only under LN. HN increased the content of umami amino acids regardless of pCO 2 condition and sweet amino acids only under HC. Our findings indicate that future ocean acidification may reduce biomass yield of P. yezoensis but increase its color and flavor, which was regulated by nitrate availability.

Antitumor bioactivity of porphyran extracted from Pyropia yezoensis Chonsoo2 on human cancer cell lines

Dan He,Siya Wu,Liping Yan,Jihui Zuo,Yang Cheng,Hanfei Wang,Jian Liu,Xu Zhang,Mingjiang Wu,Jong-il Choi,Haibin Tong

BACKGROUND Pyropia yezoensis , rich in porphyran, is a medicine-edible red alga. In the present study, the physicochemical characteristics, conformational states and antitumor activities of a novel porphyran extracted from the high-yield algal strain Pyropia yezoensis Chonsoo2 and its two degraded derivatives by gamma irradiation were investigated. RESULTS Pyropia yezoensis porphyran is a water-soluble, triple-helical sulfated hetero-galactopyranose, named PYP. PYP was degraded by gamma irradiation at 20 kGy and 50 kGy, giving two low molecular weight derivatives comprising PYP-20 and PYP-50, respectively. PYP with a higher molecular weight has a solution conformation different from PYP-20 and PYP-50. Three porphyrans had no toxicity in normal human liver cells (HL-7702) and showed antitumor effects on Hep3B, HeLa and MDA-MB-231. They had better antitumor against HeLa cells, exhibiting a similar inhibition ratio compared to 5-fluorouracil, with PYP especially exhibiting a higher inhibition ratio than 5-fluorouracil. With respect to HeLa cells, the different antitumor activities might be related to porphyran molecular weight and solution conformation. Furthermore, the HeLa cell cycle was blocked in the G2/M phase after PYP treatment, leading to cell proliferation inhibition. The induction of cell cycle arrest was related to the changes in the expression of p21, p53, Cyclin B1 and cyclin-dependent kinase 1. CONCLUSION Pyropia yezoensis porphyran, as applied to medicine and functional food, could potentially be used as a non-toxic natural adjuvant in cancer therapy. © 2019 Society of Chemical Industry

Inhibitory activity of a sulfated oligo-porphyran from Pyropia yezoensis against SARS-CoV-2

Lihua Geng,Quanbin Zhang,Qishan Suo,Jing Wang,Yingxia Wang,Cong Wang,Ning Wu

COVID-19 caused by SARS-CoV-2 has spread around the world at an unprecedented rate. A more homogeneous oligo-porphyran with mean molecular weight of 2.1 kD, named OP145, was separated from Pyropia yezoensis . NMR analysis showed OP145 was mainly composed of →3)-β- d -Gal-(1 → 4)-α- l -Gal (6S) repeating units with few replacement of 3,6-anhydride, and the molar ratio was 1:0.85:0.11. MALDI-TOF MS revealed OP145 contained mainly tetrasulfate-oligogalactan with Dp range from 4 to 10 and with no more than two 3,6-anhydro-α- l -Gal replacement. The inhibitory activity of OP145 against SARS-CoV-2 was investigated in vitro and in silico. OP145 could bind to Spike glycoprotein (S-protein) through SPR result, and pseudovirus tests confirmed that OP145 could inhibite the infection with an EC50 of 37.52 μg/mL. Molecular docking simulated the interaction between the main component of OP145 and S-protein. All the results indicated that OP145 had the potency to treat and prevent COVID-19.

Overexpression of Rboh enhances inorganic carbon acquisition through coordinating with carbonic anhydrase in Pyropia yezoensis

Zhizhuo Shao,Menglin Guo,Hong Wang,Wenhui Gu,Xiujun Xie,Guangce Wang

Pyropia yezoensis is an important intertidal economic macroalgae, which is periodically affected by various stresses, such as the limitation of inorganic carbon (C i ) deficiency. Under such environment, the redox homeostasis within the cells of Pyropia yezoensis is seriously affected, and the reactive oxygen species (ROS) signal transduction system would be activated to regulate the photosynthetic activity. Therefore, how P. yezoensis manage ROS to maintain effective photosynthetic carbon fixation has aroused great interest. Here, we characterize transformants overexpressing respiratory burst oxidase homolog ( Rboh ), an important gene that can actively produce ROS, at the levels of cellular physiology, biochemistry, and transcriptomics. Our data indicated the expression of Rboh significantly increased, accompanied by a significant upregulated expression of alpha-type carbonic anhydrase 3 (α CA3 ) and increased extracellular carbonic anhydrase activity in the Rboh overexpressing strains. Interestingly, compared with the wild type, the photosynthetic activity of transgenic strains was significantly higher under the low C i and high light condition, implying that the ROS signal triggered by overexpression of Rboh was involved in regulating the C i absorption and utilization in P. yezoensis when the C i source was limited. In summary, this study provided evidence supporting the correlation between the ROS production and the Ci utilization under stress environments in P. yezoensis .

Pyropia yezoensis porphyran alleviates metabolic disorders via modulating gut microbiota in high-sucrose-fed Drosophila melanogaster

Dan He,Qifang Wu,Chenying Lu,Jinlan Wu,Peichao Chen,Mingjiang Wu,Jong-il Choi,Haibin Tong

BACKGROUND Prebiotics, such as algal polysaccharides, can be used to manage metabolic diseases by modulating gut microbiota. However, the effect of Pyropia yezoensis porphyran (PYP), a red algal polysaccharide, on gut microbiota has not been reported. Thus, the objective of this study was to determine effects of PYP on metabolic disorders caused by high sucrose (HS) and underlying mechanisms involved in such effects. RESULTS Biochemical analysis demonstrated that an HS diet increased triglyceride and circulating sugar contents (metabolic abnormalities) in Drosophila larvae. It also increased the relative abundance of harmful microbiota within the larvae as identified by 16S ribosomal DNA analysis. PYP supplementation at 25 and 50 g kg −1 equivalently reduced metabolic abnormalities in the HS group. Therefore, 25 g kg −1 PYP was selected to investigate its effects on the metabolic pathway and gut microbiota of larvae in the HS group. The activity of PYP in ameliorating metabolic abnormalities by reverse transcription quantitative real-time polymerase chain reaction analysis was consistent with the expression trend of key factors involved in metabolism regulation. PYP reduced the relative abundance of bacteria causing metabolic abnormalities, such as Escherichia–Shigella and Fusobacterium , but increased the relative abundance of beneficial bacteria such as Bacillus and Akkermansia . However, PYP had no effect on triglyceride and circulating sugar contents in HS-fed larvae treated with a mixture of antibiotics designed to remove gut microbiota. CONCLUSION PYP exhibits anti-metabolic disorder activity by modulating gut microbiota, thereby supporting the development of PYP as a functional prebiotic derived from red algae food. Non-targeted metabolomic analysis of Pyropia yezoensis metabolites using GC-MS and its regulation under high temperature

Zeb Aurang,Yang Xiuwen,Khan Yasmin,He Hongyan,Fu Caiwei,Zhang Dongren,Qu Jing,Shen Songdong

Pyropia yezoensis (red algae) or commonly known as nori, is highly regarded for its nutritional benefits and distinct taste, leading to its widespread consumption. The bio-activity and sensory characteristics of P. yezoensis are attributed to the metabolites it contains. In this study, identification and quantification of the diverse range of metabolites of P. yezoensis and metabolomic analysis were conducted using gas chromatography-mass spectrometry (GC-MS). Furthermore, the impact of high temperature on its metabolites regulation was also investigated. Due to metabolomic analysis, a diverse range of metabolites were identified in P. yezoensis , including lipids, amino acids, carbohydrates, and secondary metabolites. Several known bioactive compounds, including alcohol and polyols, amines, amino acids-peptides-analogues, beta hydroxy acids and derivatives, carbohydrates and carbohydrate conjugates, cholestane steroids, dicarboxylic acid and derivatives, and fatty acids and conjugates were detected in abundance, highlighting the nutritional and functional properties of P. yezoensis . Additionally, the metabolites composition of P. yezoensis was significantly affected in high temperatures, which led to up-regulation of considerable primary metabolites and few were down-regulated, and suggested a potential response and adaptation mechanism of P. yezoensis to elevated temperature conditions. This research highlighted the metabolomics of P. yezoensis , provided insights into its metabolite composition and regulatory responses to high temperature conditions, enhanced our knowledge of the biochemical pathways and adaptive mechanisms of P. yezoensis , which can assist the improvement strategies of utilization and cultivation to promote this valuable alga in response to fluctuating environmental conditions.

Yanli Du,Soon-Mi Shim,Lei Wang,Xin Gao,Xiaoting Fu

Fermentation can promote various properties of food. This study investigated the impact of fermentation by Monascus purpureus combined with Lactobacillus plantarum and Saccharomyces cerevisiae on the physicochemical and flavor characteristics of Pyropia yezoensis . Results indicate that the protein and free amino acid content increased threefold. A total of 62 volatile flavor compounds were identified by GC-IMS. Alcohols, ketones and esters were enhanced with 2- to 6-fold, while aldehydes were reduced by 76.56 %, contributing to the enhancement of pleasant fragrances and the reduction of fishy and seawater odors. As for the non-volatile properties, the content of organic acids and free amino acids increased from 2.18 mg/g and 0.19 mg/g to 46.03 mg/g and 4.93 mg/g, respectively, as assessed by HPLC and amino acid analyzer. The metabolic pathways of non-volatile substances were speculated upon. This work provides a basic theory and a practical reference for fermentation food processing of P. yezoensis .
