Integration of Trehalose Lipids with Dissociative Trehalose Enables Cryopreservation of Human RBCs

Yan Wang,Shuhui Gao,Kongying Zhu,Lixia Ren,Xiaoyan Yuan

Cryopreservation of red blood cells (RBCs) is imperative for transfusion therapy, while cryoprotectants are essential to protect RBCs from cryoinjury under freezing temperatures. Trehalose has been considered as a biocompatible cryoprotectant that naturally accumulates in organisms to tolerate anhydrobiosis and cryobiosis. Herein, we report a feasible protocol that enables glycerol-free cryopreservation of human RBCs by integration of the synthesized trehalose lipids and dissociative trehalose through ice tuning and membrane stabilization. Typically, in comparison with sucrose monolaurate or trehalose only, trehalose monolaurate was able to protect cell membranes against freeze stress, achieving 96.9 ± 2.0% cryosurvival after incubation and cryopreservation of human RBCs with 0.8 M trehalose. Moreover, there were slight changes in cell morphology and cell functions. It was further confirmed by isothermal titration calorimetry and osmotic fragility tests that the moderate membrane-binding activity of trehalose lipids exerted cell stabilization for high cryosurvival. The aforementioned study is likely to provide an alternative way for glycerol-free cryopreservation of human RBCs and other types of cells.

Core–Shell Microfiber Encapsulation Enables Glycerol-Free Cryopreservation of RBCs with High Hematocrit

Qin Xianhui,Chen Zhongrong,Shen Lingxiao,Liu Huilan,Ouyang Xilin,Zhao Gang

Cryopreservation of red blood cells (RBCs) provides great potential benefits for providing transfusion timely in emergencies. High concentrations of glycerol (20% or 40%) are used for RBC cryopreservation in current clinical practice, which results in cytotoxicity and osmotic injuries that must be carefully controlled. However, existing studies on the low-glycerol cryopreservation of RBCs still suffer from the bottleneck of low hematocrit levels, which require relatively large storage space and an extra concentration process before transfusion, making it inconvenient (time-consuming, and also may cause injury and sample lose) for clinical applications. To this end, we develop a novel method for the glycerol-free cryopreservation of human RBCs with a high final hematocrit by using trehalose as the sole cryoprotectant to dehydrate RBCs and using core–shell alginate hydrogel microfibers to enhance heat transfer during cryopreservation. Different from previous studies, we achieve the cryopreservation of human RBCs at high hematocrit (> 40%) with high recovery (up to 95%). Additionally, the washed RBCs post-cryopreserved are proved to maintain their morphology, mechanics, and functional properties. This may provide a nontoxic, high-efficiency, and glycerol-free approach for RBC cryopreservation, along with potential clinical transfusion benefits.

Protocol for isolation and ATAC-seq library construction of zebrafish red blood cells

Yanyan Ding,Feng Liu

Summary Understanding chromatin dynamics in red blood cells (RBCs) is critical for exploring the differentiation process and homeostasis maintenance during erythropoiesis. Here, we describe a protocol for isolation of zebrafish erythrocytes labelled with gata1 :dsRed by fluorescence-activated cell sorting. We detail steps for ATAC-seq library construction from the isolated RBCs and describe how to analyze the quality of the library. The library can then be used to assay genome-wide chromatin accessibility in these RBCs. For complete details on the use and execution of this protocol, please refer to Ding et al. (2021). 1

Biomimetic platelet membrane-camouflaged liposomes with RBCs hitchhiking enable efficient targeting and suppression of lung metastasis

Kehui Zhu,Xunyi You,Ke Zhang,Yee Shan Wong,Hong Wang,Rui Zhong,Jiaxin Liu,Ye Cao

Activated platelet membranes upregulate tumor adhesion markers. • Liposomes with activated PLMs target CTCs efficiently. • RBC-hitchhiking technique enhances liposomal lung targeting and circulation. • The RBC-PLM-lipo system integrates biomimetic and physical targeting strategies. • A novel DDS shows promising efficacy against lung cancer CTCs and metastasis. Circulating tumor cells (CTCs) drive metastasis via bloodstream dissemination and are recognized as a leading cause of cancer-related mortality. Current targeting strategies using single surface receptors often suffer low efficiency and off-target toxicity. CTCs induce thrombosis to form a protective cloak against immune attack. Inspired by this, we used activated platelet membranes (PLMs), which show upregulated adhesion-related markers, to prepare pH-sensitive paclitaxel-loaded liposomes, offering superior tumor targeting. The RBC-hitchhiking technique further enhanced liposomal lung targeting and circulation time. Integrating these advantages, we developed an RBC-PLM-lipo drug delivery system (DDS) and evaluated it against lung cancer CTCs-induced metastases. Our results showed that RBC-PLM-lipo DDS targets lung cancer, inhibits tumor growth, reduces the M2/M1 macrophage ratio, and reverses the immune-suppressive microenvironment. Thus, this DDS improves conventional chemotherapy efficacy, reduces off-target toxicity, and maximizes clinical benefit for lung cancer patients.

Cheap and easily separate Cs(I) and Rb(I) from high salty solution by forming RbCs–MgHCF

Kang Li,Junjie Xie,Zhuonan Shi,Changli Min,Zichen Yin,Ruixin Ma,Shina Li

Rubidium and cesium are highly-priced and important rare alkali elements but they always co-exist with large amount of sodium and potassium in a complex system resulting very difficult and expensive to recover. Hexacyanoferrates (MHCFs) is one of the most efficient methods to separate Rb + and Cs + if we don't consider certain drawbacks such as too small particle size and too viscous liquids to be precipitated. In this study, a new kind of hexacyanoferrate, magnesium (II) hexacyanoferrate (RbCs–MgHCF) has been fabricated at room temperature and applied for the first time to recover Rb + and Cs + from high-salty solutions. Compared with traditional divalent transition elements, such as CuHCFs, ZnHCFs, NiHCFs, CoHCFs or FeHCFs, RbCs–MgHCF had excellent precipitation property and large particle size of about 300 nm. The structure, morphology and composition were characterized by Fourier transform infrared (FTIR), X-ray diffraction (XRD), Scanning electron microscopy (SEM), Inductively coupled plasma (ICP), and Energy dispersive spectroscopy (EDS). The effects of various factors (divalent metal cations, surfactant, Mg 2+ /(Rb + +Cs + ) mole ratio, pH value and titration rate) on the recovery of Rb + and Cs + were explored in 50 mL batch experiments. Scale-up experiment were conducted in 6 and 18 L. Results identified that deposition ratio for Cs + and Rb + were 99% and 95% among pH ranges of 4.0–11.0. The composition of RbCs–MgHCF was confirmed to be Rb 0.61 Cs 1.20 K 0.17 Mg 1.01 Fe(CN) 6 by ICP and EDS and the synthesis mechanism of RbCs–MgHCF is proposed.

RBCs as a Bioinspired Drug Delivery System for Co-delivery of Irinotecan and Nanoalumina Enhances Colorectal Cancer Therapy

Hongyu Zhong,Yichen Li,Faisal Raza,Jiyuan Xie,Ruonan Rong,Mingfeng Qiu,Jing Su

Colorectal cancer (CRC) is a malignant epithelial tumor with high morbidity and mortality. In CRC treatment, irinotecan (CPT-11) as a chemotherapeutic drug is widely applied. However, its half-life is short, leading to large dosages and severe side effects. Red blood cells (RBCs) are biocompatible drug carriers with high capacity, avoiding premature drug degradation and achieving slow drug release. Nanoalumina (AN) is an emerging immune adjuvant that can enhance the immune response. Here, we used RBCs as carriers and absorbed AN to construct AN–CPT-11–RBCs. CPT-11 would induce tumor cell death, releasing much tumor antigen, while AN would activate immune cells to recognize newly released antigens and induce lymphocyte proliferation, enhancing the antitumor effect simultaneously. With loading amounts of 4 mg of CPT-11 and 3 mg of AN per 109 RBCs, AN–CPT-11–RBCs had similar properties to natural RBCs. In vivo, AN–CPT-11–RBCs could circulate for 9 days and stimulate the proliferation of lymphocytes in the spleen and tumor tissue, having a higher tumor growth inhibition rate of 74.01% and a lower frequency of administration. In conclusion, AN–CPT-11–RBCs attain the co-delivery of CPT-11 and AN for the synergistic treatment of CRC.

Microfluidics-Generated Cryo-Fracture Resistant CNCs-Reinforced Composite Microcapsules for Enhanced Cryopreservation of Encapsulated Cells

Marlene Davis Ekpo,George Frimpong Boafo,Yanmin Yang,Yimer Seid,Chuanpin Chen,Songwen Tan,Rongrong Wang,Zhen Huang

Immunoisolated cells hold great promise for advancing cell therapeutics; however, their long-term storage remains a critical challenge due to ice formation and mechanical damage to both cells and the encapsulating matrix during cryopreservation. In this study, a cryo-fracture-resistant composite microcapsule has been developed using microfluidic technology, where sodium alginate was reinforced with cellulose nanocrystals (CNCs). The composite microcapsules were assessed for their ability to provide structural, mechanical, morphological, chemical, and thermal stability during cryopreservation. Additionally, red blood cells (RBCs) were used as a model to evaluate the ability of the optimized microcapsules to improve cell viability post-thaw. Results revealed that the microfluidic system allowed for precise control over microcapsule size and uniformity, which is crucial for even freezing and thawing. CNCs reinforced microcapsules exhibited delayed ice formation, improved ice recrystallization inhibition, and superior protection against freeze-induced deformations. Furthermore, the composite microcapsules enhanced post-thaw recovery of encapsulated RBCs and enabled the incorporation of a biocompatible cryoprotectant (trehalose). This study demonstrates a novel approach to optimizing cryopreservation techniques by leveraging the enhanced properties of composite microcapsules to mitigate cryo-injury and improve the functional recovery of encapsulated cells. These findings present a promising strategy for advancing microencapsulation-based cell therapies and broader biomedical applications.

Unraveling Potential Causative Components for the Deleterious Effect of Atmospheric Fine Particulate Matter on Red Blood Cells

Yuzhu Zhang,Laijin Zhong,Jing Zhan,Zhipeng Yin,Yao Pei,Dong Cao,Qian S. Liu,Qunfang Zhou,Qian Liu,Guibin Jiang

Atmospheric fine particulate matter (PM2.5) poses threats to the cardiovascular system. Red blood cells (RBCs) are the most abundant cells in blood, which are actively involved in multiple hematological diseases, such as blood clot formation and thrombosis. Exploring how PM2.5 with spatiotemporal heterogeneity influences the hematological system by targeting RBCs would help gain insights into the deleterious effects of PM2.5 and provide clues for finding the causative components therein. Herein, the PM2.5 samples collected from 3 urban sites in Beijing (i.e., Chaoyang, Shunyi, and Yanqing districts) during 4 seasons of 2022 were studied for their toxicities to mouse RBCs, and the main contributing components were further explored through chemical analysis and correlation measure. The results showed that exposure to PM2.5 samples decreased adenosine triphosphate (ATP) levels and increased phosphatidylserine (PS) externalization of RBCs, causing cell morphological deformity. The Pearson correlation analysis showed that the aromaticity of the dissolved organic matter (DOM) in PM2.5 samples was positively correlated with PS exposure of RBCs, showing that the lignin-like compounds were the potential contributors. The negative correlation of zeta potentials of PM2.5 samples with PS exposure of RBCs showed the particle-derived bioactivities of this airborne pollutant. The simulative test based on artificial nanomaterials of carbon black (CB) and oxidized CB (OCB) confirmed the crucial role of particulate carbon in PM2.5-induced effects on RBCs, and soot with a certain oxidation degree was, thus, recognized as another contributor, given its ubiquitous existence in PM2.5 samples. This study, for the first time, revealed PM2.5-induced PS exposure of RBCs, and the causative components of DOM and soot were unraveled. Considering the inevitable contact of airborne PM2.5 with RBCs in the blood circulatory system, the findings obtained herein would help bridge the gap between PM2.5 exposure and the risk of cardiovascular diseases, like thrombogenesis.
