A novel strategy employing lignin biochar to simultaneously promote remediation and safe crop production in Cd-contaminated soil

Yanying He,Baobin Mi,Cheng Luo,Honglin Tao,Xin Zhang,Jinwen Yu,Xueying Mo,Jian Hu,Long Chen,Naimei Tu,Fangfang Wu

The scarcity of arable land and the problem of food security urgently called for a strategy of Remediation-by-Production (RBP). Biochar is a promising method for addressing Cd-contaminated soils, but the time required for remediation and the variability of long-term performance have a direct impact on the realization of the RBP strategy. We have developed a novel approach for remediating cadmium-contaminated flooded soil by utilizing lignin biochar (LBC) in combination with safe rice cultivation. The addition of 1 % LBC achieves efficient Cd passivation (CP: 90.89 %) before rice Cd uptake peaks. Meanwhile, LBC can increase soil organic matter and alter microbial community structure, decreasing the relative abundance of soil pathogenic bacteria from 5.12 % to 4.03 % while raising nutritional bacteria from 15.18 % to 28.82 %. Furthermore, LBC successfully reduced the accumulation of Cd in rice grains by 31.3 % ∼ 54.6 %, making rice dwarf, sturdy, and greener, and lowering the health risk coefficients of Cd across various age groups by Monte Carlo simulations. This study elucidated the critical role of humic substances of LBC in remediation time and performance and laid a solid theoretical foundation for promoting the application of biochar in soil contamination remediation and simultaneously realizing the safe production of crops.

Involvement of dysregulated RNA binding protein and alternative splicing regulatory networks in diabetic nephropathy from type 2 albuminuric cohorts

Wang Yu,Zhang Jingjing,Tang Qian

Background Diabetic nephropathy (DN) is a primary contributor to end-stage renal disease, yet the underlying molecular mechanisms remain incompletely understood. This study aims to elucidate the role of RNA-binding proteins (RBPs) and RBP-alternative splicing (AS) regulatory networks in the pathogenesis of DN. Methods Two RNA-seq datasets (GSE117085 and GSE142025) were retrieved from the Sequence Read Archive (SRA) database. Regulated alternative splicing events (RASEs) and genes (RASGs) of RASEs, along with differentiated RBPs, were identified. Validated differentiated RBPs were correlated with clinical features using the Nephroseq v5 online platform. Using the DN mouse model and RT-qPCR, validated the alternative splicing of RNA. Results Our analysis revealed 15 differentiated RBP genes and 423 RASEs in the kidney cortex of DN rats compared to controls. Enrichment analysis highlighted lipid metabolism pathways for RASGs. Seven of the identified RBPs were validated in kidney biopsy samples from DN patients versus controls. A co-deregulatory network was constructed based on dysregulated RBPs and RASEs, with select RASGs identified. In vivo experiments, compared to normal mice, the mRNA levels of RPS19 were significantly elevated in the renal tissues of DN mice, while the levels of CPEB4 and CRYZ were markedly decreased. Conclusion In conclusion, this study provides evidence implicating dysregulated RBPs and RBP-AS regulatory networks in the development of diabetic nephropathy. The validated RBPs exhibited close associations with clinical biomarkers, reinforcing their potential as therapeutic targets for DN. These findings enhance our understanding of the molecular basis of DN and offer new insights for future research and intervention strategies. Clinical trial Not applicable.

Immunomodulatory mechanism of Radix Bupleuri polysaccharide on RAW264.7 cells and cyclophosphamide-induced immunosuppressed mice

Xueyan Wu,Jun Peng,Chunyu Zhang,Yifan Liao,Xinran Zhang,Yilin Hou,Tiantang Fan,Tianyun Fan,Jigang Wang

Radix Bupleuri polysaccharides (RBP) are heteropolysaccharides with a molecular weight distribution ranging from 1.10 to 121.21 kDa, extracted from the traditional Chinese medicinal herb Radix Bupleuri . Using DEAE-cellulose column chromatography, one neutral polysaccharide (RBP-1) and two acidic polysaccharides (RBP-2 and RBP-3) were isolated. These polysaccharides exhibited the capability to activate RAW264.7 macrophages, with the acidic fractions demonstrating greater effects than the neutral ones. RNA sequencing revealed that differentially expressed genes (DEGs) were significantly enriched in MAPK and NF-κB signaling pathways following RBP-3 treatment. Mechanistic studies further demonstrated that RBP-3 activates these pathways through toll-like receptors TLR2 and TLR4. Additionally, RBP effectively alleviated immunosuppression by modulating gut microbiota and restoring the integrity of the intestinal mucosal barrier in cyclophosphamide (CTX)-induced immunosuppressed mice. RBP influenced the generation of colonic short-chain fatty acids (SCFAs), regulated CD4+ T cell differentiation, and enhanced intestinal immune function. These findings highlight RBP potential as a natural immunomodulator and establish a theoretical foundation for the formulation of immune-supportive botanical products derived from Radix Bupleuri .

Engineered poly(A)-surrogates for translational regulation and therapeutic biocomputation in mammalian cells

Shao Jiawei,Li Shichao,Qiu Xinyuan,Jiang Jian,Zhang Lihang,Wang Pengli,Si Yaqing,Wu Yuhang,He Minghui,Xiong Qiqi,Zhao Liuqi,Li Yilin,Fan Yuxuan,Viviani Mirta,Fu Yu,Wu Chaohua,Gao Ting,Zhu Lingyun,Fussenegger Martin,Wang Hui,Xie Mingqi

Here, we present a gene regulation strategy enabling programmable control over eukaryotic translational initiation. By excising the natural poly-adenylation (poly-A) signal of target genes and replacing it with a synthetic control region harboring RNA-binding protein (RBP)-specific aptamers, cap-dependent translation is rendered exclusively dependent on synthetic translation initiation factors (STIFs) containing different RBPs engineered to conditionally associate with different eIF4F-binding proteins (eIFBPs). This modular design framework facilitates the engineering of various gene switches and intracellular sensors responding to many user-defined trigger signals of interest, demonstrating tightly controlled, rapid and reversible regulation of transgene expression in mammalian cells as well as compatibility with various clinically applicable delivery routes of in vivo gene therapy. Therapeutic efficacy was demonstrated in two animal models. To exemplify disease treatments that require on-demand drug secretion, we show that a custom-designed gene switch triggered by the FDA-approved drug grazoprevir can effectively control insulin expression and restore glucose homeostasis in diabetic mice. For diseases that require instantaneous sense-and-response treatment programs, we create highly specific sensors for various subcellularly (mis)localized protein markers (such as cancer-related fusion proteins) and show that translation-based protein sensors can be used either alone or in combination with other cell-state classification strategies to create therapeutic biocomputers driving self-sufficient elimination of tumor cells in mice. This design strategy demonstrates unprecedented flexibility for translational regulation and could form the basis for a novel class of programmable gene therapies in vivo.
