Pt–Se Hybrid Nanozymes with Potent Catalytic Activities to Scavenge ROS/RONS and Regulate Macrophage Polarization for Osteoarthritis Therapy

Hong Wei,Hongjun Huang,Haoqiang He,Yuanming Xiao,Lu Chun,Zhiqiang Jin,Hanyang Li,Li Zheng,Jinmin Zhao,Zainen Qin

The activation of pro-inflammatory M1-type macrophages by overexpression of reactive oxygen species (ROS) and reactive nitrogen species (RONS) in synovial membranes contributes to osteoarthritis (OA) progression and cartilage matrix degradation. Here, combing Pt and Se with potent catalytic activities, we developed a hybrid Pt–Se nanozymes as ROS and RONS scavengers to exert synergistic effects for OA therapy. As a result, Pt–Se nanozymes exhibited efficient scavenging effect on ROS and RONS levels, leading to repolarization of M1-type macrophages. Furthermore, the polarization of synovial macrophages to the M2 phenotype inhibited the expression of pro-inflammatory factors and salvaged mitochondrial function in arthritic chondrocytes. In vivo results also suggest that Pt–Se nanozymes effectively suppress the early progression of OA with an Osteoarthritis Research International Association score reduction of 68.21% and 82.66% for 4 and 8 weeks, respectively. In conclusion, this study provides a promising strategy to regulate inflammatory responses by macrophage repolarization processes for OA therapeutic.

Melanin-Deferoxamine Nanoparticles Targeting Ferroptosis Mitigate Acute Kidney Injury via RONS Scavenging and Iron Ion Chelation

Jinghua Sun,Hao Shen,Jie Dong,Jin Zhang,Tao Yue,Ruiping Zhang

Rhabdomyolysis (RM)-induced acute kidney injury (AKI) involves the release of large amounts of iron ions from excess myoglobin in the kidneys, which mediates the overproduction of reactive species with the onset of iron overload via the Fenton reaction, thus inducing ferroptosis and leading to renal dysfunction. Unfortunately, there are no effective treatments for AKI other than supportive care. Herein, we developed a multifunctional nanoplatform (MPD) by covalently bonding melanin nanoparticles (MP NPs) to deferoxamine. The nanoplatform has good dispersion and physiological stability, excellent chelating performance to iron ions, and broad-spectrum reactive species scavenging activity. Furthermore, cellular experiments showed that the NPs possessed high biocompatibility, antiapoptotic activity, antioxidant properties, and strong scavenging capacity of Fe2+ to mitigate iron overload, protecting the intracellular mitochondria from oxidative stress. Meanwhile, the intrinsic photoacoustic imaging capability of melanin allows the real-time monitoring of MPD NPs' target uptake and metabolic behavior in healthy and AKI mice. Most importantly, MPD NPs led to downregulation of the antioxidant pathway by targeting ferroptosis, thus effectively rescuing renal function in vivo, mitigating oxidative stress and inflammatory responses, and inhibiting renal tubular cell apoptosis. The nanoplatform offers a novel therapeutic strategy for RM-induced AKI.

Fluidic-manipulation-enabled multiplexed dose delivery of RONS by a CAP chip for dose optimization enhancement

Wang Fang,Zhou Liangyu,Guo Wei,Lin Haisong,Zhang Ruotong,Kuang Shaolong,Liu Yuan,Fan Xiaoxue,Chan Yau Kei,Deng Hui,Shum Ho Cheung

The plasma-derived reactive oxygen and nitrogen species (RONS) enable cold atmospheric plasma (CAP) to combat cancer and infectious wounds. Achieving therapeutic outcomes with CAP necessitates precise treatment doses. Current CAP devices are constrained by their capability of delivering a single dose to a single sample, limiting dose optimization. We propose a novel “one exposure, multiple-dose delivery” strategy by programming gas flows. This approach facilitates efficient screening of optimal CAP dose by distributing feed gas through boundary-conditioned transport channels to generate multiple, flux-varied gas streams, which ignite plasmas with diverse chemical compositions and dose gradients across samples. Our developed demonstration device, capable of administering three doses to sixteen samples, significantly reduces experimental complexity, particularly when handling large candidate doses or samples for treatment. Leveraging multiplexed treatment, we capably optimize the CAP dose to effectively eradicate the liver cancer cell line of Huh7 and bacteria of S. aureus within one exposure. Furthermore, we find manipulating gas flow velocities allows targeted generation of short-lived species. This approach disentangles the roles of short-lived and long-lived RONS in therapeutic applications, offering critical insights into their bio-functional mechanisms. The concept of multiplexed dose treatment with fluidic manipulation promises to catalyze the development of high-efficiency CAP devices and advance research in CAP-based therapies.

Tungsten disulfide atomic crystals with RONS scavenging and liver targeting capabilities for acetaminophen-induced acute liver injury therapy

Xiao Ziwen,Liu Yu,Tao Zhenchao,Zhang Yu,Chen Qian,Miao Zhaohua,Zha Zhengbao,Ma Yan,Wang Hua,Gong Deyan

The ongoing progression of acetaminophen (APAP)-induced liver injury (AILI) resulting from excessive APAP intake can lead to acute liver failure (ALF). This process involves an imbalanced antioxidant system, upregulated inflammatory responses, and excessive accumulation of reactive oxygen and nitrogen species (RONS), which ultimately trigger apoptosis. In the present study, tannic acid-capped tungsten disulfide nanosheets (WS 2 @TA NSs), which are atomic crystals with a 2D structure, a high specific surface area, and abundant phenolic groups, were synthesized using a liquid-phase exfoliation method. WS 2 @TA NSs were investigated as highly effective anti-inflammatory nanomedicines for targeted enrichment therapy of AILI lesions. Enriched WS 2 @TA NSs in the liver were able to scavenge excess RONS, exhibiting potent antioxidant properties. Importantly, in addition of oxidative stress levels reducing, bioactive WS 2 @TA NSs, which are rich in variable valence states, were able to modulate multiple biosignaling pathways including Nrf2-Keap1, NF-κB, and apoptosis in advanced-stage AILI mice model, thereby enhancing liver tolerance to AILI. The present study reveals the clinical potential of WS 2 @TA NSs atomic crystals in targeted enrichment therapy for late-stage AILI lesions. Graphical abstract

Erythrocyte membrane camouflaged celastrol and bilirubin self-assembly for rheumatoid arthritis immunotherapy based on STING inhibition and RONS clearance

Su Yanguo,Chen Rui,Wang Bingjie,Wang Teng,Tao Jiaojiao,Diao Qijie,Jiang Tianze,Zhao Xia

Activation of cGAS-STING signaling pathway and accumulation of reactive oxygen and nitrogen species (RONS) are important issues facing the treatment of rheumatoid arthritis (RA). Here, we report a biomimetic nano-Chinese medicine (HA-RM-Cel-BR) for RA immunotherapy based on STING inhibition of celastrol (Cel) and RONS clearance of bilirubin (BR). HA-RM-Cel-BR is constructed by the carrier-free self-assembly of active ingredients Cel and BR from traditional Chinese medicine, and then camouflaged by hyaluronic acid (HA)-modified red blood cell membranes (RM). HA-RM-Cel-BR prolongs circulation time through RM camouflage, targets inflamed joints by HA modification, and remodels the joint immune microenvironment by STING inhibition and RONS clearance. More importantly, HA-RM-Cel-BR shows excellent therapeutic effects on RA rat model, and significantly reduces hepatotoxicity associated with Cel. Our work provides a new strategy for RA immunotherapy with traditional Chinese medicine ingredients.

The H2S-releasing multifunctional composite ionic liquid facilitates topical melittin delivery for psoriasis treatment through RONS scavenging and inflammation inhibition

Meng Zeng,Ping Deng,Qin Li,Qi Tang,Jixiang Li,Liangke Zhang

Psoriasis, an immune-mediated disease, commonly utilizes topical drug administration in clinical treatments, yet the thickening of skin presents a challenge to conventional topical drug delivery methods. In this study, we have successfully developed a multifunctional composite ionic liquid (CIL) that enhances the local delivery of melittin (CIL@Mel) by modifying the fluidity of the stratum corneum and the structure of keratin. In addition, CIL@Mel can transport H 2 S to the psoriatic microenvironment, effectively regulating the redox imbalance within it. The in vivo experimental results demonstrated that CIL@Mel effectively mitigates the progression of psoriasis, provides relief from oxidative stress, reduces the production of inflammatory factors, and promotes the repolarization of macrophages. Computational simulations and experimental validation revealed that CIL@Mel exerts its effects by inhibiting the expression of EGFR, STAT3, IL-6, SRC, and HSP90AA1. In conclusion, this study unveils the transdermal promotion mechanism of CIL@Mel and validates its efficacy in psoriasis.

Shaofang Zhang,Ying Liu,Si Sun,Junying Wang,Qifeng Li,Ruijuan Yan,Yalong Gao,Haile Liu,Shuangjie Liu,Wenting Hao,Haitao Dai,Changlong Liu,Yuanming Sun,Wei Long,Xiaoyu Mu,Xiao-Dong Zhang

Traumatic brain injury (TBI) is a sudden injury to the brain, accompanied by the production of large amounts of reactive oxygen and nitrogen species (RONS) and acute neuroinflammation responses. Although traditional pharmacotherapy can effectively decrease the immune response of neuron cells via scavenging free radicals, it always involves in short reaction time as well as rigorous clinical trial. Therefore, a noninvasive topical treatment method that effectively eliminates free radicals still needs further investigation. Methods: In this study, a type of catalytic patch based on nanozymes with the excellent multienzyme-like activity is designed for noninvasive treatment of TBI. The enzyme-like activity, free radical scavenging ability and therapeutic efficacy of the designed catalytic patch were assessed in vitro and in vivo . The structural composition was characterized by the X-ray diffraction, X-ray photoelectron spectroscopy and high-resolution transmission electron microscopy technology. Results: Herein, the prepared Cr-doped CeO 2 (Cr/CeO 2 ) nanozyme increases the reduced Ce 3+ states, resulting in its enzyme-like activity 3-5 times higher than undoped CeO 2 . Furthermore, Cr/CeO 2 nanozyme can improve the survival rate of LPS induced neuron cells via decreasing excessive RONS. The in vivo experiments show the Cr/CeO 2 nanozyme can promote wound healing and reduce neuroinflammation of mice following brain trauma. The catalytic patch based on nanozyme provides a noninvasive topical treatment route for TBI as well as other traumas diseases. Conclusions: The catalytic patch based on nanozyme provides a noninvasive topical treatment route for TBI as well as other traumas diseases.

W-GA nanodots restore intestinal barrier functions by regulating flora disturbance and relieving excessive oxidative stress to alleviate colitis

Qingrong Li,Cong Zhang,Mengmei Zhu,Jie shan,Haisheng Qian,Yan Ma,Xianwen Wang

Inflammatory bowel disease (IBD) may arise due to disruption of mucosal barriers as a result of dysregulation of the intestinal flora and excessive oxidative stress. The creation of nanomaterials with only microbiota-regulating effects often leads to inadequate therapeutic outcomes caused by the disruption of a healthy microbial balance and the emergence of tissue harm caused by excessive oxidative stress. This report describes the multifunctional activity of ultrasmall W-GA nanodots, which can precisely regulate the intestinal microbiome by inhibiting the abnormal expansion of Enterobacteriaceae during colitis and alleviating the damage caused by oxidative stress to the reconstructive microflora, ultimately restoring intestinal barrier function. W-GA nanodots have been synthesized through a simple coordination reaction and can be dispersed in various solvents in vitro , demonstrating favorable safety profiles in cells, significant clearance of reactive oxygen and nitrogen species (RONS), and increased cell survival in models of oxidative stress induced by hydrogen peroxide (H 2 O 2 ). Through oral or intravenous administration, the W-GA nanodots were shown to be highly safe when tested in vivo , and they effectively reduced colon damage in mice with DSS-induced colitis by restoring the integrity of the intestinal barrier. W-GA nanodots have enabled the integration of microflora reprogramming and RONS clearance, creating a potent therapeutic strategy for treating gut inflammation. Consequently, the development of W-GA nanodots represents a promising strategy for enhancing the formation and preservation of the intestinal barrier to treat IBDs by suppressing the growth of Enterobacteriaceae , a type of facultative anaerobic bacterium, and facilitating the effective removal of RONS. Ultimately, this leads to the restoration of the intestinal barrier's functionality. Statement of Significance An increasing number of nanoparticles are under development for treating inflammatory bowel disease. Although they can alleviate inflammation symptoms by regulating reactive oxygen and nitrogen species (RONS) and microbiota, their understanding of the mechanism behind microbiota regulation is limited. This study synthesized W-GA nanodots using a straightforward one-pot synthesis method. Simple synthesis holds significant promise for clinical applications, as it encompasses multiple nanoenzyme functions and also exhibits Enterobacteriaceae inhibitory properties. Thus, it contributes to ameliorating the current medical landscape of inflammatory bowel disease.

Hydrogen Therapy and Lipopolysaccharide Trapping with MgB2@Inulin Gel for Reversing Colitis and Its Associated Psychiatric Complications via the Gut–Brain Axis

Yaoyu Hu,Pengfei Zhu,Junwei Deng,Yi Yu,Qian Chen,Haitao Wu,Zhengbao Zha,Hua Wang,Yan Ma

Individuals diagnosed with inflammatory bowel disease (IBD) frequently encounter dysbiosis of the intestinal microbiota and are predisposed to psychiatric comorbidities, such as depression and anxiety. The development of oral therapeutics that concurrently attenuate intestinal inflammation, modulate gut microbiota, and ameliorate psychiatric symptoms is imperative for effective IBD management. This study introduced an orally administered Inulin gel incorporating magnesium diboride nanosheets (MgB2@Inulin gel). The hydrolysis of MgB2 nanosheets could generate reductive hydrogen gas (H2), which is critical for scavenging excess reactive oxygen/nitrogen species (RONS), and boron ions that neutralize harmful lipopolysaccharides (LPS). Prebiotic Inulin could effectively protect MgB2 nanosheets from gastric acid degradation, ensuring their persistence in the intestinal tract, where it modulated the gut microbiota and preserved intestinal barrier integrity. Notably, MgB2@Inulin gel markedly alleviated anxiety and depressive behaviors in the model of colitis by regulating the gut–brain axis. Thus, this work underscores the pivotal role of the gut–brain axis in the therapeutic approach to IBD and its associated psychiatric manifestations, proposing a straightforward, safe, and efficacious oral hydrogen treatment strategy for IBD and concurrent mental health disorders.

Carbon dot targeting to nitrogen signaling molecules for inhibiting neuronal death

Lufei Ouyang,Xiaoyu Mu,Junying Wang,Qifeng Li,Yalong Gao,Haile Liu,Si Sun,Qinjuan Ren,Ruijuan Yan,Jingya Wang,Qiang Liu,Yuanming Sun,Changlong Liu,Hua He,Wei Long,Xiao-Dong Zhang

Free radical-induced oxidative damage and nitrosative stress have been identified as key factors in neuroinflammation responses after traumatic brain injury (TBI), with which reactive oxygen and nitrogen species (RONS), especially nitrogen signaling molecules, are strongly associated. Here, we prepared ultrasmall carbon dot (CD) by using a simple and facile method. In vitro assessment experiments show that the antioxidative CD exhibits an ultrahigh target-scavenging effect for nitrogen signaling molecules, especially the highly reactive ˙NO and ONOO−. However, CD can only partially eliminate conventional oxygen radials such as O2˙− and ˙OH, indicating CD has a preference for RNS modulation. Moreover, in vitro cell experiments and in vivo mice experiments reveal that CD can reduce the reactive oxygen species (ROS) level and lipid peroxidation, enhance superoxide dismutase (SOD) activity and GSSG level, and further improve the survival rate of neuron cells and TBI mice. These results declare that antioxidative CD could serve as an effective therapeutic for TBI.

Multifunctional two-dimensional Bi2Se3 nanodiscs for anti-inflammatory therapy of inflammatory bowel diseases

Cong Zhang,Qingrong Li,Jie Shan,Jianghao Xing,Xiaoyan Liu,Yan Ma,Haisheng Qian,Xulin Chen,Xianwen Wang,Lian-Ming Wu,Yue Yu

The overexpression of reactive oxygen and nitrogen species (RONS) in the colonic mucosa destroys the mucosa and its barrier, accelerating the occurrence of inflammatory bowel disease (IBD). The elimination of RONS from the inflammatory colon has proven effective in alleviating IBD. Although many nanoantioxidants have been developed, preparing robust and efficient nano-antioxidants remains challenging. Herein, by modifying bismuth selenide (Bi 2 Se 3 ) nanodiscs with polyvinylpyrrolidone (PVP), a multifunctional nanozyme based on 2D nanomaterials was developed for the treatment of IBD. By eliminating multiple RONS, such as hydroxyl radicals ( OH), superoxide anions (O 2 − ), nitric oxide (NO), and Bi 2 Se 3 nanodiscs enhanced cellular survival after H 2 O 2 stimulation. As evidenced by colonic injury, reduced body weight, spleen index, and proinflammatory cytokine levels in mice, RONS clearance alleviated intestinal inflammation in a prevention and delay model of acute colitis. 16S rDNA amplicon sequencing reveals that Bi 2 Se 3 nanodiscs had the potential to regulate intestinal flora , increase the proportion of Firmicutes to Bacteroidetes , inhibit Proteobacteria bacteria, and restore intestinal homeostasis . This study highlights the use of Bi 2 Se 3 nanodiscs with excellent biocompatibility, multienzyme functionality, and RONS scavenging ability as treatments for IBD without apparent adverse effects. Statement of Significance RONS were efficiently scavenged by Bi 2 Se 3 nanodiscs. Bi 2 Se 3 nanodiscs could be as a promising and potentially safe theraeputic agent for IBD. The gut microbiota could be modulated by Bi 2 Se 3 nanodiscs.
