High intracellular reactive oxygen species (ROS) level is characteristic of cancer cells and could act as a target for the efficient targeted drug delivery for cancer treatment. Consequently, biomaterials that react to excessive levels of ROS are essential for biomedical applications. In this study, a novel ROS-responsive polymer based on D-α-Tocopheryl polyethylene glycol 1000 succinate (TPGS) and poly (β-thioester) (TPGS-PBTE) was synthesized for targeted delivery of the first-line antineoplastic drug, paclitaxel (PTX). The resultant TPGS-PBTE NPs showed good ROS-responsive capability in size change and drug release. Compared to PTX, PTX-loaded nanoparticles (PTX@TPGS-PBTE NPs) showed enhanced cytotoxicity and higher level of apoptosis toward squamous cell carcinoma (SCC-7) cells. Tumor-targeted delivery of the NPs was also observed, especially after being modified with a tumor-targeting peptide, cRGD. Enhanced tumor growth inhibition was also observed in head and neck cancer SCC-7 murine models. In summary, PTX@TPGS-PBTE NPs can achieve good therapeutic effects of PTX against head and neck cancer both in vitro and in vivo, especially when modified by cRGD for active targeting, which enriched the application of ROS responsive system utilized in the delivery of anticancer drugs.

As a clinically prevalent condition, dysphagia substantially compromises the quality of life of patients and imposes substantial economic burdens on healthcare systems. In this study, a reactive oxygen species (ROS)-responsive cationic liposome and inulin hydrogel composite system was developed to deliver blueberry anthocyanins (ACNs). Mitochondria-targeting ACN-loaded liposomes with triphenylphosphonium (TPP) (TPP-Lip@ACNs) were prepared using the thin-film hydration method, and a hydrogel with 60 % inulin concentration, International Dysphagia Diet Standardisation Initiative (IDDSI) Level 5, was screened as the optimal carrier. Transmission electron microscopy (TEM) and dynamic light scattering (DLS) confirmed that TPP-Lip@ACNs had a particle size of 101.5 nm, with structural disintegration under H₂O₂ stimulation due to the cleavage of thioketal (TK) bonds, demonstrating ROS-responsive characteristics. Scanning electron microscopy (SEM) revealed that the inulin hydrogel had a honeycomb-like porous structure, encapsulating liposomes and slowing down drug release, reducing the 12-h cumulative release rate from 90.20 % (free liposomes) to 65.67 %. The IDDSI testing classified the hydrogel as Level 5 (pureed) for safe swallowing. Rheological analysis highlighted the superior viscoelastic and self-healing capabilities of the system. In vitro digestion simulations confirmed the stability of the system in simulated gastric fluid (SGF) and intestinal fluid (SIF), with a triggered controlled release in simulated colon fluid (SCF). Cytotoxicity and hemocompatibility tests showed no significant impact on Caco-2 cell viability and hemolysis rates of <5 %, indicating high biocompatibility. These findings provide a novel strategy for designing functional pigment delivery systems and developing dysphagia-oriented diets.

Alzheimer's disease (AD) has displayed complex pathological features, in which oxidative stress and ferroptosis play important roles. The development of collaborative treatment methods aimed at multiple pathological factors is particularly important for effective alleviation of AD. In our research, a multifunctional delivery system (CART-CeO 2 )@HA was developed for the co-delivery of neuroprotective Cocaine- and amphetamine-regulated transcript (CART) peptides and antioxidant CeO 2 nanozymes encapsulated within an ROS-responsive nanogel. Under the stimulation of Aβ, (CART-CeO 2 )@HA exhibited effective relief for cell apoptosis, mitochondrial dysfunction and synaptic loss on account of ROS removal and ferrous ions chelation. With sufficient accumulation and sustained drug release in the brain of APP/PS1 mice, (CART-CeO 2 )@HA significantly reduced the levels of oxidative stress and ferroptosis, which facilitate the reduction of Aβ deposition and the repair of neuronal damage, jointly rescuing the cognitive impairment in APP/PS1 mice. These results verified the clinical potential of the multi-target delivery system in AD synergistic therapy.

Traditional photodynamic therapy (PDT) suffers from poor tissue penetration due to using external light as excitation source. Cerenkov radiation (CR) from radionuclides can serve as an internal light source to activate photosensitizers inside lesion tissues. Using CR instead of external light in PDT is thought to be an effective way to overcome the shortcoming of traditional PDT. However, the weakness of CR emission limits the outcomes of CR-induced PDT (CR-PDT). We anticipated that CR-induced combined therapy could effectively enhance the therapeutic outcome of CR-PDT. Herein, a ROS-responsive Chemo-PDT agent ( TTC ) was synthesized by conjugation of tetraphenylporphyrin (photosensitizer) with camptothecin (chemotherapeutic drug) through a ROS-cleavable thioketal linker. Under activation by CR emission from gallium-68 ( 68 Ga), the porphyrin moiety could generate ROS , which then cleaves the thioketal linker to induce the subsequent release of CPT. Using [ 68 Ga]Ga-NOTA-Nb109 as an intracellular CR emitter, in comparison with its non-cleavable analogue ( TUC ) only displaying phototoxicity, TTC exhibited much higher cytotoxicity toward A375-hPD-L1 cells due to its combined effect of PDT and chemotherapy. Our work presents a new strategy to use ROS-responsive Chemo-PDT agent to improve the outcomes of CR-PDT.

Due to the high oxidative stress of the tumor microenvironment, more and more researchers have been devoted to reactive oxygen species (ROS)-responsive nanodrug delivery systems for anticancer therapy. Herein, a ROS-responsive moiety, thioacetal, was synthesized, and cinnamaldehyde (CA) was introduced in the polymer chain to trigger the generation of ROS to expect the enhancement of the ROS-responsive effect. The poly(ester-thioacetal) mPEG2k-b-(NTA-HD)12 polymer, its self-assembled micelles, and the ROS-responsive behavior were characterized by 1H NMR and DLS. The anticancer drug doxorubicin (DOX) was adopted to prepare DOX-loaded poly(ester-thioacetal) micelles. The intracellular ROS detection indicated that the mPEG2k-b-(NTA-HD)12 polymer could degrade via the high concentration of ROS in cancer cells, and the released CA stimulated mitochondria to regenerate additional ROS. The flow cytometry results indicated that the ROS-responsive polymeric micelles showed faster cellular uptake compared to the control mPEG2k-b-PCL5k micelles. The ROS responsive DOX/mPEG2k-b-(NTA-HD)12 micelles exhibited much better anticancer efficiency on both 4T1 and HeLa cancer cells than DOX/mPEG2k-b-PCL5k micelles.

Burns catastrophically compromise the skin barrier, driving impaired healing, infection susceptibility, and regenerative failure, primarily through dysregulated inflammation, immune dysfunction, and overwhelming oxidative stress. While reactive oxygen species (ROS) mediate essential host defense, their pathological excess necessitates targeted modulation for burn repair. We engineered a ROS-responsive nanoassembly (PTC@Cu NPs) for co-delivering chlorogenic acid (CGA) and copper ions (Cu 2+ ). Synthesizing the amphiphilic polymer mPEG-TK-CGA (PTC) via a ROS-cleavable thioketal linker enabled self-assembly with Cu 2+ into NPs. This system achieves triggered release specifically within high-ROS burn sites. CGA and Cu 2+ act synergistically to potently scavenge ROS, polarize macrophages to a pro-regenerative M2 phenotype, and thereby ameliorate oxidative damage, infection, and healing deficits in deep partial-thickness burns. In vitro and in vivo studies demonstrate PTC@Cu NPs significantly suppress apoptosis, enhance cell migration, exert potent antibacterial effects, and markedly accelerate healing. These comprehensive therapeutic benefits are mechanistically underpinned by dual ROS reduction and macrophage M2 polarization. Collectively, this platform presents a promising strategy for infected deep burn wounds by simultaneously targeting oxidative damage, microbial infection, and impaired tissue repair.

In the context of prodrug nanomedicines for cancer therapy, one of the great challenges is the slow and variable release of the parent drug in tumors. Recently, many smart redox-sensitive nanocarriers have been developed to address this problem. However, due to significant tumor heterogeneity, some redox-sensitive nanomedicines still show poor selectivity in drug release. Herein, we report the design and synthesis of a ROS-triggered prodrug nanoplatform fabricated with oxidation-responsive cabazitaxel (CTX) prodrugs for synergistic chemo-photodynamic therapy, thioether-/selenoether-linked conjugates of CTX and oleic acid (OA). These prodrugs can be readily self-assembled into nanoparticles, with pyropheophorbide a (PPa) co-encapsulated into the prodrug-nanosystem for combination therapy. The dual-source ROS-responsive prodrug nanosystems selectively and rapidly release CTX not only in response to endogenous ROS overproduced in tumor cells, but also to exogenous PPa-generated ROS under laser irradiation. Moreover, the selenium-containing linkage demonstrates significant advantages over a sulfur-containing linkage in terms of ROS-triggered drug release and cytotoxicity. The prepared prodrug-nanosystems significantly prolong the systemic circulation and tumor distribution of both CTX and PPa, thereby demonstrating synergistic chemo-photodynamic therapy in vivo. All these drug delivery advantages render the nanosystem extremely promising for cancer treatment.

Ischemic stroke is a leading cause of mortality and disability, with ischemia-reperfusion injury exacerbating neuronal damage and neuroinflammation due to pathologically activated reactive oxygen species (ROS) production in the ischemic penumbra. Edaravone, a free radical scavenger, is approved for the clinical treatment of ischemic stroke. However, edaravone directly decreases ROS even at low concentrations, which may inhibit the physiological effects of ROS and largely limit the clinical application of edaravone. To overcome this challenge, a ROS-response nanoparticle system, edaravone-loaded DEX-CDIPBE (DEX-CDIPBE-ED), is developed for the selectively releasing edaravone in the ischemic penumbra during reperfusion, mitigating oxidative stress while minimizing off-target effects. DEX-CDIPBE-ED produces potent antioxidant and neuroprotective effects in neuronal cells mimicking ischemic and/or reperfusion phases of ischemic stroke. DEX-CDIPBE-ED further prevents behavioral deficits, reduces oxidative stress, and inhibites neuroinflammation with a higher potency than edaravone in the ischemic penumbra in middle cerebral artery occlusion/reperfusion (MCAO/R)-treated rats. These findings highlight the potential of DEX-CDIPBE-ED as a pathological activated antioxidative therapy for ischemic stroke, offering enhanced neuroprotection and reduced systemic toxicity.

The clinical indications reveal that traumatic brain injury (TBI) accelerates the process of limb bone regeneration. However, the mechanisms underlying this phenomenon remain ambiguous. In this inquiry, the activation of endogenous neural stem cells in the hippocampus post TBI led to the secretion of extracellular vesicles (NSC-EV) that targeted bone marrow mesenchymal stem cells, ultimately promoting bone healing. The enrichment of miR-9-5p in NSC-EV facilitated osteogenesis through the activation of the MAP3K3 pathway. We also present a novel hydrogel formulation for clinical application of NSC-EVs by means of ROS-responsive EV delivery. The hydrogel was cross-linked with EVs to create a water-based gel encapsulation, which was applied to a 3D-printed β-TCP scaffold for controlled long-term release that corresponded to those of physiological bone tissue. Our investigation represents the inaugural usage of EVs derived from neural stem cells in bone tissue engineering. These EVs were found to encourage information exchange between the central nervous system and bone tissue regeneration. Our composite scaffold demonstrated impressive bone regeneration qualities in rats with cranial defects, highlighting its potential for pioneering approaches in bone tissue engineering and innovative medical technologies for clinical applications.

Rationale: As a potentially life-threatening disorder, cerebral ischemia-reperfusion (I/R) injury is associated with significantly high mortality, especially the irreversible brain tissue damage associated with increased reactive oxygen radical production and excessive inflammation. Currently, the insufficiency of targeted drug delivery and “on-demand” drug release remain the greatest challenges for cerebral I/R injury therapy. Bioengineered cell membrane-based nanotherapeutics mimic and enhance natural membrane functions and represent a potentially promising approach, relying on selective interactions between receptors and chemokines and increase nanomedicine delivery efficiency into the target tissues. Methods: We employed a systematic method to synthesize biomimetic smart nanoparticles. The CXCR4-overexpressing primary mouse thoracic aorta endothelial cell (PMTAEC) membranes and RAPA@HOP were extruded through a 200 nm polycarbonate porous membrane using a mini-extruder to harvest the RAPA@BMHOP. The bioengineered CXCR4-overexpressing cell membrane-functionalized ROS-responsive nanotherapeutics, loaded with rapamycin (RAPA), were fabricated to enhance the targeted delivery to lesions with pathological overexpression of SDF-1. Results: RAPA@BMHOP exhibited a three-fold higher rate of target delivery efficacy via the CXCR4/SDF-1 axis than its non-targeting counterpart in an in vivo model. Additionally, in response to the excessive pathological ROS, nanotherapeutics could be degraded to promote “on-demand” cargo release and balance the ROS level by p -hydroxy-benzyl alcohol degradation, thereby scavenging excessive ROS and suppressing the free radical-induced focal damage and local inflammation. Also, the stealth effect of cell membrane coating functionalization on the surface resulted in extended circulation time and high stability of nanoparticles. Conclusion: The biomimetic smart nanotherapeutics with active targeting, developed in this study, significantly improved the therapeutic efficacy and biosafety profiles. Thus, these nanoparticles could be a candidate for efficient therapy of cerebral I/R injury.
