Enhanced photodynamic therapy efficacy of Ni-doped/oxygen vacancy double-defect Ni-ZnO@C photosensitizer in bacteria-infected wounds based on ROS damage and ATP synthesis inhibition

Rui Zhang,Zhiling Chen,Yi Li,Delun Chen,Tao Wang,Bingrong Wang,Qionglin Zhou,Shaowen Cheng,Dan Xu,Xiaohong Wang,Lina Niu,Jinchun Tu,Qiang Wu

The use of zinc oxide (ZnO) photosensitizers (PSs)-mediated photodynamic therapy (PDT) against bacterial wound infections is greatly restricted by diminished photocatalytic efficiency caused by the rapid recombination of photogenerated electrons and holes. In this work, ZnO PSs with a Ni-doped/oxygen vacancy and a protective carbon shell were successfully synthesized by calcinating a Ni-doping zeolitic imidazolate framework-8 precursor. The double-defect structure and the carbon-based substrates significantly promoted the efficiency of photogenerated electron–hole pair separation, meanwhile, the optimal proportion of 3% Ni doping endows it with the best photocatalytic performance as elucidated by photodegradation assays of methylene blue (MB) and density functional theory (DFT) calculations, reinforcing the generation of reactive oxygen species (ROS) by ZnO and showing obvious advantages in antibacterial properties. As the ROS damage that facilitates the photogenerated electron transfer underwent localized accumulation, the PSs disturbed the bacterial membrane integrity and caused bacterial ATP synthesis inhibition, further leading to bacterial lysis and promoting bacterial deaths. Additionally, the PSs showed outstanding efficacy in eradicating bacterial biofilms. Simultaneously, the significantly enhanced PDT antibacterial performance of the PSs in vivo could initiate wound tissue repair and trigger anti-inflammatory reactions by significantly regulating the expression levels of regeneration- and inflammation-related genes or proteins. Furthermore, the PSs consistently exhibited favorable compatibility both in vitro and in vivo . In summary, this study offers evidence of the remarkably efficient and biologically safe performance of Ni-ZnO@C PSs, with antibacterial properties advancing wound healing, both in controlled laboratory environments and living organisms, further underscoring their substantial potential for biomedical applications.

Fisetin Exerts Antioxidant and Neuroprotective Effects in Multiple Mutant hSOD1 Models of Amyotrophic Lateral Sclerosis by Activating ERK

T.H. Wang,S.Y. Wang,X.D. Wang,H.Q. Jiang,Y.Q. Yang,Y. Wang,J.L. Cheng,C.T. Zhang,W.W. Liang,H.L. Feng

Oxidative stress exhibits a central role in the course of amyotrophic lateral sclerosis (ALS), a progressive neurodegenerative disease commonly found to include a copper/zinc superoxide dismutase (SOD1) gene mutation. Fisetin, a natural antioxidant, has shown benefits in varied neurodegenerative diseases. The possible effect of fisetin in ALS has not been clarified as of yet. We investigated whether fisetin affected mutant hSOD1 ALS models. Three different hSOD1-related mutant models were used: Drosophila expressing mutant hSOD1 G85R , hSOD1 G93A NSC34 cells, and transgenic mice. Fisetin treatment provided neuroprotection as demonstrated by an improved survival rate, attenuated motor impairment, reduced ROS damage and regulated redox homeostasis compared with those in controls. Furthermore, fisetin increased the expression of phosphorylated ERK and upregulated antioxidant factors, which were reversed by MEK/ERK inhibition. Finally, fisetin reduced the levels of both mutant and wild-type hSOD1 in vivo and in vitro , as well as the levels of detergent-insoluble hSOD1 proteins. The results indicate that fisetin protects cells from ROS damage and improves the pathological behaviors caused by oxidative stress in disease models related to SOD1 gene mutations probably by activating ERK, thereby providing a potential treatment for ALS.

Rapid on-demand in-situ gelling and dissociation of PEG bottlebrush hydrogel via light-mediated grafting-through polymerization for full-thickness skin wound repair

Chong Chen,Siwei Bi,Changyuan He,Ruiqi Liu,Xueshan Zhao,Jialin Liu,Sheng Chen,Jun Gu,Bin Yan

Developing light-triggered in situ forming dynamical hydrogels with on-demand dissolvable and antioxidation capacities to promote wound healing is highly desired yet challenging. Herein, we prepared an in situ forming bottlebrush hydrogel with on-demand dissolution capacity from two thioctic acid (TA)-functionalized poly(ethylene glycol)s (PEGs) telechelic (TA-PEGs) prepolymers via a light-mediated grafting-through polymerization strategy. In response to light irradiation , TA-PEGs could undergo rapid ring-opening polymerization to generate a highly branched bottlebrush network with dynamic disulfide bonds and simultaneously load water-soluble curcumin complexes with excellent antioxidant activity , enabling fast in situ gelation in 3 min to achieve perfect coverage of wound sites with different shapes. Furthermore, the resulting bottlebrush hydrogel dressing can achieve facile and painless removal of the dressing via rapid on-demand dissolution of the hydrogel during dressing changes. Meanwhile, thanks to its dynamical feature of the disulfide-based network, the hydrogel possesses excellent self-healing ability and shape adaptability that can maintain the interiority of the dressing during application and thus prolong its service life. In vitro evaluation demonstrates that this PEG bottlebrush hydrogel possesses favorable hemocompatibility, cytocompatibility, and antioxidant capacity. More importantly, in vivo experiments confirmed that the hydrogel promotes angiogenesis, dermal collagen synthesis, and alleviates the excessive ROS damage, thus providing more beneficial conditions to accelerate full-thickness skin wound healing.
