A Curcumin-Decorated Nanozyme with ROS Scavenging and Anti-Inflammatory Properties for Neuroprotection

Feng Gao,Wenyu Liang,Qixin Chen,Bairu Chen,Yuchen Liu,Zhibo Liu,Xu Xu,Rongrong Zhu,Liming Cheng

Disordered reactive oxygen/nitrogen species are a common occurrence in various diseases, which usually cause cellular oxidative damage and inflammation. Despite the wide range of applications for biomimetic nanoparticles with antioxidant or anti-inflammatory properties, designs that seamlessly integrate these two abilities with a synergistic effect in a simple manner are seldom reported. In this study, we developed a novel PEI-Mn composite nanoparticle (PM NP) using a chelation method, and the curcumin was loaded onto PM NPs via metal–phenol coordination to form PEI-Mn@curcumin nanoparticles (PMC NPs). PMC NPs possessed excellent dispersibility and cytocompatibility, was engineered to serve as an effective nanozyme, and exhibited specific SOD-like and CAT-like activities. In addition, the incorporation of curcumin granted PMC NPs the ability to effectively suppress the expression of inflammatory cytokines in microglia induced by LPS. As curcumin also has antioxidant properties, it further amplified the synergistic efficiency of ROS scavenging. Significantly, PMC NPs effectively scavenged ROS triggered by H2O2in SIM-A9 microglia cells and Neuro-2a cells. PMC NPs also considerably mitigated DNA and lipid oxidation in Neuro-2a cells and demonstrated an increase in cell viability under various H2O2concentrations. These properties suggest that PMC NPs have significant potential in addressing excessive ROS and inflammation related to neural diseases. Keywords:nanozyme;ROS scavenging;anti-inflammatory;neuroprotective

Chitosan-Derived Carbon Quantum Dots with Dual ROS Scavenging and Anti-inflammatory Functionalities for Accelerated Wound Repair

Miaomiao Yao,Qi He,Yameng Tao,Xiaoyang Kang,Xinru Wu,Fangkai Shi,Yujia Wei,Jingjing Liu,Zhiyun Meng,Ruolan Gu,Hui Gan,Guifang Dou,Shuchen Liu,Yunbo Sun

Blue fluorescent carbon quantum dots (CDs) were synthesized by a one-step hydrothermal method using chitosan as the only raw material. This material overcomes multifunctional integration limitations in traditional dressings, achieving coordinated wound repair across all phases. During inflammation (0–3 days), CDs scavenge reactive oxygen species and suppress inflammatory factors (TNF-α↓49.63%, IL-6↓70.8%). In proliferation (3–7 days), they promote angiogenesis (CD31↑56.89%) and cell proliferation (Ki67↑74.3%). During remodeling (7–14 days), functional parallel collagen deposition occurs (collagen deposition 1.26× Celox group). The seamless “anti-inflammation/prohealing/remodeling” mechanism yielded 59% wound closure by day 3 postirradiation (vs Celox 37.34%). CDs synergistically activate Nrf2 while inhibiting NF-κB pathways, enabling combined anti-inflammatory/antioxidant regulation in irradiated environments. Nanoscale reconstruction retains chitosan’s hemostasis and avoids gene modification/vector loading via one-step synthesis (cost-reduced), with biosafety confirmed by acute toxicity tests. This study provides a prohealing nanomaterial and establishes a paradigm for traditional biomaterial redevelopment.

Targeting m6A demethylase FTO to heal diabetic wounds with ROS-scavenging nanocolloidal hydrogels

Xinyao Zheng,Shaohui Deng,Yuan Li,Zhipeng Luo,Ziqi Gan,Zhaoping Zheng,Rui Xu,Shan Xiao,Yuxiong Cai,Jianfu Meng,Li Li,Changxing Li,Xiaowen Xue,Wei Dai,Si Qin,Mengying Wang,Kang Zeng,Zecong Xiao,Laixin Xia

Chronic diabetic wounds are a prevalent and severe complication of diabetes, contributing to higher rates of limb amputations and mortality. N6-methyladenosine (m 6 A) is a common RNA modification that has been shown to regulate tissue repair and regeneration. However, whether targeting m 6 A could effectively improve chronic diabetic wound healing remains largely unknown. Here, we found a significant reduction in mRNA m 6 A methylation levels within human diabetic foot ulcers, and the expression level of fat mass and obesity-associated protein (FTO) was significantly increased. We identified that m 6 A modifies the RNA of matrix Metalloproteinase 9 (MMP9), a key factor in diabetic wound healing, to regulate its expression. Importantly, we developed a ROS-scavenging nanocolloidal hydrogel loaded with an FTO inhibitor to increase the m 6 A level of MMP9 RNA in wounds. The hydrogel can effectively accelerate wound healing and skin appendage regeneration in streptozotocin-induced type I diabetic rats at day 14 (approximately 98% compared to 76.98% in the control group) and type II diabetic db/db mice at day 20 (approximately 93% compared to 60% in the control group). Overall, our findings indicate that targeting m 6 A with ROS-scavenging hydrogel loaded with FTO inhibitor may be an effective therapeutic strategy for diabetic wound healing.

Application of Mg-MOF-loaded gelatin microspheres with osteogenic, angiogenic, and ROS scavenging capabilities in bone defect repair

Zhiheng Luo,Jianxiong Ma,Yan Wang,Yuhan Du,Yujie Liu,Wei Zhang,Yuxiao Lai,Xinlong Ma

The management of bone defects, particularly those with irregular geometries resulting from osteoporotic fractures, remains fraught with challenges. Microspheres have emerged as a promising vehicle for tissue engineering, distinguished by their controlled release, safety, and ease of application. Various bioactive components are integrated into microspheres to improve their performance. Metal-organic frameworks, formed from metal ions interconnected by organic ligands, are increasingly utilized in tissue engineering. Specifically, magnesium-based MOFs are notable for their broad applicability; Mg 2+ ions are instrumental in bone reconstruction and repair, facilitating osteogenesis, angiogenesis, antibacterial effects, and anti-inflammatory properties. Mg-MOF was synthesized using magnesium chloride and gallic acid, and it was incorporated into gelatin microspheres to create Gel@Mg-MOF composite microspheres. Leveraging gelatin's biocompatibility, controlled release, and biodegradability, the composites' biocompatibility was evaluated through toxicity and adhesion assays. Moreover, the osteogenic and angiogenic potentials of the Gel@Mg-MOF microspheres were assessed, alongside their capacity for ROS scavenging. Results suggest that controlled Mg 2+ release from Gel@Mg-MOF microspheres promotes osteogenic activity in RBMSCs and enhances angiogenic potential in HUVECs. Additionally, the gallic acid-containing composite microspheres exhibited antioxidative properties. Collectively, the findings suggest that Gel@Mg-MOF microspheres could provide effective support for bone defect repair, with potential for clinical deployment.

Bioactive glasses-based nanoenzymes composite double-network hydrogel with ROS scavenging for bone tissue engineering

Hao Deng,Xiaolu Chen,Wei Zhang,Hulin Yang,Xinyue Yang,Qiyi Zhang,Yonggang Yan

Reactive oxygen species (ROS) plays a crucial role in normal physiological activities. But hypoxia-induced oxidative stress leads to excessive accumulation of ROS in the microenvironment of bone defects, which could hinder tissue repair. Therefore, the regulation of ROS is essential for promoting bone repair. Herein, a mesoporous bioactive glass-based nanoenzyme (MBGNs@PDA@MnO 2 ) with the property to manage ROS was designed and prepared. The MBGNs@PDA@MnO 2 powders displayed excellent mineralization properties and antioxidation activity. The hydrogel with double-network structure were constructed and synthesized using oxidized chitosan (OCS) and dopamine-grafted gelatin (Gel-DA) by Schiff-base and oxidative coupling reaction, and then MBGNs@PDA@MnO 2 was incorporated into the hydrogel. Cell experiments demonstrated that the composite hydrogels could effectively reduce intracellular ROS level, and had good cytocompatibility and osteogenesis promotion capability. Hence, the composite hydrogels with ROS regulation had the potential to be a new strategy for the treatment of bone defects.

Reactive oxygen species-responsive and scavenging polyurethane nanoparticles for treatment of osteoarthritis in vivo

Haolan Zhang,Hao Xiong,Wajiha Ahmed,Yuejun Yao,Shuqin Wang,Cunyi Fan,Changyou Gao

Excessive reactive oxygen species (ROS) were closely associated with the progression of osteoarthritis (OA) in terms of symptoms and aggravation. To combine the advantages of ROS-stimuli responsiveness with scavenging ability, a novel polythioketal urethane (PTKU) was synthesized by simple and direct reaction between polythioketal (PTK) and diisocyanate. Apart from the excellent mechanical properties, the PTKU could be fabricated into various formulations, for example, nanoparticles (NPs) loaded with anti-inflammatory drug, dexamethasone. The [email protected] NPs were capable of scavenging several kinds of ROS, accompanying with the degradation of polymers. In vitro evaluation with macrophages revealed that the PTKU and [email protected] NPs had low cytotoxicity, and could suppress the intracellular ROS and expression of hypoxia inducible factor-1α (HIF-1α). Injection of the PTKU and [email protected] NPs to OA in vivo could significantly reduce the ROS level in articular cavity and alleviate destruction of oxidative stress, and resulted in a lower ratio of inflammatory M1 macrophages and a higher level of anti-inflammatory M2 macrophages. The synergistic effects of ROS-responsive drug delivery and ROS scavenging PTKU promoted significantly the therapeutic outcome of OA with a best score close to the normal cartilage, revealing the great promise of PTKU in treatment of OA in vivo .

Ruthenium Nanozyme@Magnesium silicate nanosheets encapsuled in microneedle patches promote repair of radiation-induced skin defects by remodeling pathological environment

Zhongyi Sun,Haibo Liu,Mengting Yin,Ningning Cheng,Wencheng Liu,Kangkang Zhao,Hua Zeng,Xuming Chen,Zijie Wang,Xuhui Zhou,Xinyu Zhao,Feng Chen

Radiotherapy induced skin defect (RISD) is a severe radiotherapy complication with persistent oxidative stress and recurrent excessive reactive oxygen species (ROS), impeding normal tissue repair processes. Nevertheless, the lack of a standardized animal model severely hinders the progress of related research work. We develop a novel strategy for repairing the RISD microenvironment, which combines initial ROS clearance, subsequent inhibition of ROS production and the repair of proliferation related cell pathways/functions. As a proof of concept, a composite microneedle (MN) patch comprising γ-polyglutamic acid as the base and ruthenium (Ru) clusters modified magnesium silicate nanosheets (MSR NSs) as the enzyme-like component is prepared. The Ru clusters have excellent ROS scavenging ability and help activate the peroxisome proliferators activated receptor signaling pathway confirmed by the sequencing analysis while the magnesium silicate is degraded under physiological conditions to release magnesium ions and silicate ions, enhancing cell proliferation, migration, and angiogenesis ability. The radiation induced skin defect animal model is established to evaluate the RISD repair efficacy of our MSR@MN patch in comparison with γPGA-MSR ointment and commercial product Orgotein. The results show that our MSR@MN patch effectively improves the pathological microenvironment of abnormal ROS accumulation, reduces inflammatory response and promotes mature angiogenesis and tissue remodeling.

Bimetallic Metal–Organic Framework for Mitigating Aseptic Osteolysis

Huajun Pan,Xinxin Miao,Jianjian Deng,Chongzhi Pan,Xigao Cheng,Xiaolei Wang

The disability rate of joint diseases can be reduced by the use of artificial joints, but joint loosening at a late state limits the lifespan and surgical efficacy of the joints. Wear particles can be recognized by macrophages and induce cells to produce reactive oxygen species (ROS) and inflammatory factors, causing persistent inflammation and decreased osteogenic activity, which ultimately leads to loosening of joint prostheses. Here, the platinum (Pt) nanozymes with excellent ROS scavenging and anti-inflammatory capabilities were encapsulated in zinc imidazolium zeolite framework-8 (ZIF-8), and then the osteogenic active element lanthanum (La) was introduced through ion exchange to finally construct a bimetallic metal–organic framework (Pt@ZIF-8@La). In vitro and in vivo experiments demonstrated that this multifunctional nanoplatform possessed the functions of efficient scavenging of ROS, immune regulation, and promotion of osteogenic differentiation. Meanwhile, the mechanism is explored that Pt@ZIF-8@La can also promote osteogenic mineralization by upregulating the ratio of the osteoprotegerin (OPG)/receptor activator of the NF-κB ligand (RANKL), which can achieve a synergistic therapeutic effect of immunomodulation and osteogenesis, thereby realizing the purpose of relieving aseptic osteolysis.

ROS-scavenging and antimicrobial polysaccharide hydrogel for methicillin-resistant staphylococcus aureus-infected diabetic wound healing

Wenshuai Liu,Litao Jia,Sichun Zhao,Yue Ma,Siyu Liu,Qian Wang,Xiaoyan Lv,Haiyue Jiang,Xia Liu

Bacterial infection can cause delay in wound healing, and reactive oxygen species (ROS) was accumulated in wounds, which threaten patients’ lives. Therefore, it needs to develop a multifunctional hydrogel for infected wound healing. Here, a multifunctional QAS-CS@GA-GM hydrogel with antibacterial and antioxidant property, established by the self-adaptive covalent bond, is prepared aiming to eradicate bacteria and eliminate ROS. It is observed that QAS-CS@GA-GM hydrogel displays remarkable ability of clearing ROS and killing bacteria in vitro. With its antibacterial and antioxidant activity, QAS-CS@GA-GM hydrogel significantly accelerates the MRSA-infected wound closure and reduces inflammatory, when QAS-CS@GA-GM hydrogel was applied to wound sites. Collectively, the research suggests that a self-adaptive QAS-CS@GA-GM hydrogel is a promising wound dressing as antibacterial and ROS-scavenging dressing for bacterial-infected wound.

Poly-antioxidants for enhanced anti-miR-155 delivery and synergistic therapy of metastatic breast cancer

Qingyan Zhang,Ying Huang,Ruoxi Yang,Junmei Mu,Zhanwei Zhou,Minjie Sun

Despite the great progress in the control of primary tumor growth, metastasis remains the major challenge of breast cancer therapy in clinics, which is highly related to the upregulation of reactive oxygen species (ROS) and overexpression of its relevant pro-survival miR-155 gene. Therefore, we fabricated a poly-antioxidant (FTP) to deliver anti-miR-155 for synergistic treatment of metastatic breast cancer by ROS scavenging and miR-155 inhibition. FTP was synthesized by the polymerization of fluorated-polyethyleneimine (FPEI) and antioxidants (TEMPOL), using a glutathione (GSH) responsive linker for controlled drug release. Notably, the poly-drug strategy could not only promote the tumoral accumulation of small molecular antioxidants but also enhance the transfection efficiency of anti-miR-155 owing to the hydrophobic property of TEMPOL. After synergistic treatment, the NF-κB pathway was significantly blocked, thereby generating strong anti-metastatic ability both in vitro and in vivo. The poly-antioxidant could be a new type of nanoplatform for highly efficient and safe miRNA delivery, which also provides a promising strategy for the synergistic treatment of metastatic breast cancer.

Crosstalk between miR-144/451 and Nrf2 during Recovery from Acute Hemolytic Anemia

Lei Yang,Sheng He,Ling Ling,Fangfang Wang,Lei Xu,Lei Fang,Fan Wu,Shuting Zhou,Fan Yang,Hongwei Wei,Duonan Yu

miR-144/451and nuclear factor (erythroid-derived 2)-like 2 (Nrf2) regulate two antioxidative systems that have been identified to maintain redox homeostasis in erythroid cells by removing excess reactive oxygen species (ROS). Whether these two genes coordinate to affect ROS scavenging and the anemic phenotype, or which gene is more important for recovery from acute anemia, has not been explored. To address these questions, we crossedmiR-144/451knockout (KO) andNrf2KO mice and examined the phenotype change in the animals as well as the ROS levels in erythroid cells either at baseline or under stress condition. Several discoveries were made in this study. First,Nrf2/miR-144/451double-KO mice unexpectedly exhibit similar anemic phenotypes asmiR-144/451single-KO mice during stable erythropoiesis, although compound mutations ofmiR-144/451andNrf2lead to higher ROS levels in erythrocytes than single gene mutations. Second,Nrf2/miR-144/451double-mutant mice exhibit more dramatic reticulocytosis thanmiR-144/451orNrf2single-KO mice during days 3 to 7 after inducing acute hemolytic anemia using phenylhydrazine (PHZ), indicating a synergistic effect ofmiR-144/451and Nrf2 on PHZ-induced stress erythropoiesis. However, the coordination does not persist during the whole recovery stage of PHZ-induced anemia; instead,Nrf2/miR-144/451double-KO mice follow a recovery pattern similar tomiR-144/451single-KO mice during the remaining period of erythropoiesis. Third, the complete recovery from PHZ-induced acute anemia inmiR-144/451KO mice takes longer than inNrf2KO mice. Our findings demonstrate that complicated crosstalk betweenmiR-144/451and Nrf2 does exist and the crosstalk of these two antioxidant systems is development-stage-dependent. Our findings also demonstrate thatmiRNAdeficiency could result in a more profound defect of erythropoiesis than dysfunctional transcription factors.Keywords:miR-144/451;Nrf2;hemolytic anemia;reactive oxygen species

Multi-enzyme mimetic iridium nanozymes-based thrombus microenvironment-modulated nanoplatform for enhanced thrombolytic therapy

Meng Huang,Yuda Zhu,Guang Xin,Yilan Wang,Fan Li,Shiyi Li,Yuman Dong,Kun Zhang,Lijuan Feng,Li Tang,Boli Zhang,Wen Huang

Intravenous injection of thrombolytic drugs is the most feasible strategy for the clinical treatment of thrombotic diseases, however, its applications are yet limited by narrow therapeutic index, short half-life, and hemorrhagic risk. Inspired by the excessive ROS and inflammatory factors at the thrombus site, we manufactured a thrombus microenvironment-modulated nanosystem , which achieved the targeted delivery of thrombolytic drugs and ROS scavenging agents. Ir monomers (Ir-PVP) with enzyme-mimetic activity were prepared and then wrapped with lumbrokinase (LK) by HA materials of thrombo-inflammatory affinity, forming multifunctional thrombolytic agents [email protected] [email protected] could prolong the biological half-life of LK and target the thrombus site to release LK with reduced bleeding risk. Moreover, Ir-NPs, as both CT imaging contrast agent and ROS scavenger, not only allowed us to observe the progression of thrombolytic treatment through CT imaging in real time, but also played an important role in elimination of excessive ROS at thrombus site. Taken together, the in vitro and in vivo experiments illustrated that [email protected] with CT function displayed an excellent efficiency in thrombolysis, anti-inflammation and ROS scavenging. Our work may encourage further exploration of thrombus microenvironment-modulated theranostic platforms.
