Real-time monitoring of Ralstonia solanacearum infection progress in tomato and Arabidopsis using bioluminescence imaging technology

Xu Cuihong,Zhong Lingkun,Huang Zeming,Li Chenying,Lian Jiazhang,Zheng Xuefang,Liang Yan

Background Ralstonia solanacearum, one of the most devastating bacterial plant pathogens, is the causal agent of bacterial wilt. Recently, several studies on resistance to bacterial wilt have been conducted using the Arabidopsis-R. solanacearum system. However, the progress of R. solanacearum infection in Arabidopsis is still unclear. Results We generated a bioluminescent R. solanacearum by expressing plasmid-based luxCDABE. Expression of luxCDABE did not alter the bacterial growth and pathogenicity. The light intensity of bioluminescent R. solanacearum was linearly related to bacterial concentrations from 104 to 108 CFU·mL−1. After root inoculation with bioluminescent R. solanacearum strain, light signals in tomato and Arabidopsis were found to be transported from roots to stems via the vasculature. Quantification of light intensity from the bioluminescent strain accurately reported the difference in disease resistance between Arabidopsis wild type and resistant mutants. Conclusion sBioluminescent R. solanacearum strain spatially and quantitatively measured bacterial growth in tomato and Arabidopsis, and offered a tool for the high-throughput study of R. solanacearum-Arabidopsis interaction in the future.

g-C3N4@CuO electrostatic self-assembly toward Ralstonia solanacearum: Insights from cytomembrane and motility disruption

Qiulan Xia,Maoyang Ran,Lihe Zhou,Zhongwei Liu,Lin Cai

BACKGROUND Ralstonia solanacearum , a notorious and refractory bacterial plant pathogen, threatens multiple vegetable crops and causes significant economic loss in agriculture. Long-term use of traditional medicines not only increases the problem of drug resistance, but also causes great environmental pollution. Therefore, there is an urgent need to develop new agents with high efficacy and low toxicity. RESULTS In this study, we have synthesized and characterized graphitic carbon nitride incorporated copper oxide composite (g-C 3 N 4 @CuO), which showed higher antimicrobial effect than graphitic carbon nitride nanosheets (g-C 3 N 4 nanosheets) and copper oxide nanoparticles (CuONPs). Ralstonia solanacearum exposed to g-C 3 N 4 @CuO exhibited higher levels of oxygen toxicity, cell membrane damage, DNA damage, motility disruption and even cell death compared to g-C 3 N 4 nanosheets and CuONPs. In addition, g-C 3 N 4 @CuO was more effective in the control of tobacco bacterial wilt than g-C 3 N 4 nanosheets and CuONPs. CONCLUSION Thus, this study provides a new perspective on g-C 3 N 4 @CuO control of bacterial diseases in crops, and the mechanism is related to the destruction of cell membrane damage and motility disruption. © 2024 Society of Chemical Industry.

A Capsid Structure of Ralstonia solanacearum podoviridae GP4 with a Triangulation Number T = 9

Jing Zheng,Wenyuan Chen,Hao Xiao,Fan Yang,Xiaowu Li,Jingdong Song,Lingpeng Cheng,Hongrong Liu

GP4, a newRalstonia solanacearumphage, is a short-tailed phage. Few structures ofRalstonia solanacearumphages have been resolved to near-atomic resolution until now. Here, we present a 3.7 Å resolution structure of the GP4 head by cryo-electron microscopy (cryo-EM). The GP4 head contains 540 copies of major capsid protein (MCP) gp2 and 540 copies of cement protein (CP) gp1 arranged in an icosahedral shell with a triangulation number T = 9. The structures of gp2 and gp1 show a canonical HK97-like fold and an Ig-like fold, respectively. The trimeric CPs stick on the surface of the head along the quasi-threefold axis of the icosahedron generating a sandwiched three-layer electrostatic complementary potential, thereby enhancing the head stability. The assembly pattern of the GP4 head provides a platform for the further exploration of the interaction betweenRalstonia solanacearumand corresponding phages. Keywords:Ralstonia solanacearumphage;icosahedral head;triangulation number T = 9;trimeric CP;Cryo-EM

The synergistic effect of biosynthesized CuONPs and phage (ϕPB2) as a novel approach for controlling Ralstonia solanacearum

Zhang Hongbao,Cai Liuti,Yuan Kai,Liu Zhongwei,Ran Maoyang,Chen Siang,Cai Wu,Rao Cheng,Cai Lin,Zhou Dong

Background As a vital soil-borne pathogenic bacterium, Ralstonia solanacearum can cause wilt disease in multiple Solanaceae plants. Several phages, such as ϕPB2, could infect R . solanacearum acting as a potential biological control agent in soil. In addition, some nanoparticles, especially copper preparation, also showed high toxicity on R. solanacearum with low toxicity on plant. However, whether they can be administered in combination and how effective they are in inhibiting the plant disease caused by R . solanacearum is known very little. Results In this work, the characterization of CuONPs using scanning electron microscope, transmission electron microscope, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, and X-ray diffraction ascertained the presence of CuONPs which were nanometer particle of 83 nm. Then it was found that combined application of CuONPs with phage (ϕPB2) was superior to that of ϕPB2 or CuONPs alone in controlling tobacco bacterial wilt, with the CuONPs (250 mg/mL) and phage (10 6 PFU/mL) ratio being the best, at 79.1%. The combination of CuONPs and ϕPB2 also showed no obvious toxicity on tobacco growth than control like single application of CuONPs or ϕPB2. Furthermore, the transcriptome changes of R . solanacearum analysis indicated that the combination application and single allocation of CuONPs could inhibit “biofilm formation”, molecular function, biological processes, cellular components, metabolic process, and so on. In addition, the combination application showed higher inhibition of motility and biofilm, and better enhancement of cell membrane permeability, protein leakage, MDA concentration, and enzyme activity of their respiratory chain dehydrogenase than single application of CuONPs or phage (ϕPB2). Transcriptomes analysis also supported that the addition of ϕPB2 enhanced the toxicity of CuONPs by influencing the ABC transporters and quorum sensing, metabolic processes, and cellular biosynthetic processes of R. solanacearum . Conclusion In total, our work not only proposed a novel way to increase the bactericidal effect of nanomaterials by adding phage, but also discovered the influence, synergistic effects, and mechanisms, which is useful to design novel way to combat phytopathogenic bacteria in the complicated environment. Response of tomato wilt pathogen Ralstonia solanacearum to the volatile organic compounds produced by a biocontrol strain Bacillus amyloliquefaciens SQR-9

Raza Waseem,Ling Ning,Yang Liudong,Huang Qiwei,Shen Qirong

It is important to study the response of plant pathogens to the antibiosis traits of biocontrol microbes to design the efficient biocontrol strategies. In this study, we evaluated the role of volatile organic compounds (VOCs) produced by a biocontrol strain Bacillus amyloliquefaciens SQR-9 on the growth and virulence traits of tomato wilt pathogen Ralstonia solanacearum (RS). The VOCs of SQR-9 significantly inhibited the growth of RS on agar medium and in soil. In addition, the VOCs significantly inhibited the motility traits, production of antioxidant enzymes and exopolysaccharides, biofilm formation and tomato root colonization by RS. The strain SQR-9 produced 22 VOCs, but only nine VOCs showed 1–11% antibacterial activity against RS in their corresponding amounts; however, the consortium of all VOCs showed 70% growth inhibition of RS. The proteomics analysis showed that the VOCs of SQR-9 downregulated RS proteins related to the antioxidant activity, virulence, carbohydrate and amino acid metabolism, protein folding and translation, while the proteins involved in the ABC transporter system, amino acid synthesis, detoxification of aldehydes and ketones, methylation, protein translation and folding, and energy transfer were upregulated. This study describes the significance and effectiveness of VOCs produced by a biocontrol strain against tomato wilt pathogen.

An environmental-friendly resistance inducer in crop protection: V2C MXene nanosheets induce plant resistance to Ralstonia solanacearum via the ET/JA signaling pathway

Yao Wang,Yijia Liang,Xingjun Ma,Yanling Dong,Matthana Klakong,Anbin Wang,Mengle Chen,Tianyu Fan,Xiaohong Xu,Peng Xiao,Wei Ding

Frequent or excessive use of synthetic pesticides can easily lead to pollution of soil and groundwater in management of soil-borne diseases. An environmental-friendly vanadium carbide MXene nanosheets (V 2 C-NSs) have been widely studied in the field of nanomedicine and biomaterials. However, few studies have shown has direct effects of V 2 C-NSs on crop protection and induces plant resistance to disease. In this study, V 2 C-NSs were first prepared by artificial synthesis and characterized, and the resulting material displayed an excellent Tyndall effect. The ability of V 2 C-NSs to promote plant growth and biosafety and its molecular mechanism of inducing plant resistance to Ralstonia solanacearum ( R. solanacearum ) were investigated. The results showed that V 2 C-NSs could increase plant fresh (or dry) weight, promote ROS accumulation, activate plant defense enzymes, and upregulate ethylene (ET) and jasmonic acid (JA) synthesis as well as the expression of ET/JA-responsive genes. Interestingly, the MAPK signaling pathway, biosynthesis of henylpropanoid and flavonoid were also activated, but the salicylic acid biosynthetic pathway was deactivated. Thus, V 2 C-NSs likely induced plant resistance to R. solanacearum via the ET/JA pathway. Furthermore, foliar application of V 2 C-NSs could delay occurrence of tobacco bacterial wilt, while the control efficiency reached 65.52 % nine days post-inoculation (dpi), followed by 57.00 % and 52.99 % at 11 dpi and 13 dpi, respectively, which were significantly greater than the control benzothiadiazole (BTH). Moreover, the biosafety of V 2 C-NSs was evaluated using tobacco seeds (Yunyan87), bacteria and earthworms. This study suggests that V 2 C-NSs can potentially be used as an environmental-friendly plant resistance inducer for controlling bacterial wilt diseases in sustainable crop protection.

Bacterial wilt pathogen induced spatial shifts of root-associated microbiome and metabolome of potatoes

Xianjun Lai,Zhouhua He,Shuyan Wang,Feng Zhang,Haiyan Wang,Xiyao Wang,Shifeng Liu,Lang Yan

Introduction: Plant root-associated microbiomes play an important role in plant health, yet their responses to bacterial wilt remain unclear poorly understood. Methods: This study investigated spatial variations in microbiome and metabolome composition across three root-associated niches—root-surrounding soil, rhizosphere, and endosphere—of healthy and Ralstonia solanacearum-infected potato plants. A total of 36 samples were analyzed, with microbial diversity assessed by full-length 16S rRNA and ITS sequencing, and metabolic profiles characterized using LC-QTOF-MS.Results: Alpha diversity analysis revealed that bacterial diversity in healthy plants was consistently higher than in diseased plants, progressively increasing from the root-surrounding soil to the rhizosphere, and most notably in the endosphere, where the Shannon index declined from 5.3 (healthy) to 1.2 (diseased). In contrast, fungal diversity was lower in diseased plants in the root-surrounding soil and rhizosphere, but significantly elevated in the endosphere, suggesting niche-specific microbial responses to pathogen stress. Beta diversity confirmed significant microbiome restructuring under pathogen stress (R² > 0.5, p = 0.001). Taxonomic analysis showed over 98% dominance of Proteobacteria in the diseased endosphere, where Burkholderia, Pseudomonas, and Massilia enriched in healthy plants were significantly reduced. R. solanacearum infection promotes the enrichment of Fusarium species in both the rhizosphere and endosphere. Metabolomic analysis revealed extensive pathogen-induced metabolic reprogramming, with 299 upregulated and 483 downregulated metabolites in the diseased endosphere, including antimicrobial metabolites such as verruculogen and aurachin A. Network analysis identified XTP as a central metabolite regulating microbial interactions, whereas antimicrobial metabolites exhibited targeted pathogen suppression. O2PLS analysis revealed that pathogen-induced antimicrobial metabolites (e.g., Gentamicin X2, Glutathionylspermine) were associated with Clostridia and Ketobacter in diseased plants, while nucleotide-related compounds (e.g., XTP) correlated with Rhodomicrobium and others, indicating infection-driven microbial adaptation and metabolic restructuring.Discussion: These findings provide insights into pathogen-driven disruptions in root microbiomes and suggest potential microbiome engineering strategies for bacterial wilt management.

pH-responsive bentonite nanoclay carriers control the release of benzothiazolinone to restrain bacterial wilt disease

Liang Yang,Yao Wang,Yijia Liang,Haibin Deng,Jun Wang,Yuhao Dai,Fuyou Guo,Hong Zhou,Shili Li,Wei Ding

Ralstonia solanacearum ( R. solanacearum ) is one of the most devastating pathogens in terms of losses in agricultural production. Bentonite (Bent) is a promising synergistic agent used in development of effective and environmentally friendly pesticides against plant disease. However, the synergistic mechanism of Bent nanoclays with benzothiazolinone (BIT) against R. solanacearum is unknown. In this work, acid-functionalized porous Bent and cetyltrimethylammonium bromide (CTAB) were employed as the core nanoclays, and BIT was loaded into the clay to form BIT-loaded CT-Bent (BIT@CT-Bent) for the control of bacterial wilt disease. BIT@CT-Bent exhibited pH-responsive release behavior that fit the Fickian diffusion model, rapidly releasing BIT in an acidic environment (pH = 5.5). The antibacterial effect of BIT@CT-Bent was approximately 4 times greater than that of the commercial product BIT, and its biotoxicity was much lower than that of BIT under the same conditions. Interestingly, R. solanacearum attracted BIT@CT-Bent into the nanocomposites and induced cytoplasmic leakage and changes in membrane permeability, indicating an efficient and synergistic bactericidal effect that rapidly reduced bacterial density. In addition, BIT@CT-Bent significantly inhibited R. solanacearum biofilm formation and swimming activity, by suppressing the expression of phcA , solR and vsrC . Indeed, exogenous application of BIT@CT-Bent significantly suppressed the virulence of R. solanacearum on tobacco plants, with control effect of 75.48%, 72.08% and 66.08% at 9, 11 and 13 days after inoculation, respectively. This study highlights the potential of using BIT@CT-Bent as an effective, eco-friendly bactericide to control bacterial wilt diseases and for the development of sustainable crop protection strategies.

Effect and mechanism of NaHS on tobacco bacterial wilt caused by Ralstonia solanacearum

Wen Dingxin,Guo Qingqing,Zhao Wan,Yang Yong,Yang Chunlei,Yu Jun,Hu Yun

Since its discovery as a third unique gaseous signal molecule, hydrogen sulfide (H 2 S) has been extensively employed to resist stress and control pathogens. Nevertheless, whether H 2 S can prevent tobacco bacterial wilt is unknown yet. We evaluated the impacts of the H 2 S donor sodium hydrosulfide (NaHS) on the antibacterial activity, morphology, biofilm, and transcriptome of R. solanacearum to understand the effect and mechanism of NaHS on tobacco bacterial wilt. In vitro, NaHS significantly inhibited the growth of Ralstonia solanacearum and obviously altered its cell morphology. Additionally, NaHS significantly inhibited the biofilm formation and swarming motility of R. solanacearum , and reduced the population of R. solanacearum invading tobacco roots. In field experiments, the application of NaHS dramatically decreased the disease incidence and index of tobacco bacterial wilt, with a control efficiency of up to 89.49%. The application of NaHS also influenced the diversity and structure of the soil microbial community. Furthermore, NaHS markedly increased the relative abundances of beneficial microorganisms, which helps prevent tobacco bacterial wilt. These findings highlight NaHS's potential and efficacy as a powerful antibacterial agent for preventing tobacco bacterial wilt caused by R. solanacearum .

A Ralstonia solanacearum effector RipAD perturbs aquaporin NbPIP oligomeric status to suppress plant immunity

Bo Nian,Sai Yuan,Yijie Liu,Ying Zhang,Pengyue Li,Yang Lin,Jiatao Xie,Jiasen Cheng,Yanping Fu,Xiao Yu,Daohong Jiang,Bo Li

SUMMARY Bacterial wilt caused by Ralstonia solanacearum is a devastating disease that affects a wide range of crops, including tobacco. The pathogen promotes infection by secreting type III effector (T3E) proteins into host plants to facilitate pathogenicity; however, the molecular mechanisms for many of these effectors remain elusive. Here, we characterize RipAD, a conserved T3E that significantly contributes to R. solanacearum virulence in Nicotiana tabacum and Nicotiana benthamiana . We demonstrate that RipAD dampens early immune responses, including pathogen-associated molecular pattern-triggered reactive oxygen species (ROS) bursts and MAPK activation. This immunosuppressive function depends on the dual localization of RipAD to the plasma membrane and the nucleus. Mechanistically, RipAD physically associates with the N-terminal domains of the plasma membrane intrinsic proteins NbPIP2;3 and NbPIP2;4. We show that these aquaporins act as positive regulators of ROS accumulation and plant immunity. Crucially, RipAD binding is associated with reduced homo- and hetero-oligomerization of NbPIP2;3 and NbPIP2;4, thereby compromising their stability and function. Furthermore, silencing NbPIP2;3 and NbPIP2;4 increases tobacco susceptibility and rescues the virulence defect of a RipAD deletion mutant. These findings reveal a novel virulence strategy wherein a bacterial effector targets the structural integrity of host aquaporin complexes to block ROS signaling and suppress immunity.

Antibacterial Activity of Ferulic Acid Ester against Ralstonia solanacearum and Its Synergy with Essential Oils

Qing-Bo Tu,Hui-Cong Shi,Ping Li,Sheng Sheng,Fu-An Wu

Ralstonia solanacearumis one of the ten most harmful plant bacteria worldwide, and traditional agrochemicals are not very effective in controlling this pathogen. Moreover, excessive pesticides always bring organic residues and resistant strains, which cause the unsustainability of the environment. In this paper, ferulic acid and essential oils are used as antibacterial materials. These compounds are natural substances with low toxicity and environmental safety. Through the structural optimization and the analysis of binary combined bacteriostatic efficiency, the MIC values of chlorobutyl ferulate (2e) and peppermint essential oil (EO1) were 0.64 mg/mL and 2.02 mg/mL, respectively, and the MIC value of2e-EO1(mass ratio 1:1.5) was 0.40 mg/mL. The growth rate of bacteria treated with2e-EO1was inhibited, the OD590nmvalue of cell membrane decreased by 57.83%, and the expression levels ofhrpB,pehC,pilT,polA,aceE,egl, andphcAwere downregulated to 18.81%, 30.50%, 14.00%, 44.90%, 86.79%, 23.90%, and 27.56%, respectively. The results showed that2e-EO1had a synergistic inhibitory effect againstR. solanacearum. It significantly affected the formation of the bacterial cell membrane and the expression of pathogenic genes. Consequently,2e-EO1provides the potential to become a green pesticide and can promote the sustainability of the agricultural ecological environment. Keywords:Ralstonia solanacearum;ferulic acid;essential oil;antibacterial activity;synergistic effect;pathogenic gene

Molecular mechanism of plant elicitor daphnetin-carboxymethyl chitosan nanoparticles against Ralstonia solanacearum by activating plant system resistance

Yao Wang,Liang Yang,Xiao Zhou,Ye Wang,Yijia Liang,Binshao Luo,Yuhao Dai,Zhouling Wei,Shili Li,Rong He,Wei Ding

The exploration of biopolymer-based materials to avoid hazardous chemicals in agriculture has gained enormous importance for sustainable crop protection. Due to its good biocompatibility and water solubility, carboxymethyl chitosan (CMCS) has been widely applied as a pesticide carrier biomaterial. However, the mechanism by which carboxymethyl chitosan-grafted natural product nanoparticles induce tobacco systemic resistance against bacterial wilt remains largely unknown. In this study, water-soluble CMCS-grafted daphnetin (DA) nanoparticles ( [email protected] ) were successfully synthesized, characterized, and assessed for the first time. The grafting rate of DA in CMCS was 10.05 %, and the water solubility was increased. In addition, [email protected] significantly increased the activities of CAT, PPO and SOD defense enzymes, activated the expression of PR1 and NPR1 , and suppressed the expression of JAZ3 . [email protected] could induce immune responses against R. solanacearum in tobacco, including increases in defense enzymes and overexpression of pathogenesis-related (PR) proteins. The application of [email protected] effectively suppressed the development of tobacco bacterial wilt in pot experiments, and the control efficiency was as high as 74.23 %, 67.80 %, 61.67 % at 8, 10, and 12 days after inoculation. Additionally, [email protected] has excellent biosafety. Therefore, this study highlighted the application of [email protected] in manipulating tobacco to generate defense responses against R. solanacearum , which can be attributed to systemic resistance.
