Lansiumamide B (LB) is a cinnamamide alkaloid isolated from Clausena lansium seeds. The compound is the main antifungal ingredient found in C. lansium . In this paper, the antifungal activity of LB against 10 plant pathogenic fungi was evaluated and found that it was the most effective to Rhizoctonia solani (EC 50 = 2.99 μg/mL). The compound also exhibits certain protective (53.06%) and curative (71.01%) activities against R. solani at 800 μg/mL. The mechanism of action of LB was further elucidated through physiological and biochemical experiments, mycelial morphology and ultrastructure observations, and transcriptomics studies with R. solani as the experimental strain. SEM and TEM observation showed that it caused abnormal mycelium growth, extravasation of contents and cell wall degradation. Physiological and biochemical tests showed that LB treatment had no significant effect on the cellular respiration of mycelium , but it could increase the membrane permeability , cause the leakage of intracellular electrolyte, reduce the content of extracellular polysaccharide and chitin in cell wall and increased the contents of intracellular reducing sugar. The results of transcriptome sequencing analysis showed that LB had significant effects on polysaccharide and oligosaccharide related metabolic pathways, such as starch and sucrose metabolic pathways, galactose metabolic pathways, and amino sugar metabolic pathways. Furthermore, LB treatment increased the specific enzyme activity of α-1, 4-glucanase, α-1,6-glucanase, cellulase and maltase of R. solani in vivo . Comprehensive analysis of the above experimental results shows that LB induces and regulates the degradation of polysaccharides and oligosaccharides of R. solani at the transcriptional level, thereby destroying the cell wall structure and ultimately leading to the death of fungi.

Copper-based nanomaterials are effective alternatives to traditional pesticides due to their antibacterial properties. However, the high cost and low dispersity limit their application. In this study, we synthesized cost-effective, eco-friendly, and stable chitosan-copper hybrid nanoflowers (CS-Cu HNFs) through facile self-assembly to manage agricultural diseases caused by the fungal pathogen (Rhizoctonia solani). The results show that CS-Cu HNFs, which utilized chitosan and copper phosphate as primary scaffolds, were formed via a series of nucleation, aggregation, self-assembly, and anisotropic growth processes. 200 mg/L CS-Cu HNFs exhibited an excellent inhibitory effect on R. solani, which was 6.11 times that of CuO nanoparticles, despite CS-Cu HNFs containing only 45% of Cu as that in CuO nanoparticles. Additionally, CS-Cu HNFs significantly reduced R. solani infection in various crops and displayed broad-spectrum antibacterial activity. This research provides new insights into the preparation and application of organic–inorganic hybrid nanoflowers as nanopesticides.

This study was conducted to purify and identify metabolites of antimicrobial activity against phytopathogens from Xenorhabdus nematophila YL001. Three dipeptide compounds were purified from its cell-free cultural broth and identified as (±)-nematophin, cyclo (L-Pro-Gly), and N, N′-dimethyl-cyclo (L-Phe-L-Leu). Nematophin demonstrated a wider antifungal spectrum than the other two compounds. It also exhibited strong inhibitory effects on mycelial growth of Rhizoctonia solani and Phytophthora infestans with EC50 values of 40.00 and 51.25 μg/ml, respectively. Its (S)-configuration structure [(+)-nematophin] was also synthesized and exhibited higher antimicrobial activity than the enantiomeric mixture. The detached leaf assay revealed that nematophin possessed significant preventive and curative efficacy against R. solani on broad bean leaves showing corresponding control efficacies of 93.01 and 94.93% at 1,000 μg/ml, comparable to those of a chemical fungicide (carbendazim) at 500 μg/ml. Additionally, the pot experiments indicated that nematophin could effectively inhibit the disease extension on rice and broad bean plants caused by R. solani. Nematophin also exerted some adverse influences on the sclerotial development of R. solani by dramatically suppressing their formation and maturation at 40.00 μg/ml, as well as their germination at 15.00 μg/ml. Morphological and ultrastructural observations showed that the hyphae of R. solani became twisted, shriveled, and deformed at the growing points after exposure to nematophin at 40.00 μg/ml, and that the subcellular fractions also became abnormal concurrently, especially the mitochondrial structure. These results indicate that nematophin has great potential to be used as a bio-pesticide in agricultural production.

In the ongoing search for more potent succinate dehydrogenase inhibitors (SDHIs) against Rhizoctonia solani , 30 novel aromatic carboxamides containing an ether group were designed and synthesized. Most compounds, especially those with a pyrazole ring, showed excellent in vitro antifungal activity. Notably, compound 14b stood out as the most promising candidate, with a half maximal effective concentration (EC 50 ) value of 0.019 µg/mL, about 24 times more potent than boscalid (EC 50 = 0.464 µg/mL) and three times more effective than fluxapyroxad (EC 50 = 0.056 µg/mL). Furthermore, the detached leaf assay revealed that 14b significantly suppressed R. solani growth on rice leaves, with protective (90.28%) and curative (88.84%) efficacy at 100 µg/mL, outperforming boscalid (71.73% and 70.33%, respectively) and fluxapyroxad (85.98% and 83.49%, respectively), indicating its potential for effective application in real-world agricultural settings. In addition, the succinate dehydrogenase (SDH) inhibition assay showed that 14b inhibited SDH with a half maximal inhibitory concentration (IC 50 ) of 2.41 µM, stronger than boscalid (4.34 µM) and fluxapyroxad (3.90 µM). Further molecular docking suggested 14b shared a similar antifungal mechanism of action with fluxapyroxad. These results highlighted compound 14b as a promising SDHI fungicide candidate for controlling R. solani , deserving further study. Graphical

Rice sheath blight is a destructive fungal disease caused by Rhizoctonia solani . To find a safe and green measure, the biological activity of six plant extracts against R. solani was determined by mycelial growth rate method. The results showed that magnolol possessed better antifungal activities against R. solani , with an EC 50 value of 7.47 mg/L. further action mechanism of magnolol against R. solani was carried out. Studies by scanning electron microscopy (SEM) showed that the morphology of R. solani mycelia was deformation and surface folds. Transmission electron microscope (TEM) observation on treated R. solani showed that magnolol could induce cytoplasmic membrane rupture and cytoplasmic membrane even disappeared completely accompanied with cellular debris was covered around this fungal, and the mycelia treated with magnolol showed fluorescence after PI staining. Further study showed that the content of malondialdehyde (MDA) and activity of chitinase , β-1,3-glucanase and relative conductivity of mycelia were increased, while the content of soluble protein and activities of catalase (CAT), polyphenol oxidase (PPO), superoxide dismutase (SOD), succinate dehydrogenase (SDH) and NAD-malate dehydrogenase (NAD-MDH) were significantly decreased. These results indicated that magnolol could significantly damage the plasma membrane of R. solani , and interfere with cell respiratory metabolism, thus inhibiting the growth of mycelium.

Developing active targeting strategies for agrochemical delivery is paramount for simultaneously enhancing fungicide efficacy and mitigating off-target environmental risks. This study reports the development of wheat germ agglutinin (WGA)-functionalized mesoporous silica nanoparticles (MSNs) as a nanocarrier system designed to actively target fungal cell walls. These MSNs were loaded with the fungicide Fluazinam (Flu) to yield Flu@MSNs-WGA, which exhibited an average hydrodynamic diameter of 44.54 nm and a Fluazinam loading content of 10.89 % ± 0.27 %. Studies investigating transport and distribution in rice plants demonstrated that the WGA-functionalized nanoparticles were absorbed by roots and leaves and exhibited specific accumulation at sites of Rhizoctonia solani infection, confirming active targeting. Furthermore, In vitro assays revealed that the antifungal activity of Flu@MSNs-WGA against R. solani was significantly enhanced compared to the non-targeted nanocarrier (Flu@MSNs-COOH) and was 2.7 times higher than that of a commercial Fluazinam suspension concentrate (Flu SC) formulation. Representing the first application of WGA for targeted pesticide delivery, this work validates the feasibility of this approach, providing essential proof-of-concept and technical support for developing novel, actively targeted nano-pesticides for improved crop protection.

Rhizoctonia solani is a significant soil-borne pathogenic fungus that poses a significant threat to the economically important agricultural crops. 4-(Diethylamino)salicylaldehyde (DSA) is a secondary metabolite produced by Streptomyces sp. KN37, which has antifungal activity, meanwhile its inhibitory mechanism is still unclear. In this study, we explored the antifungal efficacy of DSA and its potential mechanism of inhibiting R. solani . It was found that DSA exhibited significant antifungal activity against six tested plant pathogenic fungi, with R. solani being the most sensitive (EC 50 = 26.904 μg/mL). Notably, DSA effectively reduced the mycelial mass and inhibited sclerotia germination, demonstrating a good control efficacy of cucumber damping-off disease. Morphological observation showed that DSA significantly disrupted the shape and ultrastructure of the mycelium. Transcriptomic and metabolomic analyses revealed that DSA impacted the integrity of the cell membrane, redox processes, and energy metabolism in R. solani . The results of fluorescence staining, relative conductivity, H 2 O 2 content, and antioxidant enzyme activity showed that the accumulation of ROS in hypha cells after DSA treatment possibly resulted in damage to cell membrane integrity. Furthermore, the reduction in ATP content, along with decreased ATPase and citrate synthase activity, indicates that energy production may be inhibited. Molecular docking analysis further showed that DSA may competitively inhibit citrate synthase, thereby inhibiting cell energy production and ultimately inducing apoptosis. Our study provides new insights into the potential mechanism by which DSA inhibits the mycelial growth of R. solani .

Cationic polymers are prospective fungicidal agents for inhibiting plant diseases because of the controllability of their structure and properties. This study investigates the effect of the hydrophobic–hydrophilic balance on the antifungal activities of antimicrobial polymers against phytopathogenic fungi (Rhizoctonia solani Kühn AG-1(IA)), the pathogen of rice sheath blight (RShB). A series of polydimethylsiloxane-polymethacrylate block copolymers containing quaternary ammonium salts (PDMS-b-QPDMAEMA, labeled as SnQm; n and m represent 1000th of the molecular weight of the PDMS and QPDMAEMA chain, respectively) were synthesized via anionic ring-opening polymerization and atom transfer radical polymerization (ATRP). The abilities of the quaternary ammonium salts to adsorb onto the surface of R. solani sclerotia and permeate the R. solani sclerotia were investigated on the basis of static water contact angles and fluorescence labeling. The results indicated that the moderately hydrophobic PDMS chain helped stabilize the attachment of the hydrophilic QPDMAEMA chain and then help it penetrate the R. solani sclerotia. Its antifungal properties toward R. solani were characterized by determining its minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) and the inhibition rate of R. solani sclerotia germination. The hydrophobic PDMS chains had a significant influence on the antifungal activities of amphiphilic SnQm against R. solani sclerotia. This work highlights the prospective application of amphiphilic antimicrobial polymers as antifungal agents for inhibiting plant diseases.

Polymeric quaternary ammonium salts (PQASs) exhibit antibacterial action and are less toxic, less stimulatory to the human body and have easier-to-modify functionalities than small molecular antibacterial agents . However, few studies on the structure-activity relationship and toxicity mechanism of PQASs against fungi have been reported. We previously described the synthesis of a novel PQAS, namely, a homopolymer of (2-methacrylamido)propyltetrabenzyldimethylammonium chloride (PQD-BC), and discovered that the polymer exhibits antifungal activities not only against Fusarium oxysporum f. sp. Cubense tropical race 4 (Foc 4), the pathogen of banana wilt, but also against Rhizoctonia solani ( R. solani ), the pathogen of rice sheath blight (RShB). Furthermore, we studied the mechanism of action of PQD-BC against Foc 4, which is markedly different from R. solani in morphology and life cycle. Therefore, the structure-antifungal activity relationship and toxicity mechanism of PQD-BC against R. solani were extensively studied in this work and compared with those of the low-molecular-weight quaternary ammonium salt benzyldimethyldodecylammonium chloride (BC), and the results play an important role in identifying long-term and low-toxicity fungicides that can suppress the sclerotia of R. solani . The results showed that PQD-BC and BC can destroy the structural integrity and morphology of a cell, such as by loss of the cell wall and plasma membrane integrity, leading to the release of intracellular contents and can induce mitochondrial dysfunction and interference with genomic DNA and inhibit the formation of sclerotia. However, PQD-BC showed a special mechanism for causing the lipid peroxidation of the cell membrane; this mechanism was not observed with BC. The newly elucidated mechanism accounts for differences between polymers and small-molecule compounds and provides a theoretical basis for further application of PQAS against fungi and sclerotia.

In the fields of agriculture and forestry, there are major unknown risks associated with the toxicity of chemical pesticide residues and pathogen resistance. To address this issue, in this study, 52 novel pyrazole monoterpene compounds were obtained by organic synthesis of highly active monoterpene natural products and evaluated for their in vitro and in vivo antifungal activities and crystal structure analysis of compound 5b. The results showed that compounds 4g and 8f exhibited remarkable antifungal activities against Rhizoctonia solani, with the corresponding EC50 values reaching 4.27 and 4.81 mg/L. At the same time, the results of in vivo and in vitro experiments showed that both compounds 4g and 8f had good control effects against rice sheath blight. The antifungal activity of these compounds was preliminarily explored by scanning electron microscopy, molecular docking, enzyme activity determination, and cytotoxicity, indicating that these compounds have the potential for development into low-toxic antifungal drugs.

To enhance the antifungal activity of 4-(diethylamino)salicylaldehyde
(DSA), 20 salicylaldehyde Schiff base compounds (L1–L20), including
nine novel derivatives, were synthesized and characterized by MS, 1 H NMR, and 13 C NMR. In vitro assays identified
compound L5 as the most active agent, with an EC 50 of 10.797
μg/mL (2-fold higher than DSA), and pot experiments confirmed
its efficacy against cotton damping-off disease. Mechanistic investigations
via electron microscopy, transcriptomics, physiological assays and
molecular docking suggest that L5 may target fatty acid synthase (FAS)
to interfere with fungal lipid biosynthesis, disrupt energy homeostasis,
and induce oxidative stress, which triggers synergistic membrane impairment
and oxidative damage and ultimately causes hyphal death. With a novel
target and a distinctive single-target, multieffect mode of action,
L5 is a promising candidate for new fungicide development.

Rhizoctonia solani Kühn is an important fungal pathogen that causes serious crop yield losses worldwide. The application of nanoscale strategies based on RNA interference (RNAi) represents an environmentally friendly and efficient approach for plant disease control. In this study, R. solani glycosyl hydrolase family 1 ( RsGH1 ), which functions as a cell wall degrading enzyme (CWDE), and was screened as a prospective RNAi target gene for managing R. solani AG3 TB. Additionally, a novel nanosystem for loading and stabilization of double-stranded RNA (dsRNA) was developed. When the mass ratio of ε-poly-L-lysine (ε-PL) to carboxymethyl chitosan (CMCS) is 1:1, ε-PL can spontaneously conjugate with CMCS to form nanoscale spherical particles by electrostatic interaction, hydrogen bonding and Van der Waals forces. Compared with ε-PL alone, the introduction of CMCS resulted in more regular and uniform nanoparticles. ds RsGH1 spontaneously binds with ε-PL@CMCS, which is referred to as ds RsGH1 @ε-PL@CMCS. ε-PL@CMCS protected dsRNA from RNase A degradation effectively. The combination of ds RsGH1 with ε-PL@CMCS remarkably improved the deposition and adhesion of ds RsGH1 droplets on Nicotiana tabacum ( N. tabacum ) leaves. The application of ε-PL@CMCS improved the RNAi efficiency of ds RsGH1 and prolonged its protective duration on crops. ds RsGH1 derived from R. solani AG3 TB also exhibited broad-spectrum activity against R. solani AG1-IA in rice and maize plants. In this study, a self-assembled multi-component nano-fungicide was designed based on dsRNA and nanocarriers. This work proposes an eco-friendly strategy to manage R. solani .
