Root rot caused by Rhizopus stolonifer significantly diminished the yield and quality of sweet potatoes. Currently, the control of root rot predominantly relied on toxic chemical pesticides, with a notable lack of effective alternative strategies. Microbial volatile organic compounds (VOCs) are regarded as an environmentally friendly and safe method for controlling postharvest diseases. This study investigated the in vitro and in vivo effects of VOCs produced by the Nocardiopsis dassonvillei MI-S24 strain against R. stolonifer . The results demonstrated that VOCs exhibit strong antifungal activity against various pathogenic fungi. They significantly inhibited hyphal growth (91.85 %) and spore germination of R. stolonifer while inducing morphological changes. Furthermore, during storage at 28 °C for 20 days, VOCs fumigation effectively mitigated the incidence of postharvest sweet potato root rot. It enhanced the activity of CAT, POD, SOD, PPO, and PAL enzymes by 84.62 %, 38.19 %, 47.98 %, 84.52 %, and 19.50 % respectively in sweet potatoes, safeguarded soluble sugars and starch from degradation, and preserved overall quality. Analysis of volatile components coupled with antifungal validation revealed that 2-methyl-3-(methylthio) furan and 2-undecanol exhibited significant antifungal activity against R. stolonifer . Further research indicated that VOCs affected cell membrane integrity in R. stolonifer , stimulated reactive oxygen species (ROS) accumulation, reduced antioxidant enzyme activity, elevated malondialdehyde levels, induced oxidative damage, and disrupted ATP synthesis as well as TCA cycle processes by diminishing mitochondrial membrane potential alongside succinate dehydrogenase activity, ultimately leading to mitochondrial dysfunction. This study presents a novel biological strategy aimed at preventing and controlling soft rot disease in postharvest sweet potatoes.

Post-harvest diseases of raspberries affect their storage and subsequently the ability to provide consumers with fresh products. The antifungal activity of microbial volatile organic compounds (MVOCs) against both pre and postharvest diseases has previously been demonstrated, but the control of raspberry postharvest diseases caused by Botrytis cinerea and Rhizopus stolonifer using MVOCs has not been extensively studied. We have thus investigated the effects of Bacillus siamensis G-3 volatile organic compounds (VOCs) on controlling the diseases caused by B. cinerea and R. stolonifer . Bioassays of the VOCs of B. siamensis G-3 exhibited significant control effects (89.4%) on the two pathogens. The VOCs composition was studied with solid phase micro extraction and GC/mass spectrometry (SPME-GC-MS), and 22 VOCs, different from the controls, were identified. Among these compounds, only 2, 6-di- tert -butyl-4-methylphenol (BHT), and 2,4-di- tert -butylphenol (2,4-DTBP) had significant control effects on the pathogens (* P < 0.05), 52.38 and 93.33%, respectively, compared with the controls in vivo . With the 2,4-DTBP treatments, the raspberries could be stored for 20 d at 0 °C, with the disease rate maintained at ∼10%. In addition, we have reported for the first time, that 2,4-DTBP may be a better preservative than BHT and could be used as a bio-fumigant for controlling raspberry postharvest diseases caused by B. cinerea and R. stolonifer in vivo .

The soft rot of sweet cherries caused by Rhizopus stolonifer ( R. stolonifer ) results in significant economic losses during storage and transportation. This study demonstrated that volatile organaic compounds (VOCs) derived from the fermentation broth of Bacillus velezensis ( B. velezensis ) Q-84 significantly inhibited the growth of R. stolonifer by 54.43%. Scanning electron microscope (SEM) micrographs of VOCs-treated fungi revealed antifungal activity, evidenced by the breakage and deformation of the mycelium. Through headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC/MS) analysis, ten VOCs were identified in the fermentation broth of B. velezensis . Notably, 5-methyl-2-heptanone, 2,5-dimethylpyrazine, 2-decanone, and benzaldehyde exhibited significant antifungal activity in vitro . Furthermore, in vivo assays demonstrated the efficacy of both individual and combined VOCs in managing soft rot diseases in cherries. These treatments also resulted in increased levels of total polyphenolic content (TPC) and total flavonoid content (TFC), as well as a significant enhancement in the activities of defense-related enzymes such as peroxidase (POD), polyphenol oxidase (PPO), phenylalanine ammonia-lyase (PAL), and chitinase (CHI). Our results further our understanding of B. velezensis antifungal metabolites on the management of R. stolonifer and may help in finding less toxic compounds to control soft rot on sweet cherries.
