Antioxidant and hepatoprotective effects of purified Rhodiola rosea polysaccharides

Yao Xu,Huiyan Jiang,Congyong Sun,Michael Adu-Frimpong,Wenwen Deng,Jiangnan Yu,Ximing Xu

In this study, two polysaccharide fractions (RRP1: Mw = 5.5 kDa, and RRP2: Mw = 425.7 kDa) were isolated from Rhodiola rosea to investigate their antioxidation and hepatoprotective effects. Physicochemical analysis showed that RRP1 was composed of mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose with a relative molar ratio of 0.69:0.11:0.15:1:0.51:7.5 and RRP2 was consisted of mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose (relative molar ratio = 0.15:0.19:1.01:0.18:0.47:1). Periodate oxidation and Smith degradation analysis revealed that, in RRP1, part of the arabinose and glucose residues were 1 → 3,6/1 → 3/1 → 2,3/1 → 3,4/1 → 2,4/1 → 2,3,4-linked, and the mannose, rhamnose and galactose residues were 1 → 2,6/1 → 6/1 → 2/1→/1 → 4,6/1 → 4-linked. In RRP2, the rhamnose, glucose and galactose residues were linked by 1 → 3,6/1 → 3/1 → 2,3/1 → 3,4/1 → 2,4/1 → 2,3,4 linkages, and the arabinose and mannose residues were 1 → 2/1 → 6/1 → 4-linked. The methylation analysis confirmed the structure information of the two fractions. Importantly, fraction RRP1 demonstrated stronger antioxidative activities than RRP2 by scavenging DPPH, hydroxyl and superoxide anion radicals in vitro. Correspondently, RRP1 showed more significant effects than RRP2 on decreasing the levels of ALT, AST and MDA, and increasing the GSH, SOD and CAT levels in the CCl 4 -treated mice. These data demonstrated that the polysaccharide RRP1 could be developed as a promising candidate for preventing and treating liver damage induced by toxic chemicals.

Application of UHPLC Fingerprints Combined with Chemical Pattern Recognition Analysis in the Differentiation of Six Rhodiola Species

Didi Ma,Lijun Wang,Yibao Jin,Lifei Gu,Xiean Yu,Xueqing Xie,Guo Yin,Jue Wang,Kaishun Bi,Yi Lu,Tiejie Wang

Rhodiola, especiallyRhodiola crenulateandRhodiola rosea, is an increasingly widely used traditional medicine or dietary supplement in Asian and western countries. Because of the phytochemical diversity and difference of therapeutic efficacy amongRhodiolaspecies, it is crucial to accurately identify them. In this study, a simple and efficient method of the classification ofRhodiola crenulate,Rhodiola rosea, and their confusable species (Rhodiola serrata,Rhodiola yunnanensis,Rhodiola kirilowiiandRhodiola fastigiate) was established by UHPLC fingerprints combined with chemical pattern recognition analysis. The results showed that similarity analysis and principal component analysis (PCA) could not achieve accurate classification among the sixRhodiolaspecies. Linear discriminant analysis (LDA) combined with stepwise feature selection exhibited effective discrimination. Seven characteristic peaks that are responsible for accurate classification were selected, and their distinguishing ability was successfully verified by partial least-squares discriminant analysis (PLS-DA) and orthogonal partial least-squares discriminant analysis (OPLS-DA), respectively. Finally, the components of these seven characteristic peaks were identified as 1-(2-Hydroxy-2-methylbutanoate) β-D-glucopyranose, 4-O-glucosyl-p-coumaric acid, salidroside, epigallocatechin, 1,2,3,4,6-pentagalloyglucose, epigallocatechin gallate, and (+)-isolarisiresinol-4′-O-β-D-glucopyranoside or (+)-isolarisiresinol-4-O-β-D-glucopyranoside, respectively. The results obtained in our study provided useful information for authenticity identification and classification ofRhodiolaspecies. Keywords:Rhodiola;UHPLC fingerprint;chemical pattern recognition;quality evaluation

Rhodiola rosea Rhizome Extract-Mediated Green Synthesis of Silver Nanoparticles and Evaluation of Their Potential Antioxidant and Catalytic Reduction Activities

Daihua Hu,Xu Yang,Wang Chen,Zili Feng,Chingyuan Hu,Fei Yan,Xiaohua Chen,Dong Qu,Zhiyuan Chen

The silver nanoparticles (AgNPs) using the rhizome extract of Rhodiola rosea have been reported. However, their antioxidant activity and whether the biogenic AgNPs could be used to catalyze the reduction of hazardous dye or used as fluorescence enhancers are unknown. This study focused on the facile green synthesis of silver nanoparticles using the rhizome aqueous extract of R. rosea (G-AgNPs). We then studied their antioxidant activity and catalytic degradation of hazardous dye Direct Orange 26 (DO26) and Direct Blue 15 (DB15). Their effects on fluorescein’s fluorescent properties were also evaluated. The chemical AgNPs (C-AgNPs) were synthesized by reducing solid sodium borohydride (NaBH4), and its above activities were compared with those of G-AgNPs. The formation of G-AgNPs was confirmed by the appearance of brownish-gray color and the surface plasmon resonance (SPR) peak at 437 nm. The biogenic AgNPs were approximately 10 nm in size with a regular spherical shape identified from transmission electron microscopy (TEM) analysis. G-AgNPs exhibited significantly improved 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity than butylated hydroxytoluene (BHT) and C-AgNPs (p < 0.05). The biogenic G-AgNPs were also found to function as an effective green catalyst in reducing DO26 and DB15 by NaBH4, which is superior to C-AgNPs. Furthermore, G-AgNPs showed better fluorescence enhancement activity than C-AgNPs, and the concentration required was lower. When the concentration of the G-AgNP solution was 64 nmol/L, the fluorescence intensity reached the maximum of 5460, with the fluorescence enhancement efficiency of 3.39, and the fluorescence activity was stable within 48 h. This study shows the efficacy of biogenic AgNPs in catalyzing the reduction of hazardous dye DO26 and DB15. Biogenic AgNPs could also be used as fluorescence enhancers in low concentrations.

Reveal the pharmacodynamic substances and mechanism of an edible medicinal plant Rhodiola crenulate in DSS-induced colitis through plasma pharmacochemistry and metabolomics

Yu Peng,Xiaoao Xiao,Tingting Ji,Xinyuan Wang,Yixuan Xu,Jianbo Xiao,Hui Cao,Zhiyong Chen,Huifan Liu,Yuanqing Gao,Hongxun Tao

Rhodiola crenulate is the edible medicinal herbal medicine widely used for altitude sickness in China. Interestingly, our previous work has found that R. crenulate extract (RCE) could significantly improve the pathology associated with dextran sulfate sodium-induced colitis. Thus, the current research aims to reveal the pharmacodynamic material basis of RCE, as well as its mechanism against colitis. The chemical characterization of RCE was performed by UHPLC-HR-MS, through which a total of 88 constituents were identified. Meanwhile, our results also found 29 constituents absorbed into blood and 8 metabolized absorbable compounds. The decreased flavonoids prototype and the elevated sulfated products of phenols were observed under pathophysiological conditions of colitis. The metabolomics study revealed that colitis caused the alternation of fatty acid metabolism, steroid hormone biosynthesis and bile acid metabolism. Correspondingly, RCE could prevent colitis by improving fatty acid metabolism and secondary bile acid metabolism.

Preparation, Characterization, and Antibacterial Activity of Rhodiola Carbon Dots

Pei S.,Lu Z.,Sun W.,Yan K.,Zhou J.,Sun C.,Huang J.,Luo K.,Yang X.

Abstract Rhodiola carbon dots (R-CDs) were prepared by simple and economical one-pot hydrothermal method using Rhodiola as raw materials. The morphology and surface functional groups were analyzed by TEM, FT-IR, and XPS. It was discovered that R-CDs had COC, C=O and OH functional groups linked to the surface and had a common particle dimension of 2.56 nm. The prepared difunctional R-CDs showed high water-solubility and favorable photostability. Additionally, it was unexpectedly discovered that R-CDs confirmed great antibacterial motion in opposition to both E. coli and S. aureus , with a MIC of 0.7 mg/mL. Consequently, this study provides a new, green, economical carbon dot with the potential application as potent antibacterial.

Zhang Cheng-cai,Chen Xiang-tao,Wang Hong-yang,Chen Wei-wu,Ciwang Jiumi,Dai Xiao-yu,Zhang Ya-fei,Li Qi,Wang Sheng,Zhang Ze-kun

Rhodiola species are highly valuable medicinal resources that hold significant importance in both traditional medicine and modern health fields. However, existing breeding techniques suffer from pain points such as low breeding efficiency and unstable active ingredients, which seriously restrict the development and utilization of their medicinal value. To develop a rapid tissue culture propagation system for endangered medicinal plants ( Rhodiola spp.) in plateau regions, and to determine the optimal conditions for seed germination, proliferation, seedling strengthening, rooting induction, and transplanting substrates across stages. L₉(3 3 ) orthogonal design and Box-Behnken response surface methodology were employed to explore the optimal tissue culture conditions. Results showed that the highest seed germination 92.67% was realized with 75% ethanol (30 s) pre-treatment, and then 1.0% NaClO (10 min). This study established a rapid tissue culture system for R. crenulate and R. rosea , achieving a proliferation coefficient of 7.38, 89% rooting rate, and enhanced active compound production. The most suitable transplanting substrate was a 2:1:1 mixture of vermiculite, sphagnum peat, and river sand, resulting in a survival rate of 68.54%. Post-transplantation, R. crenulate rhizomes accumulates higher salidroside and gallic acid, while R. rosea L. shows superior rosavin and tyrosol accumulation, revealing distinct interspecific patterns in active compound production. In conclusion, a tissue culture system for Rhodiola species was established providing a solid foundation for their sustainable resource utilization.

Rosavin derived from Rhodiola alleviates colitis in mice through modulation of Th17 differentiation

Wang Yi,Jiang Yu,Li Mingxing,Xiao Yaqin,Zhao Qianyun,Zeng Jiuping,Wei Shulin,Chen Shuhan,Zhao Yueshui,Du Fukuan,Chen Yu,Deng Shuai,Shen Jing,Li Xiaobing,Li Wanping,Wang Fang,Sun Yuhong,Gu Li,Xiao Zhangang,Wang Shengpeng,Wu Xu

Background Rosavin (RSV) is a naturally occurring compound isolated from Rhodiola species. While RSV has been reported with pharmacological activities of anti-oxidation, anti-inflammation, anti-stress and immunomodulation, its effect on colitis and the underlying mechanisms remain unclear. Purpose This study aims to investigate whether and how RSV alleviated colitis in mice. Study design and methods The protective effect of RSV (50, 100, 200 mg/kg, p.o.) was investigated in dextran sulfate sodium (DSS) mediated mouse models of acute and chronic colitis. Alterations in fecal microbiota were evaluated by 16S rRNA sequencing. Pseudo germ-free mice achieved by antibiotics treatment were applied to assess the RSV-mediated functional role of gut microbiota in colitis. RNA sequencing was performed to determine RSV-induced colonic response. Primary T cell culture was conducted to examine the effect of RSV on Th17 and Treg differentiation. Whole blood assay, dual luciferase reporter assay, and molecular docking methods were applied to investigate the mechanisms and targets of RSV in Th17 regulation. Results Oral RSV significantly relieved DSS-mediated acute and chronic colitis in mice, which recovered body weight loss, reduced disease activity index, alleviated colon injury, inhibited inflammation, suppressed the apoptosis of intestinal epithelia, and maintained intestinal barrier function. Moreover, RSV specifically regulated intestinal microbiota by recovering DSS-mediated microbial changes and elevating beneficial microbes such as Lactobacillus and Akkermansia . Antibiotics treatment experiment showed that the protective role of RSV was at least partially dependent on gut microbiota; however, in vitro incubation showed that RSV did not directly promote the growth of Lactobacillus and Akkermansia strains. Further analysis showed that RSV-mediated genetic alterations in colon were enriched in pathways related to lymphocyte regulation. Additionally, RSV regulated the balance of Th17/Treg in colitis mice. Importantly, RSV inhibited the differentiation of Th17 cell in vitro , suppressed the production of IL-17 by Th17 cells, and downregulated Rorc encoding RORγt and its downstream Il17 . RSV significantly inhibited the RORγt transcription activity and bound to its ligand binding domain. Conclusion RSV alleviates murine colitis through regulating intestinal immunity. Notably, RSV is identified as a novel regulator of Th17 cells that inhibits RORγt-mediated Th17 differentiation. These findings potentiate the Rhodiola -derived natural chemicals as novel anti-colitis agents.

Anti-senescence effects of Rhodiola crenulate extracts on LO2 cells and bioactive compounds

Tisong Liang,Jiexin Zhou,Pu Jing,Zhengjun He,Shunshan Jiao,Wenji Zhao,Qi Tong,Guofu Jia

Ethnopharmacological relevance Rhodiola crenulata (Rc) is a traditional herb, used in Tibetan medicine, has shown promise efficacy in physical performance improvement, work capacity enhancement, fatigue elimination, and altitude sickness prevention. Also, Rc exhibited therapeutic effects on aging-related diseases. However, relevant researches on Rc and their bioactive components are quite few and needs further investigation. Aim of the study : The objective of this study was to understand the relationship between phytochemical profiles and their activities of Rc extracts. Materials and methods Rc extracts prepared by solvents with various hydrophilicity (i.e. aqueous ethanol (70%, v/v), water, and ethyl acetate), and their chemical compositions and specific compounds were analyzed by chemical analysis method and ultra-performance liquid chromatography quadruple time-of-flight mass spectrometry (UPLC-QTOF-MS). The regulate effects of Rc extracts on senescence and antioxidant activity were evaluated using the models of LO 2 cells and Caenorhabditis elegans . Results The 70% ethanol extracts exhibited better regulating effects on senescence via the assays of senescence -associated β-galactosidase (SAβG) staining and lifespan, which was consistent with the higher antioxidant activities observed based on the results of antioxidant assays. A total of 14 phytochemicals have been identified in 70% ethanol extracts, whereas the other two extracts contained much fewer compounds in varieties. Phytochemical profile of water extract was similar to the first half (polar compounds, running time: 0–6 min) of 70% ethanol extract profile, while those of ethyl acetate extract was consistent with its second half (more nonpolar compounds, running time: 6–12 min). Conclusions The 14 phytochemicals in Rc might exhibit additive or synergistic effects on senescence regulating and antioxidant activities, providing theoretical basis for daily administration of Rc.

A Novel UDP-Glycosyltransferase of Rhodiola crenulata Converts Tyrosol to Specifically Produce Icariside D2

Zhihua Liao,Fei Qiu,Junlan Zeng,Li Gu,Bangjun Wang,Xiaozhong Lan,Min Chen

Rhodiola crenulata is a Tibetan native herbal plant belonging to the family of Crassulaceae, which produces the pharmaceutical icariside D2 with the activities of inhibiting angiotensin-converting enzyme and killing leukemia cancer cells. In this study, we functionally characterized a novel UDP-glycosyltransferase (RcUGT1) that converted tyrosol to specifically produce icariside D2 from R. crenulata at molecular and biochemical levels. RcUGT1 was highly expressed in flowers and roots, while the icariside D2 content was much higher in stems than that in other organs, suggesting the potential translocation of icariside D2 from flowers and roots to stems. The high production of icariside D2 in stems provided a reasonable suggestion to farmers to harvest stems instead of roots for icariside D2 production. Enzymatic assays of recombinant RcUGT1 indicated that it converted tyrosol to specifically form icariside D2, with the values of Km 0.97±0.10 mM, Vmax 286±8.26 pKat/mg, Kcat 0.01552 s−1, and Kcat/Km 159.55 s−1 M−1. Functional identification of RcUGT1 facilitated the icariside D2 production through metabolic engineering in plants or synthetic biology in microbes.

Multifunctional Chitosan Composite Films Reinforced With Rhodiola rosea Extract/ZnO/Palygorskite Nanocomposites for Fruit Preservation

Wenting Gao,Bin Mu,Fangfang Yang,Yalong Li,Yu Tang,Zhihe Luo,Aiqin Wang

The potential of chitosan (CS) films for the preservation has been explored, and it is recognized as one of the promising approaches to enhancing the comprehensive capability of CS films for food control after incorporation of functional nanofillers. Herein, Rhodiola rosea extract/ZnO/palygorskite (RRE/ZnO/Pal) nanocomposites were facilely prepared and incorporated into CS matrix to obtain RRE/ZnO/Pal/CS (RZPC) composite films, in which ZnO/Pal nanocomposites were synthesized by a green phyto-mediated method followed by loading of R. rosea extract via a semi-dry grinding process to form organic/inorganic bioactive nanocomposites. The composite films exhibited significantly enhanced antioxidant, antibacterial, mechanical and UV-shielding properties compared with the pure CS film, as well as the maximum scavenging activities on hydroxyl radicals and 2,2-diphenyl-1-picrylhydrazine radicals of composite films were 92.85% ± 1.59% and 56.89% ± 0.56%, respectively. The antibacterial ratios of composite films containing 6.25% of RRE/ZnO/Pal nanocomposites were 94.92% ± 2.67% and 95.48% ± 2.08% for E. coli and S. aureus , respectively. In addition, the RZPC films effectively delayed browning and deterioration of bananas. This study provides a green method for construction of bioactive nanocomposites and new strategy to design multifunctional CS films for food preservation based on the synergistic effect of organic and inorganic components.

A Rhodiola-derived acidic glycopeptide maintains sleep homeostasis by regulating brain lipid metabolism in Drosophila

Rong Xu,Yingna Li,Xinyue Chen,Peiyao Gao,Chunlei Ji,Yiming Zhao,Chuang Xue,Hui Li,Changjiu Cai,Fangbing Liu,Liwei Sun

Background Sleep deprivation is closely associated with metabolic dysregulation, yet effective preventive interventions remain limited. Although Rhodiola rosea ( R. rosea ) exhibits promising sleep-modulating potential, the specific active components mediating its effects and the underlying mechanisms remain poorly elucidated. Purpose This study aimed to identify the sleep-regulatory component of R. rosea and elucidate its molecular mechanism. Methods A bioactivity-guided fractionation strategy was applied using a sleep-deprived Drosophila melanogaster model. The active fraction was purified and structurally characterized by chromatographic separation and spectroscopic analyses. Sleep behavior, circadian rhythmicity, oxidative stress indices, neurotransmitter levels, lipid metabolic parameters, and brain transcriptomic profiles were systematically evaluated. Mechanistic validation was performed using neuronal genetic manipulation and dual-luciferase reporter assays targeting lipid metabolism-related pathways. Results An acidic glycopeptide fraction derived from R. rosea (WRPA, 76 kDa) markedly increased total sleep duration by approximately 20%, reduced sleep fragmentation by approximately 20%, and improved circadian stability, locomotor activity, and cognitive performance in sleep-deprived flies. WRPA significantly alleviated oxidative stress by reducing reactive oxygen species (ROS) and malondialdehyde (MDA) levels and restoring antioxidant capacity, while normalizing gamma-aminobutyric acid (GABA) and 5-hydroxytryptamine (5-HT) levels in the brain. Transcriptomic analysis revealed lipid metabolism as the primary pathway modulated by WRPA, with acetyl-CoA carboxylase (ACC) identified as a key target. WRPA suppressed ACC expression by inhibiting sterol regulatory element-binding protein (SREBP)-mediated transcription, leading to decreased triglyceride, cholesterol, and free fatty acid accumulation. Importantly, WRPA effectively rescued sleep disturbances induced by neuronal ACC overexpression, confirming an SREBP/ACC-dependent mechanism. Conclusion WRPA is identified as a novel sleep-regulatory acidic glycopeptide derived from R. rosea , which restores sleep homeostasis by maintaining lipid metabolic stability through the SREBP-ACC signaling axis. These findings expand the pharmacological repertoire of R. rosea beyond small molecules and highlight glycopeptides as promising lead candidates for metabolic sleep disorder intervention.

Fermentative Production of Phenolic Glucosides by Escherichia coli with an Engineered Glucosyltransferase from Rhodiola sachalinensis

Qinglin He,Hua Yin,Jingjie Jiang,Yanfen Bai,Ning Chen,Shaowei Liu,Yibin Zhuang,Tao Liu

Three rosmarinic acid analogs produced by recombinant Escherichia coli, two xanthones from fungi and honokiol from plants, were explored as the substrates of E. coli harboring a glucosyltransferase mutant UGT73B6FS to generate phenolic glucosides. Six new and two known compounds were isolated from the fermentation broth of the recombinant strain of the feeding experiments, and the compounds were identified by spectroscopy. The biotransformation of rosmarinic acid analogs and xanthones into corresponding glucosides was presented for the first time. This study not only demonstrated the substrate flexibility of the glucosyltransferase mutant UGT73B6FS toward aromatic alcohols but also provided an effective and economical method to produce phenolic glucosides by fermentation circumventing the use of expensive precursor UDP-glucose.
