This study investigated the regulatory role of Rhodopseudomonas palustris RP11 in alleviating TBBPA-induced harmful effects in soybean seedlings. In this study, the characteristics of growth promotion by strain RP11 were studied by analysing 5-aminolevulinic acid (ALA) and indole-3-acetic acid (IAA) production, as well as phosphorus-solubilizing and potassium-solubilizing ability. In the pot culture conditions, we tested whether strain RP11 improved soybean seedlings tolerance against TBBPA by measuring the root length and physiological parameters of the seedlings treated with strain RP11 and different concentration of TBBPA (0, 5, 50, 100, and 1000 mg/kg) together. The results showed that strain RP11 secreted IAA and ALA, and solubilized phosphate and potassium. In pot trials, strain RP11 increased the root length, chlorophyll content, carotenoid content, soluble sugar and protein content of soybean seedlings treated with TBBPA, in comparison with the seedlings treated only with TBBPA. Furthermore, strain RP11 induced the activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD), decreased the malondialdehyde (MDA) content in soybean seedlings under TBBPA stress. It was concluded that strain RP11 alleviated TBBPA-induced harmful effects in soybean seedlings by the secretion of IAA and ALA, the accumulation of carotenoid, soluble sugar and soluble protein, and the induction of SOD, CAT and POD as well as nutrient adjustment of phosphorus and potassium levels.

Utilizing solar energy for hydrogen production by combining light-activated materials and biocatalysts has become a promising alternative to fossil fuels. Herein, a feasible and simple bio-hybrid complex based on water-soluble oligofluorene (OF), polythiophene (PTP) and Rhodopseudomonas palustris (R. palustris), one kind of photosynthetic bacteria, was constructed for enhancing photocatalytic hydrogen production. Under the irradiation of visible light, there was fluorescence resonance energy transfer (FRET) between OF and PTP, which amplified photoelectron signals and transferred electrons via PTP to methyl viologen (MV2+), an electron mediator that established electron transfer pathways between organic materials and R. palustris. Hydrogen production involving R. palustris was catalyzed by nitrogenase. It is noteworthy that MV2+ was reduced to MV+˙ and transferred electrons to nitrogenase in R. palustris, converting protons and electrons to molecular hydrogen. Besides, triethanolamine (TEOA) was added as an electron donor to provide electrons to PTP. The proposed OF/PTP/MV2+/TEOA/R. palustris system provided a feasible strategy for photocatalytic hydrogen production with low-cost and easily implemented techniques.

Electrochemical technology is a promising technique for separating ammonia from mature landfill leachate . However, the accompanying migration and transformation of coexisting pollutants and strategies for further high-value resourceful utilization of ammonia have rarely received attention. In this study, an electrochemical separation- Rhodopseudomonas palustris electrolysis cell coupled system was initially constructed for efficient separation and conversion of nitrogen in mature landfill leachate to microbial protein with synchronously tracking the transport and conversion of coexisting heavy metals accompanying the process. The results revealed that ammonia concentration in the cathode increased from 40.3 to 49.8% with increasing the current density from 20 to 40 mA/cm 2 , with less than 3% of ammonia transformation to NO 2 − -N and NO 3 − -N. During ammonia separation, approximately 95% of HM-DOMs (Cr, Cu, Ni, Pb, and Zn) were released into the anolyte due to humus degradation and further diffused to the cathode. A significant correlation was observed between the releases of HM-DOMs. Cu-DOMs accounted for 70.2% of the total Cu content, which was the highest proportion among the heavy metals (HMs). Among the HMs in anolyte, 57.4% of Pb, 52.5% of Ni, and 50.6% of Zn diffused to the cathode, and most of the HMs were removed in the form of hydroxide precipitations due to heavy alkaline catholyte . Compared with the open-circuit condition, the utilization efficiency of NH 4 + -N in the R. palustris electrolysis cell increased by 445.1% with 47% and 50% increases in final NH 4 + -N conversion rate and R. palustris biomass, respectively, due to bio-electrochemical enhanced phototrophic metabolism and acid generation for buffering the strong alkalinity of the electrolyte to maintain suitable growth conditions for R. palustris .

Background Whole-cell biocatalysis has been exploited to convert a variety of substrates into high-value bulk or chiral fine chemicals. However, the traditional whole-cell biocatalysis typically utilizes the heterotrophic microbes as the biocatalyst, which requires carbohydrates to power the cofactor (ATP, NAD (P)H) regeneration. Results In this study, we sought to harness purple non-sulfur photosynthetic bacterium (PNSB) as the biocatalyst to achieve light-driven cofactor regeneration for cascade biocatalysis. We substantially improved the performance of Rhodopseudomonas palustris -based biocatalysis using a highly active and conditional expression system, blocking the side-reactions, controlling the feeding strategy, and attenuating the light shading effect. Under light-anaerobic conditions, we found that 50 mM ferulic acid could be completely converted to vanillyl alcohol using the recombinant strain with 100% efficiency, and > 99.9% conversion of 50 mM p -coumaric acid to p -hydroxybenzyl alcohol was similarly achieved. Moreover, we examined the isoprenol utilization pathway for pinene synthesis and 92% conversion of 30 mM isoprenol to pinene was obtained. Conclusions Taken together, these results suggested that R. palustris could be a promising host for light-powered biotransformation, which offers an efficient approach for synthesizing value-added chemicals in a green and sustainable manner. Antibiotic residues in effluents discharged from wastewater treatment plants (WWTPs) have been considered high-risk organics due to biorefractory property and potential toxicity. Secondary pollution and unsustainability existed in advanced treatment of secondary effluent are currently in urgent need of improvement. In this study, a sustainably green strategy based on Rhodopseudomonas palustris ( R.palustris ) by regulating the structure of extracellular polymeric substances (EPS) was proposed for the first time to achieve efficiently removal of sulfadiazine (SDZ). Results showed that 0.2 V was the optimal external potential for R.palustris to efficiently remove SDZ, where the biodegradation rate constant obtained at this potential was 4.87-folds higher than that in open-circuit mode and a complete removal was achieved within 58 h in the presence of EPS extracted at this potential. Three-dimensional excitation-emission matrix (3D-EEM) spectra analysis suggested that tryptophan protein-like, tyrosine protein-like, humic acid-like and fulvic acid-like substances present in EPS were the main effective components which was responsible for the indirect photodegradation of SDZ. The quenching experiments showed that 3 EPS* was the dominant reactive species which accounted for 90% of SDZ removal. This study provides new implications for the advanced treatment of secondary effluent organic matters by developing eco-friendly bioaugmentation technology and biomaterials.

Ammonium (NH 4 + ) transport across cell membranes plays an important role in assimilation or removal of the environmental nitrogen. The membrane protein AmtB has been considered to be the NH 4 + transporter that is responsible for the NH 4 + uptake. The phototrophic diazotroph Rhodopseudomonas palustris harboring two amtB genes has been widely used in wastewater treatment and bioremediation. However, the role of AmtB in NH 4 + uptake remains unclear in R. palustris . Here, we employed an innovative approach combining stable isotope probing (SIP) with Raman spectroscopy to determine the physiological functions of AmtB1 and AmtB2 in R. palustris. This powerful technique allowed us to investigate NH 4 + uptake at the single-cell level. The generated R. palustris Δ amtB1 Δ amtB2 mutant lacking AmtB1 and AmtB2 proteins was still capable of utilizing 15 NH 4 + even the 15 NH 4 + concentration was as low as 5 μM. These data demonstrate that both of the AmtB proteins are not essential for R. palustris to take up NH 4 + regardless of environmental NH 4 + levels. However, both AmtB1 and AmtB2 can contribute to NH 4 + uptake under nitrogen-limiting conditions. Given that R. palustris primarily expresses AmtB2 in these conditions, AmtB2 plays a more important role in NH 4 + uptake compared to AmtB1. In addition, transcriptomic analysis showed that the deletion of the amtB1 and amtB2 genes resulted in the upregulation of many transporter genes, providing potential targets for future investigation of alternative NH 4 + uptake systems.
