A study on the anti-corrosion mechanism of the eco-friendly inhibitor Ribavirin on Q235 steel in an acidic chloride environment

Hongmei Dong,Lifeng Ding,Boyuan Ran,Yongbo Song,Ruihong Wang,Lu Zhao,Yulan Niu

Developing efficient and eco-friendly corrosion inhibitors to replace traditional toxic and harmful ones is necessary for protecting the ecological environment. Herein, we have developed an eco-friendly drug corrosion inhibitor, Ribavirin (RBVR), which could effectively inhibit Q235 steel corrosion in 1 M HCl solution. EIS results showed that RBVR slowed down the corrosion rate of steel, and η could reach up to 97.7 % when the concentration of RBVR was only 10 mM. The potentiodynamic polarization curve results indicated that RBVR was a modest mixed-type corrosion inhibitor that could inhibit steel anodic and cathodic reactions. The adsorption isotherm and theoretical calculations revealed that the adsorption of RBVR on the steel surface was in the form of parallel adsorption, which maximized the protection of steel. The adsorption of RBVR molecules on the steel was mainly based on the N and O heteroatoms as the active adsorption centers. A monolayer protective film was formed on the steel surface, which retarded the dissolution of active sites. This work will provide a theoretical basis for developing novel, efficient, eco-friendly organic inhibitors and promoting their industrial application.

Occurrence and risk assessment of typical PPCPs and biodegradation pathway of ribavirin in wastewater treatment plants

Qixin Liu,Xuan Feng,Ning Chen,Fei Shen,Haichuan Zhang,Shuo Wang,Zhiya Sheng,Ji Li

A large number of pharmaceuticals and personal care products (PPCPs) persist in wastewater, and the consumption of PPCPs for COVID-19 control and prevention has sharply increased during the pandemic. This study investigated the occurrence, removal efficiency, and risk assessment of six typical PPCPs commonly used in China in two wastewater treatment plants (WWTPs). Ribavirin (RBV) is an effective pharmaceutical for severely ill patients with COVID-19, and the possible biodegradation pathway of RBV by activated sludge was discovered. The experimental results showed that PPCPs were detected in two WWTPs with a detection rate of 100% and concentrations ranging between 612 and 2323 ng L −1 . The detection frequency and concentrations of RBV were substantially higher, with a maximum concentration of 314 ng L −1 . Relatively high pollution loads were found for the following PPCPs from influent: ibuprofen > ranitidine hydrochloride > RBV > ampicillin sodium > clozapine > sulfamethoxazole. The removal efficiency of PPCPs was closely related to adsorption and biodegradation in activated sludge, and the moving bed biofilm reactor (MBBR) had a higher removal capacity than the anoxic-anaerobic-anoxic-oxic (AAAO) process. The removal efficiencies of sulfamethoxazole, ampicillin sodium, ibuprofen, and clozapine ranged from 92.21% to 97.86% in MBBR process and were relatively low, from 61.82% to 97.62% in AAAO process, and the removal of RBV and ranitidine hydrochloride were lower than 42.96% in both MBBR and AAAO processes. The discrepancy in removal efficiency is caused by temperature, hydrophilicity, and hydrophobicity of the compound, and acidity and alkalinity. The transformation products of RBV in activated sludge were detected and identified, and the biodegradation process of RBV could be speculated as follows: first breaks into TCONH 2 and an oxygen-containing five-membered heterocyclic ring under the nucleosidase reaction, and then TCONH 2 is finally formed into TCOOH through amide hydrolysis. Aquatic ecological risks based on risk quotient (RQ) assessment showed that PPCPs had high and medium risks in the influent, and the RQ values were all reduced after MBBR and AAAO treatment. Ranitidine hydrochloride and clozapine still showed high and medium risks in the effluent, respectively, and thus presented potential risks to the aquatic ecosystem.

Crystallization Selectivity of Ribavirin Solution and Amorphous Phase

Fuying Li,Shiying Chen,Haoxin Hu,Chengfeng Liang,Shiyu Sun,Can Jin,Fenghua Chen

Crystallization selectivity is an important principle in polymorph control. Ribavirin Form I, Form II, DMSO solvate, and amorphous ribavirin are prepared, and the short-range order similarities between these solid forms and ribavirin aqueous solution and DMSO solution are compared via mid-frequency Raman difference spectra (MFRDS). The crystallization process from amorphous ribavirin to Form I and from solution to amorphous phase is explained. Reasons for the difficulty in preparing the DMSO solvate are proposed. The rationale provided for the crystallization selectivity provides a foundation for the synthesis of metastable phases with a robust and convenient method. Keywords:ribavirin;crystallization selectivity;solution;amorphous phase;solvate;mid-frequency Raman difference spectra

Ribavirin inhibits peste des petits ruminants virus proliferation in vitro

Weifeng Zhang,Hualong Deng,Yanfen Liu,Shaohong Chen,You Liu,Yuntao Zhao

Peste des petits ruminants virus (PPRV), a member of the family Paramyxoviridae, belongs to the genus Morbillivirus. It causes devastating viral diseases in small ruminants and has been rapidly spreading over various regions in Africa, the Middle East, and Asia. Although vaccination is thought to be an effective management strategy against PPR infections, the heat sensitivity of PPRV vaccines severely restricts their use in regions with hot climates. In this research, we studied the antiviral activities of ribavirin and aimed to understand the potential mechanisms of action of ribavirin in the African green monkey kidney cells (Vero cells). In brief, the adsorption, intrusion, replication, and release of PPRV, as well as the mRNA expression level of RNA-dependent RNA polymerase ( RdRp ), were significantly inhibited in the ribavirin-treated Vero cells compared to those in the PPRV-infected cells that were not treated with ribavirin. Additionally, ribavirin has potential as an antiviral drug against PPRV, and its antiviral activity is mediated by the Janus kinase signal transducer and activator of transcription (JAK/STAT) and PI3K/AKT pathways.

Pharmaceutical Cocrystals of Ribavirin with Reduced Release Rates

Jia-Mei Chen,Song Li,Tong-Bu Lu

Cocrystals have been extensively utilized to improve drugs’ properties. Ribavirin is a water-soluble broad-spectrum antiviral drug and its application is severely limited by the peak-to-trough fluctuation in plasma drug concentrations and some undesirable side-effects. We show here that formation of cocrystals may be a useful approach to overcome this problem by reducing the release rate of ribavirin. Three cocrystals of ribavirin with 3,5-dihydroxybenzoic acid (1), gallic acid (2), and barbituric acid (3) were successfully prepared and characterized by powder and single crystal X-ray diffraction, infrared spectroscopy, differential scanning calorimetry, and thermogravimetric analysis, as well as dynamic vapor sorption measurement. The dissolution process revealed that 1–3 showed a reduced release rate as compared to ribavirin in the buffer solution representing intestinal pH 6.8. This study indicates that the release rate of ribavirin can be manipulated over a wide range by the formation of cocrystals, which may subsequently help lower its peak-to-trough fluctuation in plasma concentrations.

Precise selection and separation of ribavirin by nanoconfined imprinted MOFs membrane

Jing Yan,Yingrui Sui,Faguang Ma,Jian Lu,Yilin Wu

Metal-organic frameworks (MOFs) are highly tunable materials with potential as porous media in membrane separations. However, traditional MOFs are usually not specific and selective. Here, we demonstrate a nanocomposite molecularly imprinted MOFs membrane material that can selectively adsorb and separate specific molecules. We demonstrated this adsorption-separation mechanism by introducing ribavirin molecules into multicomponent MOFs by molecular imprinting strategy and performing permeation and dynamic permeation tests. Due to the previously formed nanocages with a specific structure inside the MOFs, combined with the synergistic effect of the subsequent secondary imprinting, excellent selectivity (more than 6) and effective adsorption (46.46 mg g −1 ) were displayed by the obtained imprinted membrane. The stability of the membrane was measured by the process of dynamic osmosis, and the results showed that the membrane always had a large factor of separation (greater than 7). The experimental results and conclusions of this paper provide new theoretical and practical support for solving the key scientific issues of the “trade-off” effect between the permeation flux and permeation selectivity of template molecules in the traditional molecularly imprinted membranes (MIMs) osmotic separation process.

Transformation of antiviral ribavirin during ozone/PMS intensified disinfection amid COVID-19 pandemic

Xiaoyu Liu,Yuntao Hong,Shunke Ding,Wei Jin,Shengkun Dong,Rong Xiao,Wenhai Chu

Due to the spread of coronavirus disease 2019 (COVID-19), large amounts of antivirals were consumed and released into wastewater, posing risks to the ecosystem and human health. Ozonation is commonly utilized as pre-oxidation process to enhance the disinfection of hospital wastewater during COVID-19 spread. In this study, the transformation of ribavirin, antiviral for COVID-19, during ozone/PMS‑chlorine intensified disinfection process was investigated. •OH followed by O 3 accounted for the dominant ribavirin degradation in most conditions due to higher reaction rate constant between ribavirin and •OH vs. SO 4 • − (1.9 × 10 9 vs. 7.9 × 10 7 M −1 s −1 , respectively). During the O 3 /PMS process, ribavirin was dehydrogenated at the hydroxyl groups first, then lost the amide or the methanol group. Chloride at low concentrations (e.g., 0.5– 2 mg/L) slightly accelerated ribavirin degradation, while bromide, iodide, bicarbonate, and dissolved organic matter all reduced the degradation efficiency. In the presence of bromide, O 3 /PMS process resulted in the formation of organic brominated oxidation by-products (OBPs), the concentration of which increased with increasing bromide dosage. However, the formation of halogenated OBPs was negligible when chloride or iodide existed. Compared to the O 3 /H 2 O 2 process, the concentration of brominated OBPs was significantly higher after ozonation or the O 3 /PMS process. This study suggests that the potential risks of the organic brominated OBPs should be taken into consideration when ozonation and ozone-based processes are used to enhance disinfection in the presence of bromide amid COVID-19 pandemic.

Antiviral effect of copper chloride on feline calicivirus and synergy with ribavirin in vitro

Li Dengliang,Cui Zhanding,Li Guohua,Zhang Liangting,Zhang Ying,Zhao Han,Zhang Shuang,Guo Yanbing,Zhao Yanli,Men Fanxing,Zhao Shihui,Shao Jiang,Du Dongju,Huang Hailong,Wang Kai,Hu Guixue,Li Tiansong,Zhao Yongkun

Background Feline calicivirus (FCV) is a common and highly prevalent pathogen causing upper respiratory diseases in kittens and felines in recent years. Due to the substantial genetic variability of the viral genes, existing vaccines cannot provide complete protection. Therefore, research on FCV antiviral drugs has received much attention. Results In this study, we found that copper chloride had dose-dependent antiviral effects on FCV in F81 cells. We also found that the combination of copper chloride and ribavirin had a synergistic protective effect against FCV in F81 cells. In contrast, the combination of copper chloride and horse anti-FCV immunoglobulin F (ab’)2 showed an antagonistic effect, likely because copper chloride has an effect on F (ab’)2 immunoglobulin; however, further research is needed to clarify this supposition. Conclusion sIn summary, we found that copper chloride had low cytotoxicity and significant antiviral effects on FCV in F81 cells, providing a new drug candidate for the prevention and treatment of FCV infection.

Degradation of ribavirin by Fe2+/PS oxidation technology: performance, mechanism and toxicity control

Xiaohui Sun,Wei Li,Zijun Dong,Yunhe Hou,Yuyang Ning,Chenyu Wang,Guo Lv

During the COVID-19 pandemic, the extensive use of ribavirin (RBV) raised concerns about its environmental residues and associated risks, necessitating remedial actions. This study investigates the degradation efficiency, mechanism, and biotoxicity of RBV using ferrous (Fe 2+ ) activated persulfate (PS) advanced oxidation technology (Fe 2+ /PS oxidation). Experiments conducted under various conditions revealed that at pH = 3, PS concentration of 4 mM, PS/Fe 2+ molar ratio of 2:1, and citric acid of 0.5 mM with Fe 2+ added twice, the degradation rate of RBV reached 98.70 %. The degradation process of RBV by Fe 2+ /PS from 0 to 5 mins followed a first-order reaction kinetics model. The presence of halide ions (Cl - , Br - , I - ) was found to inhibit the degradation efficiency. The active species involved were identified as SO 4 - ·, ·OH, 1 O 2 , and FeO 2+ , with SO 4 - · and ·OH being the most significant. Moreover, the products of ribavirin were determined and their degradation pathways were proposed. Dehydration, deamidation, C-N bond breaking and hydroxylation were the major pathways. Additionally, toxicity assessment showed that Fe 2+ /PS oxidation reduced the inhibition rate of bioluminescence in Vibrio fischeri from 31.58 % to 3.88 %, effectively controlling RBV toxicity during the reaction. This study provides insights into managing water environmental risks associated with pharmaceutical residues in the post-pandemic era.

Effect of Hydrochemical Factors on the Retention and Transport of Ribavirin in Saturated Sand and Limestone Porous Media

Shi Yanfeng,Li Bingxu,Shi Yuzhi,Sun Yuanyuan,Ding Botao,Zhang Yanhao,Zhang Xu,Zhang Zhibin,Li Fulin

The antiviral drug ribavirin is widely used and continuously released into the environment through various pathways, which will cause serious water pollution and virus resistance. However, only few works have examined ribavirin retention and transport groundwater environment. This study investigates the transport behavior of ribavirin in saturated quartz sand and limestone porous media, considering ionic strength (IS), ionic type and humic acid (HA), through column experiments and mathematical modeling. The findings reveal that, ribavirin exhibits strong mobility within saturated quartz sand media is less affected by different hydrochemical factors, posing a high risk of migration. In limestone porous media, the migration of ribavirin decreased. In comparison to Na + , the presence of Ca 2+ causes the effluent recovery rate of ribavirin decreases. And as the IS of Ca 2+ background solution increases, the effluent recovery rate of ribavirin also decreases. The presence of HA causes more ribavirin to flow out of the column, and its effect becomes more pronounced with increasing concentration. Furthermore, simulated precipitation scenarios, the ribavirin adsorbed on the of media surface is released again and transport with the water flow. These findings enhance our understanding about the environmental fate of ribavirin and facilitating ecological restoration efforts.

Effects of moroxydine hydrochloride and ribavirin on the cellular growth and immune responses by inhibition of GCRV proliferation

Xiao-Bo Yu,Kai Hao,Jing Li,Xiao-Hui Chen,Gao-Xue Wang,Fei Ling

Moroxydine hydrochloride (Mor) and ribavirin (Rib) are known for their multi-antiviral activities against DNA and RNA viruses but little information is available about the pharmacological impact in aquaculture. The present study was undertaken to investigate the response of host cells to antiviral compounds during the anti-GCRV treatment. The scanning electron microscope results showed that Ctenopharyngodon idella kidney (CIK) cells have a higher death rate at 72   h post virus infection. At the concentration of 40   μg   mL −   1 , Mor and Rib had a significant protective effect on virus-infected cells. Moreover, the gene expressions of vp5, vp6 and NS66 were significantly inhibited by treatment with Mor or Rib, especially gene expression of the vp5 . For the immunoregulatory action, no distinct induction of the expression of immune genes was observed after the addition of Mor and Rib to the virus-free cells. However, the compounds significantly decreased the virus-induced gene overexpression of Myd88, Mx1, IL-1β, IL-8, I-IFN and TNFα in CIK cells. Moreover, Mor and Rib significantly inhibited the immune genes upregulation which was induced by GCRV in kidney, liver, muscle and gill of grass carp, despite greater partial gene expressions were detected than the virus-free control group. Besides, Mor and Rib blocked cell cycle changes, cytopathic effects, cellular death and virus proliferation in CIK cells thereby maintaining normal morphological structure. Overall, Mor and Rib as antiviral compounds are effective for the control of GCRV replication and the indirectly regulation of cellular immune response.

The isolation of high-affinity ssDNA aptamer for the detection of ribavirin in chicken

Mingyan Song,Chen Lyu,Nuo Duan,Shijia Wu,Imran Mahmood Khan,Zhouping Wang

The abuse of ribavirin, an antiviral drug, in poultry breeding can cause quality degradation and drug resistance. So it is of great importance to establish a simple and effective method for detecting ribavirin in foods. In this work, aptamers that could especially bind to ribavirin with high affinity were obtained by the Capture-SELEX method. After 15 rounds of enrichment, ssDNA library pool was enriched and then analyzed by high-throughput sequencing. The seven most enriched sequences were selected as candidate aptamers for affinity and specificity characterization. Among the candidate aptamers, APT-1 was proved to be the optimal aptamer. The dissociation constant (Kd) values of APT-1 obtained by the two methods of colorimetric and fluorescence were 34.34 ± 6.038 nmol L−1, 61.19 ± 21.48 nmol L−1, respectively. To study the binding mechanism of the selected aptamer, molecular docking was conducted and results indicated that hydrogen bonds were formed at binding sites located at G37, T38, A40, T53 and A54. Furthermore, to confirm the practicability of the selected aptamer, a fluorescence assay was designed, showing the liner range within 1.0–50 ng mL−1 and the low detection limit of 0.67 ng mL−1. Besides, the aptamer was applied for the detection of ribavirin in chicken samples and the recoveries ranged from 87.26% to 105.57%, which showed great application potential in food safety.
