Leaf defense system of Robinia pseudoacacia L. seedlings exposed to 3 years of elevated atmospheric CO2 and Cd-contaminated soils

X. Jia,Y.H. Zhao,T. Liu,Y.H. He

Short-term exposure to elevated CO 2 increases cadmium (Cd) uptake in some plant species (wheat, poplars, and willows), which triggers an increase in antioxidative system activity to deal with additional reactive oxygen species that are generated. Here, we examined leaf defenses in Robinia pseudoacacia L. seedlings exposed to elevated CO 2   +   Cd for 3   years. Three years of elevated CO 2 decreased Cd uptake into leaves and the Cd content in soils and increased the pH of rhizosphere soil relative to ambient CO 2 . In plants exposed to Cd stress , leaf chlorophyll content was greater under elevated CO 2 than under ambient CO 2 . Superoxide dismutase, peroxidase, and catalase activity increased, glutathione content increased, and malondialdehyde and phytochelatins contents decreased under elevated CO 2   +   Cd relative to Cd alone. Proline, soluble sugars, flavonoids, saponins, and phenolic acids contents were greater under elevated CO 2   +   Cd than under Cd alone, and condensed tannin content was lower. Overall, long-term elevation of CO 2 enhanced the leaf defense system of R. pseudoacacia exposed to Cd by stimulating antioxidant enzyme activity , osmotic adjustment, and the production of glutathione, flavonoids and phenolic acids. Future research should focus on understanding the mechanisms involved in the decrease in Cd uptake into leaves and Cd content in soils and the increase in rhizosphere soil pH under long-term exposure to elevated CO 2 .

Three years of exposure to lead and elevated CO2 affects lead accumulation and leaf defenses in Robinia pseudoacacia L. seedlings

Xia Jia,Chunyan Zhang,Yonghua Zhao,Tuo Liu,Yunhua He

Few studies have explored the long-term effects of elevated atmospheric CO 2 combined with lead (Pb) contamination on plants. The objective of this study was to examine the effects of 3 years of elevated CO 2 (700 ± 23 μmol mol –1 ) on Pb accumulation and plant defenses in leaves of Robinia pseudoacacia L. seedlings in exposed to Pb (500 mg kg −1 soil). Elevated CO 2 increased Pb accumulation in leaves and Pb removal rate in soils. In plants exposed to Pb stress, total chlorophyll and carotenoid contents in leaves were lower under elevated CO 2 than under ambient CO 2 , but seedling height and width increased under elevated CO 2 relative to ambient CO 2 . Elevated CO 2 significantly ( p  < .01) stimulated malondialdehyde content in leaves under Pb exposure. Superoxide dismutase and catalase activity increased significantly ( p  < .01), peroxidase activity decreased significantly ( p  < .01), and glutathione , cystine, and phytochelatin contents increased under elevated CO 2  + Pb relative to Pb alone. Elevated CO 2 stimulated the production of soluble sugars, proline, flavonoids , saponins, and phenolics in plants exposed to Pb stress. Ove rall, long-term elevation of CO 2 increased Pb-induced oxidative damage in seedlings, but enhanced the phytoextraction of Pb from contaminated soils.

Fermentation of Robinia pseudoacacia flower for improving the antioxidation: optimized conditions, active composition, mechanism, and biotransformation process

Yi Yang,Yunci Zhang,Dongyu Gu,Chang Liu,Yi Wang,Shanshan Tang,Yuxin Yin,Jing Tian

Robinia pseudoacacia flower is a natural product with many biological activities, including antioxidation. To further develop its antioxidation, the extract was fermented by Aspergillus niger FFCC 3112 in the medium with carbon to nitrogen ratio of 1.4:1 and initial pH of 4.2 for 3.5 days to form the best antioxidant activity of the fermentation product by strain screening, single factor optimization, and response surface methodology. Further analysis, isolation and activity determination showed that a main chemical component, kaempferol-3-O-α-L-rhamnopyranosyl-(1→6)-β-D-galactopyranosyl-7-O-α-L-rhamnopyranoside, in the extract was completely hydrolyzed to kaempferol-7-O-α-L-rhamnopyranoside and kaempferol with better antioxidant activity through biotransformation, which was the basis for improving the antioxidant activity of fermentation products. Moreover, the mechanism of antioxidant and the contribution of phenolic hydroxyl groups were investigated by density functional theory. The result indicated that the antioxidant capacity of kaempferol-7-O-α-L-rhamnopyranoside and kaempferol increased with the increase of solvent polarity. In high-polarity solvents, they mainly scavenge free radicals through single electron transfer followed by proton transfer.

Evaluation of antiviral effect and toxicity of total flavonoids extracted from Robinia pseudoacacia cv. idaho

Hao Guo,Xinhuan Wan,Fengju Niu,Jujie Sun,Chenxiao Shi,Jessica Meng Ye,Changzheng Zhou

In this study, we aimed to evaluate the antiviral effect of total flavonoids extracted from Robinia pseudoacacia cv. idaho (RPTF) in vivo and its toxicity on rats with oral gavage. RPTF was prepared by percolation with 70% ethanol for 24 h and its antiviral effect on different kinds of viruses was evaluated in vitro by MTT staining. The long-term toxicity of RPTF on rats was evaluated through the detection of general behavior, body weight, food intake and related organ tissue sections of experimental animals. We found that RPTF produced significantly inhibitory effects on HSV-1 and EV-71 viruses with the therapeutic index TI values 113.8 and 46.2, respectively. Moreover, toxicity evaluation in vivo showed no significantly adverse effects in rats, indicating that RPTF was safe in use. In conclusion, we demonstrated that RPTF, natural compounds in the Chinese traditional medicine, could act as promising and effective antiviral therapeutics with relative safety in use.

Induction and early identification of tetraploid black locust by hypocotyl in vitro

Li Xiuyu,Zhang Zijie,Ren Yunhui,Feng Yue,Guo Qi,Dong Li,Sun Yuhan,Li Yun

Tetraploid black locust ( Robinia pseudoacacia L.) has advantages over diploid varieties in growth and stress resistance. Previous studies had shown that tetraploid black locust can be induced only by treating shoot tips of the germinating seeds with colchicine. In order to establish a simpler and more efficient tetraploid induction method in black locust, we induced tetraploid plants by treating the hypocotyls from in vitro –grown seedlings with different concentrations of colchicine, and then provided a method for tetraploid identification based on stomatal size and density in this study. The highest induction efficiency was 53.33% by treating hypocotyl explants with 70 mg·L −1 colchicine for 2 d. Tetraploid black locust was significantly different from diploid in morphological characteristics. Leaf area, leaf thickness, chlorophyll content, stomatal length, and stomatal width were greater in tetraploid plants than in diploid plants, whereas the opposite was true for leaf aspect ratio, stomatal index, and stomatal density. The differences in stomatal size and density between diploid and tetraploid were nearly two times; therefore, size and density of stomata can be used as a stable and distinct marker to rapidly screen tetraploids. The results of this study suggested that the hypocotyl was an ideal material to induce tetraploid black locust in vitro and confirmed that stomatal size and density could be an effective and reliable marker to identify ploidy level in black locust.

Leaf Age-Dependent Volatile Cues Influence Host Location and Oviposition Preference of Obolodiplosis robiniae on Robinia pseudoacacia

Weihan Xu,Jiaqiang Zhao,Qike Wang,Zhashenjiacan Bao,Yuan Xu,Haiwei Wu,Juan Shi

Simple SummaryObolodiplosis robiniae(Haldeman) is an important pest ofRobinia pseudoacaciaL., with damage mainly occurring on tender leaves. To better understand why females prefer these young tissues, we compared egg-laying behavior and odor responses of gravid females between tender leaves and mature leaves. Females laid many more eggs on tender leaves and were more attracted to odors released from tender leaves than to those from mature leaves. Chemical analysis further showed that the two leaf ages released different volatile blends. These results suggest that odors associated with leaf age may help females locate suitable oviposition sites. This study provides useful information for understanding host selection inO. robiniaeand may support the future development of environmentally friendly control strategies based on host plant volatile cues.Obolodiplosis robiniae(Haldeman) is a specialist herbivore ofRobinia pseudoacaciaL., and its infestation is closely associated with tender leaf tissues. The ability of gravid females to recognize suitable host tissues is essential for successful oviposition and subsequent population development. Here, we assessed whether leaf age affects the host-selection behavior ofO. robiniaeand whether volatile organic compounds are associated with this process. Laboratory oviposition assays were used to compare egg deposition on tender leaves and mature leaves ofR. pseudoacacia, and Y-tube olfactometer bioassays were performed to evaluate female responses to odors from the two leaf ages. Volatiles released from healthy tender leaves and mature leaves were collected using dynamic headspace sampling and characterized by thermal desorption–gas chromatography–mass spectrometry. Principal component analysis, orthogonal partial least squares discriminant analysis, and variable importance in projection scores were used to compare volatile profiles between leaf ages. Gravid females deposited significantly more eggs on tender leaves than on mature leaves in both choice and no-choice assays. Females also showed a significant olfactory preference for tender-leaf odors when directly offered a choice between volatiles from tender leaves and mature leaves, with 76.47% of responding individuals selecting tender-leaf odors and 23.53% selecting mature-leaf odors. Chemical profiling identified 28 volatile compounds across the two leaf ages, and their composition and relative abundance differed markedly. Among shared compounds, (Z)-3-hexen-1-ol and α-farnesene differed significantly between tender leaves and mature leaves. Multivariate analyses further identified several candidate compounds contributing to leaf age-related volatile differences. These results indicate that leaf age influences both oviposition behavior and odor-mediated host location inO. robiniae. Leaf age-dependent volatile blends may serve as important chemical cues associated with host selection by gravid females and provide a basis for future studies on volatile-mediated management strategies.

Hand-Held Electrostatic Spray Ionization Mass Spectrometry Enables On-Site Metabolomic Analysis of Sophora japonica and Robinia pseudoacacia

Song Mengyang,Luo Haiyan,Ji Zhenyang,Ye Zhihao,Yang Qinqin,Liu Xiaoyi,Shi Buyin,Li Yuze,Nie Zongxiu

Mass spectrometry plays a key role in agricultural and food chemistry, yet direct ionization of plant samples is complicated by endogenous salts and other matrix components that destabilize conventional direct-current nanoelectrospray ionization (DC-nanoESI). To address this limitation in point-of-need analysis, we developed hand-held electrostatic spray ionization (HESSI), which produces stable ionization from nanoliter sample volumes without direct electrical contact with the solution. A compact pulsed-discharge module and dielectric barrier limit electrochemical reactions and reduce the risk of discharge at the emitter while remaining compatible with salt-containing plant extracts. Approximately 50 nL of the sample sustained MS signals for up to 5 min. In a proof-of-concept application, 208 compounds were tentatively annotated from Sophora japonica and Robinia pseudoacacia extracts in negative-ion mode, and a neural network model distinguished the two groups with an accuracy of 83.3%. These results support HESSI as a compact and sample-efficient platform for the rapid mass-spectrometric characterization of botanical materials.

Growth under elevated air temperature alters secondary metabolites in Robinia pseudoacacia L. seedlings in Cd- and Pb-contaminated soils

Y.H. Zhao,X. Jia,W.K. Wang,T. Liu,S.P. Huang,M.Y. Yang

Plant secondary metabolites play a pivotal role in growth regulation, antioxidant activity, pigment development, and other processes. As the global climate changes, increasing atmospheric temperatures and contamination of soil by heavy metals co-occur in natural ecosystems, which alters the pH of rhizosphere soil and influences the bioavailability and mobility of metals. Elevated temperatures in combination with heavy metals are expected to affect plant secondary metabolites, but this issue has not been extensively examined. Here, we investigated secondary metabolites in Robinia pseudoacacia seedlings exposed to elevated temperatures using a passive warming device in combination with Cd- and Pb-contaminated soils. Heavy metals significantly stimulated the accumulation of saponins, phenolic compounds , and flavonoids in leaves and stems; alkaloid compounds increased in leaves and decreased in stems, and condensed tannins fluctuated. Elevated temperatures, alone and in combination with Cd and Pb, caused increases in secondary metabolites in the plant tissues. Phenolic compounds showed the greatest changes among the secondary metabolites and significant interactive effects of temperature and metals were observed. These results suggest that slightly elevated temperature could enhance protective and defense mechanisms of Robinia pseudoacacia seedlings exposed to heavy metals by stimulating the production of secondary metabolites.

Chemical Constituents Comparison Between the Flowers of Sophora japonica L. and Robinia pseudoacacia L. by UPLC-Q-TOF-MS/MS and HPLC

Cui-Cui Sun,Yi-Ting Chen,Hai-Xia Xu,Yu-Xian Guo,Qing-Feng Zhang

The flowers ofSophora japonicaL. (SJF) andRobinia pseudoacaciaL. (RPF) are edible and similar in appearance. The chemical constituents of SJF and RPF were compared by UPLC-Q-TOF-MS/MS and HPLC analysis in this study. A total of 29 and 19 constituents were identified in SJF and RPF, respectively. Flavonoid glycosides were the main constituents found in both flowers. The main aglycon moieties found in SJF were quercetin, kaempferol and isorhamnetin, whereas acacetin and kaempferol were the main ones found in RPF. Additionally, the content of flavonoids in SJF was significantly higher than that in RPF, as determined by HPLC. Rutin was the most dominant flavonoid in SJF with a content range of 72.31~88.15 mg/g, followed by quercetin (13.05~20.30 mg/g). Kaempferol-di(rhamnoside)-hexoside was the most dominant flavonoid in RPF with a content range of 25.94~30.00 mg/g. The distinct flavonoid profiles indicated the chemical non-equivalence of SJF and RPF. Therefore, RPF should not be considered a direct substitute for SJF in herbal medicine without further pharmacological and clinical validation.

Elevated CO2 increases glomalin-related soil protein (GRSP) in the rhizosphere of Robinia pseudoacacia L. seedlings in Pb- and Cd-contaminated soils

Xia Jia,Yonghua Zhao,Tuo Liu,Shuping Huang,Yafei Chang

Glomalin-related soil protein (GRSP), which contains glycoproteins produced by arbuscular mycorrhizal fungi (AMF), as well as non-mycorrhizal-related heat-stable proteins, lipids, and humic materials, is generally categorized into two fractions: easily extractable GRSP (EE-GRSP) and total GRSP (T-GRSP). GRSP plays an important role in soil carbon (C) sequestration and can stabilize heavy metals such as lead (Pb), cadmium (Cd), and manganese (Mn). Soil contamination by heavy metals is occurring in conjunction with rising atmospheric CO 2 in natural ecosystems due to human activities. However, the response of GRSP to elevated CO 2 combined with heavy metal contamination has not been widely reported. Here, we investigated the response of GRSP to elevated CO 2 in the rhizosphere of Robinia pseudoacacia L. seedlings in Pb- and Cd-contaminated soils. Elevated CO 2 (700 μmol mol −1 ) significantly increased T- and EE- GRSP concentrations in soils contaminated with Cd, Pb or Cd + Pb. GRSP contributed more carbon to the rhizosphere soil organic carbon pool under elevated CO 2  + heavy metals than under ambient CO 2 . The amount of Cd and Pb bound to GRSP was significantly higher under elevated (compared to ambient) CO 2 ; and elevated CO 2 increased the ratio of GRSP-bound Cd and Pb to total Cd and Pb. However, available Cd and Pb in rhizosphere soil under increased elevated CO 2 compared to ambient CO 2 . The combination of both metals and elevated CO 2 led to a significant increase in available Pb in rhizosphere soil compared to the Pb treatment alone. In conclusion, increased GRSP produced under elevated CO 2 could contribute to sequestration of soil pollutants by adsorption of Cd and Pb.

Lipase immobilization on multi-walled carbon nanotubes used as a target fishing tool and followed by molecular docking technique to analyze lipase inhibitor from Robinia pseudoacacia L.

Shuai Wang,Yunxiao Wang,Shunxin Wang,Shuang Guo,Dongyu Gu,Jifeng Wang,Yi Yang

In this study, R. pseudoacacia flowers exhibited potential lipase ( Aspergillus niger ) inhibitory activity. To quickly explore the active compound, ligand fishing was adopted for screening the ligand from R. pseudoacacia flowers by lipase immobilized on magnetic polyamidoamine dendrimer-coated multi-walled carbon nanotubes . After ligand fishing, the target compound was separated purposefully by optimizing the separation conditions of high-speed counter-current chromatography (HSCCC). As results, kaempferol with the IC 50 of 327.64 ± 15.33 μg/mL was separated and identified. The further molecular docking results indicated that kaempferol was interacted with amino acid residue in the active site of lipase through van der Waals force, hydrogen bonds and Pi-Pi T-shaped interactions, yielding the binding energy of − 6.43 kcal/mol. The present method not only revealed the lipase inhibitory substances in R. pseudoacacia , but also provided an effective means for the screening of other natural substances.
