The alleviating mechanism of Lycium barbarum polysaccharide on rheumatoid arthritis mediated by Romboutsia lituseburensis

Rongrong Sun,Yanghanxiu Liu,Mengyao Xia,Yihui Yang,Hao Meng,Qing Hu,Xichun Peng,Liu Liu,Jianming Luo

Our previous studies have shown that Lycium barbarum polysaccharide (LBP) alleviates rheumatoid arthritis (RA) symptoms in gut microbiota-dependent manner, with the key bacterium and the underlying mechanism remain unidentified. In this study, LBP was fermented in vitro by Romboutsia lituseburensis , Lactobacillus johnsonii , or a combination of both strains, and were administered to pseudo-germ-free RA rats. Only R. lituseburensis- fermented LBP significantly alleviated RA and markedly enriched osteoclast differentiation pathway-related genes. Twenty-six metabolites produced by R. lituseburensis , including glutamic acid-proline-glutamine (EPQ), that reduced inflammation and inhibited osteoclast differentiation by downregulating Rankl expression and its downstream factors. Notably, EPQ was also proposed to bind RANKL and potentially interfere with the RANKL/RANK pathway, thereby suppressing the expression of osteoclastogenesis-related genes. These findings identify R. lituseburensis as the key bacterium mediating the anti-RA effects of LBP, with its fermentation products, particularly EPQ, serving as critical active substances that inhibit inflammation and modulate osteoclast differentiation via the RANKL-RANK signaling.

GABAFG isolated fom ginseng ameliorates type 2 diabetes mellitus by modulating gut microbiota and autophagy-lysosome pathway

Meng-han Qi,Hai-yan Zhang,Yun-yi Hou,Ivan Steve Nguepi Tsopmejio,Wei Liu,Wen-guang Chang,Chen Chen,Zi Wang,Wei Li

Introduction Hyperglycemia and hyperlipidemia are the hallmarks of type 2 diabetes mellitus (T2DM). T2DM is a systemic metabolic disease caused by insulin resistance and malfunctioning pancreatic β-cells. Although ginseng (the roots of Panax ginseng C.A. Meyer) can be used to treat T2DM, the underlying mechanism is unclear. Objectives To assess the role and mechanism of, γ-aminobutyric acid-fructosyl-glucose (GABAFG), a maillard reaction product of ginseng, in T2DM treatment. Methods The metabolism of GABAFG in serum and tissues was analyzed via ultra-high performance liquid chromatography-Q exactive-mass spectrometry (UHPLC-QE-MS). The molecular mechanisms of GABAFG in pancreatic β-cells (in vivo and in vitro) were investigated via Western blotting, qPCR and immunofluorescence. In addition, the results were validated via high-throughput sequencing and serum metabolomics. Results GABAFG alleviated the elevation of blood glucose and blood lipids in HFD/STZ-induced T2DM mice. Also, GABAFG reduced the insulin resistance-associated IRS-1 signaling axis in pancreatic β-cells in vitro . Mechanistically, GABAFG targeted the nuclear translocation of TFEB inhibited apoptosis of pancreatic β-cells by enhancing autophagolysosome function. In addition, GABAFG remodeled the gut microbiota. Specifically, GABAFG increased Akkermansia, decreased Romboutsia abundance, and decreased serum glycerophospholipid metabolism, thus alleviating T2DM-induced dyslipidemia. Conclusion This is the first study to assess the pharmacological effects of ginseng-derived GABAFG in T2DM. Therefore, this study provides a new theoretical basis for understanding ginseng effect in metabolic diseases.

Naoxintong Capsule Inhibits the Development of Cardiovascular Pathological Changes in Bama Minipig Through Improving Gut Microbiota

Zhang Wei-jian,Su Wei-wei,Li Pei-bo,Rao Hong-yu,Lin Qing-wei,Zeng Xuan,Chen Tao-bin,Yan Zeng-hao,Liu Hong,Yao Hong-liang

Naoxintong capsule (NXT), a Chinese medicine, has performed excellent effects on the prevention and treatment against cardiovascular diseases. NXT is a fine powder mixture without any herb extraction, and there must be lots of ingredients hard to be absorbed. However, little is known about the correlation between the NXT’s cardioprotective effects and gut microbiota. Herein, we report the effect of NXT on the development of cardiovascular diseases and clarify the correlation between NXT’s cardioprotective effects and gut microbiota. In the current study, minipigs were selected and fed with high-fat diet and NXT daily for successive 8 months. During the process, up to 18 biomedical parameters were monthly determined to observe the dynamic changes after NXT treatment. At the end of experimental process, pathological examinations of heart, coronary artery, carotid artery, thoracic aorta, and abdominal aorta were conducted by HE staining and 16SrDNA sequencing, and analyzing of gut microbiota were conducted. Our results showed that NXT’s effects against cardiovascular diseases were through regulating blood lipid profiles, inhibiting vascular inflammation, enhancing antioxidant capacity, and alleviating myocardial injury, without damages on liver and kidney particularly. Concurrently, we also found that long-term administration of NXT increased the diversity of gut microbiota, influenced the microbiome structure and composition stably, and revered the increase of the ratio of the Firmicutes to Bacteroidetes (F/B ratio) in relative abundance. Specifically, our results revealed some key bacterium of Caproiciproducens (enhanced), Sutterella (enhanced), Erysipelotrichaceae (enhanced), and Romboutsia (decreased) that were closely involved in NXT’s effects. Taken together, our study demonstrates that NXT can inhibit the development of cardiovascular diseases by ameliorating high-fat diet–induced metabolic disorders and partly through improving gut microbiota.

Ethanol extract of Sargarsum fusiforme alleviates HFD/STZ-induced hyperglycemia in association with modulation of gut microbiota and intestinal metabolites in type 2 diabetic mice

Siya Wu,Jihui Zuo,Yang Cheng,Ya Zhang,Zhongshan Zhang,Mingjiang Wu,Yue Yang,Haibin Tong

Type 2 diabetes mellitus (T2DM) is considered a rapidly growing chronic disease that threatens human health worldwide. Extracts of various seaweeds have been shown to have anti-diabetic activity. Sargarsum fusiforme, an edible brown seaweed, has been shown to possess anti-inflammatory, anti-diabetic and anti-obesity activities. In this study, we investigated the beneficial effect of an ethanol extract of S. fusiforme (EE) on type 2 diabetes in mice induced with high-fat diet (HFD) and streptozotocin (STZ). Administering EE to the diabetic mice significantly reduced food intake, water intake and fasting blood glucose (FBG), while improving glucose tolerance, lipid profile and ameliorating hepatic oxidative stress. Furthermore, these animals also exhibited significantly diminished epididymal fat deposition, as well as less pathological changes in the heart and liver tissues, while displaying some highly enriched benign gut bacteria (e.g., Intestinimonas , Oscillibacter , Lachnoclostridium, unidentified_Lachnospiraceae , Roseburia and Anaerotruncus ) and a lower abundance of bacteria associated with diabetes or other metabolic diseases (e.g., Enterorhabdus and Romboutsia ). Metabolomic analysis revealed reduced levels of branched-chain amino acids (BCAA), such as l -valine and l -isoleucine, aromatic amino acids (AAA), such as l -tyrosine and l -phenylalanine, and increased levels of 4-hydroxyphenylacetic acid (4-HPA) in the gut content, suggesting that EE may impact T2DM through modulation of these compounds in the gut of the animals. Taken together, the results implied that S. fusiforme may contain valuable active components other than polysaccharides that have potential benefit in alleviating T2DM.
