Dissipation, residue and dietary risk assessment of difenoconazole in Rosa roxburghii

Qingshan Feng,Lei Han,Qiong Wu,Xiaomao Wu

Rosa roxburghii is a medicinal and edible plant, which is favored by consumers due to its rich vitamin C content. Residues and potential health risks of difenoconazole in the R. roxburghii ecosystem has aroused a concern considering its extensive use for controlling the powdery mildew of R. roxburghii. In this study, the residue of difenoconazole in R. roxburghii and soil was extracted by acetonitrile, purified by primary secondary amine and detected by liquid chromatography-tandem triple quadrupole mass spectrometry. The average recoveries in R. roxburghii and soil matrix varied from 82.59% to 99.63%, with relative standard deviations (RSD) of 1.14%–8.23%. The limit of quantification (LOQ) and detection (LOD) of difenoconazole in R. roxburghii and soil samples were 0.01 mg/kg. The dissipation of difenoconazole followed well the first-order kinetic, with a half-life of 3.99–5.57 d in R. roxburghii and 4.94–6.23 d in soil, respectively. And the terminal residues were <0.01–2.181 mg/kg and 0.014–2.406 mg/kg, respectively. The chronic and acute risk quotient values of difenoconazole were respectively 0.42% and 4.1%, which suggests that the risk was acceptable and safe to consumers. This study provides a reference for the safe and reasonable use of difenoconazole in R. roxburghii production.

Anti-browning effect of Rosa roxburghii on apple juice and identification of polyphenol oxidase inhibitors

Kaibo Yu,Lei Zhou,Yuefang Sun,Zicong Zeng,Huiwen Chen,Junping Liu,Liqiang Zou,Wei Liu

Enzymatic browning control of cloudy fruit juice with natural substances has received much attention for improving its nutritional and commercial value. This study explored the anti-browning potential of Rosa roxburghii in apple juice. The anti-browning effects and mechanisms were evaluated by serial measurements of appearance, browning index, polyphenol oxidase (PPO) activity, UPLC-QE-Orbitrap-MS identification, inhibition kinetics and molecular docking. The results showed that Rosa roxburghii juice (0.25%-1.25% w/w) could effectively inhibit browning and PPO activity of apple juice. Ascorbic acid (1.67 g/100 g) as a reducing agent was a main anti-browning factor. Furthermore, seven phenolic compounds in Rosa roxburghii were screened as PPO inhibitors. Representative phenolic inhibitors induced mixed or competitive inhibition of PPO, mainly driven by hydrophobic forces and hydrogen bonds. This work demonstrates that Rosa roxburghii is a promising natural anti-browning ingredient to control the browning of cloudy apple juice due to abundant ascorbic acid and PPO inhibitors.

CO2-responsive ionic liquid for the sustainable extraction and separation of Rosa roxburghii polysaccharides

Zhonghai Tang,Yunlan Chen,Changyong Cai,Shaoping Ma,Zhijian Tan

The extraction of Rosa roxburghii polysaccharides using conventional methods suffers from low extraction efficiency and poor selectivity. Recently, green and efficient methods of polysaccharide extraction have received increasing attention. In this study, a CO 2 -responsive ionic liquid (IL) was developed for the extraction of Rosa roxburghii polysaccharides. This IL can achieve efficient extraction, and can be recycled, and reused in the extraction process. The maximum polysaccharide extraction yield of the IL was 253 mg/g under optimal single-factor experiment conditions. Three fractionated polysaccharides, RTFP-1, RTFP-2, and RTFP-3, were obtained by further purification, and their preliminary structures were characterized. RTFP-2 and RTFP-3 showed higher antioxidant and hypoglycemic activities than RTFP-1. This IL maintains high extraction efficiency after 5 cycles and reuse. This extraction method based on a CO 2 -responsive IL provides an efficient and sustainable method for polysaccharide extraction.

Dynamic Changes of Active Components and Volatile Organic Compounds in Rosa roxburghii Fruit during the Process of Maturity

Su Xu,Junyi Deng,Siyao Wu,Qiang Fei,Dong Lin,Haijiang Chen,Guangcan Tao,Lingshuai Meng,Yan Hu,Fengwei Ma

Rosa roxburghii (R. roxburghii), native to the southwest provinces of China, is a fruit crop of important economic value in Guizhou Province. However, the changes in fruit quality and flavor duringR. roxburghiifruit ripening have remained unknown. Here, this study investigated the changes of seven active components and volatile organic compounds (VOCs) during the ripening of theR. roxburghiifruit at five different ripening stages including 45, 65, 75, 90, and 105 days after anthesis. The results indicated that during the ripening process, the levels of total acid, vitamin C, and soluble sugar significantly increased (p< 0.05), while the levels of total flavonoids, superoxide dismutase (SOD), and soluble tannin significantly decreased (p< 0.05). Additionally, the content of total phenol exhibited a trend of first decreasing significantly and then increasing significantly (p< 0.05). A total of 145 VOCs were detected by HS-SPME-GC-MS at five mature stages, primarily consisting of aldehydes, alcohols, esters, and alkenes. AsR. roxburghiimatured, both the diversity and total quantity of VOCs in the fruit increased, with a notable rise in the contents of acids, ketones, and alkenes. By calculating the ROAV values of these VOCs, 53 key substances were identified, which included aromas such as fruit, citrus, green, caramel, grass, flower, sweet, soap, wood, and fat notes. The aromas of citrus, caramel, sweet, and wood were predominantly concentrated in the later stages ofR. roxburghiifruit ripening. Cluster heatmap analysis revealed distinct distribution patterns of VOCs across five different maturity stages, serving as characteristic chemical fingerprints for each stage. Notably, stages IV and V were primarily characterized by a dominance of alkenes. OPLS-DA analysis categorized the ripening process ofR. roxburghiifruit into three segments: the first segment encompassed the initial three stages (I, II, and III), the second segment corresponded to the fourth stage (IV), and the third segment pertained to the fifth stage (V). Following the variable importance in projection (VIP) > 1 criterion, a total of 30 key differential VOCs were identified across the five stages, predominantly comprising ester compounds, which significantly influenced the aroma profiles ofR. roxburghiifruit. By integrating the VIP > 1 and ROAV > 1 criteria, 21 differential VOCs were further identified as key contributors to the aroma changes inR. roxburghiifruit during the ripening process. This study provided data on the changes in quality and aroma ofR. roxburghiifruit during ripening and laid the foundation for the investigation of the mechanism of compound accumulation during ripening.

Screening and validation of active components in Rosa roxburghii Tratt for anti-pulmonary fibrosis based on a spectrum-effect relationship

Xiaomeng Wang,Ting Zhou,Shaolin Huang,Heting Zhou,Yihan Ling,Tao Chen,Shuwen Zhang,Wenxi Wang,Chuan Wu,Wenya Yin

Rosa roxburghii Tratt (RRT), a fruit with dual medicinal and nutritional applications, exhibits therapeutic potential against pulmonary fibrosis, yet the specific bioactive constituents underlying this effect remain uncharacterized. This study employed an integrated spectrum-effect relationship to systematically identify RRT's principal anti-pulmonary fibrosis components. Our findings demonstrate that five different polar extracts of RRT (RRTEs) differentially attenuated bleomycin-induced pulmonary fibrosis in murine models, with the ethyl acetate fraction (EAE) showing superior therapeutic efficacy. HPLC-Q-Exactive Orbitrap MS identified 56 compounds, and screened out four active ingredients related to anti-pulmonary fibrosis by spectrum-effect relationship. In vitro experiments revealed that ellagic acid, gallic acid and syringic acid inhibited fibroblast migration, attenuated intracellular ROS overproduction, and downregulated the expression levels of α-SMA and collagen I. In summary, we established for the first time a spectrum-effect relationship between RRT and pulmonary fibrosis, elucidated the key components, and provided a foundation for future clinical applications.

The Dissipation Kinetics, Residue Level and Dietary Risk of Kresoxim-Methyl in Rosa roxburghii and Soil Based on the QuEChERS Method Coupled with LC–MS/MS

Zheng Handinghong,Wu Qiong,Wu Xiaomao

This study aimed to investigate the dissipation, residues and dietary assessment of kresoxim-methyl in the application of Rosa Roxburghii and soil field using liquid chromatography-tandem mass spectrometry (LC–MS/MS). The results show that kresoxim-methyl in R. roxburghii samples was extracted by acetonitrile and purified by ethyl enediamine-N-propylsilane (PSA), while kresoxim-methyl in soil samples was extracted by acetonitrile and purified by octadecylsilyl solid phase dispersant (C 18 ). 0.1% formic acid (v/v)-water–methanol solution was used as the mobile phase, LC–MS/MS exhibited a good linearity in the range of 0.001–10 mg L −1 . The recoveries of R. roxburghii and soil matrix were 82.48%–102.55%, and the relative standard deviation ( RSD) were 1.13%–4.21%. The limit of detection ( LOD ) and quantification ( LOQ ) of kresoxim-methyl in R. roxburghii and soil samples was 0.50 and 0.60 µg kg −1 , respectively. The dissipation dynamics of kresoxim-methyl in R. roxburghii and soil followed the first-order kinetics, with the half-life of 4.28 and 4.41 days, respectively. The terminal residual amount of kresoxim-methyl in R. roxburghii and soil samples was 0.003–1.764 and 0.007–2.091 mg kg −1 , respectively. The dietary intake risk assessment indicates that a risk quotient ( RQ ) for kresoxim-methyl based on the national estimated daily intake ( NEDI ) of 0.1995 mg was 0.79%, suggesting that the use of kresoxim-methyl on R. roxburghii at recommended dosage was safe to consumers. This study provides the theoretical basis for guiding the rational use of kresoxim-methyl in the production of R. roxburghii .

Rosa roxburghii Fermentation Broths Attenuate Bleomycin-Induced Pulmonary Fibrosis by Activating the Nrf2/HO-1/NQO1 Signaling Pathway and Modulating Gut Microbiota

Heting Zhou,Xinyue Zheng,Shaolin Huang,Xiaomeng Wang,Ting Zhou,Shuwen Zhang,Yihan Ling,Wenxi Wang,Xingjie Li,Shouqian Li,Yongmei Xie,Wenya Yin

Pulmonary fibrosis (PF) is a chronic and progressive lung disease, and oxidative stress plays a critical role in its pathogenesis. Rosa roxburghii Tratt, known for its anti-inflammatory and antioxidant properties, has been shown to alleviate fibrosis. This study aimed to explore whether two Rosa roxburghii fermentation broths (RRFBs) (with different proportions) could attenuate bleomycin (BLM)-induced PF in mice and to elucidate the molecular mechanisms. The results revealed that RRFBs reduced structural lung damage, collagen deposition, and lung inflammation. RRFBs also suppressed fibrotic markers (Collagen I, Vimentin, and α-SMA) while enhancing epithelial marker E-cadherin expression. Additionally, RRFBs alleviated BLM-induced oxidative stress and apoptosis by activating the Nrf2/HO-1/NQO1 signaling pathway and facilitating Nrf2 nuclear translocation. Furthermore, RRFBs attenuated the BLM-induced changes in the gut microbiota; in particular, they decreased the abundance of the pathogenic bacterium Proteus and increased the abundance of the probiotics Ileibacterium and Dubosiella . Spearman correlation analysis revealed a strong association between oxidative stress inhibition and gut microbiota composition. These results indicated that RRFBs could exert lung-protective effects by inhibiting oxidative stress and alleviating intestinal disturbances.

Phytochemical profile and antioxidant characteristics of bound and free phenolics from Rosa roxburghii Tratt

Xiaoli Zhou,Yumeng Qin,Yanli Wang,Yaping Wang,Zhicheng Qin

Rosa roxburghii Tratt (RRT) is renowned for its abundance of phenolics, various methods have been used to extract free phenolics from RRT, there remains a paucity of research on the comparative distinctions between bound phenolic compounds and their free counterparts in RRT, especially in vitro and in vivo antioxidant potential of RRT-BP necessitates further exploration. This study sought to optimize the process parameters for extracting bound polyphenols through alkaline hydrolysis . Among the bound phenolic compounds, 20 phenolic acids and 34 flavonoids were identified, while the free phenolic compounds comprised 19 phenolic acids and 42 flavonoids. The IC 50 values for DPPH and ABTS radical scavenging were 44.92 μg/mL and 60.92 μg/mL for free phenols , 87.02 μg/mL and 95.02 μg/mL for bound phenols, respectively. Compared with the control group, the lifespan of Caenorhabditis elegans fed with 1 mg/mL of free and bound phenol extended from 11.63 d to 16.77 d and 13.88 d, respectively, with no adverse impact on the reproductive capability of the nematodes. The longevity of C. elegans was observed to persist even under thermal and oxidative stress , primarily attributed to elevated superoxide dismutase and catalase activity, coupled with reduced malondialdehyde content in C. elegans . These findings underscore that free polyphenols exhibit superior polyphenolic components and antioxidant properties compared to their bound counterparts. Nevertheless, the potential health effects of bound polyphenols as a supplement to free polyphenols in RRT should not be ignored.

Antioxidative and Cytoprotective Effects of Rosa Roxburghii and Metabolite Changes in Oxidative Stress-Induced HepG2 Cells Following Rosa Roxburghii Intervention

Yangchen Mao,Ruyi Sha,Yuhao Sun,Zhenzhen Wang,Jun Huang

Rosa Roxburghii(RR), a traditional Chinese medicinal fruit, is rich in bioactive substances that make it a potential natural antioxidant resource. This research aimed to study the antioxidant properties of RR by in vitro experiments and through intracellular assessment in H2O2-induced HepG2 cells. A non-targeted metabolic analysis was conducted to indicate changes in intracellular and extracellular metabolites. Differential metabolites and metabolic pathways were explored using PCA, PLS-DA, and KEGG pathway analysis. The results showed that RR rich in bioactive substances exhibited a significant antioxidative property in vitro and intracellularly. This property may be achieved by scavenging free radicals, increasing the activity of catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and the levels of bicinchoninic acid (BCA) while reducing the reactive oxygen species (ROS) generation. This study identified 13 differential metabolites intracellularly and 7 extracellularly, among which the key differential metabolites included D-glucopyranose, D-mannose, fructose, citric acid, malic acid, cholesterol, and cholestenone. These key metabolites primarily regulated glucose-related metabolism, the citrate cycle, and the primary bile acid biosynthesis pathway in H2O2-induced HepG2 cells. These findings provide potential application evidence of RR in the development of natural resources for functional foods.

Synthesis of Novel Fluorescent Carbon Quantum Dots From Rosa roxburghii for Rapid and Highly Selective Detection of o-nitrophenol and Cellular Imaging

Zhang Qianchun,Liang Junyi,Zhao Li,Wang Yan,Zheng Yuguo,Wu Yun,Jiang Li

A novel carbon quantum dots (CQDs) were successfully synthesized by one-step hydrothermal reaction using Rosa roxburghii as a biomass-based precursor. The CQDs have an average size of 2.5 nm and a narrow size distribution. They display strong blue fluorescence with a quantum yield of 24.8% and good biocompatibility. Notably, these CQDs were capable of detecting trace o-nitrophenol in surface water and sewage with high sensitivity and specificity. The linear range is 0.08–40 μmol/L, and the limit of detection is 15.2 nmol/L. Furthermore, this CQDs was successfully applied for o-nitrophenol analysis in river water and sewage samples. Additionally, Hep3B cells, a human hepatocellular carcinoma cell line, can be easily imaged with high resolution using the as-prepared CQDs as nanoprobes. These results reveal that the as-prepared CQDs have potential applications for detecting o-nitrophenol and cell imaging.
