A photo/electrochemical coupling interface of Ru[dcbpy] 3 2+ -AMT/Au (AMT; 5-Amino-1,3,4-thiadiazole-2-thiol) was fabricated using a dehydration condensation sulfhydrating method. For the interface functional properties, a combined dual-signal recording (CDSR) method was applied to characterize the response characteristics, and a scanning electrochemical microscopy-electrochemiluminescence (SECM-ECL) imaging was developed to assess the interface distribution uniformity. The interface biosensing compatibility was validated by constructing a simple DNA sensor. The research results show that the interaction between the two functional parameters follows a synergistic effect mechanism in the coupling conditions and an interference effect mechanism in the detection condition. Under optimized conditions, the saturation dual-signal response values are 156.0 and 86.8 μA, respectively. The statistics and imaging comparison analysis validate good interface distribution uniformity and stability performance. The DNA sensor's dual-signal detection limits to the signal probe (SP) are ∼30 fM and 0.3 pM with linear ranges of 100.0 fM ∼ 1.0 nM and 1.0 pM ∼ 10.0 nM, respectively. The fabricated interface exhibits an effective bi-functional response performance compatible with biosensing. The proposed imaging method has a high technical fit for studying photo/electrochemical coupling interfaces and can also provide a reference for other similar coupling interface analyses.

The detection of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is crucial for preventing and controlling infectious diseases and disease treatment. In this work, a Au@Ti3C2@PEI-Ru(dcbpy)32+ nanocomposite-based electrochemiluminescence (ECL) biosensor was rationally designed, which realized sensitive detection of the RNA-dependent RNA polymerase (RdRp) gene of SARS-CoV-2. In addition, a DNA walker was also used to excise the hairpin DNAs under the action of Nb.BbvCI endonuclease. Furthermore, model DNA–Ag nanoclusters (model DNA–AgNCs) were used to quench the initial ECL signal. As a result, the ECL biosensor was used to sensitively detect the SARS-CoV-2 RdRp gene with a detection range of 1 fM to 100 pM and a limit of detection of 0.21 fM. It was indicated that the ECL biosensor had a great application potential for clinical medical detection. Furthermore, the DNA walker amplification also played a reliable candidate strategy for other detection methods.

Urokinase (UK), an enzymatic protein predominantly isolated from the urine of healthy humans , serves as a crucial biomarker for evaluating human renal function , cardiovascular diseases, and inflammatory states. In this study, a novel sandwich - type ratiometric electrochemiluminescence (ECL) biosensor was constructed to achieve accurate and sensitive detection of UK. This biosensor was constructed with titanium dioxide magnetic beads (P25) as the cathode luminophore and a terbium monophosphate - guanosine infinite coordination polymer network (Tb - GMP ICPn) encapsulating tris(4,4' - dicarboxylic acid - 2,2' - bipyridyl) ruthenium dichloride (Ru(dcbpy) 3 2+ /ICPn) as the anode luminophore. Under optimized experimental conditions, when potassium persulfate (K 2 S 2 O 8 ) and tri - n - propylamine (TPA) were separately utilized as co - reactants, a favorable linear correlation was observed between the variations in ECL intensities and the concentration of UK within the range of 10 −2 to 10 4 ng mL −1 . The limits of detection (LODs) were determined to be 7.2 and 5.8 pg mL −1 , respectively. Moreover, the application of this biosensor for the detection of UK in real - world samples yielded satisfactory results. These findings suggest that the developed ratiometric ECL biosensor holds significant promise in the realm of clinical detection and diagnosis.

Signal amplification provides an effective way to improve detection performance. Herein, an ultrasensitive electrochemiluminescence (ECL) aptasensor for Pb 2+ detection was developed based on a dual signal-amplification strategy of the abscission of a quencher and the generation of a G-quadruplex by one-step and simultaneous way. Nitrogen-doped carbon quantum dots linked with complementary DNA (cDNA-NCQDs) at the sensing interface was applied as the quencher of a tris(4,4′-dicarboxylic acid-2,2′-bipyridyl)ruthenium(II) (Ru(dcbpy) 3 2+ )/tripropylamine system to minimize the ECL signal due to the intermolecular hydrogen bond-induced energy-transfer process. Upon the addition of Pb 2+ , its specific binding with the aptamer triggered the abscission of cDNA-NCQDs, accompanied by the formation of G-quadruplex on the surface of the electrode, both of which amplified the intensity of the light emission. The ECL amplification efficiency induced by the above two mechanisms (78.6%) was valuably greater than that of their sum value (69.3%). This synergistic effect resulted in high detection sensitivity of the ECL aptasensor, which allowed to thereby obtain Pb 2+ measurements in the range of 1 fM - 10 nM with an ultra-low detection limit of 0.19 fM. The Pb 2+ -mediated synergistic signal-amplification ECL strategy can provide a new approach for integrating various amplification strategies.
