Rubisco accumulation is important for the greening of the fln2-4 mutant in Arabidopsis

Chao Huang,Qing-Bo Yu,Xin-Bo Yuan,Zi-Ran Li,Jing Wang,Lin-Shan Ye,Ling Xu,Zhong-Nan Yang

The fructokinase-like protein2 (FLN2) is a component of the PEP complex. FLN2 knockout mutants displayed a delayed greening phenotype on sucrose-containing medium. Our previous work indicated that partial PEP activity is essential for its greening phenotype. In this study, we further report that sufficient Rubisco accumulation is critical for fln2-4 greening. Sugar serves many important functions, such as an energy source and signaling molecule. Through pharmacological experiments using a sugar analog and sugar signaling inhibitor, we demonstrate that sugar serves as energy to support the fln2-4 greening. Seed-reserve and photosynthetic CO 2 -fixation are the primary energy sources for early seedling growth . No obvious differences were observed in the seed-reserve of the wild-type and fln2-4 by comparing their seed size and dark-germination, indicating that the defective carbon fixation may account for the energy deficit in fln2-4 during its early seedling growth. The Rubisco content was low in fln2-4 , but it rapidly accumulated during the greening of fln2-4 . Expression of a nuclear-encoded rbcL gene facilitates Rubisco accumulation and partially complements the mutant defects. These results suggest that the Rubisco accumulation is critical for fln2-4 greening. In summary, the rapid Rubisco accumulation that depends on sufficient PEP activity is important for normal seedling growth.

The production of succinate with more CO2 fixation reactions facilitated by RuBisCO-based engineered Escherichia coli

Xiuyuan Zhou,Linqing Li,Shengjie Sun,Peng Xiong,Xiutao Liu

Redesigning metabolic pathways to enhance the efficiency of carbon fixation during chemical biosynthesis is a promising approach for achieving cleaner and greener production of multi-carbon compounds. In this study, we established a model of cell growth in Escherichia coli that is dependent on the RuBisCO-Prk pathway by regulating its central metabolism. This rewiring ensures that growth depends on RuBisCO's carboxylation, allowing heterotrophic growth to rely on carbon fixation. This model was verified by detecting the growth curve, and it was used to screen four RuBisCO genes, of which the gene from Rhodospirillum rubrum ATCC 11170 serves as a growth advantage for E.coli . In addition, this model was applied to construct an efficient succinate biosynthetic pathway that can produce two moles of succinate from one mole of xylose and three moles of CO 2 . Compared to conventional succinate biosynthesis, this strategy has a CO 2 fixation capacity that is 1.5 times greater. Furthermore, to optimize succinate production, various approaches were employed, including the optimization of key enzymes, substrate transport, and the supply of inorganic carbon. The resulting strain was capable of producing succinate at a level of 2.09 ± 0.14 g/L, which is nearly 22.4 times that of the original strain. In conclusion, this study was developed for the production of two moles of succinate by implementing three moles of carbon fixation reactions and demonstrated the feasibility of various optimization strategies in biological carbon fixation.

Biomimetic reusable microfluidic reactors with physically immobilized RuBisCO for glucose precursor production

Yujiao Zhu,Qingming Chen,Chi Chung Tsoi,Xiaowen Huang,Abdel El Abed,Kangning Ren,Shao-Yuan Leu,Xuming Zhang

The chloroplast of plants is a natural microfluidic reactor for natural photosynthesis, in which the multi-enzymatic Calvin cycle is the key. In the chloroplast, the Calvin cycle enzymes are reportedly attached to the thylakoid membrane by physical interactions. To mimic this process, we physically immobilized the first enzyme of the Calvin cycle, D-ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO), into microfluidic reactors by injecting 2 μg μL−1 RuBisCO for 4 h and demonstrated the successful production of the glucose precursor, 3-phosphoglycerate, at 0.145 ± 0.008 nmol min−1. Hydrophobic interactions play an important role in the physical immobilization, making the process simple and fast. The physical immobilization presents a 5.7-fold thermal stability as compared to the free RuBisCO, and shows performances inferior but close to that of chemical immobilization in enzyme kinetics, production rate and stability. Although the reactors can retain only 40% of the initial activity after 10 cycles of reusing, the physical immobilization has an interesting special feature that the enzyme can be desorbed to refresh the reactor for new immobilization. Experiments show that >95% activity can be restored after 5 cycles of refreshing. With the merits of reusing and refreshing, up to 5 mL of 3-PGA can be produced by continuously injecting the reactant mixture with great sustainability and cost-effectiveness. The reactors are also scaled out to two and six parallel reactors as a proof of concept of large-scale synthesis. Physical immobilization in microfluidic reactors is highly suitable for the multi-enzymatic, cascaded reactions in the Calvin cycle and facilitates the future study of artificial synthesis of glucose.

Enhanced Carbon Sequestration Performance and Mechanism of Adsorption-Catalytic Bifunctional ZIF-8 Coupled Microalgae

Luna Wang,Yan Gong,Yanming Miao,Zhifeng Zhang

To support the development of a low-carbon economy and contribute to the achievement of carbon neutrality, this study synthesized bifunctional zeolitic imidazolate framework-8-based nanozymes (ZNZs) with dual functionality: CO2 adsorption and carbonic anhydrase (CA)-mimetic catalytic activity. The influence of ZNZs at varying concentrations on the growth rate of microalgae, chlorophyll and carbohydrate content, and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and CA activity were explored. Results indicated that after the addition of 5 mg/L ZNZs, the growth rate of microalgae increased by 7%. Furthermore, the maximum chlorophyll and carbohydrate contents reached 4.55 and 43.59 mg/L, respectively, which were 12.6 and 4.2% higher than those in the control group. Additionally, Rubisco and CA activity (intra- and extracellular) increased by up to 21.26, 10, and 8.3%, respectively. These findings demonstrate that ZNZs enhance the photosynthetic efficiency of microalgae and increase microalgae’s affinity for CO2, highlighting their promise as a strategy for improving carbon capture and sustainable bioenergy production.

Preparation of nitrogen-doped carbon dots and their enhancement on lettuce yield and quality

Jieqiang Tan,Shili Zhao,Junyu Chen,Xiaoqin Pan,Chen Li,Yingliang Liu,Caijuan Wu,Wei Li,Mingtao Zheng

Nanotechnology is an effective way to stimulate the yield potential of crops. Various nano-fertilizers and nano-carriers are gradually being developed to bring about a technological revolution in the agricultural industry. As a biocompatible water-soluble nanomaterial, carbon dots (CDs) have attracted the attention of researchers for applications in agriculture. In this study, we prepared nitrogen-doped CDs (N-CDs) as a type of water-soluble carbon nanofertilizer by a one-pot hydrothermal method, and investigated its effects on lettuce biomass and quality. 100 and 200 mg L−1 of N-CDs substantially promoted lettuce biomass accumulation (41.70%), elevated lettuce nutrient content, as well as promoted the accumulation of major nutrients. Moreover, 100 mg L−1 N-CDs increased the chlorophyll a content by 12.68%, significantly increased the electron transport rate (ETR) by 38.61%, significantly increased the light energy conversion efficiency (Y(II)) by 31.24% and increased the Rubisco activity by 60.61%, which are important reasons for its increase in actual photosynthesis rate. N-CDs also have a positive effect on plant nitrogen metabolism by promoting the activity of glutamine synthetase. The significant benefits of N-CDs on lettuce make them have great potential for agricultural yield increase and quality improvement.

Molecular mechanism of dissolvable metal nanoparticles-enhanced CO2 fixation by algae: Metal-chlorophyll synthesis

Ruohua Qu,Na Liu,Qiong Wen,Jingyi Guo,Fei Ge

Algae-driven photosynthetic CO 2 fixation is a promising strategy to mitigate global climate changes and energy crises. Yet, the presence of metal nanoparticles (NPs), particularly dissolvable NPs, in aquatic ecosystems introduces new complexities due to their tendency to release metal ions that may perturb metabolic processes related to algal CO 2 fixation. This study selected six representative metal NPs (Fe 3 O 4 , ZnO, CuO, NiO, MgO, and Ag) to investigate their impacts on CO 2 fixation by algae ( Chlorella vulgaris ). We discovered an intriguing phenomenon that bivalent metal ions released from the metal NPs, especially from ZnO NPs, substituted Mg 2+ within the porphyrin ring. This interaction led to 81.8% and 76.1% increases in Zinc-chlorophyll and Magnesium-chlorophyll contents within algal cells at 0.01 mM ZnO NPs, respectively. Integrating metabolomics and transcriptomics analyses revealed that ZnO NPs mainly promoted the photosynthesis-antenna protein pathway, porphyrin and chlorophyll metabolism, and carbon fixation pathway, thereby mitigating the adverse effects of Zn 2+ substitution in light harvesting and energy transfer for CO 2 fixation. Ultimately, the genes encoding Rubisco large subunit ( rbcL ) responsible for CO 2 fixation were upregulated to 2.60-fold, resulting in a 76.3% increase in carbon fixation capacity. Similar upregulations of rbcL expression (1.13-fold) and carbon fixation capacity (76.1%) were observed in algal cells even at 0.001 mM ZnO NPs, accompanied by valuable lipid accumulation. This study offers novel insights into the molecular mechanism underlying NPs on CO 2 fixation by algae and potentially introduces strategies for global carbon sequestration.

The mechanisms by which phenanthrene affects the photosynthetic apparatus of cucumber leaves

Liqiao Jin,Xingkai Che,Zishan Zhang,Yuting Li,Huiyuan Gao,Shijie Zhao

Phenanthrene is a polycyclic aromatic hydrocarbon (PAH) that is widely distributed in the environment and seriously affects the growth and development of plants. To clarify the mechanisms of the direct effects of phenanthrene on the plant photosynthetic apparatus, we measured short-term phenanthrene-treated cucumber leaves. Phenanthrene inhibited Rubisco carboxylation activity, decreasing photosynthesis rates (Pn). And phenanthrene inhibited photosystem II (PSII) activity, thereby blocking photosynthetic electron transport. The inhibition of the light and dark reactions decreased the photosynthetic electron transport rate (ETR) and increased the excitation pressure (1−qP). Under high light, the maximum photochemical efficiency of photosystem II (F v /F m ) in phenanthrene-treated cucumber leaves decreased significantly, but photosystem I (PSI) activity (Δ I/I o ) did not. Phenanthrene also caused a J-point rise in the OJIP curve under high light, which indicated that the acceptor side of PSII Q A to Q B electron transfer was restricted. This was primarily due to the net degradation of D1 protein, which is caused by the accumulation of reactive oxygen species (ROS) in phenanthrene-treated cucumber leaves under high light. This study demonstrated that phenanthrene could directly inhibit photosynthetic electron transport and Rubisco carboxylation activity to decrease net Pn. Under high light, phenanthrene caused the accumulation of ROS, resulting in net increases in D1 protein degradation and consequently causing PSII photoinhibition .

Effects of multifunctional cerium-doped carbon dots on photosynthetic capacity and nutritional quality of lettuce

Shili Zhao,Chen Li,Caijuan Wu,Juan Hu,Zhiwei Zhang,Bingfu Lei,Wei Li,Chaofan Hu,Yingliang Liu,Mingtao Zheng

As biocompatible water-soluble nanomaterials, carbon dots (CDs) have attracted the interest of researchers in agricultural production. In this work, cerium-doped carbon dots (Ce-CDs) have been prepared as a water-soluble carbon nano-fertilizer using a one-pot hydrothermal method. The effects on lettuce yield and quality are investigated. The application of lower concentrations of Ce-CDs (50–150 mg L−1) all contributes to lettuce yield and quality enhancement. 100 mg L−1 of Ce-CDs demonstrates the most significant effect, increasing biomass by about 83.3% and promoting the accumulation of soluble sugars (87.5%) and soluble proteins (52.3%). Further investigation reveals that Ce-CDs can promote the electron transfer rate (33.35%), photosynthetic activity (30.28%), photoconversion efficiency (37.90%) and Rubisco activity (37.38%), and have a promoting effect on glutamine synthetase (GS, 3.10%) and glutamate synthase (GOGAT, 10.09%) activities. Regarding the net photosynthetic rate (54.99%), Ce-CDs elevate and induce stomatal opening and promote nutrient accumulation. This work not only evaluates the effects of rare earth-doped CDs as nano-fertilizers on plants but also provides strong technical support for the application of nano-fertilizers in practical production and life sciences.
