Dental caries is one of the most common diseases affecting more than 2 billion people’s health worldwide. In a clinical setting, it is challenging to predict and proactively guard against dental cavities prior to receiving a confirmed diagnosis. Streptococcus mutans (S. mutans) in saliva has been recognized as the main causative bacterial agent that causes dental caries. High sensitivity, good selectivity, and a wide detection range are incredibly important factors to affect S. mutans detection in practical applications. In this study, we present a portable saliva biosensor designed for the early detection of S. mutans with the potential to predict the occurrence of dental cavities. The biosensor was fabricated using a S. mutans-specific DNA aptamer and S. mutans-imprinted polymers. Methylene blue was utilized as a redox probe in the sensor to generate current signals for analysis. When S. mutans enters complementarily S. mutans cavities, it blocks electron transfer between methylene blue and the electrode, resulting in decreases in the reduction current signal. The signal variations are associated with S. mutans concentrations that are useful for quantitative analysis. The linear detection range of S. mutans is 102–109 cfu mL–1, which covers the critical concentration of high caries risk. The biosensor exhibited excellent selectivity toward S. mutans in the presence of other common oral bacteria. The biosensor’s wide detection range, excellent selectivity, and low limit of detection (2.6 cfu mL–1) are attributed to the synergistic effect of aptamer and S. mutans-imprinted polymers. The sensor demonstrates the potential to prevent dental caries.

Dental plaque, a kind of natural biofilm, has attracted much attention because of its role in caries and periodontal disease. The pathogenicity of the dominant bacterial species of S. mutans is based on the formation of extracellular polysaccharides (EPS) and its survival in acidic environments. In addition, the formed plaque biofilm has the characteristics of water insolubility, retention, and low membrane permeability. This is why there are no effective antibiotics or other ways to treat dental caries. Chemodynamic therapy (CDT) is an ideal therapeutic modality with biofilm microenvironment as stimulus. In this paper, a unique nanocomposite, ZnO 2 -Cu@RB NPs, has been prepared which is composed of copper-doped zinc peroxide nanoparticles (ZnO 2 -Cu NPs) and antibacterial agent Rose Bengal (RB). It can quickly (<10 min) and efficiently kill S. mutans (greater than 8.0 log 10 CFU mL −1 ) in acidic biofilm via the in situ enhanced CDT, and significantly reduce the demineralization of apatite. ZnO 2 -Cu@RB NPs can also weaken the acid production of S. mutans and inhibit the formation of EPS. More significantly, excellent therapeutic efficacy against S. mutans in the rat model was observed. Our findings reveal the great potential of metal peroxide nanoparticles as a new strategy for combating biofilm-related diseases.

Enzyme-mediated bacterial active endocytosis enhanced the permeability of NG/NO to biofilms. • The chemical structures of Schiff base and NONOates provided dual proton-responsive of gas release. • NO induces S-nitrosylation of PDEs in S. mutans 。, thereby reducing lactate production. Dental caries in permanent teeth have remained one of the most prevalent oral diseases over the past three decades, with their global incidence exhibiting a steady upward trajectory. Streptococcus mutans ( S. mutans ) colonization on dental surfaces, coupled with its acidogenic metabolic activity, constitutes a primary etiological factor in caries pathogenesis. In this work, a NO-loaded polysaccharide nanogel, NG/NO was developed to address this issue. Dextran component of the NG/NO demonstrated selective targeting capabilities, promoting active internalization by S. mutans through enzyme-mediated endocytosis. This mechanism significantly enhanced biofilm penetration of the NG/NO (>70% biofilm penetration depth) within a 4-h exposure. The Schiff base bonds and N-diazeniumdiolates conferred the dual-acid-responsive NO release kinetics, achieving mM concentration of NO liberation within several minutes under cariogenic conditions (pH 4.5–5.5), thereby triggering rapid biofilm disrupting (>80% biomass reduction). Moreover, lower concentration of NO was shown to inhibit glycolysis and synthesis of lactic acid in S. mutans , thus improving acidic environment and demonstrating the potential to prevent enamel demineralization and dental caries. In conclusion, the NG/NO has promising applications in daily oral hygiene maintenance and caries prevention.

Dentin bonding is the most common form of human tissue repair among tissue-biomaterial adhesions, concerning billions of people's oral health worldwide. However, insufficient adhesive infiltration in the demineralized dentin matrix (DDM) always produces numerous defects in the bonding interface termed the hybrid layer, which causes high levels of bacteria-related secondary dental diseases, and less than 50% of the bonding lasts more than 5 years. Therefore, it is urgent and vital to construct an antibacterial low-defect hybrid layer to solve the durability-related problems. A DDM with a hydrogel-like surface formed by the hydration of highly-anionic non-collagenous proteins (NCPs) is firstly used as a template to electrostatically assemble polyethyleneimine (PEI). The formation of a stable antibacterial polyelectrolyte complex of PEI/NCPs rapidly eliminates NCP hydration capacity and significantly improves the infiltration of various adhesives. Simultaneously, both the PEI during the assembly and the PEI-assembled DDM can directly destroy a biofilm of S. Mutans on the DDM. Consequently, a long-term antibacterial and low-defect hybrid layer is successfully created, which greatly improves the bonding effectiveness. This helps to improve the clinical treatment of bacteria-based dental diseases and the tooth-restoration repair effect and prevent secondary dental diseases, having significance in clinical dentistry and providing insights for other tissue-biomaterial adhesions.

Bacterial infection and the resulting inflammation response not only affect the functions of osteogenic-relative cells but also lead to the failure of titanium (Ti)-based implantation. It is urgent to develop multifunctional (antimicrobial, anti-inflammation, and pre-osteogenesis) titanium implants to address this tissue. Herein, an osteogenic growth peptide (OGP) loaded zeolitic imidazolate frameworks-67 (ZIF-67) coating was fabricated on titanium dioxide nanotubes (TNTs) to prepare a [email protected] surface. The [email protected] coating was rapidly dissolved under an acidic environment with pH-sensitive property. Moreover, the [email protected] displayed a strong antibacterial activity against Escherichia coli ( E. coli ), Staphylococcus aureus ( S. aureus ), Streptococcus mutans ( S. mutans ), and methicillin-resistant Staphylococcus aureus ( MRSA ) owing to the hydrolysis of ZIF-67 nanoparticles and the formation of an alkaline microenvironment. The implants had good biocompatibility toward mesenchymal stromal cells (MSCs) and macrophages. Notably, the [email protected] could suppress the inflammatory response and simultaneously improve the cellular differentiation of MSCs under an inflammatory environment. In vivo study further revealed that [email protected] implants presented strong antimicrobial and anti-inflammatory properties at the early stage of implantation, thereby enhancing the osteointegration of bone-implant at the late stage. Thus, this multifunctional Ti implant combining osteoimmunomodulatory and antibacterial effects is a promising candidate for implant-associated infectious bone regeneration.
