Nanozymes, the nanomaterials with enzyme-like characteristics which exhibit lower cost, easier synthesis and functionalization, better stability compared with natural enzymes, have been widely developed for biosensing, disease therapy and environmental governance. However, the lack of catalytic efficiency of nanozymes compared to natural enzymes makes it difficult to completely replace natural enzymes to achieve higher sensitivity and lower detection limit in biosensing. Herein, magnetism-controlled technology was used to form nanozyme array consisted of stacked Fe3O4/Au NPs at the bottom of the microchannel as a spatially confined microreactor for the catalytic reaction. By enhancing the mass transfer process of substrate towards nanozyme which was mediated by the corresponding V-structure, a higher local concentration of substrate and more efficient utilization of active sites of nanozyme were obtained to increase the catalytic efficiency (kcat/KM) of the nanozyme array consisted of the Fe3O4/Au NPs by 95.2%, which was two orders of magnitude than that of the open reactor. Based on this, a colorimetric method on an integrated microfluidic platform was proposed for sensitive biosensing of Salmonella typhimurium. The entire detection could be completed within 30 minutes, yielding a linear range from 102 to 107 CFU/mL and a detection limit as low as 5.6 CFU/mL.

The rapid detection of Salmonella is an essential precautionary measure to ensure food safety and prevent foodborne illness. In this study, based on the specific binding between mannan oligosaccharides (MOS) and type I fimbriae of Salmonella typhimurium ( S. typhimurium ), an efficient sensing strategy was developed. Obtained by enzymatic hydrolysis of konjac glucomannan, MOS were grafted to mannan-oligosaccharide modified carbon dots (g-CDs-MOS). It is worth noting that the introduction of MOS reduced the dissociation constant (K D ) between fabricated g-CDs-MOS and Con A from 25.4 µM (g-CDs and ConA) to 6.22 µM. This enhanced binding affinity stem from elevated density of MOS on surface of g-CDs. Furthermore, to construct recovery-type fluorescent probe, g-CDs-MOS was bonded onto two-dimensional quencher such as 3-aminobenzeneboronic acid-functionalized graphene oxide (GO-PBA), MnO 2 or MoS 2 . The g-CDs-MOS/MoS 2 exhibited the best sensitivity due to the high electron mobility of MoS 2 . Its fluorescent intensity was logarithmically responsive to concentration of S. typhimurium within the range of 10 2 -10 8 CFU·mL − 1 , and the corresponding limit of detection was as low as 11.79 CFU·mL − 1 . This fluorescent sensor shows promise for the detection of S. typhimurium in food safety. Graphical

An electrochemical biosensor using gold nanoparticles (AuNPs)-doped bimetallic-organic framework (BMOF) with enhanced peroxidase-like activity was constructed to detect Salmonella Typhimurium ( S. typhimurium ). The BMOF of CuZr-MOF was synthesized via a two-step method and used as carrier to in situ immobilize AuNPs. Due to the stability of Zr-MOF, the good electrocatalytic ability of Cu (II), and the synergetic effects of AuNPs, Cu (II) and Zr (IV), the prepared AuNPs@CuZr-MOF nanozyme showed improved stability and catalytic activity to H 2 O 2 oxidation. The oxidation reaction was found to be a surface-controlled process of electron transfer and a pH-dependent electron transfer process of oxidation reaction involving two electrons. Further, AuNPs@CuZr-MOF was biofunctionalized with signal DNA probe, forming sDNA-AuNPs@CuZr-MOF nanotags. The biosensing platform was constructed on a glassy carbon electrode modified sequentially with electrodeposited AuNPs, capture DNA probe (cDNA), and BSA. Finally, a sandwich-type detection structure was formed by hybridization reactions between cDNA and target invA gene of S. typhimurium , as well as between invA gene and the sDNA of sDNA@AuNPs@CuZr-MOF nanotags. Under optimized experimental conditions, the biosensor achieved a linear range of 1 × 10 −16 to 1 × 10 −8 mol L −1 for the target invA gene with a detection limit (LOD) of 6.2 × 10 −17 mol L −1 using differential pulse voltammetry measurement (DPV). It was successfully applied to the direct and quantitative detection of invA gene segments in total DNA extracts of S. typhimurium , showing a linear range from 3.5 to 3.5 × 10 6 CFU mL −1 and a LOD of 0.82 CFU mL −1 . The fabricated biosensor exhibited good selectivity, reproducibility, and storage stability, enabling its use for the detection of invA gene segments in contaminated milk, with recoveries between 95.9% and 103.1%. Salmonella typhimurium (S. typhimurium) is a major foodborne pathogen, posing significant public health risks and leading to substantial economic losses. Traditional lateral flow immunoassay (LFIA) technology for foodborne pathogen detection faces limitations, such as the challenging and costly process of screening paired antibodies. To address these issues, we developed an innovative label-free LFIA utilizing carboxyl-functionalized Fe3O4 nanoparticles for sensitive detection of S. typhimurium. Fe3O4 nanoparticles offer unique properties, including magnetic separation and peroxidase-like catalytic activity, which enhance both bacterial capture and colorimetric signal amplification. The proposed label-free magnetic separation LFIA (LFMS-LFIA) achieved a detection limit of 103 cfu mL−1 for S. typhimurium using catalytic signal amplification, with a sensitivity 10 times greater than that achieved with colorimetric signal alone. This approach demonstrates promising application potential in S. typhimurium contamination detection in samples such as drinking water, eggs, and skim milk. By utilizing probes that integrate efficient binding, magnetic enrichment, and signal amplification capabilities, this Fe3O4-enhanced LFIA represents a major advancement in S. typhimurium detection with high sensitivity and without the need for complex equipment. This work offers a cost-effective and highly sensitive solution in portable devices of food safety and environmental monitoring fields for the on-site detection of pathogens.

In this study, a novel electrochemical biosensor based on exonuclease III (Exo III)-aided autonomous cascade signal amplification for ultrasensitive and highly specific detection of Salmonella typhimurium (S. typhimurium) has been reported. Herein, two duplex DNA probes were ingeniously designed. An Ap–Pr duplex probe was constructed as a recognition element by hybridizing an S. typhimurium aptamer (Ap) with a primer (Pr). A P–S duplex probe has been designed to contain a protruding DNA fragment at 5′-termini that has an identical sequence with Pr (the secondary Pr), a complementary duplex DNA that is used for blocking a signal-triggered probe (S), and an overhung nucleic acid segment at 3′-end that can anneal with Pr and form double-stranded DNA with a blunt 3′-terminus, thus initiating Exo III-aided amplification reaction. Upon sensing of the target S. typhimurium, the specific recognition of S. typhimurium and aptamer releases Pr, which anneals with the P–S duplex and triggers the Exo III cleavage reaction, accompanied by release of Pr, S, and secondary Pr. After this, the released Pr and secondary Pr anneal with other P–S duplex, which in turn triggers the Exo III-assisted cycling cleavage. Thus, large amounts of S are liberated, which hybrids with S* (DNA probe complementary to S); thereby, 5′-methylene blue (MB)-labelled DNA probe (MBP) folds into a hairpin structure with 5′-terminus close to electrode surface for efficient electronic transfer. Moreover, the S–S* hybrid duplex is specifically hydrolyzed by Exo III to liberate S, which accordingly results in the cycling cleavage of S*. Therefore, numerous MBP transform into a hairpin structure with electroactive MB close to the electrode surface; this generates significantly amplified electrochemical signal towards S. typhimurium detection. The present developed strategy exhibited high sensitivity towards S. typhimurium with a detection limit of 2.8 × 10 cfu mL−1. Furthermore, the proposed autonomous cascade signal amplification strategy can be easily extended for detection towards a wide spectrum of analytes including nucleic acid, proteins, and small molecules possessing their corresponding aptamers. Therefore, the developed Exo III-aided autonomous cascade signal amplification strategy might indeed provide a facile and universal tool for bioanalysis and clinical diagnosis.

Simultaneous detection of multiple foodborne pathogens is of great importance for ensuring food safety. Herein, we present a sensitive dual-channel electrochemical biosensor based on copper metal organic frameworks (CuMOF) and lead metal organic framework (PbMOF) for simultaneous detection of Salmonella typhimurium ( S. typhimurium ) and Listeria monocytogenes ( L. monocytogenes ). The MOF-based nanotags were prepared by functionalizing gold nanoparticles loaded CuMOF (Au@CuMOF) and PbMOF (Au@PbMOF) with signal DNA sequences 1 (sDNA1) and sDNA2, respectively. By selecting invA of S. typhimurium and inlA gene of L. monocytogenes as targe sequences, a sandwich-typed dual-channel biosensor was developed on glassy carbon electrodes (GCE) through hybridization reactions. The sensitive detection of S. typhimurium and L. monocytogenes was achieved by the direct differential pulse voltametric (DPV) signals of Cu 2+ and Pb 2+ . Under optimal conditions, channel 1 of the biosensor showed linear range for invA gene of S. typhimurium in 1 × 10 −14 −1 × 10 −8 M with low detection limit (LOD) of 3.42 × 10 −16 M ( S/N = 3), and channel 2 of the biosensor showed linear range for inlA gene of L. monocytogenes in 1 × 10 −13 −1 × 10 −8 M with LOD of 6.11 × 10 −15 M (S/N = 3). The dual-channel biosensor showed good selectivity which were used to detect S. typhimurium with linear range of 5−1.0 × 10 4 CFU mL −1 (LOD of 2.33 CFU mL −1 ), and L. monocytogenes with linear range of 10 − 1.0 × 10 4 CFU mL −1 (LOD of 6.61 CFU mL −1 ).

Foodborne pathogens are a leading cause of food safety incidents. Here, we developed a highly sensitive dual-channel biosensor for the parallel electrochemical detection of Salmonella typhimurium ( S. typhimurium ) and Escherichia coli ( E. coli ) O157:H7 using bimetallic metal–organic framework (BMOF)-based signal nanotags. The BMOF-based nanotags were synthesized by functionalizing gold nanoparticle-doped CuZr-MOF with signal DNA sequences sDNA1 (forming sDNA1-AuNPs@CuZr-MOF nanotag) and PbZr-MOF with signal DNA sequence sDNA2 (forming sDNA2-AuNPs@PbZr-MOF nanotag). The biosensing platform was constructed using two glassy carbon electrodes (GCEs). Each GCE was sequentially modified, first with electrodeposited gold nanoparticles (depAu) and then with the corresponding capture DNA sequences (cDNA1 for channel 1 or cDNA2 for channel 2). Target detection was achieved on the sensing interface via sandwich hybridization: in channel 1, the invA gene of S. typhimurium (tDNA1) was recognized by both cDNA1 on the sDNA1-AuNPs@CuZr-MOF nanotag; similarly, in channel 2, the eaeA gene of E. coli O157:H7 (tDNA2) was recognized by both cDNA2 and sDNA2 on the sDNA2-AuNPs@PbZr-MOF nanotag. Leveraging the high stability and ease of surface modification of UiO-66 (Zr-MOF), along with the enhanced electrochemical oxidation signals from the introduced secondary metal ions (Cu 2+ and Pb 2+ ) in Zr-MOF framework, the biosensor achieved parallel detection with wide linear ranges (3.5 to 3.5 × 10 5 CFU mL −1 for S. typhimurium , and 2.2 to 2.2 × 10 5 CFU mL −1 for E. coli O157:H7) and low detection limit (LODs of 0.54 CFU mL −1 and 0.80 CFU mL −1 , respectively). Furthermore, the fabricated biosensor demonstrated excellent reproducibility, selectivity, and stability, highlighting great potential for practical use in monitoring foodborne pathogens. Graphical

Sensitive and simultaneous detection of multiple foodborne pathogens has become an urgent need in the food industry. This work presents a signal-on fluorescence biosensor using DNA-templated metal nanoclusters as signal nanotags and an Exonuclease III (Exo III)-assisted target-recycling (EATR) strategy for the detection of Cronobacter sakazakii ( C. sakazakii ) and Salmonella typhimurium ( S. typhimurium ). The detection system consists of carboxylated polypyrrole nanoparticles functionalized with hairpin DNA sequences HP1 and HP2 (HP1-cPPy-HP2 NPs), the target ompA gene (tDNA1) of C. sakazakii and invA gene (tDNA2) of S. typhimurium , DNA-templated gold and silver nanocluster nanotags (sDNA1-AuNCs and sDNA2-AgNCs), and Exo III. In the presence of the targets, HP1 and HP2 on cPPy surface are opened through hybridization, forming tDNA1-HP1-cPPy-HP2-tDNA2 complex. These opened HP1 and HP2 then hybridize with their respective sDNA1 (sDNA1-AuNCs) and sDNA2 (sDNA2-AgNCs), leading to the increased fluorescence intensity of both AuNCs and AgNCs in the detection system. Furthermore, with EATR amplification strategy, the targets can be released from the complex through cleavage by Exo III and proceed to the next cycle of HP1/HP2 opening. As a result, the fluorescence signals of AuNCs and AgNCs can be significantly increased. Under optimal conditions, the fluorescence biosensor shows wide linear ranges for C. sakazakii (1.64 ∼ 1.64×10 5 CFU mL −1 ) and S. typhimurium (2.56 ∼ 2.56×10 5 CFU mL −1 ), with LODs of 1.4 CFU mL −1 and 2.2 CFU mL −1 , respectively. The biosensor was used for the detection of C. sakazakii and S. typhimurium in real samples, with recovery ranges of 88.8% to 110.6% and 90.4% to 114.4%, respectively.

The tumor microenvironment, characterized by hypoxia, supports the efficacy of anaerobic bacteria like attenuated S. typhimurium in cancer therapies. These bacteria target and penetrate deep tumor regions, significantly reducing tumor size but often lead to tumor regrowth due to limited long-term efficacy. To enhance the therapeutic impact, a novel biohybrid system, S@UIL, has been developed by coating S. typhimurium with a zirconium-based nanoscale metal–organic framework (UiO-66-NH 2 ) loaded with indocyanine green (ICG) and luteolin (LUT). This system maintains the bacteria’s tumor-targeting ability while increasing the penetration and therapeutic effectiveness through excessive autophagy and mild hyperthermia. In a subcutaneous colon cancer model, the integration of LUT and ICG promotes autophagic cell death and photothermal sensitization, leading to the release of damage-associated molecular patterns (DAMPs). These DAMPs activate immune responses through dendritic cells and T -cells, enhancing immunogenic cell death (ICD) and potentially reducing immune evasion by tumors. This single-administration approach also integrates multimodal imaging capabilities, providing a promising strategy for improved tumor ICD induction and cancer progression inhibition.
