Cyclodextrin-based electrospun nanofibers are promising for encapsulating and preserving unstable compounds, but quick dissolution of certain nanofibers hinders their delivery application. In this study, hydroxypropyl-β-cyclodextrin (HPβCD) was used as an effective carrier of resveratrol (RSV) to obtain the RSV/HPβCD inclusion complex (HPIC), which was then incorporated into pullulan nanofibers. For enhancement of RSV release toward colon target, multilayer structure with a pullulan/HPIC film sandwiched between two layers of hydrophobic Eudragit S100 (ES100) nanofibers was employed. The relationship between the superiority of the ES100-pullulan/HPIC-ES100 film and its multilayer structure was verified. The intimate interactions of hydrogen bonds between two adjacent layers enhanced thermal stability , and the hydrophobic outer layers improved water contact resistance. According to release results, multilayer films also showed excellent colon-targeted delivery property and approximately 78.58 % of RSV was observed to release in colon stage. In terms of release mechanism, complex mechanism best described RSV colonic release. Additionally, ES100-pullulan/HPIC-ES100 multilayer films performed higher encapsulation efficiency when compared to the structures without HPIC, which further increased the antioxidant activity and total release amount of RSV. These results suggest a promising strategy for designing safe colonic delivery systems based on multilayer and HPIC structures with superior preservation for RSV.

Psoralidin (PL) has recently been attracting more attention as a new anticancer agent candidate. Nevertheless, peroral administration of PL is largely challenged by its insoluble nature and intestinal efflux. This article aimed to develop a nanoencapsulation formulation of PL using water-soluble chitosan and Eudragit S100 and to evaluate its potential for bioavailability enhancement. PL-loaded nanocapsules (PL-NCs) were prepared by a solvent diffusion and high-pressure homogenization technique with Poloxamer 188 as a stabilizer. The resultant PL-NCs were approximately 132.5 nm in particle size and possessed a high entrapment efficiency (98.1%). In vitro release showed that PL was released less from the nanocapsules due to electrostatic complexation. A lipolytic experiment demonstrated that our prepared PL-NCs were not degraded by lipase , in contrast with the most commonly used lipid nanoparticles. Furthermore, PL-NCs appeared to have less affinity for intestinal mucins. Following oral administration, the bioavailability of PL was significantly enhanced via the PL-NCs, with a value of 339.02% relative to the reference (suspensions). Excellent intestinal adhesion and transepithelial permeability accounted for the enhancement of oral bioavailability. Taken together, these results indicate that nanoencapsulation of PL with chitosan and Eudragit S100 is a promising strategy for improved PL oral delivery.

A surface-enhanced Raman scattering (SERS)-based lateral flow assay (LFA) is described for the quantitative analysis of the proteinic stroke biomarker S100-β that has to be detected at very low concentration levels. The Raman reporter 5,5′-dithiobis-2-nitrobenzoic acid (DTNB) on gold nanoparticles (GNPs) was employed as the SERS tags. They are shown to perform much better than bare GNPs in LF strips. The S100-β protein can be detected by this method with very low detection limits by monitoring the intensity of the characteristic Raman peak of the S100-β protein-conjugated GNPs at 1332 cm −1 . Under optimized conditions, the assay works in the 1 pg·mL −1 to 40 ng·mL −1 S100-β concentration range, and the detection limit is as low as 0.14 pg·mL −1 . This is lower by a factor of 3 compared to colorimetric or fluorimetric methods.
Schematic illustration of the configuration (A) and the principle of the SERS-based lateral flow assay for quantification of S100-β (B).

Background: S100 calcium-binding protein A6 (S100A6) is a calcium-binding protein that is involved in a variety of cellular processes, such as proliferation, apoptosis, and the cellular response to various stress stimuli. However, its role in NAFLD and associated metabolic diseases remains uncertain. Methods and Results: In this study, we revealed a new function and mechanism of S100A6 in NAFLD. S100A6 expression was upregulated in human and mouse livers with hepatic steatosis, and the depletion of hepatic S100A6 remarkably inhibited lipid accumulation, insulin resistance, inflammation, and obesity in a high-fat, high-cholesterol (HFHC) diet-induced murine hepatic steatosis model. In vitro mechanistic investigations showed that the depletion of S100A6 in hepatocytes restored lipophagy, suggesting S100A6 inhibition could alleviate HFHC-induced NAFLD. Moreover, S100A6 liver-specific ablation mediated by AAV9 alleviated NAFLD in obese mice. Conclusions: Our study demonstrates that S100A6 functions as a positive regulator of NAFLD, targeting the S100A6-lipophagy axis may be a promising treatment option for NAFLD and associated metabolic diseases.

Probiotics, which play a crucial role in regulating intestinal homeostasis and maintaining host health, face significant challenges in maintaining their survival rate after processing, storage, and oral administration, posing a substantial hurdle in probiotic use. In this study, electrospinning technology was employed to prepare polyvinyl alcohol (PVA)/Pullulan (PUL) nanofibers and Eudragit® S100 (ES100)/PVA/PUL core-shell nanofibers. The effects of different PVA, PUL, and ES100 solution concentrations, applied voltage, collector distance, and feed rate on the morphology and diameter of nanofibers were investigated by scanning electron microscopy. The fibers were characterized using fourier transform infrared spectroscopy, X-ray diffraction, and thermal analysis. Additionally, the storage stability and resistance to gastrointestinal stress of the nanofibers loaded with Lactiplantibacillus plantarum KLDS1.0386 were investigated. The results indicate that the prepared PVA/PUL nanofibers and ES100/PVA/PUL core-shell nanofibers exhibit smooth surfaces, uniform diameters, and excellent thermal stability. Compared with free L. plantarum KLDS1.0386, the survival rate of PVA/PUL nanofiber-encapsulated bacteria increased by 25.07% after 21 days of storage at 4 °C and by 33.21% at 25 °C, respectively. Experiments simulating gastrointestinal digestion showed that the survival rates of PVA/PUL nanofibers and ES100/PVA/PUL nanofiber-coated L. plantarum KLDS1.0386 were 9.11% and 15.73% higher in simulated gastric juice compared to free L. plantarum KLDS1.0386, respectively. Additionally, 13.15% and 13.92% higher in simulated intestinal fluid, respectively. The nanofibers prepared in this study can be used as delivery carriers for probiotics. They enhance the storage stability and improve the tolerance of probiotics to the harsh gastrointestinal environment.

This study aimed to develop polymer Eudragit S100 for preparing pH-responsive liposomes-loaded betulinic acid (pH-BA-LP) to improve the therapeutic index of chemotherapy for colorectal cancer. BA-loaded liposomes were coated with Eudragit S100 by a thin film dispersion and easily scalable pH-driven method. The prepared liposomes were evaluated for size, surface morphology, entrapment efficiency, stability, in vitro drug release, and antitumor activity. In particular, pH-BA-LP showed advantages such as lower size (<100 nm), encapsulation efficiency of 90%, high stability, and stably cumulative release. By detecting the antitumor effects of pH-BA-LP in vivo, it showed that the tumor proliferation and cell migration were significantly inhibited in colorectal cancer. The pH-BA-LP also inhibited tumor growth via the regulation of Akt/TLR-mediated signalling and significantly down-regulated the expression of NFAT1 and NFAT4 proteins. It was found that pH-BA-LP can increase NK cells and CD3+ cells in tumor tissues, and the proportion of CD8+ cells in CD3+ cells was also increased, which proved that pH-BA-LP can play an antitumor effect by enhancing the autoimmunity level in tumor-bearing mice. The positive infiltration rates of CD8 and CD68 were increased and CD163 was relatively decreased by using pH-BA-LP, which proved that pH-BA-LP can regulate the immune infiltration levels in tumor-bearing mice. Therefore, the present work provides an effective method to prepare pH-responsive polymer-coated liposomes for colonic delivery with biologically active compounds.

Luteolin (Lu) is a kind of flavonoid that has been proved to treat non-alcoholic fatty liver disease by alleviating intestinal microbiota disorder. In this study, luteolin was coated with methoxy poly(ethylene glycol)–poly(dl-lactide-co-glycolic acid) (mPEG-PLGA) using an emulsion solvent evaporation method, and the optimum preparation process was determined by a single-factor experiment combined with response surface methodology (RSM). Methacrylic acid-methyl methacrylate (1:2) copolymer (Eudragit S100) was then used to coat the surface of Lu/mPEG-PLGA nanoparticles. The physical parameters of Eudragit S100-coated Lu/mPEG-PLGA nanoparticles (Lu-NPs), such as appearance, particle size, potential, particle size distribution and drug release, and stability in vitro, were evaluated. In addition, its cytotoxicity in vitro , pharmacokinetics, tissue distribution, and toxicity in vivo were also studied. The results showed that the prepared Lu-NPs had uniform particle size distribution, high encapsulation efficiency, and good stability. Normal colonic epithelial cells showed good tolerance to Lu-NPs. After oral administration, the blood concentration of luteolin peaked at 8 h, and the main tissue distribution was within the colon, confirming its colon-targeted profile. Safety assessments also indicated that no significant changes were observed in main organs after administration of Lu-NPs. The use of Eudragit S100-coated Lu/mPEG-PLGA nanoparticles is a new strategy for colon-targeted delivery of luteolin that encourages luteolin to fulfill its role in the colon. Graphical abstract

Vitamin C, a key antioxidant in the central nervous system, cycles between ascorbic acid and dehydroascorbic acid under pathophysiological conditions. Clinical evidence supports that the absence of vitamin C may be linked to depressive symptoms, but much less is known about the mechanism. Herein, we show that chronic stress disrupts the expression of ascorbic acid transporter, sodium-dependent vitamin C transport 2, and induces a deficiency in endogenous ascorbic acid in the medial prefrontal cortex, leading to depressive-like behaviors by disturbing redox-dependent DNA methylation reprogramming. Attractively, ascorbic acid (100 mg/kg-1000 mg/kg, intraperitoneal injection, as bioequivalent of an intravenous drip dose of 0.48 g–4.8 g ascorbic acid per day in humans) produces rapid-acting antidepressant effects via triggering DNA demethylation catalyzed by ten-eleven translocation dioxygenases. In particular, the mechanistic studies by both transcriptome sequencing and methylation sequencing have shown that S100 calcium binding protein A4, a potentially protective factor against oxidative stress and brain injury, mediates the antidepressant activity of ascorbic acid via activating erb-b2 receptor tyrosine kinase 4 (ErbB4)-brain derived neurotrophic factor (BDNF) signaling pathway. Overall, our findings reveal a novel nutritional mechanism that couples stress to aberrant DNA methylation underlying depressive-like behaviors. Therefore, application of vitamin C may be a potential strategy for the treatment of depression.

The therapeutic effect of 5-amino salicylic acid (5-ASA), a first-line therapeutic agent for the treatment of ulcerative colitis (UC), is limited by the modest bioavailability afforded by its oral administration. In this study, a 5-ASA oral delivery system was developed using Eudragit S100-coated iron oxide-chitosan nanocomposites (ES-IOCS/5-ASA) to address this issue. According to drug release studies in vitro, ES-IOCS/5-ASA only released a small amount of drug in simulated gastric fluid with a pH of 1.2. However, in a medium with a pH of 7.5, a relatively rapid and complete release was noted. 5-ASA-loaded iron oxide-chitosan nanocomposites (IOCS/5-ASA) could be effectively taken up by NCM460 cells and performed better anti-inflammatory effects than free 5-ASA. At the same time, IOCS/5-ASA improved barrier damage in DSS-induced NCM460 cells. In vivo models of dextran sulphate sodium (DSS)-induced colitis were used to assess the therapeutic efficacy of oral administration of ES-IOCS/5-ASA. ES-IOCS/5-ASA significantly relieved DSS-induced colitis and enhanced the integrity of the intestinal epithelial barrier. ES-IOCS/5-ASA also reduced the expression of NLRP3, ASC and IL-1β. Additionally, iron oxide nanoparticles used as nanozymes could alleviate inflammation. In summary, this study indicates that ES-IOCS/5-ASA exert anti-inflammatory effects on DSS-induced colitis by improving intestinal barrier function and inhibiting NLRP3 inflammasome expression, presenting a viable therapeutic choice for the treatment of UC.

This study aims to achieve co-delivery of paeoniflorin (PF) and butyric acid (BA) to the colon through Eudragit S100 (EUS100)-coated halloysite nanotube/o-butyryl chitosan microspheres (EUS100-HNT-BUCHNPs). O-butyrylated chitosan (BUCH) was prepared by the acylation reaction of chitosan (CH) and butyric anhydride with methane sulfonic acid as a catalyst. PF-loaded halloysite nanotube/o-butyrylated chitosan microspheres (HNT-BUCHNPs) were prepared by emulsion crosslinking with glutaraldehyde as the crosslinking agent and then coated with EUS100 using the emulsion solvent evaporation method. X-ray photoelectron spectroscopy (XPS) and scanning electron microscopy results indicated the presence of EUS100 coating on the surface of HNT-BUCHNPs. In vitro release studies demonstrated that the EUS100 coating effectively prevented the release of PF and BA at pH 1.2, while the halloysite nanotube slowed down PF release at pH 6.8. Sequential release experiments in different pH media showed lower release of PF and BA at pH 1.2 and pH 6.8, and higher release at pH 7.4, reaching approximately 80 % and 85 %, respectively. Gastrointestinal tract retention studies in mice confirmed that EUS100-HNT-BUCHNPs reduced the release of PF and BA in the upper digestive tract, facilitating delivery to the colon. In vivo and in vitro results indicated efficient transport of PF and BA to the colon, suggesting the potential of EUS100-HNT-BUCHNPs as colon-targeted carriers for PF and BA.

Postoperative delirium (POD) is a severe postoperative neurocognitive complication associated with neuroinflammation and blood-brain barrier dysfunction. As a clinically relevant circulating biomarker for POD, S100-β detection is of great importance for accurate diagnosis and progression evaluation. Traditional approaches generally rely on converting antigen-antibody recognition events into the changes in labeled signal molecules, while neglecting the intrinsic charge properties of S100-β. Herein, a bioinspired microchannel-based ion current rectification (ICR) sensor was developed for sensitive S100-β detection, where S100-β antibodies were immobilized on the microchannel surface to simulate natural molecular recognition processes. The low isoelectric point of S100-β (∼4.7) renders the protein negatively charged under neutral conditions. Specific antigen and antibody recognition increased the interfacial negative charge density of the microchannel, thereby facilitating cation transport and enhancing the rectification response under an applied electric field. This process mimics the biological processes of channel opening and ion influx in living systems. The detected rectification ratio (RR) values was linearly correlated with the logarithm of S100-β concentration in the range of 0.01 ∼ 5.0 ng/mL with a detection limit of 4.0 pg mL −1 (S/N = 3). The sensor also showed good selectivity, as well as high stability with small RSD of 2.45%. The proposed sensor was successfully applied to the clinical serum analysis, showing good agreement with enzyme-linked immunosorbent assay and high recovery rates of 97.8% - 104.0%. This work provides a promising strategy for POD-related biomarker analysis and broadens the application of bioinspired microchannel-based ICR sensors in clinical diagnostics and bioanalytical sensing.

The vicious cycle of reactive oxygen species (ROS) overload and bacterial infection critically impedes diabetic wound healing. While nanozyme-antibiotic dressings offer therapeutic potential, their uncontrolled agent release remains a key challenge. In this study, a functional nanofiber membrane (EIPR) based on the pH-sensitive polymer Eudragit S100 was fabricated via coaxial electrospinning to load iridium nanozymes (Ir NPs-PVP) and roxithromycin (Rox), enabling on-demand release of therapeutic agents for combined antibacterial and antioxidant wound therapy. The obtained dressing displays highly precise and controllable therapeutic agent release characteristics. Rox shows approximately 90 % cumulative release at pH 7.4, while release is suppressed to <8 % at pH 6.0 and 4.5. Notably, the release trend of Ir NPs-PVP mirrors that of Rox, with significantly higher release under alkaline conditions. The results exhibit >97 % eradication of both Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) coupled with >75 % ROS clearance at pH 7.4. This study presents a novel dual-function system, highlighting the simplicity and efficiency of electrospinning technology, which are key advantages enhancing its scalability for large-scale manufacturing and suitability for clinical wound care translation.
