Subarachnoid hemorrhage (SAH) is a common cerebrovascular disease that can lead to cognitive impairment. Although tetrandrine (Tet) has been proposed as a potential therapeutic agent, its efficacy in the treatment of SAH has not been fully explored. To investigate Tet effects on SAH, a rat model was established and divided into an SAH + vehicle and an SAH + Tet group. Cognitive function and behavioral performance were assessed using the Morris water maze and open field tests. Inflammatory cytokine levels were measured by ELISA, and hippocampal injury was evaluated by hematoxylin-eosin (HE) staining. Neuronal loss was quantified using Nissl staining, while neuronal density was assessed via immunofluorescence. The expression of the TLR4/NF-κB signaling pathway related-proteins was examined by Western blotting, and 16 S rRNA sequencing was conducted to determine differences in gut microbiota composition across the groups. Tet treatment significantly reduced mNSS and SAH assessment scores in SAH rats, suggesting an improvement in neurological and cognitive function. Behavioral analysis demonstrated that Tet increased escape latency, movement speed, and total distance in the Morris water maze. Histological staining revealed attenuated hippocampal damage and decreased neuronal death in the Tet-treated group. These neuroprotective effects were accompanied by reduced expression of TLR4 and NF-κB pathway components, as well as decreased secretion of LPS, TNF-α, IL-1β, and IL-6. Notably, Tet modulated the gut microbiota, restoring microbial diversity and abundance, and this modulation was associated with changes in the CYP51 metabolic pathway. Tet improves cognitive function and reduces neuronal injury in SAH rats by regulating the gut microbiota and its associated CYP51 metabolic pathway, thereby suppressing activation of the TLR4/NF-κB signaling cascade.

BACKGROUD Zinc (Zn) is an essential catalytic element in the human health system but its absorption in the intestinal system can be strongly affected by gastrointestinal (GI) digestion. In this study, the food-derived potential Zn carrier, scallop adductor hydrolysates (SAHs), was produced and characterized. RESULTS During temporary storage at 4 °C, SAH decreased in Zn-chelating capacity in the aqueous phase, whereas the SAH–Zn complex exhibited high stability. Moreover, the secondary structure of SAH had no significant alteration. Zn morphologically altered the surface structures of SAH, which was involving in carboxyl group of SAH. Results of in vitro GI digestion suggested that the SAH–Zn maintained good stability in GI system and only proportion of high molecular weight cleaved. In addition, SAH could successfully carry and transport Zn while the fluorescence staining revealed free Zn accumulation inside the tissue. Finally, three representative absorbed peptides (around 600 Da) were identified and synthesized. Three synthetic peptides exhibit higher Zn-chelating capacity than SAH and could also successfully transported through the intestine. CONCLUSION This study provided a theoretical basis for the investigation of digestion and absorption of marine animal-derived peptides as Zn carriers. © 2021 Society of Chemical Industry.

Selective chemical modification of cellulose in lignocellulosic
fibers without disrupting the native hierarchical structure remains
a central challenge in polysaccharide chemistry. Herein, we report
a reactive succinic anhydride-based deep eutectic solvent (SAH-DES)
strategy. The SAH-DES system, comprising choline chloride, succinic
anhydride, and water (1:1:1 molar ratio), forms a robust hydrogen-bonded
network to stabilize anhydride reactivity and suppress hydrolysis.
Under optimized conditions (100 ° C, 3 h), carboxylated bamboo
fibers (C-BFs) achieved a high carboxyl content of 1.01 ± 0.03
mmol/g while retaining excellent tensile strength (2104 ± 237
MPa). The introduced carboxyl groups enabled uniform deposition of
conductive materials (Ag/Ni nanoparticles, polypyrrole), resulting
in a high electrical conductivity of 34,271 ± 1266 S/m. This
study demonstrates that water-regulated reactivity in deep eutectic
solvents provides a pathway for selective cellulose esterification,
offering mechanistic insight into structure–reactivity relationships
of polysaccharides in confined solvent systems.

Objective Subarachnoid hemorrhage (SAH) is associated with high mortality and poor outcomes, which are closely related to white matter injury (WMI). (-)-Epigallocatechin-3-gallate (EGCG) exerts neuroprotective effects by inhibiting oxidative stress-related ferroptosis in astrocytes and improving neurological function. However, the role and mechanism of EGCG in regulating reactive astrocytes (RAs) to alleviate WMI after SAH remain unclear. Methods A mouse model of SAH was used to evaluate the effects of EGCG-loaded nanoparticles (EGCG-NPs). Western blot, qPCR, and immunofluorescence were performed for biochemical analysis. Neurological function was assessed using neurological deficit scores and the Morris water maze test. Results Following SAH, HO-1-mediated iron accumulation and ROS production promoted A1/A2 reactive astrocyte polarization, resulting in myelination damage and aggravated WMI. Knockdown of S100A10 inhibited iron-dependent oxidative stress in RAs and attenuated WMI. EGCG-NPs significantly suppressed HO-1/S100A10-mediated iron overload and oxidative stress in both A1 and A2 RAs, thereby alleviating WMI after SAH.Conclusion EGCG-NPs attenuate SAH-induced WMI by inhibiting the iron overload-activated HO-1/S100A10 axis in RAs, representing a promising therapeutic strategy.

Quantitative analysis of microRNAs (miRNAs) in a noninvasive manner is of vital importance for disease diagnosis and prognosis evaluation. However, conventional strategies for realizing accurate, simple, and sensitive detection of target molecules are still a challenge, especially for miRNAs due to their low abundance and susceptibility in the complex biological environment. Here, a novel surface-enhanced Raman scattering (SERS) strategy was established for quantitative detection and monitoring of miRNA-21-5p (miR-21-5p) in living cells and in vivo cerebrospinal fluid (CSF) by applying hairpin DNA (hpDNA)-conjugated gold nanostars (GNSs) SERS probes combined with acupuncture-based technology. This strategy enabled ultrasensitive exploration toward miR-21-5p in a wide range from 1 fM to 100 pM in cell lysates. Moreover, SERS analysis facilitated the detection and long-term monitoring for in vivo miR-21-5p noninvasively. This developed strategy promises to offer a powerful method for the analysis of multiple biomolecules in single cells and living bodies.

Mitochondrial dysfunction and subsequent autophagy, which are common features in central nervous system (CNS) disorders, were found to contribute to neuronal cell injury after subarachnoid hemorrhage (SAH). (-)-Epigallocatechin-3-gallate (EGCG), the main biological active of tea catechin, is well known for its beneficial effects in the treatment of CNS diseases. Here, the ability of EGCG to rescue cellular injury and mitochondrial function following the improvement of autophagic flux after SAH was investigated. As expected, EGCG-protected mitochondrial function depended on the inhibition of cytosolic Ca 2+ concentration ([Ca 2+ ] i ) influx via voltage-gated calcium channels (VGCCs) and, consequently, mitochondrial Ca 2+ concentration ([Ca 2+ ] m ) overload via mitochondrial Ca 2+ uniporter (MCU). The attenuated [Ca 2+ ] i and [Ca 2+ ] m levels observed in the EGCG-treated group likely lessened oxyhemoglobin (OxyHb)-induced mitochondrial dysfunction, including mitochondrial membrane potential depolarization, mitochondrial membrane permeability transition pore (mPTP) opening, reactive oxygen species (ROS), and cytochrosome c (cyt c) releasing. Subsequently, EGCG can restore the disrupted autophagy flux after SAH both at the initiation and formation stages by regulating Atg5 , LC3B , and Becn-1 ( Beclin-1 ) mRNA expressions. Thus, precondition EGCG resulted in autophagosomes and more autolysosomes compared with SAH group. As a result, EGCG pre-treatment increased the neurological score and decreased cell death. This study suggested that the mitochondrial dysfunction and abnormal autophagy flux synergistically contribute to SAH pathogenesis. Thus, EGCG can be regarded as a new pharmacological agent that targets both mitochondria and altered autophagy in SAH therapy.

Background
Subarachnoid hemorrhage (SAH) is a common acute condition in neurosurgery, with microglial function playing a crucial role in determining patient outcomes. However, the involved mechanisms are complex and demand thorough investigation. In our study, we combined transcriptomic and metabolomic approaches to identify key regulators of microglial function, offering novel insights for potential therapeutic strategies in SAH treatment.
Methods
The neurological function of SAH mice was evaluated using the open field test, rotarod test, and nissl staining. In an in vitro SAH model, key pathways and genes that regulate microglial function were identified through the integration of transcriptomics and metabolomics. Enzyme-Linked Immunosorbent Assay, Immunofluorescence, Western blot, Real-Time PCR, pHrodo, and Lyso-Tracker were employed to assess lysosomal and microglial function.
Results
In SAH mice, both neurological and microglial functions were significantly impaired on the first day but showed recovery over time. The combined results from transcriptomics and metabolomics identified 5-lipoxygenase (5-LOX) as a key target in regulating microglial function post-SAH. Administering resolvin D1 (RvD1) exogenously to inhibit 5-LOX nuclear translocation or directly supplementing lipoxin A4 (LXA4) to adjust the leukotriene B4 (LTB4)/LXA4 ratio, can improve lysosomal acidification, mitigate impaired lysosomal function, and enhance phagocytosis while reducing the inflammatory response. Protecting microglial function can also be achieved through JNJ or P2X purinoceptor 7 (P2X7) siRNA, which inhibit calcium/calmodulin-dependent protein kinase II (CaMKII) activation, prevent 5-LOX nuclear translocation, and improve lysosomal function. Furthermore, P2X7 siRNA also improved neurological function, lysosomal function and microglial function in SAH mice.
Conclusion
Activation of P2X7-CaMKII after SAH promotes 5-LOX nuclear translocation, increases the LTB4/LXA4 ratio, elevates intracellular Ca2+ concentration, and impairs lysosomal and microglial function.

Halide methyltransferase (HMT)-catalyzed synthesis of alkyl donors from S -adenosyl- L -homocysteine (SAH) and haloalkanes is a promising method for biocatalytic alkylation. However, the reported halide methyltransferases show limited catalytic capacity towards unnatural substrates such as ethyl iodide, which limits their application in biocatalytic ethylation. In this work, we performed structure-based directed evolution starting from the wild-type Aspergillus clavatus halide methyltransferase ( Ac HMT). The best variant, Ac HMT M3 , was identified with an activity of up to 421.5 mU mg –1 towards ethyl iodide, 38.7-fold higher than that of the wild type Ac HMT. Subsequently, molecular dynamics simulations provide structural insights into how mutations improve the catalytic activity towards iodoethane. Finally, Ac HMT M3 was utilized in one-pot cascade with the reported O -methyltransferase Rn COMT+Y200L to synthesize ethyl vanillin from 3,4-dihydroxybenzaldehyde (79% conversion). Our work demonstrated the potential of the engineered HMT in further biocatalytic alkylation.

A surface-enhanced Raman scattering (SERS)–based immunoassay with gold nanostars (GNSs) is utilized for determination of the subarachnoid hemorrhage (SAH) biomarker glial fibrillary acidic protein (GFAP) at very low concentration levels, which allows for early diagnosis and guides clinical decision-making to treat SAH-induced complications. The Raman reporter 5,5′-dithiobis-2-nitrobenzoic acid (DTNB) modified on GNSs was selected as the SERS tags. The SERS immunoassay was assembled by SERS tag and GFAP probe-immobilized ITO substrate. Therefore, the level of GFAP can be detected by monitoring the characteristic Raman peak intensity of GFAP-conjugated GNSs at 1332 cm −1 with a very low detection limit. Under optimized conditions, the assay can work in the GFAP concentration range from 1 pg⋅mL −1 to 1 μg⋅mL −1 , with a detection limit as low as 0.54 fg⋅mL −1 . The performance of the SERS immunoassay proven by the detection of GFAP is equivalent to that of the conventional enzyme-linked immunosorbent assay (ELISA). Graphical abstract Scheme 1. Schematic illustration of GNSs SERS immunoassay for ultrasensitive dynamic change detection of GFAP.
