Environmental pollution and medicine safety have aroused increasing public concerns due to human health. Amongst various contaminants, mercury is of special attention owing to their environmental persistence and biogeochemical recycling and ecological risks. Herein, a simple and highly parallel electrochemical biosensor for Hg determination was designed and investigated. The proposed biosensor was prepared and compared between (1) DTT/MB-DNA/Au with configuration occupation approach and (2) MCH/MB-DNA/Au with passivation approach. According to the combined results of scanning electrochemical microscope (SECM) and Randles-Sevcik equation, the DTT modified electrode exhibited high uniformity on DNA distribution and superb stability on electron transfer in Hg 2+ detection. Evidentially, the response value of proposed DTT/MB-DNA/Au was increased from 57.518% to 97.607%, while RSD% between duplicate runs had dropped from 22.658% to 0.223% (n = 3). Moreover, the increased proportion of effective working area was 467.380% compared with general sensors. Besides, DTT concentration, DNA concentration as well as assembly time were optimized, utilizing electrochemical impedance spectroscopy (EIS), Cyclic Voltammetry (CV) and Square Wave Anode Stripping Voltammetry (SWASV). This optimized biosensor exhibited an excellent selectivity toward Hg 2+ over Cu 2+ , As 2+ , Cd 2+ , Pb 2+ , Cr 3+ , Ni 2+ and Zn 2+ etc., and the stability of DTT/MB-DNA/Au were at least two times better even after 3 days under room temperature. Also, a linear relation was observed between the peak current and Hg 2+ concentrations in a range from 0.25 nM to 2.00 μM with a detection limit of 53.00 pM under optimal conditions. Finally, DTT/MB-DNA/Au was applied for plants and medical products analysis. In all, this optimized DTT/MB-DNA/Au with advantages of high repeatability and sensitivity would provide a new insight into the design and application of biosensor for reliable sensing in safeguarding plant protection and medicinal safety.

The degradation behavior of AZ91D magnesium alloy with micro-arc oxidation (MAO) coating in NaCl solutions was studied by scanning electrochemical microscopy (SECM). The investigation was carried out at SECM with 10 micron tip using ferrocenemethanol (FcMeOH) as the redox mediator in feedback mode. The variation of localized reactivity of the MAO coating are reflected by tip currents in SECM mapping images, and the generation and evolution of active site/sites on MAO coating are correlated to the concentration of Cl - in the solution and microstructure of the coating. With Cl - concentration increasing, the rate of generation of active sites is accelerated and meanwhile the reactivity of the active sites is enhanced. For the relatively thick and porous coating, there are lots of independent active sites incubating, generating and evolving on coating with the extension of immersion time. While the coating is relatively thin and smooth, the coexistence of slender and lower current peaks and pieces of active regions appears. The reactivity of the artificial line scratch and surrounding areas varied with the increase of immersion time, indicating that the generation, accumulation and dissolution of corrosion product affect the corrosion behavior of AZ91 alloy.

Understanding phase-specific corrosion behavior in multiphase alloys is critical for elucidating localized corrosion mechanisms. In this study, an integrated microelectrochemical cell technique (MECT) and scanning electrochemical microscopy (SECM) approach was developed to investigate in-situ electrochemical activity and local chemical environments on 2205 duplex stainless steel. Using a custom-fabricated dual-channel pipet, spatially resolved potentiodynamic polarization (PDP) and electrochemical impedance spectroscopy (EIS) measurements were performed at the α (ferrite) and γ (austenite) phases. Austenite exhibited higher corrosion current density i corr (60.4 μA·cm -2 ), lower charge transfer resistance R ct (39.2 kΩ·cm²), and reduced low-frequency impedance ( |Z| 0.1 Hz = 19.4 kΩ·cm²), indicating its preferential dissolution as the local anode in micro-galvanic coupling. Complementary in-situ pH mapping using a solid-state W/WO x micro-sensor revealed localized acidification and alkalization zones after 22, 32, and 56 h of immersion in acidic NaCl solution (pH = 1.5), coinciding with the onset and propagation of pitting. This dual-technique strategy provides unprecedented phase-resolved insight into corrosion dynamics, linking structural, electrochemical, and chemical heterogeneities across microdomains. The findings underscore the effectiveness of MECT-SECM integration for mechanistic studies of localized corrosion in duplex stainless steels and other complex alloys.

Cathode-electrolyte interphase (CEI) is critical for inhibiting the cathode degradation to maintain cell life. However, the evolution of the CEI is still unclear due to its complex and slow dynamic process. Here we used scanning electrochemical microscopy (SECM) for in situ investigation of CEI formation process on LiFePO4 cathode. Feedback images and probe scan curves showed a heterogeneous passivation that was gently generated on the LiFePO4 particles during both charging and discharging. Besides, a LiFePO4 composited electrode was also used to investigate the CEI formation to simulate the condition of real battery system. The composited cathode does not show obvious CEI formation within first two cycles. The SECM results between the pristine LiFePO4 particles and the composited LiFePO4 indicated the dynamic accumulation of CEI, which is influenced by the ability to charge transfer kinetics of cathode materials. This approach provided a feasible consideration for the connections between the dynamic evolution of the CEI and changes in charge transfer capability of cathode during cycling.

Hexamethylene diisocyanate trimer (HDIt) microcapsules were synthesized by in situ polymerization and were characterized via field emission-scanning electron microscopy, fluorescence microscopy, and Fourier transform infrared spectroscopy. We added HDIt microcapsules into epoxy resin coatings as functional additives and then assessed the resulting self-healing performance of the epoxy resin coatings using a scratched crevice on a 5083 aluminum surface as a test scenario. The protective coating with the HDIt microcapsules had robust self-healing after the samples were immersed in a solution of 0.6 M NaCl for 48 h. Healing efficiencies of epoxy resin coatings as high as 45.3 ± 2.9%, characterized by a fracture test, were achieved for 16 wt% HDIt microcapsules. The self-healing ability was further shown by using scanning electrochemical microscopy (SECM) and alternating current scanning electrochemical microscopy (AC-SECM). We found that AC-SECM offered additional local electrochemical topographies of the self-healing surface at different AC frequencies. Comparing the two topographical tests obtained from both SECM and AC-SECM revealed that AC-SECM achieved better spatial resolution and was less influenced by the scanning step size and the cleanliness of the tip surface.

The electrochemical carbon dioxide reduction reaction (CO 2 RR) often yields gaseous and liquid products simultaneously, making in situ, rapid, and distinguishable quantitative detection highly challenging due to product coexistence and signal overlap. To address this, we developed an electrochemical “fingerprint” collection and calibration framework suitable for multi-product CO 2 RR systems. Using Pt/Au ultramicroelectrodes, we record cyclic voltammograms across multiple scan rates to probe multicarbon alcohols in alkaline media and carboxylic acids in acidic media. The scan rate dependent current voltage responses were analyzed to identify characteristic potential and current features for each target species. After constructing calibration fingerprints for individual compounds, we resolved and validated the composite electrochemical features of binary and ternary multi-product systems with scanning electrochemical microscopy (SECM). By progressively advancing from single product systems to binary mixtures and ultimately to more realistic ternary product environments, we achieved effective discrimination of gas-liquid products under multi-component coexistence. This work establishes a detection and analysis workflow spanning from ultramicroelectrode-based CV calibration to SECM mixed-gas validation, providing a methodological foundation for process monitoring, rapid product identification, and qualitative analysis in complex, multiphase CO 2 RR systems.

A label-free method that can doubly image both the physical patterns and the electrochemical information of latent fingerprints (LFPs) on nitrocellulose (NC) membrane has been achieved here by water and scanning electrochemical microscopy (SECM). In this approach, the NC membrane with LFP (LFP/NC) sample was first placed in water, where the different absorption of water onto relatively hydrophobic ridge residues of LFP versus NC membrane background resulted in a high-resolution physical ridge pattern of the fingerprint to naked eyes within just 1–3 s and could further be photographed by a digital camera. Thereafter, the LFP/NC sample was electrochemically imaged by recording the current variations of SECM tip scanning over the label-free fingerprint in the surface-interrogation (SI) mode. The methyl viologen (MV 2+ ) was chosen as the redox mediator to react selectively with the electroactive species in the fingerprint ridge residues rather than furrow regions, which could cause the sharp contrast of the SECM tip current for imaging. Both the collected physical and electrochemical images of LFPs can provide high resolution up to level 2 and level 3 features required for personal identification. In addition, for the first time commercial NC membrane instead of adhesive forensic tape was discovered here for lifting the LFPs from various surfaces, which can then be imaged by this electrochemical approach. Taken together, this method demonstrates a powerful strategy for directly imaging the electrochemical information in LFPs without damaging the fingerprint physical ridge pattern on various substrates, so it has great potentiality in individual identity related applications.

The electron transfer process of protein on the surface of biomembrane is closely related to various physiological phenomena occurring on the biomembrane. Calcium cations (Ca 2+ ), as a ubiquitous messenger, induced electron transfer of negatively charged protein on the surface of negatively charged biomembrane is a problem worth exploring. In this work, the 3-Mercaptopropionic acid self-assembled monolayer (MPA SAM) with terminal carboxyl groups was constructed on gold surface as a simplified model for simulated surface of negatively charged biomembrane. The electron transfer of negatively charged redox ion induced by Ca 2+ on the surface of MPA SAM was investigated using scanning electrochemical microscopy. It was found that Ca 2+ can induce electron transfer and enhance the electron transfer of negatively charged redox ions. The result is attributed to the reduction of the energy barrier of the electron transfer of negatively charged redox ion on the surface of MPA SAM because Ca 2+ neutralizes the electrostatic repulsion interaction between negatively charged redox ion and negatively charged carboxylate surface of MPA SAM. The conditions of lower pH also benefit the above-mentioned electron transfer, which further confirms the mechanism of Ca 2+ -induced electron transfer. This work may provide a reference for studying the electron transfer process on the surface of simulated biomembrane.

Myocardial fibrosis progression and imbalanced redox state are closely associated with increased extracellular matrix (ECM) stiffness. Candesartan (CAN), an angiotensin II (Ang II) receptor inhibitor, has shown promising anti-fibrosis and antioxidant efficacy in previous cardiovascular disease studies. However, the effect of ECM stiffness on CAN efficacy remains elusive. In this study, we constructed rat models with three different degrees of myocardial fibrosis and treated them with CAN, and then characterized the stiffness, cardiac function, and NADPH oxidase-2 (NOX2) expression of the myocardial tissues. Based on the obtained stiffness of myocardial tissues, we used polyacrylamide (PA) gels with three different stiffness to mimic the ECM stiffness of cardiac fibroblasts (CFs) at the early, middle, and late stages of myocardial fibrosis as the cell culture substrates and then constructed CFs mechanical microenvironment models. We studied the effects of PA gel stiffness on the migration, proliferation, and activation of CFs without and with CAN treatment, and characterized the reactive oxygen species (ROS) and glutathione (GSH) levels of CFs using fluorometry and scanning electrochemical microscopy (SECM). We found that CAN has the best amelioration efficacy in the cardiac function and NOX2 levels in rats with medium-stiffness myocardial tissue, and the most obvious anti-fibrosis and antioxidant efficacy in CFs on the medium-stiffness PA gels. Our work proves the effect of ECM stiffness on CAN efficacy in myocardial anti-fibrosis and antioxidants for the first time, and the results demonstrate that the effect of ECM stiffness on drug efficacy should also be considered in the treatment of cardiovascular diseases. Keywords:myocardial fibrosis;candesartan (CAN);extracellular matrix (ECM);oxidative stress;scanning electrochemical microscopy (SECM)

Investigating the conversion of various kinds of weak interactions on protein surfaces may provide new insights in developing rational drugs. Herein, conversion from coordination bonds to electrostatic interactions occurring on the cysteine self-assembled monolayer (Cys SAM), a simplified protein surface, was investigated using scanning electrochemical microscopy (secm). On the Cys SAM electrode surface, the coordination bonds between Cys and copper (Cu [II]) were characterized by positive feedback. With the introduction of aspartic acid (Asp) on the electrode surface, the trend of positive feedback declined until it eventually presented negative feedback, indicating the conversion from the original coordination bonds to electrostatic interactions formed by Cys and Asp. This conversion phenomenon was driven by a higher binding affinity between Cys and Asp than between Cys and Cu (II). The conversion ratio was assessed to be about 89 %, based on the changes of the integral area of the Cu (II) reduction peak presented by cyclic voltammetry. Moreover, the binding specificity of Asp in electrostatic interaction with Cys compared to the other three amino acids was deemed to be structural matching. This work may facilitate the comprehension of the underlying mechanism of conversion phenomena on the biological surface at a molecular level.

The behaviors and kinetics of hydrogen evolution reaction (HER) on pure titanium spontaneously passivated in air (P-Ti), activated by electrochemical method (EA-Ti) and fluoride ion (FA-Ti) were investigated by impedance and scanning electrochemical microscopy (SECM). During cathodic polarization, the passive film and proton were simultaneously reduced on P-Ti, only HER current was detected on EA-Ti, while both HER and active/passive transition behavior presented on FA-Ti. Corresponding transfer coefficients and standard rate constants of HER are 0.48/6.00 × 10 −12 cm/s, 0.25/7.60 × 10 −7 cm/s and 0.24/1.14 × 10 −6 cm/s, respectively. These differences originate from the change in electronic structure of passive film by pretreatments.

As the new type of natural material, Basalt scales (BS) exhibited flaky micron-sized structure. Due to the acid and alkali resistance and stable chemical properties, it was suitable to act as the filler for anti-corrosion coatings. However, BS showed unsatisfactory compatibility with epoxy resins, and its application was limited. This paper combined the basalt scales with the organosilane by covalent grafting, and prepared organosilane modified basalt scales. The modification of organosilane promoted the combination of basalt scales and epoxy resin and the stability of BS in the coatings by covalent bonds, simultaneously improved the anti-corrosion performance of the coatings. Electrochemical impedance spectroscopy (EIS) experiments showed that organosilane significantly improved the anti-corrosion performance of Basalt scales epoxy coatings, especially the organosilane containing amino groups. The promoting effect of organosilane on the stable dispersion of BS was evaluated by the barrier ability of the coatings to corrosive solutions, and the diffusion dynamics analysis implied the existence of the promoting effect. The enhancement of shielding anticorrosion was result of the optimized dispersion and stability of modified basalt scales.
