Rapid Detection of Dezocine in Biological Fluids Based on SERS Technology

Siqingaowa HAN,Chen ZHANG,Zhijie CHEN,Xuanyu SHA,Wuliji HASI

This paper describes a method based on surface enhanced Raman spectroscopy (SERS) technology for rapid detection of dezocine in urine and serum. Firstly, an Ag colloid substrate was prepared and characterized. Then the Raman characteristic peaks of dezocine were assigned from both theoretical and experimental aspects. Finally, the Raman peak at 661 cm−1 was selected as its characteristic peak to perform SERS detection on dezocine in urine and serum, and the detection limits of dezocine in urine and serum were determined. The relationships between the characteristic peak intensity and the concentration of dezocine in urine and serum were fitted and the recovery rates were calculated. This rapid, accurate and non-destructive method establishes a good foundation for rapid on-site detection of dezocine in biological samples.

Fano enhancement of SERS for rapid early diagnosis of colorectal cancer

Tianxun Gong,Zhenjiang Wei,Libin Huang,Yan Hong,Yuan Li,Ke-ling Chen,Wen Huang,Xiaojing Zhong,Jinzhao He,Ming-Yi Lee,En-Chi Chang,Kien Voon Kong,Xiaosheng Zhang,Zongguang Zhou

Patients benefit greatly from early detection of colorectal cancer, but present diagnostic procedures have high costs, low sensitivity, and low specificity. However, it is still difficult to develop a strategy that can effectively detect cancer early using high-throughput blood analysis. Fano resonance-boosted SERS platform label-free serum creates an effective diagnostic system at the point of care. We obtained 220 high-quality SERS serum spectral datasets from 88 healthy volunteers and 132 patients with colorectal cancer. The biomarker detected in serum was further evaluated using 100 colorectal cancer tissues and adjacent normal intestinal tissues collected from West China Biobanks, West China Hospital, Sichuan University. The results showed that in 97 out of 100 paired samples, the biomarkers were successfully detected using the SERS platform. This demonstrates that Fano resonance-based SERS is highly effective for diagnosing colorectal cancer.

Rapid simultaneous detection of multi-pesticide residues on apple using SERS technique

Yizhi Zhang,Zhuyuan Wang,Lei Wu,Yuwei Pei,Peng Chen,Yiping Cui

A rapid and straightforward method has been employed to simultaneously detect two pesticides (thiram and methamidophos (MTD)) on apple surface using surface enhanced Raman scattering (SERS) technique. In the experiment, ethanol was dropped onto the contaminated apple surface for pesticide extraction and then gold@silver core–shell nanorods (Au@Ag NRs) were added to generate the SERS signals of the pesticides. Under a laser excitation at 632.8 nm, prominent SERS peaks of blended contaminants were observed, which were chosen to characterize and quantify their concentration. It was found that the SERS intensity of these two peaks changed as a function of the concentration ratio of thiram to MTD. In addition, a better SERS enhancement performance of Au@Ag NRs was demonstrated compared with that of gold nanorods. Our experimental results show that the lowest detectable concentration on apple surfaces is ∼4.6 × 10−7 M for thiram and ∼4.4 × 10−4 M for MTD. This study provides a straightforward method for the simultaneous detection of multiple pesticides on fruit surfaces, which is important for food safety and human health.

Establishment of a reliable scheme for obtaining highly stable SERS signal of biological serum

Hongmei Li,Qing Wang,Jun Tang,Ningning Gao,Xiaxia Yue,FuRu Zhong,Xiaoyi Lv,Jihong Fu,Tao Wang,Cailing Ma

As a rapid and non-destructive biological serum detection method, SERS technology was widely used in the screening and medical diagnosis of various diseases by combining the analysis of serum SERS spectrum and multivariate statistical algorithm. Because of the high complexity of serum components and the variability of SERS spectra, which often resulted in the phenomenon that the SERS spectrum of the same biological serum was significantly different due to the different test conditions. In this experiment, through the dilution treatment of the serum and the systematic test of the serum of all concentration gradients with lasers of wavelength of 785, 633 and 532 nm, the most suitable conditions for detecting the serum were investigated. The experimental results showed that only when the serum is diluted to low concentration (10 ppm), the SERS spectrum with high reproducibility and stability could be obtained, furthermore, the low concentration serum had weak tolerance to laser, and 532 nm laser was not suitable for serum detection. In this paper, a set of test scheme for obtaining highly stable serum SERS spectra was established by using high-performance gold nanoparticles (Au NPs) as the active substrate of SERS. Through comparative analysis of SERS spectrum of serum of normal people and cervical cancer, the reliability of the established low-concentration serum test program was verified, as well as its great potential advantages in disease screening and diagnosis.

Nasopharyngeal cancer screening and immunotherapy efficacy evaluation based on plasma separation combined with label-free SERS technology

Ruiying Lin,Zhangying Jiang,Jinyong Lin,Feifei Tong,Yangmin Wu,Jinquan Hong,Sufang Qiu,Qiong Wu

Background In recent years, significant progress has been made in the treatment of nasopharyngeal carcinoma (NPC). The application of immunotherapy, especially the use of Programmed Cell Death Protein 1 inhibitors, has demonstrated excellent therapeutic effects. However, inconsistent immunotherapy outcomes due to individual differences in patients remain a major challenge, making the search for specific biomarkers to screen for the population who may benefit from immunotherapy a priority. Surface-enhanced Raman spectroscopy (SERS) has shown great potential as a highly sensitive and specific optical analytical tool for identifying and monitoring tumor-related markers in NPC. Results In this study, plasma samples from Nasopharyngeal cancer patients before and after immunotherapy, along with those from healthy volunteers, were tested using label-free SERS combined with plasmapheresis. Especially, the components with varying molecular weight sizes were analyzed via the separation process, thereby preventing the potential loss of diagnostic information that could result from competitive adsorption. Subsequently, a robust machine learning algorithm based on principal component analysis and linear discriminant analysis (PCA-LDA) was used to extract features from plasma SERS data and establish an effective discriminant model. The results showed that in the upper plasma layer, the most optimal discrimination was found between normal patients and those post-treatment, with sensitivity and specificity of 88.5% and 92.3%, respectively. In the lower plasma layer, the most optimal discrimination was found between the pre-treatment and post-treatment patients, with sensitivity and specificity of 84.6% and 80.8%, respectively. Significance and novelty Plasmapheresis can reveal latent diagnostic information in the plasma and holds promise for treating NPC, the screening of the population who may benefit from immunotherapy, and the postoperative evaluation of immunotherapy. Further exploration of the feasibility of using SERS to detect tumor markers in the population who may benefit from immunotherapy for NPC will help improve the therapeutic efficacy, optimize clinical practice, and promote the development of individualized treatment strategies.

Ag NP-filter paper based SERS sensor coupled with multivariate analysis for rapid identification of bacteria

Rong Wang,Jiamin Luo

Rapid and accurate identification of bacteria is essential to ensure food safety and prevent pathogenic bacterial infection. In this study, a highly efficient method was established for accurately identifying bacterial species by applying Ag NP-filter paper based Surface enhanced Raman spectroscopy (SERS) analysis and Partial Least Squares-Discriminant Analysis (PLS-DA) statistical methods. The flexible Ag NP filter paper substrate with high sensitivity and uniformity was prepared by a facile and low-cost silver mirror reaction at room temperature, which exhibited desirable SERS activity in bacteria detection. Furthermore, PLS-DA was successfully employed to distinguish SERS spectra from S. aureus CMCC 26003, E. faecalis ATCC29212 and L. monocytogenes ATCC 19115 with a sensitivity of 93.3–100%, specificity of 96.7–97%, and overall predicting accuracy of 95.8%. This exploratory study demonstrates that a Ag NP-filter paper based SERS sensor coupled with PLS-DA has great potential for rapid and effective detection and identification of bacteria.

Advanced SERSome-Based Artificial-Intelligence Technology for Identifying Medicinal and Edible Homologs

Shuang Jiang,Yue Zhao,Qingyu Meng,Rongheng Ma,Xiaomeng Sun,Xiaoming Lyu,Li Zhang,Guangrun Wu,Xiaotong Wang,Yiyang He,Jiayue Liang,Yuxin Qiao,Zhibin Wang,Yanli Wu,Jing Wu,Chunjuan Yang,Huimin Yu,Yang Li

Medicinal and edible homologs (MEHs) offer significant preventive and therapeutic benefits for various diseases and health functions. However, the widespread application of MEHs faces significant challenges, particularly in quality control, rapid identification, and authentication. In this study, we present a novel approach that combines surface-enhanced Raman spectroscopy (SERS) based on spectral set, referred to as “SERSome”, with deep learning to develop an identification model for analyzing MEHs. The platform uses silver nanoparticles prepared via reduction with NaBH 4 , activation with sodium borohydride, and aggregation with calcium ions. The method avoids the use of additional protective agents during the reaction process, thereby reducing interference from the protective agents and other materials. Additionally, the method also overcomes the fluorescence interference from MEHs. SERSomes provide comprehensive molecular fingerprint recognition, significantly enhancing the system's detection accuracy. The introduction of Ca 2+ as aggregator promotes the aggregation of silver nanoparticle, significantly enhancing the electromagnetic field of the nanoparticle system. The study achieves up to 98% accuracy in identifying specific MEHs. The integration of SERSomes with deep learning offers a promising methodology for the rapid and modern detection of MEHs, advancing the MEHs industry and contributing to the protection of human health.

A SERS aptasensor for simultaneous multiple pathogens detection using gold decorated PDMS substrate

Nuo Duan,Mofei Shen,Shuo Qi,Wenyue Wang,Shijia Wu,Zhouping Wang

An aptamer-based sensitive method was developed here for detection of multiple foodborne pathogens in food matrix by surface-enhanced Raman scattering (SERS) technology. Polydimethylsiloxane (PDMS) film was first prepared and then coated with gold nanoparticles (AuNP) to act as an active substrate for the enhancement of Raman scattering. The as-prepared Au-PDMS film was functionalized with specific pathogen aptamers (Apt) to capture the targets. In addition, aptamers functionalized AuNP integrated with Raman reporters (4-Mercaptobenzoic acid (4-MBA)/Nile blue A (NBA)) were fabricated as pathogen-specific SERS probes. In this scheme, pathogens were first captured by Apt-Au-PDMS film and then bind with SERS probes to allow the formation of a sandwich assay to complete the sensor module for the detection of multiple pathogens. With Vibrio parahaemolyticus and Salmonella typhimurium as model targets, this protocol can selectively detect 18 cfu/mL and 27 cfu/mL, respectively. Furthermore, this platform can be successfully applied to detect pathogens in seafood samples with recoveries ranging from 82.9% to 95.1%.

Tailored SiO2-TiO2 Aerogel/Ag Flexible Films as Stable SERS Substrates for Hazardous Materials Detection

Wei Liu,Xue Wang,Zhiyang Zhao,Xin Zhou,Shiliang Huang,Longjin Huang,Zihan Wang,Jingru Li,Junjie Fu,Yu Liu,Sheng Cui,Xuan He

Flexible surface-enhanced Raman spectroscopy (SERS) substrates have received more and more attention for their non-invasive sample collection ability. However, majority of them have poor stability and reproducibility because noble metal is easily oxidized and led to lose SERS activity. Herein, SiO 2 -TiO 2 -Ag hybrids aerogel (STAHA) material is first applied as a flexible SERS substrate for explosives and dyes long-term sensitive detection. The sensitive non-invasive detection of 3-nitro-1, 2, 4-triazol-5-one (NTO) residues can be achieved with a limit of 1 × 10 −6 m by wiping sample collection method. Meanwhile, similar SERS sensitivity from long-term (50 days) kept substrates is observed compared with fresh-made substrates. This long-term sensitivity is contributed to electronic transfer compensation from TiO 2 aerogel. This proposed flexible STAHA enrich the type of SERS substrates, and their long-term sensitivity can provide a prospective application for trace analyses of explosives and other hazardous residues in criminal forensic, military, and security check.

Bicycled Hairpin Assembly-Mediated SERS/Colorimetric Dual-Mode Sensor for the Detection of Dexamethasone in Milk and Pork

Rui Tan,Ziji OuBu,Qun Zhou,Juan Mu,Chenxi Yi,Xiaojun Luo

The presence of dexamethasone (DEX) residues and the illegal addition of DEX to food have garnered widespread public concern over recent decades. Based on the bicycled hairpin assembly (BHA) mechanism and the catalytic oxidase effect of urchin-shaped Au–Ag alloy loaded with Pt nanoparticles (Au@Ag@Pt), an accurate and ultrasensitive surface-enhanced Raman scattering (SERS)/colorimetric dual-mode method for DEX detection is pioneered. When DEX is introduced into the sensor system, the BHA cycle begins. The Fe3O4 nanosphere loaded with Au nanoparticles (Fe3O4@Au) surface facilitates the anchoring of a large amount of Au@Ag@Pt, resulting in synergistic signal amplification and facile magnetic separation from the complex system. Finally, the collected product effectively catalyzes the conversion of 3,3′,5,5′-tetramethylbenzidine (TMB) to the oxidized product, producing a strong SERS response, an ultraviolet–visible absorption signal, and color deepening. Our dual-mode platform achieves ultrasensitive detection of DEX, achieving a detection limit (LOD) of 6.61 × 10–17 M within the broad range of 10–16 to 10–9 M for SERS and a LOD of 8.81 × 10–13 and 6.98 × 10–13 M within the range of 10–12–10–5 M for the colorimetric method. Additionally, this platform shows high selectivity and satisfactory recovery rates for DEX in milk and pork samples, with recoveries of 92.3%–106.5% (RSDs <8.35%) for SERS and 95.2%–107.6% (RSDs <8.81%) for colorimetric analysis. This study demonstrates that by combining the BHA amplification process with multifunctional Au@Ag@Pt and Fe3O4@Au substrates, the DEX content in actual food samples can be accurately determined using a dual-mode approach that combines the high sensitivity of SERS with the simplicity of colorimetric analysis.

RETRACTED ARTICLE: A SERS-based lateral flow assay for the stroke biomarker S100-β

Wang Ying,Hou Yajun,Li Hanxia,Yang Mingfeng,Zhao Peng,Sun Baoliang

A surface-enhanced Raman scattering (SERS)-based lateral flow assay (LFA) is described for the quantitative analysis of the proteinic stroke biomarker S100-β that has to be detected at very low concentration levels. The Raman reporter 5,5′-dithiobis-2-nitrobenzoic acid (DTNB) on gold nanoparticles (GNPs) was employed as the SERS tags. They are shown to perform much better than bare GNPs in LF strips. The S100-β protein can be detected by this method with very low detection limits by monitoring the intensity of the characteristic Raman peak of the S100-β protein-conjugated GNPs at 1332 cm −1 . Under optimized conditions, the assay works in the 1 pg·mL −1 to 40 ng·mL −1  S100-β concentration range, and the detection limit is as low as 0.14 pg·mL −1 . This is lower by a factor of 3 compared to colorimetric or fluorimetric methods. Schematic illustration of the configuration (A) and the principle of the SERS-based lateral flow assay for quantification of S100-β (B).

Preparation of SERS substrate with 2D silver plate and nano silver sol for plasticizer detection in edible oil

Haonan Wang,Chuyi Wang,Jiali Huang,Yunzhen Liu,Yang Wu,Ruiyun You,Jian-Han Zhang,Yudong Lu,Huiying Shen

As a widely used industrial additive of plastic products, phthalate ester (PAE) plasticizers can easily migrate into food, threatening human health. In this work, we proposed a rapid, precise, and reliable method to detect PAE plasticizers in edible oils by using surface-enhanced Raman spectroscopy (SERS) technology. A two-dimensional (2D) silver plate synergizing with a nanosilver sol was prepared as a substrate for SERS to detect potassium hydrogen phthalate (PHP), a hydrolysate of a PAE plasticizer. Detection conditions, such as pH values, drying times, and hydrolysate interference, were optimized. The working curve was well fitted with a linear parameter R 2 of 0.9994, and the minimum detection limit was evaluated as 10 −9  mol/L. Furthermore, the detection accuracy was supported by five edible oil samples. Therefore, using SERS technology to detect PHP is expected to provide an avenue for PAE plasticizer detection in oils and fats, and it features promising potential applications in food safety.
