Synthesis of novel silica-encapsulated core-satellite nanotags for enhanced SERS detection

Haoquan Wang,Qi Yang,Yan Zhang,Bo Peng,Feng Wu,Zewei Quan

Developing a SERS nanotag with high signal intensity, excellent signal stability, and biocompatibility is of significant importance in the fields of food safety, drug testing, and virus detection. In this work, silica-encapsulated, indocyanine green (ICG)-modified Au octahedral core-small Au spherical nanoparticles (NPs) satellite nanostructures (Au/Au@ICG@SiO 2 ) are uniformly synthesized as surface-enhanced Raman scattering (SERS) tags, which provide abundant “hot spots” and also increase the stability of Raman reporter molecule ICG to greatly facilitate subsequent functionalization and detection. Both experimental and numerical results demonstrate that this Au/Au@ICG nanostructure achieve a detection limit of 10⁻⁷ M for ICG in aqueous solution ascribed to the increased number of hot spots compared with Au octahedral NPs. As-prepared Au/Au@ICG@SiO 2 SERS nanotags are further integrated with a lateral flow immunoassay (LFIA) for the detection of the SARS-CoV-2 nucleocapsid protein. Compared to the traditional bio-recognition method on LFIA strips with the detection limit of 8 pg/mL, the detection capability for the SARS-CoV-2 nucleocapsid protein is improved approximately 16 times (500 fg/mL) based on this novel SERS tag and custom-built adaptive Raman mapping spectrometer. This Au/Au@ICG@SiO 2 SERS tag is expected to play a significant role in early detection, prevention, and control of similar infectious viruses in the future.

Dual-mode of magnetic assisted Au@Ag SERS tags and cationic conjugated UCNPs for qualitative and quantitative analysis of multiple foodborne pathogens

Yi Xu,Md Mehedi Hassan,Afang Zhu,Huanhuan Li,Quansheng Chen

A new dual-mode optical-nano-sensor was designed to effectively isolate, discriminate and inactivate multiple foodborne pathogens through the combined effort of magnetic SERS tags and functionalized upconversion nanoparticles. The positively charged quaternary ammonium salt (QAS) functionalized upconversion nanoparticles (UCNPs@QAS) were used as a fluorescent probe for the analysis of total bacteria count and inactivation of bacteria, and 4-mercaptophenylboronic acid (4-MPBA) modified magnetic Au@Ag NPs (MAu@Ag@MPBA) were applied as a SERS tag for isolation and classification of bacteria. Various types of bacteria can be captured by UCNPs@QAS through electrostatic interactions and by MAu@Ag@MPBA via the reversible recognition interaction of a boronic acid group of 4-MPBA and peptidoglycan from bacteria simultaneously. And then the formed complex structural assemblies were isolated rapidly from the sample matrix by employing the magnetic force for fast and simple dual-model optical detection. In addition, the novel biosensor was successfully applied to discriminate and detect Escherichia coli , Salmonella , Staphylococcus aureus, and their mixture in contaminated milk with the classification rate of 97.2–100 %. The proposed multi-functional nano-platform was able to sensitively capture, detect, and kill foodborne pathogens in a complex system, which could be deployed successfully towards the monitoring of food safety.

Polystyrene Encapsulated SERS Tags as Promising Standard Tools: Simple and Universal in Synthesis; Highly Sensitive and Ultrastable for Bioimaging

Qian Yu,Yunqing Wang,Rongchao Mei,Yingchao Yin,Jinmao You,Lingxin Chen

Surface coating determined the sensitivity and stability of surface-enhanced Raman scattering (SERS) tags in bioanalysis. The reported various coatings suffered from the drawbacks of a lack of rigidity, stability, or synthesis versatility. Herein, we demonstrated robust polystyrene (PS) coated SERS tags that could be prepared by an easy and universal approach. Taking advantages of biocompatible, transparent, compact properties of PS shell, the coated tags showed satisfactory sensitivity, biocompatibility, and superior structural stability in cell and in vivo imaging applications. More importantly, the PS coating strategy allowed for the encapsulation of SERS tags encoded with not only thiolated but also nonthiolated Raman reporters without loss of sensitivity, as exemplified in the synthesis of 9 different resonant dye-encoded tags. Moreover, the coating of SERS tags with various kinds of substrates was achieved via the same standard protocol. Comparing with widespread silica coated tags, the PS coated ones were more stable in harsh conditions and had an easily expanded ultrasensitive (resonant) tags library with much lower cost (no need of expensive sulfhydryl/isothiocyano reporters with limited types), illustrating great promise as standard analytical tools of commercialized value for bioanalysis, medical diagnostics, and environmental science studies.

Gold-nanorod-enhanced Raman spectroscopy encoded micro-quartz pieces for the multiplex detection of biomolecules

Wang Bei,Guan Tian,Jiang Jingying,He Qinghua,Chen Xuejing,Feng Guangxia,Lu Bangrong,Zhou Xuesi,He Yonghong

The rapid analysis and detection of biomolecules has become increasingly important in biological research. Hence, here we propose a novel suspension array method that is based on gold nanorod (AuNR)-enhanced Raman spectroscopy and uses micro-quartz pieces (MQPs) as microcarriers. AuNRs and Raman reporter molecules are coupled together by Au–S bonds to obtain surface-enhanced Raman scattering labels (SERS labels). The SERS labels are then assembled on the surfaces of the MQPs via electrostatic interactions, yielding encoded MQPs. Experimental results showed that the encoded MQPs could be decoded using a Raman spectrometer. A multiplex immunoassay experiment demonstrated the validity and specificity of these encoded MQPs when they were used for bioanalysis. In concentration gradient experiments, the proposed method was found to give a linear concentration response to the target biomolecule at target concentrations of 0.46875–30 nM, and the detection limit was calculated to be 1.78 nM. The proposed method utilizes MQPs as carriers rather than conventional microbeads, which allows the interference caused by the background fluorescence of microbeads to be eliminated. The fluorescence of the encoded MQPs can be simply, rapidly, and inexpensively quantified using fluorescence microscopy. By dividing the quantitative and qualitative detection of biomolecules into two independent channels, crosstalk between the encoded signal and the labeled signal is averted and high decoding accuracy and detection sensitivity are guaranteed. Synthesis of MBA-Encoded Silver/Silica Core-Shell Nanoparticles as Novel SERS Tags for Biosensing Gibberellin A3 Based on Au@Fe3O4 as Substrate

Qingmin Wei,Jianjuan Lin,Fa Liu,Changchun Wen,Na Li,Guobao Huang,Zhihui Luo

A surface-enhanced Raman scattering (SERS) tag is proposed for high-sensitivity detection of gibberellin A3(GA3). Silver nanoparticles (AgNPs) were synthesized using citrate reduction. 4-Mercaptobenzoic acid (MBA) was used for the Raman-labeled molecules, which were coupled to the surface of the AgNPs using sulfydryls. MBA was coated with silica using the Stöber method to prevent leakage. GA3antibodies were attached via the active functional groups N-Hydroxysuccinimide (NHS) and N-Ethyl-N’-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC) to construct a novel[email protected]2SERS tags. The captured SERS substrates were fabricated through Fe3O4nanoparticles and gold nanoparticles (AuNPs) using chemical methods. These nanoparticles were characterized using ultraviolet-visible spectroscopy (UV–Vis), dynamic light scattering, Raman spectroscopy, transmission electron microscope (TEM), and X-ray diffraction (XRD). This[email protected]2SERS tags has a strong SERS signal based on characterizations via Raman spectroscopy. Based on antigen-antibody reaction, the[email protected]3O4nanoparticles can capture the GA3and[email protected]2SERS tags. Due to the increasing number of captured nanoprobes, the SERS signal from MBA was greatly enhanced, which favored the sensitive detection of GA3. The linear equation for the SERS signal was y = −13635x + 202211 (R2= 0.9867), and the limit of detection (LOD) was 10−10M. The proposed SERS tags are also applicable for the detection of other food risk factors.Keywords:surface-enhanced Raman scattering;[email protected]2core-shell nanoparticles;[email protected]3O4nanoparticle;gibberellin

A SERS bioassay based on vancomycin-modified PEI-interlayered nanocomposite and aptamer-functionalized SERS tags for synchronous detection of Acinetobacter baumannii and Klebsiella pneumoniae

Clement Yaw Effah,Lihua Ding,Longlong Tan,Sitian He,Xiang Li,Huijie Yuan,Yi Li,Shaohua Liu,Tongwen Sun,Yongjun Wu

Klebsiella pneumoniae ( KP ) and Acinetobacter baumannii ( AB ) are two important gram-negative bacteria that cause pneumonia and have been recently known to be associated with food. The rapid detection of these pathogens in food is important to minimize their colonization of the gut and stop new threats of the disease from spreading across the food chain. Herein, a double-edged sword aptasensor was developed for the synchronous detection of KP and AB in food and clinical samples. A highly sensitive, selective, specific, and synchronous detection of the target bacteria was achieved, and the limit of detection (LOD) was 10 cells/mL with a liner range of 50 to 10 5 cells/mL. The total assay time was 1.5 h. This study does not only provide a new tool for the detection of the target bacteria, but also serves as a promising tool for food safety and pneumonia diagnosis.

A boronate-modified molecularly imprinted polymer labeled with a SERS-tag for use in an antibody-free immunoassay for the carcinoembryonic antigen

Feng Jun,Li Xuan,Cheng Hao,Huang Wenyi,Kong Hongxing,Li Yanqing,Li Lijun

An antibody-free immunoassay that makes use of a boronate affinity molecularly imprinted polymer (MIP) and surface enhanced Raman scattering (SERS) tags is described. It was applied to the specific determination of the carcinoembryonic antigen (CEA) in human serum. For the preparation of the boronate affinity array, a polymer capable of adsorbing glycoproteins was first synthesized on the surface of a glass slide with four spots using 4-vinylbenzeneboronic acid (VPBA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the crosslinking agent, and ethylene glycol and cyclohexanol as porogens. The surface of the VPBA-Co-EGDMA can bind target glycoproteins. After specific capture of the glycoprotein, a “MIP-target glycoprotein-SERS tag” sandwich structure was formed by covalent interaction between the SERS nanotag (consisting of gold nanoparticles and 4-mercaptophenylboronic acid [MPBA]). CEA can be quantified in spiked serum with a detection limit of 0.1 ng·mL −1 via the specific Raman band at 1098 cm −1 . Schematic representation of the boronate affinity molecularly imprinted polymer (MIPs) array-based SERS sensor for rapid and sensitive detection of the carcinoembryonic antigen (CEA) from human serum. The boronate affinity MIPs array are used as capture probes, and MPBA@AuNPs are used as SERS tags.
