Oral ulcer (OU) is one of the most common mucosal diseases, yet current drug treatments yield unsatisfactory outcomes. Persistent inflammatory responses and insufficient angiogenesis are the two major obstacles to OU healing. Recently, stem cell-based therapies, particularly mesenchymal stem cells (MSCs), have shown great regenerative potential through their anti-inflammatory and proangiogenic properties in OU treatment. However, they still face challenges, such as low cell survival rates, uncontrolled differentiation, and immune rejection. Meanwhile, a humid and highly dynamic oral environment degrades and dilutes biological drugs via saliva, thereby reducing their bioavailability during OU repair. To address these limitations, we developed an injectable fibrinogen/thrombin (FT) hydrogel encapsulating cell lysate (CL) derived from the stem cells of human exfoliated deciduous teeth (SHEDs). The SHED-derived CL retained the therapeutic properties of SHEDs while eliminating risks associated with cell transplantation. The FT hydrogel exhibited excellent biocompatibility, controlled CL release, and strong adhesion to oral wounds (> 24 h). In vitro, the FT/CL hydrogel polarized macrophages toward the anti-inflammatory M2 phenotype (upregulating CD206 and Arg1 expression) and suppressed pro-inflammatory M1 markers (iNOS and TNF-α) secretion. It also significantly enhanced tube formation, with a 2.5-fold increase in luminal length and 3.7-fold increase in the number of tubes compared with that in the control group. In a rat OU model, the FT/CL group showed accelerated ulcer healing, clearly reducing the inflammatory response on day 3 and nearly restoring epithelial integrity by day 5. Additionally, the FT/CL hydrogel effectively inhibited inflammatory infiltration and alleviated pain at the wound site, with effects similar to those of the positive control (the watermelon frost group). Furthermore, the FT/CL hydrogel promoted cell proliferation in the epithelial tissue and enhanced vascular remodeling near the basement membrane, thereby accelerating ulcer healing. Therefore, the FT/CL hydrogel promotes ulcer healing by regulating inflammation, promoting cell proliferation, and enhancing angiogenesis. This study provides a safe and efficient method for enhancing the therapeutic application of SHED-derived CL. The findings suggest that CL-based cell-free therapy may be a promising strategy for future clinical use in ulcer and chronic wound healing. Graphical abstract

Micro-nano bioactive glass (MNBG) has bone regenerative potential. However, the difficulty of MNBG molding restricts its clinical applications in bone regeneration. Exosomes carry proteins, lipids, and nucleic acids for communication between cells. In this study, we adhered human exfoliated deciduous teeth (SHED)-derived exosomes (SHED-Exos) to electrospun micro-nano bioactive glass/polycaprolactone (MNBG/PCL) membrane to control inflammation and promote bone regeneration. MNBG/PCL membrane showed biocompatibility and osteogenic ability. Next, we demonstrated that SHED-Exos internalized into macrophages attenuated LPS-induced expression of M1-macrophage markers iNOS, IL-6, and IL-1β and upregulated M2-macrophage marker IL-10 expression, indicating a switch from M1 to M2 macrophage phenotype. MNBG/PCL-loaded SHED-Exos membrane supported the survival and growth of mouse bone marrow stromal cells (mBMSCs). When M1 macrophages were co-cultured with mBMSCs on the membrane, the SHED-Exos-loaded membrane showed higher Runx2, Alp, and Ocn gene expression. Our findings indicate that SHED-Exos-functionalized MNBG/PCL membrane has anti-inflammatory and osteoinductive potential, suggesting its potential as a candidate material for bone regeneration.

Fruit colour is mostly determined by the composition and content of carotenoids . In this study, a metabolic transcription analysis of carotenoids in the pericarp of ‘Shatangju’ at three different growth periods under two cultivation methods, open-air and netting, was performed. The results showed that the lightness value, red-green value and yellow-blue value indices of the pericarp varied greatly. Fourteen carotenoids were detected in the citrus pericarp. The content of lycopene was 3.21 μg/g and 3.43 μg/g, and the content of β-citraurin was 6 μg/g and 7.27 μg/g under open-air and net shed cultivation, respectively. Shading affected the synthesis of key genes such as phytoene synthase ( PSY ), carotenoid cleavage dioxygenase 4 ( CCD4 ), and zeaxanthin epoxidase ( ZEP ), thereby inhibiting the accumulation of lycopene and β-citraurin. Studies have shown that the degreening of the ‘Shatangju’ pericarp was accelerated, but the accumulation of carotenoid biosynthesis genes was inhibited, which was not conducive to the synthesis of carotenoids.

Introduction Cellular senescence drives aging and age-related diseases, posing a critical barrier to stem cell-based regeneration. While selective elimination of senescent cells (senolysis) remains a theoretically promising strategy, safe natural senotherapeutic agents are limited. Human exfoliated deciduous tooth stem cells (SHED) possess high regenerative potential yet are vulnerable to senescence, compromising their therapeutic utility. Methods We performed a phenotypic screen of 800 natural compounds in replicatively senescent SHED. The hit compound, the phytochemical melissic acid, was then validated in two independent senescence models—replicative exhaustion and doxorubicin-induced stress. Melissic acid (5–20 μM) was assessed for cytotoxicity and anti-senescence efficacy via SA-β-Gal staining and senescence marker expression. Transcriptomic profiling and mechanistic assays were performed to dissect the underlying molecular pathways. Results Melissic acid exhibited minimal cytotoxicity but significantly reduced SA-β-Gal positivity and senescence markers in both models. Transcriptomic profiling revealed marked upregulation of ADCY5 and coordinated changes in cAMP signaling, autophagy, and lipid metabolism genes. Mechanistically, melissic acid increased CREB Ser133 phosphorylation, enhanced Beclin-1 expression, and reduced p62/SQSTM1, indicating activation of cAMP/CREB-dependent autophagy. It also decreased lipid peroxidation, restored mitochondrial membrane potential, and relieved G0/G1 arrest via upregulation of Cyclin D1 and Rb Ser780 phosphorylation. Discussion Collectively, melissic acid alleviates SHED senescence through the ADCY5/cAMP/CREB-autophagy axis, promoting autophagic clearance, mitochondrial recovery, and cell-cycle re-entry—thereby ameliorating senescence-associated phenotypes in senescent stem cells. This study identifies melissic acid as a natural senomorphic candidate for stem cell function restoration and provides a mechanistic link between cAMP-dependent autophagy and amelioration of stem cell aging, offering a potential strategy to overcome stem cell exhaustion and regenerative limits imposed by cellular senescence.

Stem cell-derived exosomes from human exfoliated deciduous Teeth (SHED-Exos) show promising treatment effects through immune modulation and angiogenic properties. However, when applied to diabetic wound healing, their inactivation and rapid clearance limit their clinical application. In this study, we aimed to develop a chitosan/dextran-based hydrogel using a biotin-avidin system that showed excellent injectability, adhesiveness, and antibacterial effects and enabled the slow and steady release of SHED-Exos to enhance diabetic wound healing. SHED-Exos were immobilized within a hydrogel through the biotin-avidin interaction and electrostatic forces of chitosan, allowing for slow release over >7 days. Additionally, the hydrogel exhibited excellent cell compatibility and supported cell spheroid growth in a 3D network. In vivo , the hydrogel significantly enhanced wound healing in diabetic rats by reducing inflammation and decreasing the ratio of Th17/Treg cells around wounds. This innovative noncellular hydrogel shows promise in facilitating the healing of diabetic wounds and may serve as a potential clinical therapy using natural bioactive materials delivery of exosomes.

The primary pathology of periodontitis involves the gradual deterioration of periodontal tissues resulting from the inflammatory reaction triggered by bacterial infection. In this study, a novel drug for periodontal pocket injection, known as the Shed-Cu-HA hydrogel, was developed by incorporating copper ions (Cu2+) and Shed-derived exosomes (Shed-exo) inside the hyaluronic acid (HA) hydrogel. Suitable concentrations of Cu2+ and Shed-exo released from Shed-Cu-HA enhanced cell viability and cell proliferation of human periodontal ligament stem cells. Additionally, the Shed-Cu-HA demonstrated remarkable antibacterial effects against the key periodontal pathogen (Aa) owing to the synergistic effect of Cu2+ and HA. Furthermore, the material effectively suppressed macrophage inflammatory response via the IL-6/JAK2/STAT3 pathway. Moreover, the Shed-Cu-HA, combining the inflammation-regulating properties of HA with the synergistic osteogenic activity of Shed-exo and Cu2+, effectively upregulated the expression of genes and proteins associated with osteogenic differentiation. The experimental findings from a mouse periodontitis model demonstrated that the administration of Shed-Cu-HA effectively reduced the extent of inflammatory cell infiltration and bacterial infections in gingival tissues and facilitated the regeneration of periodontal bone tissues and collagen after 2 and 4 weeks of injection. Consequently, it holds significant prospects for future applications in periodontitis treatment.

Introduction: Cleidocranial dysplasia (CCD) is an autosomal-dominant, heritable skeletal and dental disease, involving hypoplastic clavicles, defective ossification of the anterior fontanelle, dentin and enamel hypoplasia, and supernumerary teeth, which can seriously affect the oral and mental health of patients. Amyloid-like protein aggregation, which is established by lysozyme conjugated with polyethylene glycol (Lyso-PEG), forms a mineralized nanofilm layer on a healthy enamel surface. However, whether it can form a remineralization layer in dental tissues from CCD remains unclear. Methods: This study evaluated deciduous teeth from healthy individuals and a patient with CCD. Because pulp and dentin are functionally closely related, stem cells from human exfoliated deciduous teeth (SHED) from CCD patients and healthy individuals were collected to compare their biological properties. Results: The results found that deciduous teeth from patients with CCD exhibited dentin hypoplasia. In addition, the proliferative ability and osteogenic potential of SHED from patients with CCD were lower than those of control individuals. Finally, Lyso-PEG was applied to dentin from the CCD and control groups, showing a similar remineralization-induced effect on the dentin surfaces of the two groups. Conclusion: These results extend our understanding of the dentin and SHED of patients with CCD, exhibiting good caries-preventive capacity and good biocompatibility of Lyso-PEG, thus providing a novel dental therapy for CCD and patients with tooth hypoplasia.

Preserving the vital pulp poses significant challenges in clinical pulp injury repair. Tricalcium silicate (C3S)-based cement has emerged as a successful pulp capping biomaterial. However, its efficacy in inducing pulp-dentin complex regeneration requires enhancement, and handling difficulties, poor anti-collapsible ability and inadequate self-setting performance remain significant obstacles, restricting its broader application in endodontics. In this study, a multifunctional SrCO 3 -doped C3S biocement (C3S-Sr/F127-nHA) reinforced by Pluronic® F127 thermoreversible hydrogel and nano hydroxyapatite (nHA) was developed to overcome the limitations of C3S-based cements. The thermoresponsive characteristic of F127 conferred the C3S pastes smart response behavior, allowing for optimal operability at room temperature and maintaining excellent anti-collapse properties at body temperature. C3S-Sr/F127-nHA exhibited an injectability rate of up to 93 %. nHA facilitated the hydration kinetics of C3S, resulting in reduced setting time and enhanced compressive strength. Furthermore, C3S-Sr/F127-nHA demonstrated excellent apatite mineralization ability, cytocompatibility, and potential for osteo/odontogenic induction. The inclusion of Sr 2+ ions and nHA promoted osteo/odontogenic differentiation of stem cells isolated from human exfoliated deciduous teeth (SHED). In vivo findings indicated that C3S-Sr/F127-nHA could stimulate the formation of a homogeneous dentin bridge. Consequently, C3S-Sr/F127-nHA is a promising biocement for use in regenerative endodontics.
