An OPRM1-SNAP-tag/CMC method to directly identify drug components in sewage

Li Chenjia,Liao Qi,Wang Rui,Zhang Xinping,Ma Mengyang,Liu Yonghong,Xiao Lei,Jiao Ying,Wang Nan

The scourge of drug addiction and abuse poses a significant challenge to society. Opioid drugs acting on μ-opioid receptor (OPRM1) make it one of the pivotal targets for drug addiction. In the past decade, sewage analysis has become a prevalent method of drug monitoring. However, traditional methods of detecting drugs in sewage are cumbersome, and rapid detection methods are relatively lacking. To address this, an innovative OPRM1-SNAP-tag/CMC method to directly identify drug components in sewage was established. Cell membrane chromatography (CMC) is an affinity chromatography technique which effectively detects receptor affinity substances. Cells constructed with high expression of specific receptor could be used to screen for compounds acting on the receptor. CMC based on OPRM1 provides a potentially convenient and effective tool for the detection of targeted drug components in sewage. In this study, the selectivity, reproducibility, column lifetime, and carryover of the CMC column had been assessed. Initially, we eluted the collected domestic sewage with methanol and acetonitrile, and the retention peaks were observed on the CMC system. Subsequently, without any preliminary sample preparation, we directly injected filtered samples of suspicious sewage into the OPRM1-SNAP-tag/CMC system, where we observed retention peaks as well. The retained components were further identified as morphine by using UPLC-MS/MS. In conclusion, the OPRM1-SNAP-tag/CMC method stands out as a reliable and robust model for the detection of drug components in sewage. It provides a valuable analytical tool for frontline drug control efforts, enhancing our capacity to monitor and mitigate the impact of drug abuse on society. Graphical Abstract

An assembly-regulated SNAP-tag fluorogenic probe for long-term super-resolution imaging of mitochondrial dynamics

Wenjuan Liu,Qinglong Qiao,Jiazhu Zheng,Jie Chen,Wei Zhou,Ning Xu,Jin Li,Lu Miao,Zhaochao Xu

Super-resolution fluorescence microscopy has emerged as a powerful tool for studying mitochondrial dynamics in living cells. However, the lack of photostable and chemstable probe makes long-term super-resolution imaging of mitochondria still a challenging work. Herein, we reported a 4-azetidinyl-naphthliamide derived SNAP-tag probe AN-BG exhibiting excellent fluorogenicity and photostability for long-term super-resolution imaging of mitochondrial dynamics. The azetidinyl group and naphthalimide fluorophore are in a flat conformation which can effectively suppress twisted intramolecular charge transfer and then effectively improve the brightness and photostability. This planarized molecular structure is conducive to the formation of fluorescence-quenched J -aggregates, and the protein labeling process will depolymerize the probes and restore fluorescence. Fluorescent labeling mitochondrial inner membrane proteins via SNAP tags overcomes the shortcomings that variations in mitochondrial inner membrane potential will release probes attached to mitochondria by electrostatic interactions . Therefore, AN-BG realized the stable labeling of mitochondria and the long-term imaging of mitochondrial dynamics under super-resolution microscopy.

Magnetic nanoparticles covalently immobilizing epidermal growth factor receptor by SNAP-Tag protein as a platform for drug discovery

Huaxin Zhou,Jia Fu,Qianqian Jia,Saisai Wang,Peida Liang,Yamin Wang,Yanni Lv,Shengli Han

Magnetic nanoparticles (NPs) cloaked with cell membranes expressing high levels of the epidermal growth factor receptor (EGFR) have been used to screen for EGFR-targeting active compounds in traditional Chinese medicine (TCM) formulations. However, previous strategies involved physical immobilization of the biomaterials on the surface of the nanocarrier , resulting in highly unstable platforms since the biological materials could dislodge easily. Chemical bonding of biomaterials to the nanoparticles surface can improve the stability of the biomimetic platforms. In this study, membrane fragments from cells expressing SNAP-Tag-EGFR (ST-EGFR) were immobilized on the surface of magnetic NPs. The ST-EGFR magnetic cell membrane nanoparticles (ST-EGFR/MCMNs) showed greater stability, and higher binding capacity, selectivity adsorption of gefitinib after 7 days compared to the un-immobilized magnetic cell membrane nanoparticles (EGFR/MCMNs). The ST-EGFR/MCMNs were used to screen for the EGFR-targeting active compounds of Zanthoxyli Radix (ZR), and identified toddalolactone and nitidine chloride. The latter significantly inhibited the proliferation of EGFR-overexpressing cancer cells, and was more effective compared to gefitinib. This innovative technology can be used to rapidly screen for active compounds from complex extracts, and aid in drug discovery.

Development and Validation of an ADRβ1-SNAP-tag/CMC Model for Screening ADRβ1 Antagonists: Application to Nebivolol and Its Byproducts

Liju Yu,Jie Gao,Tingting Deng,Chang Gao,Jiaxing Wang,Nan Wang

Heart failure is a global epidemic characterized by high mortality and morbidity rates. β1-Adrenoceptors (ADRβ1) antagonists are the main drugs used in the treatment of HF. A simple and convenient method for screening ADRβ1 antagonists is necessary. Nebivolol has four chiral centers, and its byproducts, which are not easy to avoid, may interact with ADRβ1. Cell membrane chromatography based on SNAP-tag technology provides a comprehensive solution. In this study, an ADRβ1 target cell membrane chromatography model based on SNAP-tag technology was established, and the selectivity, specificity, repeatability, and column life of the system were validated. Under optimized conditions, this model was used to screen for active components targeting ADRβ1 in nebivolol and its byproducts. The bioactivity of nebivolol and its byproducts was detected by calcium influx. The results showed that the ADRβ1 SNAP-tag/CMC model was successfully established and could be used to screen active drugs targeting ADRβ1. Among nebivolol and its byproducts, D-nebivolol (SRRR-enantiomer) had the best affinity for ADRβ1. Three byproducts of nebivolol also exhibited retention characteristics on ADRβ1. In vitro calcium influx experiments verified the interaction of these components with ADRβ1, showing good inhibitory effects on ADRβ1. In conclusion, the ADRβ1 SNAP-tag/CMC model developed in this study can be used for the comprehensive screening of active components targeting ADRβ1 in complex samples.

Platelet membrane coated Cu9S8-SNAP for NIR-II photoacoustic imaging guided NO repairing acute kidney injury

Xiaojuan Hu,Danping Wu,Shijie Yao,Gongning Chen,Ping Li,Yang Chen,Wancheng Gong,Weijiao Fan,Xiaoyi Chen,Xiaozhou Mou,Yu Cai,Xianghong Yang

Acute kidney injury (AKI) is a prevalent critical condition in hospitalized patients with an elevated risk of death. Challenges in AKI management persist due to the absence of efficient early diagnostic modalities and the lack of effective therapeutic interventions, imposing significant burdens on medical resources for prolonged treatment. Here, we proposed a second near-infrared (NIR-II) photoacoustic imaging (PAI) guided triggered on-demand release of (nitric oxide) NO from the injury zone to monitor and treat AKI through the anti-inflammatory, anti-apoptotic, and oxidative stress-damage-reducing cell- or organ-protective effects of NO. We engineered hollow copper sulfide nanoparticles (Cu 9 S 8 NPs) loaded with platelet membrane-encapsulated NO-releasing precursors (S-Nitroso-N-acetylpenicillamine, SNAP), as a nanoplatform for early AKI diagnosis and precise spatiotemporal NO release Besides acting as a drug carrier, the inherent PAI compatible Cu 9 S 8 also enables targeted renal injury localization, exploiting the natural propensity of platelets to accumulate at vascular injury sites, facilitating real-time AKI monitoring in murine models. Experiments demonstrated that nitric oxide delivery nanoparticles can specifically target and accumulate in the kidney, reduce inflammation, and promote damage repair. Overall, this gas therapy strategy represents a new therapeutic pathway for the clinical AKI management.

Chitosan/PVA nanofibrous membrane loaded with cerium manganese nanoparticles/SNAP promotes wound healing in diabetes

Zhonghao Jiang,Zhuo Yan,Shumin Yan,Zhihao Chen,Lianhai Li,Guoliang Huang,Li Li,Jiantao Lin

Diabetic wounds are one of the most problematic complications among diabetic patients, often progressing to chronic, persistent ulcers that are a leading cause of amputation and disability. The constant accumulation of reactive oxygen species (ROS) at diabetic wound sites, along with bacterial infection and an excess of pro-inflammatory M1-type macrophages, are key factors contributing to the difficulty of wound healing. Nanofibrous membranes can be used directly in diabetic wounds with the dual function of adsorbing tissue exudate and releasing drugs. Cerium oxide nanoparticles can be used in nanotherapy to reduce the accumulation of chronic inflammatory substances due to their excellent antioxidant properties. The antioxidant properties can be further improved by doping with other metal elements. In this paper, we constructed a nanofibrous membrane based on cerium manganese nanoparticles (CeMn NPs) for the treatment of diabetic wounds. The nanofibrous membrane was loaded with cerium manganese nanoparticles and nitric oxide donor SNAP, named PVA-CS-CeMn NPs-SNAP nanofibrous membrane. The PVA-CS-CeMn NPs-SNAP nanofibrous membrane was able to cover the wounds of diabetic mice with slow release of CeMn NPs and SNAP. The nanoparticles could scavenge ROS accumulated at the wound site and regulate the macrophage phenotype at the wound site, increasing the proportion of anti-inflammatory M2-type macrophages. In addition, SNAP had antimicrobial and vascular regenerative effects at the wound site. Together, these synergistic effects resulted in faster wound healing and reduced tissue inflammation in diabetic mice, and no organ toxicity was observed. Overall, our results indicate that PVA-CS-CeMn NPs-SNAP nanofibrous membranes have significant potential for the treatment of diabetic wounds and provide a strong scientific basis for the development of novel diabetic wound treatment materials.

Platelet Membrane Coated Cu9S8-SNAP for Targeting NIR-II Mild Photothermal Enhanced Chemodynamic/Gas Therapy of Triple-Negative Breast Cancer

Danping Wu,Xiaoyi Chen,Shijie Yao,Yichen He,Gongning Chen,Xiaojuan Hu,Yang Chen,Zhenye Lv,Jing Yu,Ketao Jin,Yu Cai,Xiaozhou Mou

Triple-negative breast cancer (TNBC) is a highly aggressive and uncommon subtype of breast cancer with a poor prognosis. It is crucial to prioritise the creation of a nanotherapeutic method that is highly selective and actively targeting TNBC. This study explores a new nanosystem, Cu 9 S 8 -SNAP@PM (C-S@P), composed of Cu 9 S 8 -SNAP coated with a platelet membrane (PM). The purpose of this nanosystem is to cure TNBC using multimodal therapy. The utilisation of PM-coated nanoparticles (NPs) enables active targeting, leading to the efficient accumulation of C-S@P within the tumour. The Cu 9 S 8 component within these NPs serves the potential to exert photothermal therapy (PTT) and chemodynamic therapy (CDT). Simultaneously, the S-Nitroso-N-Acetylvanicillamine (SNAP) component enables nitric oxide (NO) gas therapy (GT). Furthermore, when exposed to NIR-II laser light, Cu 9 S 8 not only increases the temperature of the tumour area for PTT, but also boosts CDT and stimulates the release of NO through thermal reactions to improve the effectiveness of GT. Both in vitro and in vivo experimental results validate that C-S@P exhibits minimal side effects and represents a multifunctional nano-drug targeted at tumors for efficient treatment. This approach promises significant potential for TNBC therapy and broader applications in oncology.

KIT-SNAP-tag/cell membrane chromatography model coupled with liquid chromatography–mass spectrometry for anti-GIST compound screening from Evodia rutaecarpa

Feng Jingting,Jiang Yuhan,Liu Sihan,Deng Linge,Lv Yanni,Chen Nanzheng,Han Shengli

Gastrointestinal mesenchymal tumors, as the most common mesenchymal tumors in the gastrointestinal tract, are adjuvantly treated with multi-targeted tyrosine kinase inhibitors, such as imatinib and sunitinib, but there are problems of drug resistance and complex methods of monitoring therapeutic agents. The pathogenesis of this disease is related to mutations in tyrosine kinase (KIT) or platelet-derived growth factor receptor α, an important target for drug therapy. In recent years, the screening of relevant tyrosine kinase inhibitors from traditional Chinese medicine has become a hotspot in antitumor drug research. In the current study, the KIT-SNAP-tag cell membrane chromatography (KIT-SNAP-tag/CMC) column was prepared with satisfying specificity, selectivity, and reproducibility by chemically bonding high KIT expression cell membranes to the silica gel surface using the SNAP-tag technology. The KIT-SNAP-tag/CMC-HPLC-MS two-dimensional coupling system was investigated using the positive drug imatinib, and the results showed that the system was a reliable model for screening potential antitumor compounds from complex systems. This system screened and identified three potential active compounds of evodiamine (EVO), rutaecarpin (RUT), and dehydroevodiamine (DEVO), which possibly target the KIT receptor, from the alcoholic extract of the traditional Chinese medicine Evodia rutaecarpa . Then, the K D values of the interaction of EVO, RUT, and DEVO with KIT receptors measured using nonlinear chromatography were 7.75 (±4.93) × 10 −6 , 1.42 (±0.71) × 10 −6 , and 2.34 (±1.86) × 10 −6 mol/L, respectively. In addition, the methyl thiazolyl tetrazolium assay validated the active effects of EVO and RUT in inhibiting the proliferation of high KIT-expressing cells in the ranges of 0.1–10 µmol/L and 0.1–50 µmol/L, respectively. In conclusion, the KIT-SNAP-tag/CMC could be a reliable model for screening antitumor components from complex systems. Graphical Abstract

Application of SNAP-tag-EGFR cell membrane chromatography model in screening antitumor active components of Silybum marianum (L.) Gaertn

Xin Zhang,Yuhan Jiang,Na Guo,Yifan Ding,Jingting Feng,Chenyang Miao,Yanni Lv

The SNAP-tag-epidermal growth factor receptor (SNAP-tag-EGFR) cell membrane chromatography (CMC) model is a powerful tool for investigating ligand-receptor interactions and screening active ingredients in traditional Chinese medicine. Most tyrosine kinase inhibitors (TKIs) target epidermal growth factor receptors. However, TKIs associated with significant side effects and drug resistance must be addressed immediately. Therefore, there is an urgent need to develop new TKIs with high efficiency and low toxicity. Because of its low toxicity and side effects, traditional Chinese medicine has been widely employed to treat various diseases, including cancer. Hence, this study aimed to use the SNAP-tag-EGFR/CMC-high-performance liquid chromatography-mass spectrometry (HPLC-MS) two-dimensional system model as the research tool to screen and identify potential EGFR antagonists from the Chinese medicine Silybum marianum (L.) Gaertn . The applicability of the system was verified using the positive control drug osimertinib. Four potential EGFR antagonists were screened from the Chinese medicine Silybum marianum (L.) Gaertn. . They were identified as silydianin, silychristin, silybin, and isosilybin. Additionally, their pharmacological activity was preliminarily verified using a CCK-8 assay. The kinetic parameters of the four active ingredients interacting with EGFR and their binding modes with EGFR were analyzed using nonlinear chromatography (NLC) and molecular docking. This study identified silydianin, silychristin, silybin, and isosilybin from Silybum marianum (L.) Gaertn. and verified their potential antitumor effects on EGFR.

Snap-Buckling Motivated Controllable Jumping of Thermo-Responsive Hydrogel Bilayers

Guorong Gao,Zhenwu Wang,Dan Xu,Liufang Wang,Ting Xu,Hua Zhang,Jing Chen,Jun Fu

Responsive hydrogel actuators have promising applications in diverse fields. Most hydrogel actuators are limited by slow actuation or shape transformations. This work reports on snap-buckling motivated jumping of thermoresponsive hydrogel bilayers. The bilayers are composed of poly(NIPAM-co-DMAPMA)/clay hydrogel with different lower critical solution temperatures in each layer, and thus undergo slow reversible curling/uncurling at temperature changes. The gels are adhesive to numerous materials including aluminum. The adhesion between the gels and an aluminum ratchet is utilized to constrain the thermoresponsive deformation of the bilayers to store elastic energy. When the accumulated elastic energy overwhelms the gel–aluminum adhesion, snap-buckling takes place to abruptly release the accumulated energy, which motivates the bilayer to jump. The jumping direction, start time, height, and distance are controlled by the geometry of the bilayers or the ratchet. This work paves a novel way for the rapid actuation of responsive hydrogels in a controlled manner and may stimulate the development of novel hydrogel devices.

Temperature-triggered inflatable hydrogel muscles with snap-through instability for untethered robots

Cui Yande,Hu Jianhua,Dong Ziyang,Li Bing,Chang Chunyu

Pneumatic artificial muscles have been widely used in the field of robotics because of their large output force and fast actuation, however, the accompanying bulky compressors and pumps limit their miniaturized applications. Despite current untethered pneumatic artificial muscles can be driven by adjusting the internal pressure, it is challenging to structurally mimic natural muscles with high water content. Here, we propose untethered pneumatic artificial muscles comprising a hydrogel actuator with snap-through instability and an air storage chamber. These hydrogel actuators can realize the conversion from hydrophobic association of octyl acrylate moieties to host-guest interaction between β -cyclodextrin and octyl acrylate under thermal stimuli, leading to the decrease of their moduli. The inflated hydrogel actuators exhibit rapid actuation with a radial expansion speed of 200% s −1 , which are powered by snap-through instability, thermal expansion of the gas inside the hydrogel actuator, and evaporation of water on its internal surface. With the pneumatic artificial muscles miniaturized, we demonstrate diving and rolling robots, exemplifying bionic robots able to adapt to and modify the environment. We expect that the design of hydrogel actuator in miniaturized pneumatic artificial muscles will facilitate rapid locomotion for future bionic robotic platforms.

Investigation on droplet dynamic snap-off process in a short, abrupt constriction

Zhang Li,Zhaolin Gu,Ran Li,Chen Wang,Chungang Chen,Chunlei Yu,Yigen Zhang,Qinglin Shu,Junwei Su

Droplet snap off is one of the most usual phenomena in oil recovery during water injection in water-wet porous media. In this work, the droplet dynamic snap-off process in a short, abrupt constriction is investigated. Three different phenomena, i.e. total breakup, partial breakup and non-breakup, of the nonwetting phase droplets are observed, the flow characteristics of droplets in each phenomenon and relevant physical mechanisms are analyzed. We find that both the initial size and velocity of the droplet greatly influence the breakup process. The viscosity of droplet also affects the breakup state. The critical conditions of each phenomenon are obtained based on dimensionless droplet size and capillary number, which could be used to predict the droplet breakup state. Under a certain capillary number, daughter droplet distribution is influenced by initial droplet size.
