Multi-modal porous scaffolds can promote bone regeneration. In the present study, they were successfully fabricated from poly (lactic-co-glycolic acid) (PLGA)/superparamagnetic iron oxide nanoparticles (SPIONs) by supercritical CO 2 foaming, where SPIONs was employed as heterogeneous nucleation agent. The effects of SPION size on pore nucleation in PLGA/SPIONs nanocomposites were investigated. It was found that the addition of smaller SPIONs size caused the reduction of pore size. Multi-modal porous scaffolds could be available by controlling the SPIONs content, soaking temperature, pressure, and depressurization rate. The porosity of the PLGA/SPIONs scaffolds ranged from (58.68 ± 0.62)% to (94.97 ± 0.14)% could be obtained via adjusting the supercritical CO 2 foaming conditions. Moreover, it was found that the compressive modulus and porosity of the scaffolds were strongly associated with the porous structure. This study provided a new green preparation method for magnetic scaffolds to realize the controllable adjustment of the multi-mode porous scaffolds.

In this research, reducible polydopamine coated magnetic nanoparticles (SPIONs@PDA) were prepared and doxorubicin (DOX) was encapsulated for both magnetic resonance imaging (MRI) detection and cell targeting drug delivery. The SPIONs@PDA exhibited excellent water-solubility and colloidal stability, because the coating of the magnetic nanoparticle was a crosslinked reducible polydopamine synthesized with N,N-bis(acrylate) cystamine (BACy) as a crosslinker, dopamine methacrylamide (DMA) as an anchor to immobilize SPIONs, and polyethylene glycol (PEG) to provide colloidal stability. The SPIONs@PDA showed reduction-sensitivity due to the reducible disulfide bond in the polymer network. Subsequently, a sustained and high cumulative drug release in glutathione (GSH) was observed, rather than the low drug release in PBS (pH 7.4). Fluorescent imaging studies by confocal laser scanning microscopy indicated that the prepared SPIONs@PDA could specifically target tumor cells. The results of the cytotoxicity assays further demonstrated that the DOX-loaded SPIONs@PDA presented efficient antitumor activity to HeLa cells. These favorable properties suggested SPIONs@PDA as promising targeted magnetic nanoparticles for MRI detection and stimuli-triggered drug delivery in cancer theranostics.

This study investigates the molecular mechanisms by which superparamagnetic iron oxide nanoparticles (SPIONs) loaded with the WSGC peptide (WSGC@FA@PEG/PEI-SPIONs)—a 40-amino acid polypeptide derived from apoC-III—modulate chemotherapy resistance in gastric adenocarcinoma (GA). Emphasis is placed on their role in regulating mitophagy and mitochondrial homeostasis via the Notch signaling pathway. The physicochemical properties of WSGC@FA@PEG/PEI-SPIONs are thoroughly characterized, demonstrating favorable biocompatibility, stable size distribution, and efficient peptide loading. In vitro experiments show that these nanoparticles significantly inhibit GA cell proliferation, migration, and invasion by downregulating mitophagy-associated proteins (LC3, PINK1, and Parkin), primarily through modulation of the Notch pathway. In vivo studies, using a GA nude mouse model, confirm the therapeutic potential of WSGC@FA@PEG/PEI-SPIONs, revealing marked tumor growth inhibition and increased apoptotic activity. Collectively, the findings highlight the WSGC peptide as a promising therapeutic agent for overcoming chemotherapy resistance in GA by targeting the Notch signaling pathway and suppressing mitophagy, thereby presenting a novel strategy for polypeptide-based cancer therapy.

In this study, magnetic nanocarriers (DU@Fe, D avg = 281.6 nm, zeta potential −28.0 mV) were fabricated using dextran, urushiol as the shell and superparamagnetic iron oxide nanoparticles (SPIONs) as the core. Subsequently, the specific ligand Lys-D-Ala-D-Ala of vancomycin (Van) was grafted onto the surface of DU@Fe, which generated nanoparticles (Lys-D-Ala-D-Ala@DU@Fe) with an average particle size of 385.5 nm and a zeta potential of −16.8 mV via specific and robust interaction. Ultimately, the immobilization capacity of Van reached up to 294.1 mg·g −1 for efficient antibacterial properties. Moreover, the assembly process adhered to the pseudo-second-order kinetics model ( R 2 = 0.998–0.999) and the Langmuir adsorption isotherm model ( R 2 = 0.999, 30 °C). Notably, V@DU@Fe effectively adhered to the cell envelopes of both Gram-negative and Gram-positive bacteria, achieving rapid bactericidal effects within 1 h. Furthermore, it maintained over 85.0 % of its initial antibacterial efficiency against S. aureus and S. epidermidis even after six recycles. Therefore, this study provides strategies and methods for the development of urushiol-dextran intelligent SPIONs nanomedicines.

The impact of the surface modification on the subcellular distribution of nanoparticles in the brain remains elusive. The nanoparticles prepared by conjugating polyethylene glycol and maleic anhydride-coated superparamagnetic iron oxide nanoparticles (Mal-SPIONs) with bovine serum albumin (BSA/Mal-SPIONs) and with Arg–Gly–Asp peptide (RGD/Mal-SPIONs) were injected into the rat substantia nigra. Observation of transmission electron microscopy (TEM) samples obtained 24 h after perfusion showed that abundant RGD/Mal-SPIONs accumulated in the myelin sheath, dendrites, axon terminals and mitochondria, and on cell membranes in the brain tissue near the injection site. For rats injected with BSA/Mal-SPIONs, a few nanoparticles accumulated in the myelin sheath, axon terminals, endoplasmic reticulum, mitochondria, Golgi, and lysosomes of neurons and glial cells while least SPIONs in rats injected with Mal-SPIONs were found. TEM pictures showed some Mal-SPIONs were expelled out of the brain. RGD/Mal-SPIONs diffused extensively to the thalamus, frontal cortex, temporal lobe, olfactory bulb, and brain stem after injection. Only a few BSA/Mal-SPIONs diffused to the afore-mentioned brain areas. This work reveals different surface modifications on the iron oxide nanoparticles play crucial roles in their distribution and diffusion in the rat brains.

The proliferation of antibiotic-resistant bacteria (ARB) and pathogens in biomedical wastewater poses a critical global health threat, underscoring the need for antibiotic-free disinfection strategies. Herein, we develop recyclable triethylene glycol-modified superparamagnetic iron oxide nanoparticles (TEG-SPIONs) as an efficient photothermal antibacterial platform. Under near-infrared (NIR) irradiation, TEG-SPIONs exhibit outstanding photothermal performance with a conversion efficiency of 30.44%, enabling the rapid inactivation of bacteria through synergistic mechanisms, including membrane disruption, ROS generation, and nucleic acid leakage. Moreover, TEG-SPIONs effectively inhibit biofilm formation and disrupt mature biofilms. Their superparamagnetism allows facile magnetic retrieval and reuse, maintaining over 60% antibacterial activity after 20 cycles, along with excellent colloidal stability for 14 days. Unlike conventional antibiotics, TEG-SPIONs do not cause the development of resistance or secondary pollution. This work highlights the promise of TEG-SPIONs as a sustainable, recyclable nanoplatform for environmental remediation and water purification, offering a practical strategy against antimicrobial resistance.

Recent research has shown that etoposide (ETP), an immunomodulatory medication, can have cytotoxic effects. When accompanied by nano-drug delivery, this possibility can lead to a more effective treatment for cancer. This study involved the delivery of ETP via PDA@SPIONs (termed as ETP-PDA@SPIONs), which are superparamagnetic iron oxide nanoparticles (SPIONs) that have been coated with polydopamine (PDA) and utilized to treat C6 and U87 glioma cancer cell lines. The particle diameter, polydispersive index (PDI), zeta potential , and shape of nanoparticles were determined. The fabricated nanoparticles had a mean particle size of 164 ± 7 nm and a − 37 ± 3 mv zeta potential. Cell lines were tested for sensitivity to PDA@SPIONs, free-ETP, and ETP-PDA@SPIONs. On the cell lines, we tested the proliferation of PDA@SPIONs, free-ETP, and PDA@SPIONs loaded with ETP. The cytotoxicity of ETP-PDA@SPIONs was substantially higher than that of free-ETP-treated cells. The cells' morphological changes were examined by dual staining (acridine orange/ethidium bromide, AO/EB) and nuclear staining methods. Interestingly, this inhibitory impact was not ascribed to the known cellular membrane destruction, cell membrane damage, and reactive oxygen species (ROS). Further tests demonstrated that the nanoparticles substantially impacted mitochondrial functioning, resulting in an aberrant mitochondrial shape, and reduced mitochondrial membrane potential (MMP). Future studies into glioma cancer therapy may want to investigate using the proposed ETP-loaded nanoparticles.

Superparamagnetic iron oxide nanoparticles (SPIONs) have been presented to regulate the migration and osteogenic differentiation of bone mesenchymal stem cells (BMSCs) under magnetic field (MF). However, the toxicity and short residence for the massively exposed SPIONs at bone defects compromises their practical application. Herein, SPIONs were encapsulated into PLGA microspheres to overcome these shortcomings. Three types of PLGA microspheres (PFe-I, PFe-II and PFe-III) were prepared by adjusting the feeding amount of SPIONs, in which the practical SPIONs loading amounts was 1.83%, 1.38% and 1.16%, respectively. The average diameter of the fabricated microspheres ranged from 160 μm to 200 μm, having the porous and rough surfaces displayed by SEM. Moreover, they displayed the magnetic property with a saturation magnetization of 0.16 emu/g. In vitro cell studies showed that most of BMSCs were adhered on the surface of PFe-II microspheres after 2 days of co-culture. Moreover, the osteoblasts differentiation of BMSCs was significantly promoted by PFe-II microspheres after 2 weeks of co-culture, as shown by detecting osteogenesis-related proteins expressions of ALP, COL I, OPN and OCN. Afterward, PFe-II microspheres were surgically implanted into the defect zone of rat femoral bone, followed by exposure to an external MF, to evaluate their bone repairing effect in vivo . At 6th week after treatment with PFe-II + MF, the bone mineral density (BMD, 263.97 ± 25.99 mg/cm 3 ), trabecular thickness (TB.TH, 0.58 ± 0.08 mm), and bone tissue volume/total tissue volume (BV/TV, 78.28 ± 5.01%) at the defect zone were markedly higher than that of the PFe-II microspheres alone (BMD, 194.34 ± 26.71 mg/cm 3 ; TB.TH, 0.41 ± 0.07 mm; BV/TV, 50.49 ± 6.41%). Moreover, the higher expressions of ALP, COL I, OPN and OCN in PFe-II + MF group were displayed in the repairing bone. Collectively, magnetic PLGA microspheres together with MF may be a promising strategy for repairing bone defects.

Nanoparticles have great potential in remote neural stimulation with physical fields, but translation of these approaches to the medical arena faces enormous challenges, especially due to the use of highly invasive brain delivery of nanoparticles. Here we explore an alternative strategy through employing non-invasive brain delivery of nanoparticles. Non-invasive brain delivery of superparamagnetic iron oxide nanoparticles (SPIONs) in rats is performed by combining several complementary methods. The thus-delivered nanoparticles (called Tat-SPIONs) are found to enhance the neural stimulation effects of transcranial magnetic stimulation (TMS), a clinically-used non-invasive brain stimulation device, as analyzed by electrophysiological, biochemical, and behavioral experiments. In particular, ischemic stroke rats treated by non-invasively delivered Tat-SPIONs coupled with TMS show significant improvement in stroke recovery in terms of general health status, motor-sensory function, and cognitive function, compared to those treated by TMS alone. Importantly, mechanistic studies by experiments and numerical simulation suggest that the neural stimulation enhancement is mediated by neuroplasticity at least in part, is caused by a magnetoelectric rather than magnetothermal effect, and is generated primarily by those Tat-SPIONs associated with neurons’ plasma membranes . This further implies that the excellent membrane-binding ability of Tat peptide is crucial for SPION-enhanced TMS. Tat-SPIONs exhibit minimal toxicity and neuroinflammation, and are clearable from major organs. The presented technology platform paves the way for medical applications of nanoparticles in remote brain stimulation with physical fields for stroke as well as other neurological disorders.

To develop an efficient probe for targeted magnetic resonance (MR) imaging of liver carcinoma, the surface modification of superparamagnetic iron oxide nanoparticles (SPIONs) was carried out by conjugating a naturally-occurring glycosaminoglycan with specific biological recognition to human hepatocellular liver carcinoma (HepG2) cells. These modified SPIOs have good water dispersibility, superparamagnetic property, cytocompatibility and high magnetic relaxivity for MR imaging. When incubated with HepG2 cells, they demonstrated significant cellular uptake and specific accumulation, as confirmed by Prussian blue staining and confocal microscopy. The in vitro MR imaging of HepG2 cells and in vivo MR imaging of HepG2 tumors confirmed their effectiveness for targeted MR imaging of liver carcinoma.

Transcranial magnetic stimulation (TMS) is a non-invasive and clinically approved method for treating neurological disorders. However, the relatively weak intracranial electric current induced by TMS is an obvious inferiority which can only produce limited treatment effects in clinical application. The present study aimed to investigate the possibility of enhancing the effects of TMS with intravenously administrated magnetic nanoparticles. To facilitate crossing of the blood–brain barrier (BBB), the superparamagnetic iron oxide nanoparticles (SPIONs) were coated with carboxylated chitosan and poly(ethylene glycol). To aid the nanoparticles in crossing the BBB and targeting the predesigned brain regions, an external permanent magnet was attached to the foreheads of the rats before the intravenous administration of SPIONs. The electrophysiological tests showed that the maximum MEP amplitude recorded in an individual rat was significantly higher in the SPIONs + magnet group than in the saline group (5.78 ± 2.54 vs. 1.80 ± 1.55 mV, P = 0.015). In the M1 region, biochemical tests detected that the number density of c-fos positive cells in the SPIONs + magnet group was 3.44 fold that of the saline group. These results suggest that intravenously injected SPIONs can enhance the effects of TMS in treating neurological disorders.

Effective attachment of magnetic nanoparticles to neuronal membranes has far-reaching significance in activating ion channels and treating neurodegenerative diseases. Superparamagnetic iron oxide nanoparticles (SPIONs) synthesized by the polyol pyrolysis method have the advantages of rich surface functional groups, excellent magnetic properties, controllable particle size and water dispersibility. We propose that perfusion of biotin into the targeted brain area should be initially performed because it tends to be adsorbed by cell membranes, followed by injection of streptavidin (SA)-modified SPIONs into the same area of the brain. By means of the strong binding force between SA and biotin, the SPIONs may subsequently adhere to the cell surfaces in the brain area. In this work, fluorescein isothiocyanate-streptavidin (FITC-SA) was modified on the surface of polyethylene imine (PEI)-SPIONs by the EDC–NHS method and stereotaxically injected into the biotin-supplemented substantia nigra of mice. The combination of fluorescence detection with transmission electron microscopy (TEM) confirmed that FITC-SA/PEI-SPIONs adhered to neuronal membranes in the substantia nigra of mice 24 h after injection. The results show that our strategy can promote the attachment of SPIONs to neuronal membranes.
