China is a major textile manufacturer in the world; as a result, large quantities of dyeing effluents are generated every year in the country. In this study, the performances of two cationic starch-based flocculants with different chain architectures, i.e., starch- graft -poly[(2-methacryloyloxyethyl) trimethyl ammonium chloride] (STC- g -PDMC) and starch-3-chloro-2-hydroxypropyl trimethyl ammonium chloride (STC-CTA), in flocculating dissolved organic matter (DOM) in dyeing secondary effluents were investigated and compared with that of polyaluminum chloride (PAC). In the exploration of the flocculation mechanisms, humic acid (HA) and bovine serum albumin (BSA) were selected as main representatives of DOM in textile dyeing secondary effluents, which were humic/fulvic acid-like and protein-like extracellular matters according to the studied wastewater’s characteristics based on its three-dimensional excitation–emission matrix spectrum. According to experimental results of the flocculation of both the real and synthetic wastewaters, STC- g -PDMC with cationic branches had remarkable advantages over STC-CTA and PAC because of the more efficient charge neutralization and bridging flocculation effects of STC- g -PDMC. Another interesting finding in this study was the reaggregation phenomenon after restabilization at an overdose during the flocculation of BSA effluents by STC- g -PDMC at a very narrow pH range under a nearly neutral condition. This phenomenon might be ascribed to the formation of STC- g -PDMC/BSA complexes induced by some local charge interactions between starch-based flocculant and the amino acid fragments of protein due to charge patch effects.

Abstract CaSR has been recognized as a potential receptor involved in kokumi peptide perception. However, the molecular basis underlying kokumi-peptide-induced cellular responses remains insufficiently understood. Herein, CaSR-associated responses induced by kokumi peptides were investigated in STC-1 cells. Compared with MSG stimulation alone, MSG combined with γ-glutamyl peptides enhanced intracellular Ca2+ mobilization and CCK secretion, with response trends consistent with sensory evaluation. MSG+γEγEV elicited the strongest cellular response, increasing CCK secretion from 31.38 ± 3.18 pg/mL in the MSG group to 60.84 ± 4.45 pg/mL, while Ca2+ fluorescence intensity increased by 133.60% relative to MSG treatment alone (p < 0.05). Pretreatment with the CaSR antagonist NPS-2143 reduced MSG+γEγEV-induced Ca2+ mobilization and CCK secretion by 51.09 and 42.90%, respectively, supporting the involvement of CaSR in these cellular responses. Metabolomic and transcriptomic analyses revealed alterations in signaling-associated metabolites and pathways, including calcium signaling, MAPK signaling, PI3K-Akt signaling, cAMP signaling, phosphoinositide-related metabolism, and glycerophospholipid metabolism. These findings suggest that signaling responses associated with CaSR involvement in STC-1 cells share certain features with taste-related signal transduction processes. Collectively, the results provide insights into kokumi-peptide-induced cellular responses and their potential relevance to umami enhancement.

Background Satiety hormone cholecystokinin (CCK) plays an important role in food intake inhibition. Its secretion is regulated by dietary components. The search for bioactive compounds that induce CCK secretion is currently an active area of research. The objective of this study was to evaluate the ability of highland barley protein digest (HBPD) to stimulate CCK secretion in vitro and in vivo and identify the responsible peptide sequences. Results HBPD was prepared by in vitro gastrointestinal digestion model. The peptides with < 1000 Da in HBPD accounted for 82%. HBPD was rich in essential amino acids Leu, Phe and Val, but lack in sulfur amino acids Met and Cys. HBPD treatment with a concentration of 5 mg/mL significantly stimulated CCK secretion from enteroendocrine STC-1 cells ( P < 0.05). Moreover, oral gavage with HBPD in mice significantly increased plasma CCK level. Chromatographic separation was performed to isolate peptide fractions involved in CCK secretion and peptide sequence was determined by UPLC-MS/MS. Two novel cholecystokinin-releasing peptides PDLP and YRIVPL were pointed out for their outstanding CCK secretagogue activity. Conclusion This study demonstrated for the first time that HBPD had an ability to stimulate CCK secretion in vitro and in vivo and determined the bioactive peptides exerting CCK secretagogue activity in HBPD. Rehmannia glutinosa (RG) as a Chinese herbal medicine can be used both in medicine and food. As the main component of RG, the polysaccharides have a hypoglycemic effect, however, the hypoglycemic activity of RG homopolysaccharides remains unknown. We isolated and purified two polysaccharides, RGP70-1-1 and RGP70-1-2 (4.9 kDa and 2.8 kDa) from RG. The structural characteristics, including monosaccharide composition, linkage, and configuration were analyzed by FT-IR, HPLC, GC-MS, NMR spectroscopy, Congo test, and SEM. RGP70-1-1 and RGP70-1-2 consist of four monosaccharides (glucose, mannose, arabinose, and galactose). RGP70-1-1 contains 14 connection modes, with the linkages including L-Araf-(1→, →3)-L-Araf-(1→, →5)-L-Araf-(1→, →3,5)-L-Araf-(1→, →2,5)-L-Araf-(1→, D-Manp-(1→, →2)-D-Manp-(1→, →4)-D-Manp-(1→, D-Galp-(1→, →4)-D-Galp-(1→, →4,6)-D-Galp-(1→, →6)-D-Glcp-(1→, →4,6)-D-Glcp-(1→, →3,6)-D-Glcp-(1→. The linkages of RGP70-1-2 is including →5)-L-Araf-(1→, →3,5)-L-Araf-(1→, →4)-D-Manp-(1→, →3,6)-D-Manp-(1→, D-Galp-(1→, →6)-D-Galp-(1→, D-Glcp-(1→, →6)-D-Glcp-(1→, →4,6)-D-Glcp-(1→. Furthermore, RGP70-1-1 and RGP70-1-2 can inhibit α-glucosidase and α-amylase. RGP70-1-1 stimulated GLP-1 secretion in STC-1 cells and was related to the up-regulation of PI3K and p-AKT protein expression. The findings revealed a natural product with potential hypoglycemic activity, which may be used as a GLP-1 secretagogue and a beneficial functional food ingredient for T2D.

Slow Transit Constipation (STC) is characterized by impaired colonic motility, but its relationship with gut microbiota remains unclear. This study investigated the correlation between specific gut microbial populations and STC, focusing on the Lactobacillus acidophilus to Lactobacillus johnsonii (A/J) ratio. We used four rat groups: Control (CON), Loperamide-induced STC (LOP), antibiotic-treated (ABX), and antibiotic plus Loperamide (ABX + LOP). Fecal samples were analyzed using 16S rRNA gene sequencing, and serum metabolites were examined through LC–MS. The LOP group showed an increased A/J ratio, while ABX and ABX + LOP groups had decreased ratios. Notably, the ABX + LOP group did not develop STC symptoms. Metabolomic analysis revealed alterations in key metabolites across groups, including changes in levels of guanidinoacetate, glycine, l -glutamine, nicotine, and nicotinate d -ribonucleotide in the LOP group, and variations in l -glutamine, l -phenylalanine, l -tyrosine, histamine, d -ornithine, and lecithin in the ABX and ABX + LOP groups. Our findings suggest a correlation between the A/J ratio and STC development, offering insights into STC pathophysiology and potential microbiome-targeted therapies.
