Hybrid chain reaction-based and Au/Bi4NbO8Cl/In2S3 layer-by-layer assembled dual-mode photoelectrochemical-electrochemical aptasensor for the detection of Salmonella enteritidis

Haoran Cheng,Yifan Wang,Yirui Zhao,Hongman Hou,Gongliang Zhang,Jingran Bi,Shuang Yan,Hongshun Hao

Salmonella enteritidis ( SE ) is a food-borne pathogens that can cause acute gastroenteritis. With the increasing social attention to food safety, the detection method of SE has attracted wide attention. In response to the demand for efficient detection methods of SE , this study constructed a novel dual-mode photoelectrochemical-electrochemical (PEC-EC) aptamer-based biosensor. The sensor was constructed using Bi 4 NbO 8 Cl/In 2 S 3 heterojunction as the electrode substrate material, the hybridization chain reaction (HCR) and dye sensitization were used as the signal amplification strategies. Bi 4 NbO 8 Cl/In 2 S 3 heterojunction could provide an excellent initial photocurrent response for the sensing platform, and the HCR was opened by the end of complementary DNA (cDNA) and generated an ultra-long DNA double-stranded (dsDNA) “super structure” on the surface of the electrode, which could be embedded with a large number of methylene blue (MB) as the bifunctional probes. Thus, dual-mode output was achieved via the PEC and EC activity of MB. Under the optimized conditions, the PEC and EC signal responses of the system were linear to the logarithm of SE concentration in a range from 1.5 × 10 2  CFU/mL to 1.5 × 10 7  CFU/mL. The detection limits were found to be 12.9 CFU/mL and 12.3 CFU/mL using the PEC and EC methods, respectively. The constructed dual-mode biosensor exhibited good performance for real sample analysis, and demonstrated great application potential in the field of SE rapid detection. Moreover, this dual-mode detection strategy provided more accurate and reliable results than the single-mode output.

Global transcriptomic analysis of ethanol tolerance response in Salmonella Enteritidis

Shoukui He,Yan Cui,Rui Dong,Jiang Chang,Hua Cai,Hong Liu,Xianming Shi

Adaptation to sublethal amounts of ethanol enables Salmonella Enteritidis to survive under normally lethal ethanol conditions, which is referred to as the ethanol tolerance response (ETR). To uncover mechanisms underlying this adaptative response, RNA-seq and RT-qPCR techniques were employed to reveal global gene expression patterns in S. Enteritidis after sublethal ethanol treatment. It was observed that 811 genes were significantly differentially expressed in ethanol-treated cells compared with control cells, among which 328 were up-regulated and 483 were down-regulated. Functional analysis revealed that these genes were enriched in different pathways, including signal transduction, membrane transport, metabolism, transcription, translation, and cell motility. Specifically, a couple of genes encoding histidine kinases and response regulators in two-component systems were up-regulated to activate sensing and signaling pathways. Membrane function was also influenced by ethanol treatment since ABC transporter genes for transport of glutamate, phosphate, 2-aminoethylphosphonate, and osmoprotectant were up-regulated, while those for transport of iron complex, manganese, and ribose were down-regulated. Accompanied with this, diverse gene expression alterations related to the metabolism of amino acids, carbohydrates, vitamins, and nucleotides were observed, which suggested nutritional requirements for S. Enteritidis to mount the ETR. Furthermore, genes associated with ribosomal units, bacterial chemotaxis, and flagellar assembly were generally repressed as a possible energy conservation strategy. Taken together, this transcriptomic study indicates that S. Enteritidis employs multiple genes and adaptation pathways to develop the ETR.

A caprylate esterase–activated fluorescent probe for sensitive and selective detection of Salmonella enteritidis

Zhang Haotian,Wang Xiaohong,Xu Zuhui,Ma Jimei,Li Zi-Long,Cheng Wan-Min,Jiang Hong

Salmonella enteritidis is one of the most common foodborne pathogens. Many methods have been developed to detect Salmonella , but most of them are expensive, time-consuming, and complex in experimental procedures. Developing a rapid, specific, cost-effective, and sensitive detection method is still demanded. In this work, a practical detection method is presented using salicylaldazine caprylate as the fluorescent probe, which could be hydrolyzed by caprylate esterase liberated from Salmonella lysed by phage, to form strong fluorescent salicylaldazine. The Salmonella could be detected accurately with a low limit of detection of 6 CFU/mL and a broad concentration range of 10–10 6  CFU/mL. Moreover, this method was successfully used for the rapid detection of Salmonella in milk within 2 h through pre-enrichment by ampicillin-conjugated magnetic beads. The novel combination of fluorescent turn-on probe salicylaldazine caprylate and phage ensures this method has excellent sensitivity and selectivity. Graphical Abstract

Inactivation of Salmonella Enteritidis by clove essential oil: Multi-pathway responses and gene-function analyses

Xiaohui Sun,Yifei Wang,Shiyang Dai,Zanhui Gu,Joanna Trafiałek,Chunlei Shi,Shoukui He

Salmonella Enteritidis ( S. Enteritidis) is a primary foodborne pathogen whose stress resistance raises major food safety concerns. Clove essential oil (CEO), a natural antimicrobial agent, has garnered considerable attention due to its broad-spectrum antibacterial activity. However, the molecular mechanisms responsible for its antibacterial effect remain poorly elucidated. In this work, an integrated strategy combining proteomic profiling, bioinformatic analysis, and mutant analysis was adopted to explore the antibacterial mechanism of CEO against S. Enteritidis. The results showed that the minimum inhibitory and bactericidal concentrations of CEO were 2 g/L and 4 g/L, respectively. Proteomic and bioinformatic analyses revealed that CEO modulated the expression of 1437 proteins involved in cellular metabolism, ABC transporters, two-component systems, global regulation, flagellar assembly, and other cellular processes. Based on the proteomics results, three proteins from these pathways (HiuH, ProX, and PmrA) were selected for further investigation. The functional relevance of these proteins was subsequently validated by phenotypic comparisons between gene knockout mutants and the wild-type strain under CEO treatment. Intriguingly, the Δ hiuH , Δ proX , and Δ pmrA mutants displayed better in vitro growth yet greater susceptibility to CEO inactivation in chicken meat, highlighting the importance of assessing CEO efficacy in food matrices. Collectively, this work reveals the multi-pathway response of S. Enteritidis to CEO exposure, offering new mechanistic insights into the interplay between bacterial stress-defense systems and essential oil-mediated bacterial inactivation.

Functionalized Persistent Luminescence Nanoparticle-Based Magnetic Separation Aptasensor for Autofluorescence-Free Determination of Salmonella enteritidis

Lixia Yan,Liufeng Yu,Ling Sun,Beibei Wang,Yi Zhang

Salmonella enteritidis(SE) is recognized as a primary etiological agent of foodborne infection and food poisoning. Selective and sensitive determination of SE in animal-derived products is of great importance for ensuring safety in the food industry. Here, we report a highly sensitive and specific competition assay for detecting SE in eggs without interference from background fluorescence, by using persistent luminescent nanoparticles (PLNPs) as luminescent probes in combination with aptamer recognition and magnetic separation. Initially, the SE-specific aptamer (SEapt), as previously reported, was conjugated onto the surface of Fe3O4magnetic nanoparticles to serve as both the recognition and separation unit. Meanwhile, the ZnGa2O4:Cr (PLNPs) were functionalized with the aptamer-complementary DNA (cDNA), serving as the PL signal generator. The constructed PL aptasensor is composed of the aptamer-conjugated MNPs (MNPs-SEapt) and cDNA-functionalized PLNPs (PLNPs-cDNA), integrating the merits of the long-lasting luminescence of PLNPs, the magnetic separation ability of MNPs and the selectivity of the aptamer. This integration offers a promising approach for autofluorescence-free determination of SE in food samples. The proposed aptasensor exhibited excellent linearity in the range from 1.0 × 102–1.0 × 107CFU mL−1with a limit of detection as low as 32 CFU mL−1. The precision for 11 replicate determinations of 1.0 × 103CFU mL−1SE was 3.4% (relative standard deviation). The developed aptasensor achieved recoveries ranging from 98.8% to 102.8% for the determination of SE in the presence of common foodborne bacterial interferents. The method was successfully applied to the analysis of Salmonella genus in egg samples. In principle, the proposed platform may be adapted to other food matrices by substituting the target-specific aptamer, pending target-dependent optimization and validation.

Induction of Salmonella Enteritidis into a Viable but Nonculturable State by Cinnamaldehyde and Its Resuscitation

Ziying Lu,Tong Xing,Zepeng Zhao,Zefeng Li,Dongping Hou,Zhuolin Ma,Siyi Chen,Yuheng Yang,Shaoting Li,Hongmei Zhang

Trans-cinnamaldehyde (TC), a typical plant-derived compound, has been widely used in the control of foodborne pathogen contamination. Nevertheless, the risk associated with the occurrence of viable but nonculturable (VBNC) bacteria induced by TC remains unclear. The results of this study showed that Salmonella Enteritidis (S. Enteritidis) entered the VBNC state after being induced by TC at a minimum inhibitory concentration of 312.5 μg/mL and survived for at least 22 days under TC treatment. Enhanced resistance was found against heat treatment (75°C, 30 s), antibiotics (i.e., ampicillin, ceftriaxone sodium, chloramphenicol), and hydrogen peroxide (3%) in VBNC S. Enteritidis. A synergistic effect against VBNC S. Enteritidis occurred when TC was combined with acid treatment, including lactic acid and acetic acid (pH = 3.5). VBNC and resuscitated S. Enteritidis by sodium pyruvate treatment (100 mM) were found to retain the infectious ability to Caco-2 cells. Relative expression levels of the stress-related genes relA, spoT, ppx, lon, katG, sodA, dnaK, and grpE were upregulated in VBNC S. Enteritidis. Accumulation of reactive oxygen species (ROS) and protein aggregates was observed in VBNC cells. Besides, the resuscitation of VBNC cells was accompanied with clearance of ROS and protein aggregates. In summary, this study presents a comprehensive characterization of stress tolerance and resuscitation of VBNC S. Enteritidis induced by cinnamaldehyde, and the results provide useful information for the development of effective control strategy against VBNC pathogenic bacteria in food production.

Ratiometric fluorescence platform based on lanthanide-functionalized hybrid organic framework for sensitive detection of Salmonella enteritidis

Shihao Huang,Hao Shen,Shaoning Yu

The fluorescent hybrid, Ln-MOF/HOF, was prepared. • A rapid and sensitive strategy for Salmonella enteritidis detection was developed. • A wide linear range (10–10 8  CFU/mL) and low limit of detection (6 CFU/mL) were obtained. • The platform can be used in real food samples. The development of rapid and accurate methods of identifying foodborne pathogens is essential for ensuring food safety and protecting public health. We rationally constructed a lanthanide-functionalized organic framework heterostructure through an Ln 3+ -induced in situ partial transformation of a hydrogen-bonded organic framework (HOF) in which metal-organic framework (MOF) nanodomains were generated while preserving the original HOF skeleton. The resulting Ln-MOF/HOF composite integrates the structural stability and high surface accessibility of HOFs with the unique luminescent properties of lanthanide centers, enabling efficient fluorescence regulation and signal transduction. Based on this hybrid architecture, a ratiometric fluorescent aptasensor was developed to quantitatively identify Salmonella enterica via an aptamer-triggered competitive displacement strategy. Ligand-centered emission acted as the responsive signal, whereas Tb 3+ emission served as an internal reference, producing a self-calibrated “ON–OFF–ON” fluorescence response. Mechanistic investigations revealed that the sensing behavior was dominated by static quenching accompanied by dynamic aptamer redistribution between the material surface and target bacteria. A satisfactory linear relationship was obtained within the concentration interval of 10–10 8  CFU/mL, whereas the minimum detectable concentration reached 6 CFU/mL, with excellent selectivity, reproducibility, and environmental stability. Furthermore, satisfactory recoveries ranging from 93.0% to 103% were achieved in real food samples, demonstrating good practical applicability. Thus, the present study demonstrates an effective route for fabricating lanthanide-functionalized MOF/HOF heterostructures through partial framework conversion and establishes a self-referencing fluorescent sensing system for dependable pathogen analysis in complex food samples. Development of heat tolerance resulted from adaptation of Salmonella Enteritidis to sublethal concentration of sodium hypochlorite

Yuan Liang,Mengjun Hu,Rui Dong,Shoukui He,Yan Cui,Jinzeng Yang,Xiangyu Deng,Xianming Shi

Sodium hypochlorite (NaClO) is a widely used disinfectant in food processing environments. However, its improper use can lead Salmonella to develop tolerance to other stresses, which threatens food safety and human health. In this study, the survival ability assays showed that adaptation of S. Enteritidis to NaClO resulted in the development of heat tolerance at 55 °C and 63 °C. NaClO-adapted S. Enteritidis also showed a survival benefit in milk at 55 °C and 63 °C. NaClO-adapted S. Enteritidis exhibited a decrease in accumulation of reactive oxygen species (ROS) compared to non-adapted S. Enteritidis after exposure to 63 °C. Moreover, compared to non-adapted S. Enteritidis, the membrane fatty acids C16:0 and cyclo-heptadecanoic acid Cyc17 were significantly increased in NaClO-adapted S. Enteritidis, whereas UFA to SFA ratio was observed to decrease from 0.52 ± 0.04 to 0.41 ± 0.04. These changes resulted in a decrease in membrane fluidity of NaClO-adapted S. Enteritidis. Furthermore, RT-qPCR analysis revealed a considerable increase in the expression levels of heat shock-related genes in NaClO-adapted S . Enteritidis with or without heat treatment, in comparison to non-adapted S . Enteritidis. It was noteworthy that upregulation was 7.30-fold for dnaK , 11.62-fold for ibpA , and 3.92-fold for rpoH under heat treatment, suggesting that these genes played important roles in the development of heat tolerance. Taken together, adaptation of S . Enteritidis to NaClO resulted in the development of heat tolerance, which might increase the risk of cross-contamination and salmonellosis in the food industry.

A rapid and facile analytical approach to detecting Salmonella Enteritidis with aptamer-based surface-enhanced Raman spectroscopy

Lei Jin,Shufang Wang,Qing Shao,Yiyu Cheng

Salmonella should be absence in pharmaceutical preparations and foods according to regulations in many countries. Up to now, rapidly detecting Salmonella at 1 CFU·[10 g (mL) ] −1 in pharmaceutical preparation or 1 CFU·[25 g (mL) ] −1 in food samples is still a challenge. Herein, we present an aptamer-based surface-enhanced Raman spectroscopy (SERS) method for rapidly detecting Salmonella Enteritidis by using a handheld Raman instrument. The aptamer could specifically recognize S. Enteritidis, and 4-MBA self-assembled on the surface of [email protected] NPs was used as a Raman reporter molecule. The method was validated to be high specific with no interference from other five pathogenic bacteria. It could identify S. Enteritidis contaminant at ∼ 1 CFU·(10 g) −1 spiked level in a real sample (Wenxin granule, a botanical drug) after 6 h of enrichment. The detection time was much shorter than that of the methods (more than 54 ∼ 96 h) in the standards of pharmaceutical preparations and foods. In addition, the method could quantitatively determinate S. Enteritidis with satisfactory results. The SERS peak intensities of 4-MBA at 1072 cm −1 showed a good linear correlation (R 2  = 0.9873) with the logarithms of S. Enteritidis concentrations ranging from 4.17 × 10 2 to 1.39 × 10 7 CFU·mL −1 . T-test result (P = 0.425) revealed that there was no significant difference between the determination results obtained by the SERS method and the plate counting method. Therefore, the study indicated that the method was practical and reliable, and it could be a promising alternative for the on-site detection of S. Enteritidis.

Inactivation of Salmonella enteritidis on the surface of eggs by air activated with gliding arc discharge plasma

Li Guo,Pengyu Zhao,Zhiqian Yao,Tianhui Li,Mengying Zhu,Zifeng Wang,Lingling Huang,Gulimire Niyazi,Dingxin Liu,Mingzhe Rong

Salmonella enteritidis , a Gram-negative foodborne pathogen , causes inflammation of the stomach and intestines. Poultry and poultry products are considered to be the main hosts of S. enteritidis , especially the most common edible eggs, and increase the risk of Salmonella infection. Therefore, developing the inactivation strategy of S. enteritidis on the surface of eggs is the key step during the egg production process. Gas plasma could effectively inactivate bacteria and fungi and is developed as a potential non-thermal sterilization method in the food industry. In this study, the plasma-activated air (PAA) prepared by the air activated with gliding arc discharge contained a variety of reactive species. When the PAA activated at the power of 50 W and 100 W was inducted into the incubation chamber, the numbers of S. enteritidis on egg fragments or the whole eggs were reduced below the detection limit. After PAA treatment, the eggshells exhibited no change in structure demonstrated by scanning electron microscope, the color of eggshells became a little decrease in red, the internal quality of both the raw egg and the cooked egg also exhibited no change, and trace reactive species remained in the albumen. Further, the PAA was also feasible for the batch treatment of stacked eggs. Therefore, this study supplied an effective and safe strategy for aseptic processing in the poultry industry.

Antibacterial properties and mechanism of 2-methoxycinnamaldehyde against Salmonella enteritidis and its potential application in treating raw pork

Kegang Wu,Tong Zhang,Xianghua Chai,Pingping Wang,Xuejuan Duan,Huiying Hu

This study investigated the antibacterial activity and mechanism of action of 2-methoxycinnamaldehyde (MCA) against Salmonella enteritidis and evaluated the efficacy of MCA as a food disinfectant. The minimum inhibitory concentration (MIC) of MCA against S. enteritidis was 0.16 mg/mL. The acquired antimicrobial resistance toward MCA was weak, with low hemolytic activity and good in vitro stability. S. enteritidis exposed to MCA exhibited the formation of both holes and grooves on the cell surface, alterations in the structure and conformation of membrane proteins, leakage of nucleic acids, production of reactive oxygen species, membrane lipid peroxidation, binding to the groove of DNA, and destruction of protein secondary structures. After treatments with 1 MIC, 2 MIC, and 4 MIC of MCA for 20 min, S. enteritidis strain in pork slices was significantly inhibited ( p  < 0.05), and the reduction amounts ranged from 1.31 to 4.98 log CFU/g. Additionally, the treated pork contained low levels of residual disinfectants and demonstrated high sensory acceptability. This study provides new insights into the antibacterial mechanism of MCA, which has broad application potential as a plant-derived antibacterial washing disinfectant to prevent foodborne pathogens.

A simple quantum dot-based fluoroimmunoassay method for selective capturing and rapid detection of Salmonella Enteritidis on eggs

Beibei Wang,Xi Huang,Meihu Ma,Qing Shi,Zhaoxia Cai

The aim of this paper was to demonstrate a rapid and selective fluorescence-linked immunoassay method based on quantum dots as the fluorescent marker for the detection of Salmonella Enteritidis on eggshell. Highly-fluorescent and water-soluble CdTe quantum dots were prepared by using thioglycolic acid and 1-thioglycerol as ligands and were then conjugated with anti- Salmonella antibodies. As a result of specific interaction, Salmonella Enteritidis were specifically captured by bioconjugated CdTe quantum dots which led to the detection of a fluorescent signal. The bacterial cell images were obtained using fluorescence microscopy. Under optimal conditions, the quenched fluorescence intensity increased linearly with the log total count of Salmonella Enteritidis ranging from 3 × 10 2 to 3 × 10 7  CFU/mL within 1–2 h. The low detection limit was 1 × 10 2  CFU/mL without sample enrichment. This method was satisfactorily applied to the analysis of egg samples, which was demonstrated as a simple scheme for quick and selective detection of Salmonella Enteritidis on eggshell.
