Target-inhibited MCOF-Apt-AuNPs self-assembly for multicolor colorimetric detection of Salmonella Typhimurium

Li Hang,Xu Hui,Yao Shuo,Wei Shengnan,Liu Yi,Shi Xuening,Zhao Wei,Zhao Chao

Pathogens detection is a crucial measure in the prevention of foodborne diseases. This study developed a novel multicolor colorimetric assay to visually detect Salmonella Typhimurium ( S. Typhimurium ), by utilizing the etching process of gold nanorods (AuNRs) with TMB 2+ . The strategy involved the construction of nanozyme by assembling magnetic covalent organic framework (MCOF) with aptamer-conjugated AuNPs (Apt-AuNPs), exhibiting remarkable peroxidase-like activity to catalyze the oxidation of TMB/H 2 O 2 and inducing the etching of AuNRs. The presence of S. Typhimurium could inhibit this process, resulting in the generation of vivid colors. The multicolor colorimetric assay could specifically determine S. Typhimurium from 10 2 to 10 8  CFU mL −1 in 60 min with visual detection limit of 10 2  CFU mL −1 , and instrumental detection limit of 2.3 CFU mL −1 . Moreover, detecting S. Typhimurium in chicken, milk, pork and lettuce samples has shown promise in practical applications.

Optimisation of treatment conditions for reducing Shewanella putrefaciens and Salmonella Typhimurium on grass carp treated by thermoultrasound-assisted plasma functionalized buffer

Okon Johnson Esua,Jun-Hu Cheng,Da-Wen Sun

A novel method of thermoultrasound-assisted plasma functionalized buffer (PFB) for decontaminating grass carp was evaluated using the Box–Behnken design (BBD) with processing variables including PFB generating voltage (P V ), ultrasound treatment time (U T ) and temperature (T P ). The predicted models were found to be significant ( p  < 0.05) and displayed sufficient fitness with experimental data as indicated by non-significant ( p  > 0.05) lack of fit and high coefficient of determination (R 2 ≥ 0.97) values. The optimum decontamination conditions for the responses of S. putrefaciens and S. Typhimurium were P V of 66 V, U T of 14.90 min and T P of 60 ℃, achieving reductions of 4.40 and 3.97 log CFU/g, respectively, with a desirability of 0.998. Among the variables, temperature presented higher significance for inactivating bacteria and the production of volatile basic nitrogen and lipid peroxidation under the optimized conditions were within the limits of freshness for grass carp. Additionally, the effects of PFB and the optimized thermoultrasound-assisted PFB decontamination were mild on the microstructure of grass carp with slight ruptures and loose myofibril structures, indicating the potential of thermoultrasound-assisted PFB for seafood products decontamination with reduced processing time.

Exonuclease III-aided autonomous cascade signal amplification: a facile and universal DNA biosensing platform for ultrasensitive electrochemical detection of S. typhimurium

Qianqian Pei,Yu Wang,Su Liu,Yifei Qin,Xueqi Leng,Xuejun Cui,Jiadong Huang

In this study, a novel electrochemical biosensor based on exonuclease III (Exo III)-aided autonomous cascade signal amplification for ultrasensitive and highly specific detection of Salmonella typhimurium (S. typhimurium) has been reported. Herein, two duplex DNA probes were ingeniously designed. An Ap–Pr duplex probe was constructed as a recognition element by hybridizing an S. typhimurium aptamer (Ap) with a primer (Pr). A P–S duplex probe has been designed to contain a protruding DNA fragment at 5′-termini that has an identical sequence with Pr (the secondary Pr), a complementary duplex DNA that is used for blocking a signal-triggered probe (S), and an overhung nucleic acid segment at 3′-end that can anneal with Pr and form double-stranded DNA with a blunt 3′-terminus, thus initiating Exo III-aided amplification reaction. Upon sensing of the target S. typhimurium, the specific recognition of S. typhimurium and aptamer releases Pr, which anneals with the P–S duplex and triggers the Exo III cleavage reaction, accompanied by release of Pr, S, and secondary Pr. After this, the released Pr and secondary Pr anneal with other P–S duplex, which in turn triggers the Exo III-assisted cycling cleavage. Thus, large amounts of S are liberated, which hybrids with S* (DNA probe complementary to S); thereby, 5′-methylene blue (MB)-labelled DNA probe (MBP) folds into a hairpin structure with 5′-terminus close to electrode surface for efficient electronic transfer. Moreover, the S–S* hybrid duplex is specifically hydrolyzed by Exo III to liberate S, which accordingly results in the cycling cleavage of S*. Therefore, numerous MBP transform into a hairpin structure with electroactive MB close to the electrode surface; this generates significantly amplified electrochemical signal towards S. typhimurium detection. The present developed strategy exhibited high sensitivity towards S. typhimurium with a detection limit of 2.8 × 10 cfu mL−1. Furthermore, the proposed autonomous cascade signal amplification strategy can be easily extended for detection towards a wide spectrum of analytes including nucleic acid, proteins, and small molecules possessing their corresponding aptamers. Therefore, the developed Exo III-aided autonomous cascade signal amplification strategy might indeed provide a facile and universal tool for bioanalysis and clinical diagnosis.

PfAgo-DNAzyme cascade amplification improves Salmonella typhimurium detection

Bingjie Zheng,Yuqing Qin,Yanyan Chai,Aizhen Guo,Yonggang Hu

A DNA preamplification-free method was developed for sensitive detection of Salmonella typhimurium ( S. typhimurium ) by combining Pyrococcus furiosus Argonaute ( Pf Ago), a programmable small DNA-guided endonuclease and label-free DNAzyme, a functional nucleic acid (FNA). The effects of 5′-phosphorylated/hydroxylated single-strand DNA (5′P/OH ssDNA), including chimeric ssDNA-RNA and ssDNA-RNA-DNA (DNAzyme) on Pf Ago-mediated cleavage activity, were systematically investigated. Two detection strategies were developed: (1) A 63 nt 5′P DNAzyme was used as a guide DNA (gDNA) to mediate Pf Ago cleavage of probe MB1, resulting in a fluorescence signal inversely proportional to S. typhimurium concentration. RNase H2 from S. typhimurium (STH2) cleaved the DNAzyme, thereby reducing the signal. The limit of detection (LOD) was 4.5 × 10 CFU mL −1 , with a linear range from 8.0 × 10 to 6.4 × 10 4  CFU mL −1 (2) A 64 nt 5′OH DNAzyme was cleaved by STH2 to generate a 50 nt 5′P ssDNA, triggering Pf Ago-mediated MB2 cleavage. The fluorescence signal positively correlated with the concentration of S. typhimurium , with the LOD reduced to 2.0 × 10 CFU mL −1 and a detection range from 3.2 × 10 to 8.0 × 10 4  CFU mL −1 . Compared with the previous LOD of 3.2 × 10 3  CFU mL −1 achieved using the same DNAzyme labeled with a fluorescence probe and quencher, but without Pf Ago, the sensitivity of these methods was significantly improved, with increases of 71- and 160-folds, respectively. This method offers advantages of simplicity, specificity, rapidity, and sensitivity, making it suitable for point-of-care testing and holding promising potential for environmental monitoring, food safety, and clinical diagnosis.

Antibiofilm potential of lactobionic acid against Salmonella Typhimurium

Qiuxia Fan,Qian He,Ting Zhang,Wei Song,Qinglin Sheng,Yahong Yuan,Tianli Yue

This study aimed to explore the antibiofilm activity and possible mechanism of lactobionic acid (LBA) against Salmonella enterica subsp. enterica serovar Typhimurium ( S. Typhimurium). The minimum inhibitory concentration (MIC) of LBA against S. Typhimurium was 8 mg/mL. At sub-inhibitory concentrations (SICs), LBA inhibited the biofilm formation of S. Typhimurium on glass surface, as evidenced by the results of microscope observations. Crystal violet staining assay and cell enumeration assay revealed that LBA was also effective in inhibiting the biofilm formation on stainless steel surface. Moreover, LBA repressed the swimming and swarming motility of S. Typhimurium and also reduced the production of extracellular polysaccharide and protein during biofilm formation. Reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR) analysis revealed that LBA down-regulated the expression of genes involved in S. Typhimurium biofilm formation. These data suggested that LBA exerted an inhibitory effect on S. Typhimurium biofilm formation through interfering with cell motility, extracellular polysaccharide and protein production as well as expression of biofilm-related genes . Additionally, LBA at 4MIC and 8MIC had the efficacy to inactivate S. Typhimurium biofilms formed on stainless steel and chicken meat.

Colorimetric determination of Salmonella typhimurium based on aptamer recognition

Changqing Zhu,Ying Hong,Zhen Xiao,Yang Zhou,Yuan Jiang,Ming Huang,Xinglian Xu,Guanghong Zhou

A sensitive method for the colorimetric determination of Salmonella typhimurium (S. typhimurium) was developed based on aptamer recognition and gold nanoparticles. First, a biotinylated specific aptamer for S. typhimurium was immobilized on avidin-coated microplate wells. Then, target bacteria, a biotinylated aptamer and avidin-catalase were successively introduced into the microplate wells. Finally, hydrogen peroxide and freshly prepared gold(III) chloride trihydrate were added, and the absorbance of the reaction product was measured with a plate reader. Under optimized conditions, there was a linear relationship between the absorbance (A550 nm) and the concentration of S. typhimurium over the range of 101 to 106 cfu mL−1 (R2 = 0.9920), with a detection limit of 10 cfu mL−1. This simple rapid method provided a promising tool for on-site screening of S. typhimurium in raw chicken samples.

Sequential gene loss promotes expansion of monophasic Salmonella Typhimurium ST34

Wu Xiaolei,Wang Teng,Jia Chenghao,Zhou Haiyang,Li Yan,Baker Stephen,Yue Min

Understanding the genetic factors facilitating emergence of infectious diseases is critical, however, mechanisms underlying expansion of pathogenic bacterial variants remain unclear. Here we performed a large-scale genomic analysis of 44,597 Salmonella Typhimurium genomes and observe that sequential gene loss in monophasic Salmonella Typhimurium (mSTM) ST34 explains its clonal expansion as an increasingly prevalent zoonotic lineage. Functional and in vivo competition experiments show that a frameshift mutation in dinB , a polymerase for translesion DNA synthesis, leads to transcriptional changes affecting flagellin gene expression and subsequent loss of the flagellin-encoding fljB , altering the requirements for gut infection. Temporal evolutionary modelling supports a role for gene loss events in a specific chronological order for mSTM ST34 expansion. Our findings reveal a stepwise pathoadaptation model underpinning clonal global spread, providing mechanistic insights relevant to forecasting future pandemics.

Milkfat influences thermal tolerance and biofilm formation of Salmonella Typhimurium during pasteurization

Ziqi Liu,Yuechong Liu,Jian He,Xiaoyan Pei,Yixin Lin,Marciane Magnani,Donghong Liu,Tian Ding,Jinsong Feng

The bacterial contamination and failure of pasteurization are leading threats to the safety of dairy productions. Milk contains a large quantity of fat which supports bacterial growth but its influence on pasteurization efficacy and biofilm formation remains poorly understood. In this study, we first demonstrate that the inactivation of Salmonella during pasteurization is significantly less effective in whole milk compared to skim milk, with the D-value increasing from 0.60 min in skim milk to 0.67 min in whole milk ( P  = 0.0047). In whole milk, Salmonella biofilms form greater biomass and display a distinct architecture from biofilms in skim milk. Specifically, Salmonella in whole milk was embedded within the extracellular polymeric substances (EPS), while cells in skim milk biofilms were sparsely distributed. Raman spectroscopy showed that biofilms formed in whole milk possess a distinctly higher lipid content. Therefore, we hypothesized that the thermal tolerance of Salmonella in whole milk was directly linked to its physical interaction with milkfat. Confocal laser scanning microscopy showed an increase from 26.1 % to 86.1 % of the fraction of Salmonella co-localizing with milkfat after 12 h of incubation in whole milk. Lipase treatment (200 units/mL) on whole milk reduced biofilm formation and improved the efficacy of thermal inactivation. Our findings demonstrate the milkfat acts as a physical scaffold, facilitating bacterial aggregation and the formation of a protective, lipid-rich biofilm matrix enhancing Salmonella survival during pasteurization. These insights should inform pasteurization strategies of high-fat dairy products to ensure food safety.

Genomic epidemiology and public health implications of zoonotic monophasic Salmonella Typhimurium ST34

Xiaolei Wu,Jiaxin Du,Xiao Zhou,Xianqi Peng,Chenghao Jia,Baikui Wang,Beibei Wu,Yan Li,Min Yue

Background: Monophasic Salmonella Typhimurium sequence type 34 (mSTM ST34) has emerged as a significant global health threat, but our understanding of its genomic epidemiology and potential public health implications in international and regional contexts remains limited. This study aims to fill this crucial gap by assessing the genomic epidemiology of multidrug resistance (MDR) mSTM ST34, as well as its clinical characteristics and virulence. Methods: To achieve the objectives of this study, we conducted a comprehensive genomic analysis of mSTM ST34 isolates. We obtained a global dataset comprising 13,844 strains from public databases, along with 339 strains from a regional surveillance collection in Zhejiang Province, China. This dataset aims to provide in-depth insights into antimicrobial resistance, mobile genetic elements, and pathogenicity. Additionally, we meticulously assessed the association between phenotypic profiles and clinical presentations. Results: Our findings revealed that the prevalence of mSTM ST34 has surpassed that of the previously dominant ST19. In addition, we observed an increase in the detection of the IncQ1 plasmid, which is responsible for disseminating MDR. The prevalence of mSTM ST34 carriage was exceptionally high among children (≤12 years old) and elderly individuals (≥65 years old), with 92.6% of the isolates exhibiting MDR, including resistance to frontline antimicrobials such as third-generation cephalosporins and ciprofloxacin. Additionally, the human mSTM ST34 strain demonstrates a remarkable capacity for biofilm formation, which increases its virulence in animal models and complicates therapeutic interventions. Conclusions: mSTM ST34 has surpassed the previously dominant ST19, and its ability to transmit across multi-species increases its potential for further human transmission. This study addresses critical gaps in our understanding of mSTM ST34 prevalence, highlighting the importance of whole genome sequencing in surveilling zoonotic pathogens.

Nanostructure-Modified Optical Fiber Sensors for Detection of Salmonella typhimurium in Food

Shuai Wang,Huibo Han,Sandeep Boddu,Ragini Singh,Ben Niu,Penglei Zhou,Fei Liu,Xian Zhou,Bingyuan Zhang,Santosh Kumar

In this study, a nanostructure-modified bulge-tapered-bulge (BTB) optical fiber sensor was developed for the rapid and label-free detection of Salmonella typhimurium (S. typhimurium) in food samples. The BTB design improves the evanescent field interaction, and immobilized gold nanoparticles (AuNPs), copper sulfide nanoflowers (CuS-NFs), and nickel ferrite (NiFe2O4) cause localized surface plasmon resonance (LSPR), which amplifies the optical response to biomolecular interactions. The sensing surface was modified with S. typhimurium-specific antibodies that selectively recognize the antigen–antibody interactions. The fabricated sensor exhibited ultrahigh selectivity for nontarget bacteria, including Escherichia coli, Listeria monocytogenes, and Vibrio cholerae, and good stability against sodium dodecyl sulfate (SDS) treatment, pH variations, and environmental fluctuations in temperature and salinity. The detection range was −101–109 CFU/mL, and a low detection limit of 1 CFU/mL was achieved. These results demonstrate that the BTB fiber sensor has the potential for the sensitive, selective, and on-site detection of foodborne pathogens and can be used in the field of food safety.

The recovery effect and mechanism of sublethally injured Salmonella Typhimurium by lactic acid

Rongrong Yang,Ye Liu,Chang Su,Yinying Luo,Lele Shao

LA-induced SI S. Typhimurium could completely repair in beef, milk, and liquid media. • Microstructure and physiological functions of SI cells regained after recovery. • Surface hydrophobicity and membrane composition slightly altered after recovery. • Multiple stress resistance genes were significantly up-regulated after recovery. Foodborne pathogens can survive in a sublethally injured (SI) state during treatment, which can repair themselves under suitable conditions, posing a potential risk to food safety. In this study, the recovery effects of lactic acid-induced SI Salmonella Typhimurium were investigated in beef, milk, and liquid media. The microstructural and physiological function measurements, as well as transcriptomic analysis were performed to elucidate the recovery mechanism of SI bacteria. The results demonstrated that SI bacteria could completely recover in beef, milk, and liquid media. SI S. Typhimurium gradually restored membrane integrity, as evidenced by the recovery of membrane potential, reduced cytoplasmic and outer membrane permeability, normalized morphology, and restored alkaline phosphatase activity. However, surface hydrophobicity remained slightly higher than the control, and FTIR revealed minor but persistent changes in membrane phospholipids and polysaccharides, indicating adaptive membrane remodeling. Metabolic activity and esterase activity progressively recovered, while the elevated intracellular reactive oxygen species were effectively eliminated, accompanied by down-regulation of antioxidant enzyme genes. DNA repair pathways, particularly homologous recombination, remained persistently activated, likely to ensure long-term genomic stability. Notably, recovered bacteria significantly up-regulated a broad range of stress resistance genes, reflecting a transition to a state of potentially enhanced resistance. These findings demonstrated that the recovery of SI S. Typhimurium was an active adaptive process that not only restored culturability on selective media but also conferred potentially enhanced environmental tolerance.

A novel nanoparticle-based fluorescent sandwich immunoassay for specific detection of Salmonella Typhimurium

Xuechen Dou,Zhiwei Zhang,Chao Li,Yaohua Du,Feng Tian

The diseases caused by foodborne pathogens have a serious impact on human health and social stability. Conventional detection methods can involve long assay times and complex pretreatment steps, making them unsuitable for rapid, large-scale analysis of food samples. We constructed a novel nano-fluorescence sandwich immunosorbent immunoassay (nano-FSIA) to rapidly detect Salmonella Typhimurium in food, based on strong covalent binding between streptavidin and biotin . We used antibodies coupled to large particle-size fluorescent microspheres as fluorescent probes for direct quantitative analysis of S. typhimurium in milk. The optimized parameters were determined, and specificity and sensitivity were validated in phosphate-buffered saline (PBS) and milk. The results demonstrated a wide dynamic detection range for S. typhimurium (10 3 –10 8 colony forming units [CFU]/mL), with the limit of detection in PBS and milk at 234 and 346 CFU/mL, respectively. The results of nano-FSIA were consistent with those of plate counts and enzyme-linked immunosorbent assays, providing an effective and promising single-bacterium counting method for the rapid detection of Salmonella .
