Sarcosine Biosensor Based on Pt/g-C3N4 Nanocomposites with High Electrocatalytic Activity

Junyuan Feng,Xiaohui Chen,Xinlin Shi,Weidong Zheng,Xiaofang Zhang,Haipeng Yang

Sarcosine has been shown to be a new biomarker for prostate cancer that is superior to prostate-specific antigen (PSA). By detecting the concentration of sarcosine in the human body, accurate screening and diagnosis of prostate cancer can be achieved in clinical practice. However, the sarcosine level is extremely low in human serum or urine, and accurate detection of sarcosine concentration is difficult. More work is needed to prepare sarcosine biosensors with good anti-interference performance. In this work, we prepared a sarcosine biosensor using a highly electrocatalytically active nanocomposite comprising platinum/carbon nitride (Pt/g-C3N4) as the electrode modification material. The porous lamellar graphite phase carbon nitride (g-C3N4) prevents the agglomeration of Pt nanoparticles and keeps the Pt nanoparticles in a highly dispersed state. The constructed sarcosine biosensor achieves high sensitivity and can selectively detect low concentrations of sarcosine. Its linear detection range is 2–70 μM, and the limit of detection (LOD) is 0.8 μM (S/N = 3). The prepared sarcosine biosensor shows relatively good performance in terms of resistance to interference caused by electroactive substances and provides the potential to be applied in serum for accurate detection of sarcosine.

Sensitive determination of the potential biomarker sarcosine for prostate cancer by LC–MS with N,N′-dicyclohexylcarbodiimide derivatization

Jing Chen,Juan Zhang,Wenpeng Zhang,Zilin Chen

Sarcosine, a potential biomarker of prostate cancer, has drawn great attention in recent years. However, controversial research keeps arising about its role as a biomarker that might come from the two isomers (α-alanine and β-alanine) of sarcosine due to their same molecular weight and similar properties, which could interfere with the accurate detection of sarcosine. In this study, a simple and sensitive method was developed for the detection of sarcosine and the two isomers by LC with ion-trap MS through a novel derivatization reagent N , N ′-dicyclohexylcarbodiimide. N , N ′-Dicyclohexylcarbodiimide is usually considered as a condensation reagent, however, it was directly used as a derivatization reagent through a rearrangement side reaction in this study. The proposed method not only improved the chromatographic retention behavior of sarcosine and its two isomers, which was a benefit to their separation, but also dramatically enhanced the detection sensitivity of sarcosine, which was more favorable for real sample analysis. The factors affecting the productivity of the derivatization reaction, such as reaction time and amount of derivatization reagent, were systematically optimized. The method shows good linearity ( R 2 > 0.99), sensitivity with LODs of sarcosine as low as 1 ng/mL, and repeatability with the RSD < 6.07%. The developed method was applied to the analysis of urine.

Construction of NM88B(Fe)-POR/Rf equipped with hyperoxia microenvironment as a cascade nanozyme for ultrasensitive electrochemical detection of sarcosine

Weixuan Wu,Weiqi Feng,Jingyu Bao,Xin Yang,Zhongfeng Shi,Jianmin Chen,Liqin Zhou,Zhenxia Zhao,Zhongxing Zhao

High detection stability makes sarcosine oxidase (SOx) nanozyme as natural enzyme substitute, a promising sensor for prostate cancer biomarker Sarcosine (Sar). However, its limited catalytic capacity towards product (H 2 O 2 ) and the following weak signal response significantly restrict the detection sensitivity of Sar. Herein, a novel NH 2 -MIL88B(Fe)-porphyrin/riboflavin (NM88B-POR/Rf) electrochemical sensor with a hyperoxia microenvironment and cascade reaction functions is proposed to promote catalytic H 2 O 2 production and cascade signal amplification (from H 2 O 2 to •OH). Fe-porphyrin implantation in NM88B-POR/Rf as bionic heme enhanced the O 2 -carrying capacity, thus effectively improving the sensor's utilization rate of dissolved oxygen and the catalytic H 2 O 2 production ability of Rf. Based on enzymatic cascade strategy, NM88B in NM88B-POR/Rf as peroxidase nanozyme further catalyzed H 2 O 2 to generate •OH. Methyl blue was used to capture •OH and enhance Sar detection sensitivity. Detection results revealed that NM88B-POR/Rf presented as low limit of detection (LOD) as 3.31 pM with detection range of 10 pM∼1 mM. The synergistic effect of the hyperoxia microenvironment and enzyme cascade make LOD about 4 orders of magnitude lower than reported state-of-the-art biosensors. Moreover, NM88B-POR/Rf has good selectivity and stability and hence also be applied to the detection of Sar in real urine.

Lysine-Sarcosine PiPo Functionalized Surface with Excellent Hemocompatibility

Kai Hao,Ronglu Cui,Lifeng Fang,Jun Ling,Baoku Zhu

Polysarcosine (PSar) is an electrically neutral and excellently hydrophilic polypeptoid showing limited interaction with proteins and cells, which possesses better biocompatibility compared with polyethylene glycol. However, the immobilization of PSar is difficult due to the high water solubility. Herein, lysine-sarcosine PiPo, which was the random copolymer of lysine and sarcosine (PLS), was synthesized via a phosgene-free and water-tolerable polymerization of N-phenyloxycarbonyl-amino acids for the first time. PLS was immobilized by tannic acid (TA) on the polysulfone (PSf) membrane for a short time to obtain a neutral surface. The modified membrane showed improved hydrophilicity, decreased protein adsorption, and low cytotoxicity. Moreover, barely any hemolysis, no platelet adhesion, prolonged clotting time, and low complement activation further suggested good hemocompatibility. In order to improve the antifouling ability of the membrane under pressure, the neutral surface was oxidized by sodium periodate, which accelerated the chemical reaction between amino groups in PLS and phenolic hydroxyl groups in TA. Meanwhile, carboxyl groups generated due to the decomposition of TA and a negatively charged surface were obtained. While maintaining the good properties of the unoxidized one, the hydrophilicity of the oxidized membrane was improved and the clotting time was further prolonged. Besides, the filtration recovery of the oxidized membrane was improved remarkably. This approach of rapid immobilization of PSar has great potential for applications in the biomedical area, especially for blood-contacting materials.

Synthesis and gelation of copolypept(o)ides with random and block structure

Yan Xiao,Jianqiang Wang,Jun Zhang,Andreas Heise,Meidong Lang

Copolypept(o)ides of polysarcosine (PSar) and poly( N -isopropyl-L-glutamine) (PIGA) with random and block sequence structures were synthesized by ring-opening polymerization (ROP) of sarcosine N -carboxyanhydrides (Sar-NCA) and γ-benzyl- l -glutamate N -carboxyanhydrides (BLG-NCA) and post modification. With different distribution of Sar along the main chain, H-bonding pattern and secondary structure of polypeptides were turned, as well as aggregation and gelation behavior. Both copolypept(o)ides formed hydrogels above their critical gelation concentrations (CGCs) without thermo-sensitivity, which was normally reserved for PEG copolypeptides (eg, PEG- b -PIGA). In particular, a different mechanism from previously reported micellar percolation or fibrillar entanglement was suggested for gelation of the random copolypept(o)ide. Therefore, hydrogels from copolymers of PSar and PIGA represented a new approach to construct easy-handling, biocompatible, biodegradable and thermo-stable gels that could potentially be applied in biomedical fields. Amperometric sarcosine biosensor based on hollow magnetic Pt–Fe3O4@C nanospheres

Qingui Yang,Na Li,Qiang Li,Shaoqing Chen,Hsing-Lin Wang,Haipeng Yang

Sarcosine is a recently identified biomarker for prostate cancer. However, the rapid detection methods for sarcosine are relatively lack because of the low concentration and the presence of complicated interfering substances in serum or urine. In this manuscript, hollow nanospheres of Fe 3 O 4 was synthesized and used as carrier to disperse Pt (Pt) nanoparticles . In order to achieve excellent electron transfer ability, we use polyaniline to coat Pt–Fe 3 O 4 nanoparticles, and pyrolyze the polyaniline to carbon (C). Thus, hollow magnetic Pt–Fe 3 O 4 @C nanocomposites with good electron transfer ability are formed. The Pt–Fe 3 O 4 @C nanocomposites have high catalytic activity and stability. The nanocomposites were immobilized on glassy carbon electrode (GCE) to construct a nonenzyme hydrogen peroxide (H 2 O 2 ) sensor (Pt–Fe 3 O 4 @C/GCE). We further construct a sensitive sarcosine biosensor by immobilizing sarcosine oxidase (SOx) on the Pt–Fe 3 O 4 @C/GCE. The high catalytic activity and good biocompatibility of Pt–Fe 3 O 4 @C nanocomposites greatly retained the bioactivity of immobilized SOx, and the prepared sarcosine biosensor has good electrocatalytic performance towards sarcosine. It has a linear detection range between 0.5 and 60 μM with a limit of detection (LOD) of 0.43 μM (the signal to noise ratio is 3), and the sensitivity is 3.45 nA μM −1 (48.8 nA μM −1  cm −2 ), which has the potential to be used for rapid screening of prostate cancer.

A novel and sensitive sarcosine biosensor based on organic electrochemical transistor

Jin Hu,Weiwei Wei,Shanming Ke,Xierong Zeng,Peng Lin

Organic electrochemical transistors (OECTs) are successfully used for sensitive and selective detection of sarcosine , which is recognized as a potential non-invasive specific biomarker for prostate cancer. The sensitivity and selectivity of OECT-based sarcosine sensors are significantly improved by modifying sarcosine oxidase (SOx) on platinum-plated anodized aluminum oxide (AAO) gate electrodes. The device with Nafion-SOx/Pt/AAO gate electrode shows the excellent sensing performance and the sarcosine can be detected in a linear range from 50 nM to 100 μM with a detection limit of 50 nM, which is satisfied with the clinical range of prostate cancer levels in human urine (20 nM–5 μM) and much lower than that of conventional electrochemical methods. Meanwhile, the selectivity of the sensor is also well proved. This work indicates a novel OECT-based sarcosine biosensor with high sensitivity and selectivity for the first time, which has the prospect of a wide range of routine testing.

Azo-capped polysarcosine-b-polylysine as polypeptide gene vector: A new strategy to improve stability and easy optimization via host–guest interaction

Jianwei Du,Ce Tian,Yajie Liu,Jun Ling,Youxiang Wang

Polypeptide has been extensively researched in gene/drug delivery system due to the good biocompatibility. Herein, we synthesized total-polypeptide copolymers, i.e. Azo(azobenzene)-capped polysarcosine- b -polylysine (ASL) with narrow molecular weight distribution by α-amino acid N-carboxyanhydride (NCA) polymerization. Although the molecular weight of PLL segment was only about 6   kDa, ASL could condense DNA effectively and form about 150   nm spherical nanoparticles at N/P ratio of 15. The surface charge was significantly reduced due to the shielding effect of polysarcosine (PSAR) shell. ASL/DNA PeptoPlexes showed good colloidal stability under physiological salt condition and complexation competition stability in the presence of counter polyanion, which might improve the circulation time in vivo . The tip design of azobenzene provided a facile way for ligand modification via host–guest interaction, which could be flexibly optimized by changing its functional tags responding to a request. Our data showed that the introduction of CD-R8 could promote the internalization of gene into cytoplasm and even nucleus owing to the membrane penetrating effect of R8. Cell culture experiments indicated as a total-polypeptide system, ASL showed good cellular viability and comparable gene transfection level as PLL with molecular weight of 50   kDa. Overall, PSAR served as an ideal alternative of PEG and this total-polypeptide system showed us a good direction for gene carrier design.

Aptamer Regulated NIR Carbon Dot/AuNP Fluorescence Sensing Platform for Selective Sarcosine Detection

Huang Haiyin,Zhou Shuping,Yang Mei,Xie Shenghui,Jiang Xiaohua,Yue Jiahui,Yang Haipeng

A near-infrared (NIR) fluorescence sensing platform based on NIR carbon dots (CDs), gold nanoparticles (AuNPs) and sarcosine-specific aptamers (SA-Apt) was developed for the sensitive and selective detection of sarcosine (SA), a potential biomarker associated with prostate cancer. Nitrogen- and sulfur-co-doped NIR-CDs were synthesized via a solvothermal method using L-glutathione and formamide, while AuNPs were prepared through a classical citrate-reduction approach. The sensing mechanism is based on an aptamer-regulated inner filter effect (IFE) between NIR-CDs and aggregated AuNPs. In the absence of SA, SA-Apt adsorbs onto the AuNP surface, stabilizing the nanoparticles against salt-induced aggregation and preserving the fluorescence of the NIR-CDs. Upon target recognition, SA-Apt preferentially binds to sarcosine and detaches from the AuNP surface, triggering salt-induced aggregation of AuNPs. This aggregation produces significant spectral overlap with the NIR-CD emission band, resulting in efficient fluorescence quenching via the inner filter effect. The incorporation of NIR-CDs enhances detection sensitivity by providing strong fluorescence emission, minimal background interference, and improved photostability. Under optimized conditions, the NIR-CDs/AuNPs/SA-Apt platform exhibited a linear response to sarcosine over the range of 1–15 µM, with a correlation coefficient (R 2 ) of 0.996 and a limit of detection of 0.32 µM (S/ N  = 3). The sensing system demonstrates high selectivity, good reproducibility, and stable NIR fluorescence performance. By integrating NIR-emissive carbon dots with plasmonic aggregation and aptamer-mediated regulation, this work provides a robust non-enzymatic strategy for sarcosine detection.

Highly specific colorimetric detection of sarcosine using surface molecular imprinted Zn/Ce-ZIF

Peng Liu,Yeping Liu,Zhexu Gai,Fei Yang,Yanzhao Yang

Despite significant progress in nanozyme research and the advancement of analytical techniques, the inherent lack of specificity for target analytes often limits their utility in analysis. Integrating specific recognition capabilities into inorganic nanomaterials, independent of biological catalysts or adaptors, represents a crucial breakthrough in the field. Detecting Sarcosine (Sar) in human urine has recently emerged as a non-invasive biomarker for prostate cancer (PCa), presenting a valuable diagnostic tool. This study introduces a novel method for embedding molecular imprinting sites directly onto the surface of a Zn/Ce-based zeolitic imidazolate framework (Zn/Ce-ZIF) nanozyme, facilitating the development of a highly specific colorimetric assay for precise Sar measurement. By utilizing the lanthanide metal cerium as the catalytic element and ZIF-8 as the structural scaffold, we synthesized spherical Zn/Ce-ZIF nanozymes with exceptional oxidase-like catalytic efficiency. The efficiency of molecular imprinting experiments and the ability of molecularly imprinted polymers (MIPs) to identify target molecules were significantly enhanced by using theortical calculations to screen suitable functional monomers. The molecularly imprinted nanozyme (Zn/Ce-ZIF@MIP) initiates a colorimetric oxidation reaction of 3,3′,5,5′-tetramethylbenzidine (TMB), wherein the presence of Sar facilitates selective recognition and capture by the MIP shell, modulating the colorimetric response by hindering TMB’s access to the catalytic site. An intelligent color extraction detection device has been developed for the rapid perception of Sar. This colorimetric sensing platform has been validated through the detection of Sar in simulated urine samples. Overall, the application of surface molecular imprinting enhances the functionality of nanozymes in analytical fields.

Amperometric Sarcosine Biosensors Based on Electrodeposited Conductive Films Contain Indole-6-carboxylic Acid

Xiaofang Zhang,Jing Chen,Qia Wang,Bing Du,Gaochao Fan,Weidong Zheng,Haipeng Yang,Tailin Xu

Sarcosine level in serum is of important clinical significance in distinguishing prostate cancer. This work depicts an amperometric sarcosine biosensor with good anti-interference performance by electro-codepositing manganese phosphate, 3,4-ethylenedioxythiophene (EDOT) and indole-6-carboxylic acid (IA) on the glass carbon electrode. The prepared sarcosine biosensor has a wide linear detection range (1–55 μM) with a low detection limit of 0.11 μM. This work provides an anti-interference approach by controlling the surface charge density of the biosensor to sarcosine sensing, which has great potential to be used as point of care testing (POCT) device for the rapid detection of prostate cancer biomarkers.

Nanoreactor of sarcosine oxidase-embedded ZIFs activates fluorescent response for diagnosis of prostate cancer

Qianqian Hu,Guoning Chen,Lu Wang,Xia Cui,Chun Chang,Qiang Fu

Sarcosine is an important biomarker in the diagnosis of prostate cancer. It is essential to explore a method that detects sarcosine in biological samples quickly and accurately for the early screening and diagnosis of prostate cancer. Considering the high catalytic efficiency and good selectivity of enzymes, there is potential for establishing enzymatic methods for the determination of sarcosine. However, the poor stability, high economic consumption, and challenging recyclability of free enzymes limit their use. Here, sarcosine oxidase was immobilized to improve its stability. The properties of the immobilized enzyme, including the morphology and stability, were characterized. Then, the enzymatic reaction between the immobilized enzyme and the substrate sarcosine was employed to establish a fluorescence method with a linear range from 0.1563 to 10.00 μg mL−1, a LOD of 0.04 μg mL−1, and a recovery of 84.7–104.0%. It has been proved that sarcosine can be detected using the established method with short time consumption, high specificity, and low cost, and which is easy to operate and is affordable for many laboratories and community-level medical institutions. These results demonstrate that this method shows potential for practical applications in the diagnosis of prostate cancer.
