In this paper, a new strategy of introducing an extra solvent mode in the three-way EEMs data to construct a four-way EX–EM-solvent-samples data set was used in order to investigate the ‘third-order advantage’ by comparing the performances of two four-way calibration algorithms with those of two three-way calibration algorithms. The results of the self-weighted alternating normalized residue fitting (SWANRF) and the parallel factor analysis (PARAFAC) algorithms were both disappointing in this work due to serious collinearity and high background interference. In contrast, the satisfactory results (average recovery, 100.0–107.4% and standard deviation, 1.0–3.0%) were obtained by the third-order calibration methods based on both four-way self-weighted alternating normalized residue fitting (four-way SWANRF) and four-way parallel factor analysis (four-way PARAFAC) algorithms. In addition, for prediction of Dulbecco's modified eagle medium (DMEM) samples, the root-mean-square error of prediction (RMSEP) values for schizandrol B obtained from two three-way calibration algorithms were 60.91 and 35.46 μg mL − 1 with 80% ethanol in the aqueous solvent , which were obviously much larger than these (0.0832 and 0.0825 μg mL − 1 ) obtained using two algorithms of the four-way calibration. The results demonstrated that third-order calibration can extract more inherent information from the data, and can easily solve the problems of serious collinearity and high background interference. In short, among the advantages of this method over the existing methods, low cost, non-toxic and non-destructive analysis can be cited.

The clinical effects of Schisandra chinensis against human disease are well-documented; however, studies on its application in controlling plant pathogens are limited. Here, we investigated its inhibitory effect on the growth of Alternaria alternata , a fungus which causes significant post-harvest losses on apples, known as black spot disease. S. chinensis fruit extract exhibited strong inhibitory effects on the growth of A. alternata with an EC 50 of 1882.00 mg/L. There were 157 compounds identified in the extract by high performance liquid chromatography-mass spectrometry, where benzocaine constituted 14.19% of the extract. Antifungal experiments showed that the inhibitory activity of benzocaine on A. alternata was 43.77-fold higher than the crude extract. The application of benzocaine before and after A. alternata inoculation on apples prevented the pathogen infection and led to mycelial distortion according to scanning electron microscopy. Transcriptome analysis revealed that there were 4226 genes differentially expressed between treated and untreated A. alternata -infected apples with benzocaine. Metabolomics analysis led to the identification of 155 metabolites. Correlation analysis between the transcriptome and metabolome revealed that benzocaine may inhibit A. alternata growth via the beta-alanine metabolic pathway . Overall, S. chinensis extract and benzocaine are environmentally friendly plant-based fungicides with potential to control A. alternata .

Green extraction of the active lignans from Schisandra chinensis has received considerable attention due to their promising applications in the biomedical field. In this study, microwave-assisted deep eutectic solvent method was used to simultaneously extract five lignans: Schizandrol A, Schizandrol B, Schisantherin A, Schisandrin A and Schizandrin B. The extraction parameters were optimized according to the calculated comprehensive score obtained from the analytic hierarchy process combined with the entropy weight method. The optimal extraction conditions were that the molar ratio of choline chloride and glycolic acid is 4:1, water content is 30% (V/V), solid–liquid ratio is 1:20 (W g :V mL ), extraction time is 20 min, and extraction temperature is 70°C, under which the extraction rate of the five lignans were 10.89, 8.616, 4.019, 4.893, and 5.318 mg/g. These results confirmed that our extraction method is efficient and feasible for green extraction of lignans from S. chinensis .

Schisandra chinensis is a hepatoprotective herb that has been used for centuries in China. Polysaccharide is one of the major active components in S. chinensis , which has been reported to improve liver injuries induced by carbon tetrachloride, alcohol, or high-fat diet. In this study, we observed the effects and corresponding mechanisms of the secondary component of Schisandra polysaccharide (acidic polysaccharide, SCAP) on a murine model of severe acute liver injury induced by acetaminophen (APAP). SCAP significantly decreased the serum alanine aminotransferase (ALT), aspartate aminotransferas (AST), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β) levels, and was found to alleviate hepatic pathological alterations in the mouse model. Meanwhile, SCAP revealed a protective effects on the liver injury-related enzymes and factors, such as significantly diminished malondialdehyde (MDA) levels and glutathione (GSH) depletion, reduced ratio of B-cell lymphoma-2 (Bcl-2)-associated X protein (Bax)/Bcl-2, prohibited cleaved caspase-3 expression, and elevated the expression of p-AMPK, p-Akt, p-glycogen synthase kinase 3β (GSK 3β), nuclear factor erythroid 2–derived-like 2 (Nrf 2) and heme oxygenase-1 (HO-1) proteins in the liver tissues of the mouse model. In conclusion, we speculated that the protective activities of SCAP on the APAP-induced mouse model of acute liver injury might be related to its antioxidation, anti-inflammation and anti-apoptosis properties.

Schisandra chinensis extract (SCE) sensitizes SKOV3 cells to doxorubicin. • UPLC-Q-TOF/MS identifies 247 components; 25 bioactive candidates are screened by network pharmacology. • SCE, not single lignans, co-inhibits PI3K-AKT, MAPK and STAT3 pathways. • SCE suppresses ovarian cancer cell migration and invasion. • SCE's multi-target synergy supports its use as a chemotherapy adjuvant. Despite the documented potential of Schisandra chinensis extract (SCE) in enhancing chemosensitivity and providing hepatorenal protection, its active components and underlying mechanisms remain unclear. Herein, this study systematically identifies the bioactive components of SCE and elucidates their role in sensitizing ovarian cancer (OC) cells. The 80% ethanol fraction, prepared by macroporous resin column chromatography, demonstrated potent chemosensitizing activity. UPLC-Q-TOF/MS analysis identifies 247 compounds, from which network pharmacology screening yielded 25 potential active molecules and 7 key protein targets. Molecular docking reveals strong binding of schisandrin C and schizandrol B to PI3K and MAPK3. Western blot analysis further confirms the superior inhibitory effect of SCE over its individual constituents on PI3K-AKT, MAPK, and STAT3 pathways, highlighting a synergistic mechanism that underlies the enhanced chemosensitivity. Furthermore, scratch assay, migration, and invasion assays collectively confirm the anti-metastatic potential of SCE. This study deciphers the multi-target mechanism of S. chinensis in enhancing chemosensitivity of OC, providing both mechanistic insights for drug development and a foundation for integrative medicine. The antioxidant action and mechanism of selenizing schisandra chinensis polysaccharide (sSCP) were investigated in chicken embryo hepatocyte (CEH) taking schisandra chinensis polysaccharide (SCP) and N-acetyl-L-cysteine (NAC) as control. The CEH was cultured and treated with sSCP, then exposed to H 2 O 2 . The CEHs’ viability, apoptosis, ROS and antioxidase contents and the protein expression in MAPKs pathway and mitochondrion-dependence apoptotic signal pathway were assayed. The results showed that sSCP could significantly raise the cell viability and the activities of SOD, CAT and GSH-Px, decrease the cell apoptosis and the content of LDH, AST, ALT and MDA, down-regulate the protein expression of p-JNK 1 , p-ERK 1/2 , p-p38, Bax, Caspase 3 and cytochrome C, and up-regulate the protein expression of Bcl-2 in comparison with H 2 O 2 control group. The action of sSCP were stronger than those of SCP and NAC. These results indicated that selenylation modification could significantly enhance the antioxidant activity of SCP, sSCP could significantly protect hepatocyte from oxidative damage induced by H 2 O 2 , and its mechanism was by regulating the protein expression in MAPKs and mitochondrion-dependence apoptotic signaling pathways.

Background: Bee pollen is a uniquely complete nutritional product that has shown promise in alleviating obesity. While existing research has largely focused on the role of gut microbiota in obesity, the mechanisms by which bee pollen influences bile acid (BA) metabolism via microbial regulation remain poorly understood. Methods: This study hypothesized thatSchisandra chinensisbee pollen extract (SCPE) could mitigate high-fat diet (HFD)-induced obesity by regulating BA metabolism. Results: In a 12-week animal experiment, SCPE supplementation significantly reduced body weight gain, lipid accumulation, and adipocyte hypertrophy, while improving insulin sensitivity and relieving hepatic oxidative stress. These benefits were attributed to an increased relative abundance of bile salt hydrolase (BSH)-producing microbes, includingBacteroides,Lachnospiraceae NK4A136 group, andAkkermansia, which modulated BA metabolism by improving the expression of BA metabolism-related genes and reducing the concentrations of various types of BAs. Conclusions: These findings provide new insights into the mechanism by which SCPE alleviates obesity through the gut microbiota-BA axis and support the potential of bee pollen as a functional food for obesity management.

Background Atherosclerosis (As) is a chronic vascular inflammatory disease, which has become a major global public health challenge. Therefore, it is urgent to find natural anti-As active ingredients with high efficacy and low toxicity and develop a suitable delivery system to prevent and treat AS. Schisandra chinensis (Sch) has attracted much attention because of its rich anti-inflammatory active compounds, but its targeted delivery and bioavailability in vivo are still problems to be solved urgently. Objective The purpose of this study is to screen the effective components of Sch with significant therapeutic effect on AS, and to construct a dual-response nano-carrier of pH‑ROS to realize accurate drug release and targeted therapy. Methods The experimental research design was employed by integrating network pharmacological prediction, molecular docking verification, in vitro cell model and in vivo animal model. Results Through network pharmacology and molecular docking technology, we first identified schizandrol A (Sch A) as the main active ingredient against As. Under the condition of a feed ratio of 6:4, the double-response nanoparticles showed excellent physical and chemical properties: the average particle size was 234.9 nm, the drug loading was 19.88%, and the encapsulation efficiency was 62.06%. In vitro biological evaluation demonstrated that the nanoparticles loaded with Sch A exerted a strong anti‑steatosis effect by inhibiting the PI3K/AKT/SREBP-1 signaling pathway, thus reducing intracellular lipid synthesis and inflammatory responses. In vivo experiments revealed that the nanoparticles displayed good biocompatibility and targeting ability, effectively reducing plaque area in the aorta, enhancing plaque stability, and significantly lowering blood lipid levels in mice. Conclusions In summary, Sch A is an effective component for anti-As, and the constructed pH-ROS dual-responsive nanoparticles demonstrate good biological safety and targeting both in vitro and in vivo , indicating their potential application in the treatment of As.

This work was aimed at the evaluation of stability of components in oleogels and providing a quick, visual description of the relationship between the composition of an oleogel and its thermal and oxidation properties. Composition stability analysis indicated that almost all oleogel components, including approximately 24 fatty acids (FAs), seven diglycerides (DAGs), 21 triglycerides (TAGs) and 19 lignans, were inherited from the precursor oil. The oil content among different oleogels ranged from 56.17 to 82.25%, and the oleogels made with candlestick wax (CW) showed higher component stability. Non-isothermal crystallization (C), melting (M) and oxidation analysis of the oleogels by differential scanning calorimetry (DSC) showed slight differences among the various oleogels. For visual analysis, seven components had significant positive relationships with k(Ts) and Ea(Ts), and 18 FAs, 10 glycerides and four lignans had significant positive relationships with k(Tp) and Ea(Tp). As for C(Tp) and C(ΔH), nine lignans, 13 glycerides and three FAs exhibited a notable positive correlation with k(Tp). These results might provide guidance for designing oleogel features and a new application perspective on the stability of solidified oils.

The effectiveness of traditional Chinese medicine (TCM) against various diseases urges more low cost, speed and sensitive analytical methods for investigating the phamacology of TCM and providing a theoretical basis for clinical use. The potential of second-order calibration method was validated for the quantification of two effective ingredients of Schisandra chinensis in human plasma using spectrofluorimetry. The results obtained in the present study demonstrate the advantages of this strategy for multi-target determination in complex matrices. Although the spectra of the analytes are similar and a large number of interferences also exist, second-order calibration method could predict the accurate concentrations together with reasonable resolution of spectral profiles for analytes of interest owing to its ‘second-order advantage’. Moreover, the method presented in this work allows one to simply experimental procedure as well as reduces the use of harmful chemical solvents.

Spinal cord injury (SCI) is a devastating condition from which there is limited recovery of motor function. Inhibiting oxidative stress and inflammation is an effective strategy for treating SCI. The polysaccharide component of Schisandra chinensis is receiving increasing attention due to its potential in neuroprotection. Based on our previous research, we synthesized an Schisandra chinensis polysaccharide-zinc complex (SPZn) for synergistic treatment of SCI. However, poor blood-spinal cord barrier permeability limited the efficacy of SPZn. Microglia are immune cells that are inherent to the spinal cord and possess intrinsic targeting ability combined with immune evasion capacity, making them an ideal drug delivery system for SCI. Therefore, in this study, we selected microglia membranes to encapsulate SPZn (M-SPZn) for the treatment of SCI. The nanovesicle (M-SPZn) designed by us can target the site of spinal cord trauma and has good biosafety. The results, both in vivo and in vitro, showed that M-SPZn is more effective at alleviating oxidative stress and inhibiting inflammation. Moreover, M-SPZn reduced cell death and promoted motor function recovery in mice after SCI. In summary, M-SPZn has multiple roles in treating SCI and has a good usage potential.

Schisandra sphenanthera and Schisandra chinensis are widely consumed either as food or for medicinal purposes. Nevertheless, no detailed comparative assessments of their physicochemical properties and biological activity have been reported. In this paper, using hot-water extraction, alcohol precipitation, and deproteinization, we obtained polysaccharidic extracts from Schisandra sphenanthera and Schisandra chinensis (denoted as SSP and SCP, respectively) and investigated their antioxidant and immunological activities. The extracts were different from each other with regard to sugar, protein, and uronic acid contents. Both extracts were mainly composed of arabinose, glucose, and galactose, but their contents varied greatly; SSP had more galacturonic acid. Compared with SCP, SSP had stronger free radical scavenging ability, protective effects on biomolecules, cellular antioxidant activity, owing to its higher protein (35.35 ± 1.73%) and uronic acid (12.81 ± 1.15%) contents. With respect to cell viability, neutral red phagocytosis, NO production, and acid phosphatase activity, SCP had stronger effects than SSP; this was largely due to its high levels of mannose, galactose, arabinose, and glucose. These results provide evidence to support the use Schisandra -derived polysaccharides for several purposes, including clinical, agricultural, and industrial applications.
