Objective: Dietary n-3 polyunsaturated fatty acids (PUFAs), especially eicosapentaenoic acids (EPA) and docosahexaenoic acid (DHA), are proved to be effective in obesity reduction. Microalgal oil (MO) is an important alternative source of n-3 PUFAs that effectively alleviates obesity. The aim of the present study was to explore the anti-obesity effects of microalgal oil from Schizochytrium sp. (SMO) and to compare the effects of 2 SMOs (SMO1 and SMO2) with different levels of purity of n-3 PUFAs on high fat diet (HFD)-induced obesity in male C57BL/6J mice. Methods: Mice were randomly divided into 5 groups: (1) regular chow (RC); (2) HFD; (3) HFD + fish oil (FO); (4) HFD + SMO1; and (5) HFD + SMO2. Body weight and food intake were weekly monitored. After 16 weeks of treatment, a glucose tolerance test (GTT) and an insulin tolerance test (ITT) were performed. Serum lipid profile, morphological changes in the liver and epididymal white adipose tissue (eWAT), and the mRNA expression of lipid metabolism–related genes were also examined. Results: SMO treatment significantly decreased HFD-induced abdominal fat accumulation, lowered the levels of triglycerides, cholesterol, and low-density lipoprotein, as did the positive control treated with FO. Morphological examination revealed a remarkable reduction in lipid droplet formation in the liver tissue and the particle size of eWAT. An alleviation of inflammation infiltration in eWAT caused by a high-fat diet was also observed. Real-time reverse transcription–polymerase chain reaction analysis examination confirmed that microalgal oil inhibited the gene expression of fatty acid synthase, sterol responsive element-binding protein-1c, and acetyl-CoA carboxylase but promoted that of hormone-sensitive lipase and lipoprotein lipase, carnitine palmitoyltransferase-1, and uncoupling proteins in the liver and eWAT. Moreover, similar anti-obesity effects were obtained with the same dosage but different purity of n-3 PUFAs.Conclusions: As an alternative n-3 PUFAs resource, dietary intake of SMO might be beneficial to prevent HFD-induced abdominal fat accumulation.

Antioxidant addition is an effective strategy to achieve docosahexaenoic acid (DHA) overproduction in oleaginous microorganisms. Nevertheless, antioxidants like phenolic compounds sometimes exert pro-oxidant activity. In this work, effects of proanthocyanidins (PAs) on fermentation performance and oxidative stress in Schizochytrium sp. were investigated. Low PAs addition (5 mg/L) reduced reactive oxygen species and enhanced lipogenic enzymes activities and NADPH, resulting in significant increase in lipid (20.3 g/L) by 33.6 % and DHA yield (9.8 g/L) by 53.4 %. In contrast, high PAs addition (500 mg/L) exerted pro-oxidant effects, aggravated oxidative damage and lipid peroxidation, leading to sharp decrease in biomass (21.3 g/L) by 35.1 %, lipid (8.2 g/L) by 46.0 %, and DHA (2.9 g/L) by 54.8 %. Therefore, the antioxidant concentration is especially crucial in DHA production. This study is the first to report concentration-dependant dual roles of PAs in oxidative stress and DHA production in Schizochytrium sp., providing new insights into microbial DHA production.

Rapeseed meal hydrolysate (RSMH), is a low-cost, nitrogen-rich substrate with high potential for microbial fermentation. This study investigated the feasibility of using RSMH as an alternative nitrogen source to enhance docosahexaenoic acid (DHA) production in Schizochytrium sp. H016. Nutritional analysis revealed that RSMH contains abundant amino acids and phenolics, which are favorable for microbial growth and stress tolerance. Fermentation results showed that replacing yeast extract with RSMH significantly improved biomass accumulation, lipid production, and DHA yield. At 120 h, the maximum biomass, lipid, and DHA yields reached 45.80 g/L, 18.62 g/L, and 7.30 g/L, respectively, corresponding to increases of 23.96 %, 97.66 %, and 94.15 % compared to the control. RT-qPCR analysis revealed that RSMH upregulated key genes involved in glycolysis, fatty acid biosynthesis, triacylglycerol assembly, and antioxidant defense, which together promoted carbon flux toward lipid synthesis and improved oxidative stress resistance. Confocal microscopy further confirmed the increased accumulation of lipid droplets in RSMH medium. Fed-batch fermentation in a 5-L bioreactor yielded DHA and lipids of 15.51 and 33.81 g/L, respectively, with a final biomass of 63.80 g/L. These findings demonstrate that RSMH effectively substitutes yeast extract and upgrades rapeseed meal into a feedstock for DHA production.

Docosahexaenoic acid (DHA) is essential
for human health, and sustainable
production of DHA lipids by Schizochytrium sp. is industrially important. Unlike conventional bottom-up engineering
of individual pathway enzymes, this study applied a top-down strategy
using heterologous transcription factors. Screening identified soybean
GmDof4 as the most potent regulator. Overexpression significantly
boosted total lipid yield by 31.7% in a 5-L bioreactor and improved
glucose conversion efficiency by 22.4%. Although the DHA proportion
decreased slightly, absolute DHA lipid titer and yield rose by 20.8%
and 14.4%, respectively. Mechanistically, GmDof4 primarily upregulated
acetyl-CoA carboxylase (ACCase) while coordinately modulating fatty
acid synthesis and triacylglycerol assembly. Chemical inhibition of
ACCase abolished this enhancement, confirming its centrality. These
findings validate transcription factor engineering as a powerful strategy
for developing high-performance microbial DHA platforms.

The global rise in unhealthy lifestyles has accelerated hyperlipidemia prevalence, increasing demand for natural lipid-lowering agents as alternatives to conventional pharmaceuticals. This study investigated defatted Schizochytrium sp. biomass as a potential source of these bioactive compounds. Schizochytrium sp. proteins were enzymatically hydrolyzed under response surface methodology (RSM)-optimized conditions: 3.90 % (w/v) substrate, 9.60 % (w/w) enzyme, 45 °C, 4 h. The <3 kDa ultrafiltrate exhibited 57.38 % pancreatic lipase inhibition. Liquid chromatography-mass spectrometry (LC-MS/MS) identified VIEIPDDVAEK (VIE-11) as the core peptide with potent activity (IC 50 = 5.10 μM), supported by molecular docking studies. Structural characterization revealed predominantly α-helical and random coil conformations with hydrophobic residues. In free fatty acids (FFA)-induced HepG2 cells modeling hepatic steatosis, VIE-11 (50, 100, and 200 μM) reduced intracellular lipid accumulation, and was associated with downregulation of lipogenic regulators (SREBP-1c, FAS, CD36) and upregulation of lipid catabolism and export markers (ATGL, PPARα, CPT1A, APOB, MTTP). These findings suggest that VIE-11 may hold potential as a nutraceutical candidate for hyperlipidemia intervention.

Schizochytrium sp. is one of the most promising marine oleaginous microorganisms for industrial production of docosahexaenoic acid (DHA). In this study, the exogenous supplementation of 1 mM sesamol to the fermentation medium effectively prevented the peroxidation of polyunsaturated fatty acids in the fermentation process, which thereby significantly increasing the lipid and DHA yield by 53.52% and 78.30%, respectively. The addition of sesamol also increased the total antioxidant capacity of cells and induce the gene expression of polyketide synthase and antioxidant enzyme system. Moreover, the supply of nicotinamide adenine dinucleotide phosphate was regulated by sesamol by inhibiting the malic enzyme activity and promoting the glucose-6-phosphate dehydrogenase activity. Finally, fed-batch fermentation showed that the addition of sesamol significantly enhanced the DHA yield by 90.76%. This study provides an important reference for enhancing the DHA productivity of Schizochytrium sp. in industrial fermentation.

Omega-3 PUFAs rich in fish oil are believed to prevent obesity by improving lipid metabolism and regulating gut microbiota. Microalgae oil is considered as an alternative source of omega-3 PUFAs owing to diminishing fish resources. Schizochytrium microalgae oil (SMO), with a high DHA proportion, is a promising source for commercial DHA production. However, its weight-loss and gut microbiota-regulating properties are not well studied. Here we compared the obesity reducing effects of SMO, commercial fish oil (FO) and a weight-loss drug, Orlistat (OL), in a high-fat diet (HFD) induced obesity mouse model. We found that SMO is comparable to commercial FO and OL with regard to weight loss, and it even exhibits the weight-loss effects earlier than FO and OL. It can efficiently inhibit the expression of lipogenesis-related genes and induce the expression of lipolysis-related genes. Moreover, SMO has different gut microbiota modulating effects from those of FO and OL. It does not influence the diversity of bacterial community, but does increase the abundance of several beneficial SCFAs-producing bacteria and inhibits obesity-promoting Desulfovibrio and several pathogens. We also found that SMO recovers the HFD-disturbed metabolic capability of gut microbiota. It can increase the abundance of several metabolism-related pathways, such as those of amino acids, SCFAs and bile acid, and decrease the level of the LPS biosynthesis pathway, which probably contributes to an improvement of lipid metabolism and restoration of the colonic mucosal barrier impaired by HFD. Our data suggest that SMO can be used as a superior dietary supplement for alleviating obesity.

Schizochytrium sp., a microalga with high lipid content, holds the potential for co-producing docosahexaenoic acid (DHA) and carotenoids. In this study, the ability of Schizochytrium sp. to naturally produce carotenoids was systematically explored. Further, by enhancing the precursor supply of geranylgeranyl diphosphate, regulating carbon source through sugar limitation fermentation and employing a combination of response surface methodology and artificial neural networks to precisely optimize nitrogen sources, a new record of 43-fold increase in β-carotene titer was achieved in the 5L bioreactor (653.2 mg/L) . Meanwhile, a high DHA content was maintained (13.4 g/L). Furthermore, the use of corn stover hydrolysate has effectively lowered the production costs of carotenoid and DHA while sustaining elevated production levels (with total carotenoid titer and DHA titer reached 502.0 mg/L and 13.2 g/L, respectively). This study offers an efficient and cost-effective method for the co-production of carotenoid and DHA in Schizochytrium sp. .

Schizochytrium sp. has become a potential feed additive due to its rich contents of functional components such as docosahexaenoic acid (DHA), but its dose effects and physiological effects on darkbarbel catfish have not yet been clarified. This study evaluated the effects of six graded levels of dietary Schizochytrium sp. powder (0, 5, 10, 15, 20, and 25 g/kg; designated D0, D1, D2, D3, D4, and D5, respectively) on 30-d post-hatching juvenile darkbarbel catfish ( Pelteobagrus vachelli ) with an initial body weight of 0.38 ± 0.011 g, using a total of 1800 fish (three replicates per group, 100 fish per replicate) over a 10-week feeding trial. The optimal inclusion level of Schizochytrium s p. powder was also determined. Results showed that fish in the D4 group exhibited the highest weight gain rate (WGR) and specific growth rate (SGR) ( P < 0.05), accompanied by significantly decreased feed conversion ratio (FCR) and feed intake (FI) compared with the D0 group ( P < 0.05). Nutrient composition analysis revealed no significant changes in crude fat or moisture ( P > 0.05), but compared with the D0 group, crude protein increased significantly in D2, D3, and D5 groups ( P < 0.001). For fatty acids, D4 group exhibited the highest polyunsaturated fatty acid (PUFA) and ∑n-6 PUFA contents, the lowest saturated fatty acids (SAFA) content and health indices (atherogenic index and thrombosis index) ( P < 0.05), while monounsaturated fatty acid (MUFA) content significantly decreased with increasing addition ( P < 0.001). Principal component analysis confirmed PUFA, C18:1n-9c and MUFA as key factors driving intergroup differences. Histologically, the D4 group had significantly increased intestinal villus length and muscularis thickness compared with the D0 group ( P < 0.05), whereas the D5 group showed obvious liver damage. Physiologically, D4 group had the highest activities of superoxide dismutase (SOD), catalase (CAT) and total antioxidant capacity (T-AOC), the lowest content of malonic dialdehyde (MDA), and the activities of digestive enzymes (lipase and protease) were also significantly enhanced in liver and intestine ( P < 0.05). In conclusion, 20 g/kg Schizochytrium sp. powder promotes growth, optimizes fatty acid profiles, and enhances intestinal health and physiological function in juvenile darkbarbel catfish, while excess supplementation (25 g/kg) causes liver damage. This study revealed the application value and dose-effect relationship of Schizochytrium sp. powder as an aquatic feed additive, and provides a comprehensive theoretical basis for its precise application in freshwater fish farming.
