Response Characteristics of Scirpus Triqueter and Its Rhizosphere to Pyrene Contaminated Soils at Different Growth Stages

X. Y. Zhang,X. Y. Liu,S. S. Liu,F. H. Liu,L. S. Chen,G. Xu,C. L. Zhong,P. C. Su,Z. N. Cao

Scirpus triqueter (Triangular club-rush), a typical wetland species, is used to study the response characteristics to pyrene. A pot experiment was conducted to investigate the growth parameters (height, diameter, shoot number, total volume, underground biomass, aboveground biomass and total biomass), and enzymes (catalase and superoxide dismutase) of S. triqueter. The characteristics of soil enzymes (catalase and polyphenol oxidase) and microorganisms (bacteria and fungi) were also assessed after pyrene treatment. Elevated pyrene concentration (80 mg·kg−1) in the soil reduced the shoot number and biomass significantly, especially at the early growth stage. In root tissue, the enzyme catalase was activated at 80 mg·kg−1 of pyrene. Compared to roots, shoots had higher enzyme activities. Catalase activities in the rhizosphere increased throughout the growth period of S. triqueter. Polyphenol oxidase activities in the rhizosphere were higher than those in the bulk soil and unplanted soil. The populations of bacteria (total bacteria, pyrene-tolerant bacteria, and actinomyces) and fungi decreased under the stress of high pyrene concentration, while that of pyrene-tolerant bacteria increased with the increasing pyrene concentration. The presence of pyrene did not benefit the growth of S. triqueter. S. triqueter and soil enzymes varied within the growth stages. The presence of S. triqueter could improve the activity of soil enzymes and facilitate the propagation of microorganisms which could help eliminate pyrene contamination.

Responses of Scirpus triqueter, soil enzymes and microbial community during phytoremediation of pyrene contaminated soil in simulated wetland

Xinying Zhang,Xiaoyan Liu,Shanshan Liu,Fahui Liu,Lisha Chen,Gang Xu,Chenglin Zhong,Pengcheng Su,Zhengnan Cao

The aim of this study was to determine the enhancement of Scirpus triqueter in the dissipation of pyrene and the interaction of pyrene with plant, soil enzymes and microbial community. The results indicated that the dissipation ratios of pyrene in the rhizospheric and non-rhizospheric soil were 64.65   ±   3.86% and 54.49   ±   2.74%, respectively, and were higher than that in the unplanted soil (42.60   ±   0.71%) at 80d after planting S. triqueter . The pyrene was toxic to S. triqueter , as evidenced by growth inhibition in height, diameter, shoot number and biomass during the planting period. The activities of dehydrogenase decreased significantly at the presence of pyrene in soils, and increased remarkably with the introduction of S. triqueter . It was found that the pyrene addition increased the ratios of fungal/total fatty acids and gram-positive/gram-negative, but the presence of S. triqueter decreased the ratios of gram-positive/gram-negative. A larger stress level was found in the pyrene treated soils without S. triqueter . The ratio of aerobic/anaerobic bacteria decreased with increasing pyrene concentration, but increased when S. triqueter was planted. The principal analysis of phospholipid fatty acid signatures revealed that microbial community structures in the rhizospheric and non-rhizospheric soil were similar, but different from those in the unplanted and control soil.

Assessment of Pb and pyrene accumulation in Scirpus triqueter assisted by combined alkyl polyglucoside and nitrilotriacetic acid application

Chen Tingru,Liu Xiaoyan,Zhang Xinying,Hu Xiaoxin,Cao Liya

To understand the accumulation and uptake of polycyclic aromatic hydrocarbons (PAHs) and heavy metals by plants is an important part of the assessment of phytoremediation for PAHs and heavy metals co-contaminated soil. This study was an investigation of the accumulation and uptake of pyrene and lead (Pb) by Scirpus triqueter under the condition of alkyl polyglucoside (APG) and nitrilotriacetic acid (NTA) combined application. The results indicated that the accumulation of Pb by S. triqueter was significantly improved by NTA and APG addition into the soil. The pyrene accumulation in plant was also increased after application of APG when compared to the control treatment. However, the pyrene accumulation was decreased when APG was applied together with NTA. SEM and TEM images of root surface suggested that more Pb in the soil transferred to the plant by combined application of APG and NTA. More importantly, TEM images of xylem cells of S.triqueter root showed that permeability of cell membrane was improved by application of APG.

Enhanced Scirpus triqueter phytoremediation of pyrene and lead co-contaminated soil with alkyl polyglucoside and nitrilotriacetic acid combined application

Chen Tingru,Liu Xiaoyan,Zhang Xinying,Hou Yunyun,Chen Xiao,Tao Kaiyun

Purpose This study aimed to evaluate the effect of combination of alkyl polyglucoside (APG) and nitrilotriacetic acid (NTA) on improving the efficiency of phytoremediation for pyrene and lead (Pb) co-contaminated soil by Scirpus triqueter . Materials and methods Seedlings of S. triqueter with a similar size and biomass (3 g/pot) were grown on 2-month aged soil contaminated with 184.5 mg kg −1 of pyrene and 454.3 mg kg −1 of Pb at pH = 8.3. After growth for 10 days, different doses of APG and NTA were added into the soil. After 60 days, the height of plants, Pb concentrations in plants, and pyrene amounts in soil were determined. Results and discussion Combined application of NTA and APG with lower dosage (1 + 1 g kg −1 soil and 1 + 2 g kg −1 soil) had no notable negative influence on the growth of S. triqueter . Moreover, significant synergy on Pb accumulation in S. triqueter was achieved with APG and NTA combined application. Besides, the dissipation of pyrene from soil after 60-day planting was increased in APG and NTA treatments when compared with the control treatments. Application of APG alone or combined with NTA had greater effect on enhancing dissipation of pyrene from soil than NTA alone. Conclusions This study demonstrated that the remediation of Pb and pyrene co-contaminated soil by S. triqueter can be enhanced by combined application of APG and NTA. Long-term evaluation of this strategy is needed in co-contaminated field sites.

Phytoremediation effect of Scirpus triqueter inoculated plant-growth-promoting bacteria (PGPB) on different fractions of pyrene and Ni in co-contaminated soils

Xiao Chen,Xiaoyan Liu,Xinying Zhang,Liya Cao,Xiaoxin Hu

At present, few reveal the mechanism of inoculation plants with PGPB to remediate PAH-metal co-contaminated soil by analyzing the chemical speciations of contaminants. This study investigated the influence of inoculation plants with PGPB on different fractions of pyrene and Ni in rhizospheric and non-rhizospheric soil. The results demonstrated that the addition of PGPB brought the extensive increase of FDA activities in pyrene-Ni co-contaminated soil. PGPB increased the resistance of plants in nickel and pyrene-Ni contaminated soil, but decreased the plant biomass in single pyrene contaminated soil. The addition of PGPB efficiently decreased bioaccessible fractions of pyrene and increased the bioavailability of Ni in both rhizospheric and non-rhizospheric soil. Although inoculation plants with PGPB significantly increased the accumulation of Ni in single Ni and pyrene-Ni co-contaminated soil, the poor bioavailability of Ni in rhizospheric soil still restricted the phytoremediation of the heavy metal. The presence of pyrene hindered the inoculated plant from accumulating Ni to some extent. On the contrary, the presence of Ni significantly promoted the degradation of pyrene in both rhizospheric and non-rhizospheric soil after inoculation plants with PGPB.

Soil microbial community response to pyrene at the presence of Scirpus triqueter

Xinying Zhang,Xiaoyan Liu,Chenglin Zhong,Zhengnan Cao,Fahui Liu,Lisha Chen,Shanshan Liu,Yan Hu

Phytoremediation of pyrene pollution by Scirpus triqueter in a stimulated wetland system has been studied. The results showed that pyrene removal was higher in the rhizospheric soils (49.8–60.8%) than that in the unplanted (17.5–41.6%) and non-rhizospheric soils (33.0–56.4%). The variation in microbial community compositions was investigated by analyzing phospholipid fatty acids (PLFAs) profiles in microbes under pyrene exposure. The microbial biomass , as revealed by PLFAs, decreased significantly with increasing pyrene concentration in all the three parts of soils. Specifically, the pyrene exposure inhibited the growth of fungi and some microorganisms (indicated by 14:0, 17:0, 16:1ω7c and 7Me17:0) to some degree. Principal component analysis (PCA) also revealed that all the fatty acid negatively correlated with the pyrene at higher concentration. However, the presence of S. triqueter enhanced the proportions of fungi, Gram-negative bacteria and aerobic bacteria, which possibly contributed to the degradation of pyrene. Total PLFAs in rhizospheric soils were profoundly higher than those in non-rhizospheric soils. The results indicated that S. triqueter might induce the colonization of bacteria in the stimulated wetland system, and led to a discrimination of microbial community, which might be one of the mechanisms on pyrene dissipation in phytoremediation system.

Effect of plant-growth-promoting rhizobacteria on phytoremediation efficiency of Scirpus triqueter in pyrene-Ni co-contaminated soils

Xinying Zhang,Chang Su,Xiaoyan Liu,Zhenguo Liu,Xia Liang,Yanming Zhang,Yuwei Feng

The aim of this study was to investigate whether the plant-growth-promoting rhizobacteria (PGPR) could enhance phytoremediation efficiency of Scirpus triqueter ( S.triqueter ) in the pyrene-Ni co-contaminated soil. We also expected to reveal the possible mechanism for the affected phytoremediation efficiency induced by PGPR. We used three kinds of contaminated soils (Ni-contaminated soil, pyrene-contaminated soil and pyrene-Ni co-contaminated soil) to conduct this pot study. After harvest, plants growth indicators, polyphenol oxidase (PPO) activity and soil microbial community structure of each treatment were investigated to explain the different dissipation rates of pyrene and removal rates of Ni between treatments with and without PGPR. The results showed that PGPR-inoculated S. triqueter increased dissipation rates of pyrene and removal rates of Ni in all three contaminated soils, among which Ni removal rates in Ni single contaminated soil was elevated most significantly, from 0.895‰ to 8.8‰, increasing nearly 9 folds. However, Ni removal rate efficiency in co-contaminated soil was weakened because more toxic and complicated co-contaminated soil restrained plant growth and Ni absorption. We also observed that co-contamination harmed the soil microbial community more severely than that in single pyrene or Ni contaminated soil through phospholipid fatty acids analysis. Furthermore, dissipation rates of pyrene and removal rates of Ni were found positively correlated to the PPO activity and the abundance of branched and saturated fatty acids reflected by Pearson correlation analysis.
